Molecular detection of Fasciola, Schistosoma and Paramphistomum species from freshwater snails occurring in Gauteng and Free State provinces, South Africa.

Trematodiases are diseases caused by snail-borne trematode parasites that infect both animals and humans. Fascioliasis, schistosomiasis and paramphistomosis are some of these diseases and they affect millions of livestock, leading to significant economic losses. The aim of the study was to document freshwater snails occurring in selected study sites in the Free State and Gauteng provinces as well as identify and detect larval trematodes that they harbour. Samples were collected from a total of five study sites within two provinces of South Africa. Morphological features were used to identify s

Schistosomiasis amongst adolescent boys in non-lakeshore southern Malawi: Investigating local risk-factors within a nested community-based cross-sectional survey.

BACKGROUND: Schistosomiasis is endemic to Malawi, where preventive chemotherapy by mass drug administration (MDA) has been the foundational public health strategy for over a decade. Despite ongoing control, our understanding of the contemporary epidemiology of schistosomiasis in rural Malawi is limited to infrequent school-based surveys, typically lacking evidence from community-based surveys particularly within non-lakeshore upland communities who may be perceived to be at lower risk. METHODS: Between July and August 2022, we conducted a cross-sectional parasitological survey amongst a commu

Development of an in vitro drug screening assay using Schistosoma haematobium schistosomula.

BACKGROUND: The development of novel antischistosomal drugs is crucial, as currently no vaccine and only a single drug is available for the treatment of schistosomiasis. Fast and accurate in vitro assays are urgently needed to identify new drug candidates and research efforts should include Schistosoma haematobium. The aim of the present study was to develop a S. haematobium drug sensitivity assay based on newly transformed schistosomula (NTS). METHODS: We first undertook comparative studies on the cercarial emergence rhythms of the intermediate host snails Biomphalaria glabrata (S. mansoni)

Molluscicidal effect of three monoterpenes oils on schistosomiasis and fascioliasis vector snails in Egypt.

Thymol, Linalool and Eugenol showed considerable molluscicidal effect against Biomphalaria alexandrina, Bulinus truncatus and Lymnneae natalensis. The thymol was the potent one at least LC50 and LC90) followed by euganol then linalool. L. natalensis were more sensitive to these compounds followed by B. truncatus and then B. alexandrina. The LC50 & LC90) of thymol were 22 and 34 ppm against B. alexaldrina, 20 and 30 ppm for B. truncatuts and 18 and 29 ppm for L. nalalensis. These values were higher with Eugenol, 28 and 48 ppm for B. alexuadrina, 24 and 44 ppm for B. truncatus and 22 and 40 ppm

Transmission of schistosomiasis in Kariba, Zimbabwe, and a cross-sectional comparison of schistosomiasis prevalences and intensities in the town with those in Siavonga in Zambia.

Given that the two communities lie only 10 km apart, on the northern shore of Lake Kariba, it is surprising that human schistosomiasis now appears to be a much less important health problem in Kariba town (Zimbabwe) than in Siavonga town (Zambia). In an attempt to explain this difference, the level and sites of Schistosoma haematobium and S. mansoni transmission in Kariba, and the prevalences and intensities of human infection with these parasites in both communities, have now been investigated. In a longitudinal study, a cohort of 378 schoolchildren, 150 subsistence fishermen and 42 commercia

Observations on Neotricula aperta (Gastropoda: Pomatiopsidae) population densities in Thailand and central Laos: implications for the spread of Mekong schistosomiasis.

BACKGROUND: The snail Neotricula aperta transmits Mekong schistosomiasis in southern Laos and Cambodia, with about 1.5 million people at risk of infection. Plans are under consideration for at least 12 hydroelectric power dams on the lower Mekong river and much controversy surrounds predictions of their environmental impacts. Unfortunately, there are almost no ecological data (such as long-term population trend studies) available for N. aperta which could be used in impact assessment. Predictions currently assume that the impacts will be the same as those observed in Africa (i.e., a worsening

A pilot control trial of schistosomiasis in central Liberia by mass chemotherapy of target populations, combined with focal application of molluscicide.

In an area of high transmission of Schistosoma haematobium and S. mansoni in Central Liberia, populations of five villages and intermediate host snails were surveyed for two years. In three of these villages focal application of molluscicide (niclosamide) in the main transmission sites was combined with mass chemotherapy of a target population representing 76 to 90% of the contamination index. In the two other villages, which served as control, the prevalence indexes remained stable or increased a little during the period of this study. A three dose metrifonate mass treatment was applied in on

Modelling climate change impact on the spatial distribution of fresh water snails hosting trematodes in Zimbabwe.

BACKGROUND: Freshwater snails are intermediate hosts for a number of trematodes of which some are of medical and veterinary importance. The trematodes rely on specific species of snails to complete their life cycle; hence the ecology of the snails is a key element in transmission of the parasites. More than 200 million people are infected with schistosomes of which 95% live in sub-Saharan Africa and many more are living in areas where transmission is on-going. Human infection with the Fasciola parasite, usually considered more of veterinary concern, has recently been recognised as a human heal

Development of a dynamical model to enhance understanding of epidemiology of schistosomiasis in school-aged-children.

School-aged-children are most vulnerable to schistosomiasis as exemplified by age-prevalence profiles although current understanding of these patterns needs improvement. Using epidemiological data from the southern shoreline of Lake Malawi, we investigated the dynamics of Schistosoma transmission and the main determinants of Schistosoma infection risk using a transmission dynamic model, considering urogenital and intestinal schistosomiasis respectively. Specifically, we assessed whether the proximity of primary schools to the immediate shoreline was a major geospatial and epidemiological deter

Transmission sites for Schistosoma haematobium and Schistosoma bovis identified in localities within the Athi River basin of Kenya using a PCR-RFLP assay.

BACKGROUND: The epidemiology of human urinary schistosomiasis caused by Schistosoma haematobium can be complicated by the presence of ruminant schistosomiasis caused, primarily by S. bovis. The two schistosome species may be transmitted by the same Bulinus species, they may occur sympatrically in the same habitat, and their cercariae are very similar in morphology and therefore, difficult to tell them apart. Screening of snails collected from freshwater habitats for schistosome infections is often used to identify transmission sites or to evaluate success or failure of interventions. However,

Geographical and behavioral risks associated with Schistosoma haematobium infection in an area of complex transmission.

BACKGROUND: Schistosoma haematobium infection in endemic areas varies depending on the nature and complexity of the transmission networks present. Studies of micro-geographical transmission of S. haematobium infection indicate that discrepancy in prevalence between households is associated with diverse water contact behaviors and transmission that is restricted to particular sites harboring snail intermediate hosts. Detection of variations in the transmission sources with complex transmission networks of water bodies is required for optimization of malacological control. Longitudinal parasitol

Characteristics of persistent hotspots of Schistosoma mansoni in western Côte d'Ivoire.

BACKGROUND: Preventive chemotherapy with praziquantel is the cornerstone of schistosomiasis control. However, in some social-ecological settings, the prevalence and/or intensity of Schistosoma infection does not lower meaningfully despite multiple rounds of preventive chemotherapy, a phenomenon termed persistent hotspot (PHS). We assessed the characteristics of PHS in a Schistosoma mansoni-endemic area of Côte d'Ivoire. METHODS: In October 2016, a cross-sectional survey was conducted in 14 schools in the western part of Côte d'Ivoire, one year after multiple rounds of preventive chemotherapy.

Study and implementation of urogenital schistosomiasis elimination in Zanzibar (Unguja and Pemba islands) using an integrated multidisciplinary approach.

BACKGROUND: Schistosomiasis is a parasitic infection that continues to be a major public health problem in many developing countries being responsible for an estimated burden of at least 1.4 million disability-adjusted life years (DALYs) in Africa alone. Importantly, morbidity due to schistosomiasis has been greatly reduced in some parts of the world, including Zanzibar. The Zanzibar government is now committed to eliminate urogenital schistosomiasis. Over the next 3-5 years, the whole at-risk population will be administered praziquantel (40 mg/kg) biannually. Additionally, snail control and b

Increased interleukin-4 and interleukin-5 production in response to Schistosoma haematobium adult worm antigens correlates with lack of reinfection after treatment.

Acquired immunity to human schistosomiasis correlates with increased serum levels of schistosome antigen-specific IgE. Since interleukin (IL)-4 stimulates IgE production, the hypothesis that Th2-associated cell-mediated immunity participates in protection to reinfection was studied in a cohort of adolescent boys 12-18 months after chemotherapeutic cure in Upper Egypt. Initial Schistosoma haematobium prevalence was 51% and posttreatment incidence was 44%. Water contact was similar between putatively resistant and susceptible patients. Resistant persons had a 3.5- to 14-fold greater frequency of

Studies on the epidemiology of schistosomiasis in Liberia: the prevalence and intensity of schistosomal infections in Bong County and the bionomics of the snail intermediate hosts.

Urine samples from 3548 individuals residing in six of the eight districts which comprise Bong County, Liberia, the project area of the Bong County Agricultural Development Project (BCADP), and fecal specimens from 3408 of these individuals were examined for schistosome ova. A total of 164 water sites, including rice paddies, were surveyed for schistosome vector snails and monthly changes in snail population density and infection rate were determined in selected water sites. Bulinus globosus was more widely distributed than Biomphalaria pfeifferi but the latter species showed a higher infectio

Contrasting patterns of trematode and host snail dispersal across clusters of tropical crater lakes.

BACKGROUND: Comparing the genetic diversity and population structure of parasites and their hosts can improve understanding of their respective methods of dispersal, demographic histories, and factors driving genetic differentiation. In parasite taxa such as trematodes which have both intermediate and final hosts, comparative population-level analyses may also inform how each host influences gene flow among parasite populations. Here we aimed to assess how intermediate and final hosts influence trematode dispersal by examining the population genetic structure of the aquatic snail Bulinus tropi

One Health monitoring reveals invasive freshwater snail species, new records, and undescribed parasite diversity in Zimbabwe.

BACKGROUND: Snail-borne trematodes afflict humans, livestock, and wildlife. Recognizing their zoonotic potential and possible hybridization, a One Health approach is essential for effective control. Given the dearth of knowledge on African trematodes, this study aimed to map snail and trematode diversity, focusing on (i) characterizing gastropod snail species and their trematode parasites, (ii) determining infection rates of snail species as intermediate hosts for medically, veterinary, and ecologically significant trematodes, and (iii) comparing their diversity across endemic regions. Concurrent Schistosoma mansoni and Schistosoma haematobium infections in a peri-urban community along the Weija dam in Ghana: A wake up call for effective National Control Programme.

Globally over 200 million people are infected with schistosomiasis, and approximately 80% are caused by just two of five species, Schistosoma haematobium and Schitosoma mansoni that are broadly distributed, and often overlap across sub-Saharan Africa. Like most neglected tropical diseases, mortality is low (an estimated 200,000 deaths annually) and morbidity is considerably high and probably underestimated. Surprisingly, little attention has been given to co-infection with these two species. We have studied co-infection with S. mansoni and S. haematobium in a peri-urban community in Ghana, one
