Muscarinic receptor modulation of slow afterhyperpolarization and phasic firing in rat supraoptic nucleus neurons.

A slow posttrain afterhyperpolarization (sAHP) was studied in rat magnocellular neurosecretory cells (MNCs) in vitro. The sAHP was isolated from other afterpotentials by blocking the depolarizing afterpotential (DAP) with Cs(+) and the medium afterhyperpolarization (mAHP) with apamin. The sAHP amplitude increased logarithmically with activity ( approximately 3 mV per e-fold increase in number of impulses) and, when firing stopped, decayed exponentially with a time constant of 2 sec. The sAHP was associated with increased membrane conductance, and its amplitude varied linearly with voltage, rev

Enhancement of purinergic neurotransmission by galantamine and other acetylcholinesterase inhibitors in the rat vas deferens.

Galantamine, a mild acetylcholinesterase inhibitor and an allosteric ligand of nicotinic receptors, enhanced in a concentration-dependent manner the amplitude of purinergic twitch contractions of the electrically stimulated rat vas deferens (0.2 Hz, 1 ms, 60 V). Other acetylcholinesterase inhibitors also increased the twitches, showing a hierarchy of potencies of galantamine>physostigmine>tacrine>rivastigmine=donepezil. The potentiations seem to be unrelated to the ability to inhibit acetylcholinesterase, since the hierarchy of potencies to block the enzyme in vas deferens was tacrine>physosti

Electrophysiological modulation of cardiomyocytic tissue by transfected fibroblasts expressing potassium channels: a novel strategy to manipulate excitability.

BACKGROUND: Traditional pharmacological therapies aiming to modify the abnormal electrophysiological substrate underlying cardiac arrhythmias may be limited by their relatively low efficacy, global cardiac activity, and significant proarrhythmic effects. We suggest a new approach, in which transfected cellular grafts expressing various ionic channels may be used to manipulate the local electrophysiological properties of cardiac tissue. To examine the feasibility of this concept, we tested the hypothesis that transfected fibroblasts expressing the voltage-sensitive potassium channel Kv1.3 can m

Peptidyl inhibitors of shaker-type Kv1 channels elicit twitches in guinea pig ileum by blocking kv1.1 at enteric nervous system and enhancing acetylcholine release.

Potent and selective peptidyl blockers of the Shaker-type (Kv1) voltage-gated potassium channels were used to determine the role of these channels in regulating the spontaneous motility of smooth muscle preparations. Margatoxin (MgTX), kaliotoxin, and agitoxin-2 at 1 to 10 nM and agitoxin-1 at 50 to 100 nM induce twitches in guinea pig ileum strips. These twitches are abolished by tetrodotoxin (TTX, 0.5 microM), atropine (1 microM), hexamethonium (10 microM), or nifedipine (0.1 microM). It is proposed that blockade of Kv1 channels by MgTX, kaliotoxin, or the agitoxins increases excitability of

Stimulation of a nicotinic ACh receptor causes depolarization and activation of L-type Ca2+ channels in rat pinealocytes.

1. Membrane voltage (Vm) recordings were obtained from isolated rat pinealocytes using the patch-clamp technique. In parallel to the electrophysiological experiments, intracellular Ca2+ measurements were performed using fura-2. 2. The resting Vm averaged -43 mV and replacement of extracellular NaCl by KCl completely depolarized the cells. This indicates that the resting Vm is dominated by a K+ conductance. Single-channel recordings revealed the presence of a large conductance Ca(2+)-activated charybdotoxin-sensitive K+ channel. 3. Application of ACh (100 microM) depolarized the pinealocytes on

The effects of K+ channel blockers on the spontaneous electrical and contractile activity in the proximal renal pelvis of the guinea pig.

PURPOSE: The role of various K+ channel populations in determining the time course and amplitude of the spontaneous action potentials and contractions in the smooth muscle cells of the guinea pig renal pelvis were investigated by standard electrophysiological and tension recording techniques. MATERIALS AND METHODS: Electrical recordings in the proximal renal pelvis were used to demonstrate the presence of 3 cell populations based on the waveform of their Ca(2+)-dependent action potentials which we have termed "pacemaker," "intermediate" and "driven" action potentials. RESULTS: Blockade of la

Two types of voltage-gated K channels in carotid body cells of adult cats.

The purpose of this study was to investigate if the oxygen-sensitive K channel is present in the carotid body cells of adult cats, and if all carotid body cells express the oxygen-sensitive K channel. A standard patch-clamp technique with a whole-cell configuration was applied to cultured carotid body cells from adult cats. The cells were continuously perfused with Krebs equilibrated with 5% CO2/air or 5% CO2/argon at room temperature. The results showed that electrophysiologically at least two types of cells existed in cultured cat carotid body cells. One type expressed the oxygen-sensitive K

Effects of cyclopiazonic acid on phenylephrine-induced contractions in the rabbit ear artery.

1. Upon stimulation with phenylephrine, the rabbit ear artery displays endothelium-regulated rhythmic contractions, which may be attributed to the periodical activation of the dihydropyridine-sensitive Ca2+ channel, presumably regulated by the Ca(2+)-activated K+ channel. The effect of cyclopiazonic acid (CPA), an inhibitor of the Ca(2+)-ATPase of the sarcoplasmic reticulum (SR), on phenylephrine-induced contractions was examined in endothelium-denuded rabbit ear arteries suspended in an organ chamber for isometric tension recordings. 2. Phenylephrine-induced tonic contractions were converted

Calcium channels and calcium-gated potassium channels at the frog neuromuscular junction.

Two mechanisms which regulate transmitter release by regulating Ca2+ entry in the presynaptic nerve terminal were studied at the frog neuromuscular junction (nmj). First, the location of Ca2+ channels in relation to transmitter release sites and, second, the regulation of Ca2+ entry by Ca(2+)-gated potassium (gKca) channels. Ca2+ channels were disclosed using fluorescent omega-conotoxin GVIA (omega-CgTX) which blocks transmitter release and Ca2+ entry at the frog nmj. Ca2+ channels were located in bands spaced at regular intervals of 1 micron. The omega-CgTX labeling was removed following mech

Potassium currents of neurons isolated from medical nucleus tractus solitarius.

Voltage-dependent potassium currents of neurons enzymatically isolated from the medial and dorsal subnuclei of the solitary tract (mNTS) of adult guinea pig have been characterized with respect to their voltage dependence, time dependence, and sensitivity to specific blocking agents. This region of the medulla receives baroreceptor afferent input and is involved in cardiovascular regulation. Our results showed the presence of three types of potassium currents. First, in all neurons studied (n = 58) a slowly developing outward current was present at potentials more positive than -30 mV. The tim

Effect of potassium channel blockade on the anti-ischemic actions of mechanistically diverse agents.

The ATP-sensitive potassium channel opener, cromakalim, protects ischemic hearts and its effect can be reversed by glyburide. It is presently unknown if glyburide can abolish the anti-ischemic effects of mechanistically different agents or if blockers of other potassium channels can abolish the protective effects of cromakalim. Thus, the effect of glyburide on previously reported cardioprotective agents was tested in globally ischemic/reperfused isolated rat hearts. Calcium antagonists, sodium channel blockers and calmodulin antagonists were found to significantly improve postischemic contract

Effect of lowered extracellular pH on Ca2(+)-dependent K+ currents in type I cells from the neonatal rat carotid body.

1. The whole-cell configuration of the patch-clamp technique was used to record K+ currents from type I cells enzymatically dispersed from the neonatal rat carotid body. The current-voltage (I-V) relationship for the K+ currents showed a prominent, outward shoulder at test potentials of between +10 and +30 mV. 2. The shoulder of the I-V curve could be enhanced by raising extracellular Ca2+ concentration or by bath application of 5 microM-Bay K 8644. It could also be suppressed by bath application of 100 microM-Cd2+ or 5 microM-methoxyverapamil (D600), indicating that a large component of the K

Mechanisms of potassium channel block in rat alveolar epithelial cells.

Block of inactivating delayed rectifier K+ currents was studied in rat alveolar epithelial cells in primary culture using the whole-cell configuration of the gigohm-seal voltage-clamp technique. Charybdotoxin was the only blocker studied which did not alter K+ current kinetics; it produced a simple block (K1 approximately 1 nM) which appeared to be independent of voltage or channel state (open, closed or inactivated). Tetraethylammonium slowed inactivation of K+ currents, consistent with the notion that blocked channels cannot inactivate. Verapamil and methoxyverapamil produced time-, voltage-

Patch-clamp studies in human macrophages: single-channel and whole-cell characterization of two K+ conductances.

Human peripheral blood monocytes cultured for varying periods of time were studied using whole-cell and single-channel patch-clamp recording techniques. Whole-cell recordings revealed both an outward K current activating at potentials greater than 20 mV and an inwardly rectifying K current present at potentials negative to -60 mV. Tail currents elicited by voltage steps that activated outward current reversed near EK, indicating that the outward current was due to a K conductance. The I-V curve for the macroscopic outward current was similar to the mean single-channel I-V curve for the large c

Molecular dynamics simulations of a K+ channel blocker: Tc1 toxin from Tityus cambridgei.

Toxins that block voltage-gated potassium (Kv) channels provide a possible template for improved homology models of the Kv pore. In assessing the interactions of Kv channels and their toxins it is important to determine the dynamic flexibility of the toxins. Multiple 10 ns duration molecular dynamics simulations combined with essential dynamics analysis have been used to explore the flexibility of four different Kv channel-blocking toxins. Three toxins (Tc1, AgTx and ChTx) share a common fold. They also share a common pattern of conformational dynamics, as revealed by essential dynamics analys

Alteration of Piezo1 signaling in type 2 diabetic mice: focus on endothelium and BKCa channel.

Piezo1 mechanosensitive ion channel plays a important role in vascular physiology and disease. This study aimed to elucidate the altered signaling elicited by Piezo1 activation in the arteries of type 2 diabetes. Ten- to 12-week-old male C57BL/6 (control) and type 2 diabetic mice (db-/db-) were used. The second-order mesenteric arteries (~ 150 μm) were used for isometric tension experiments. Western blot analysis and immunofluorescence staining were performed to observe protein expression. Piezo1 was significantly decreased in mesenteric arteries of type 2 diabetic mice compared to control mic

17α-ethinylestradiol modulates endothelial function in ovariectomized rat carotid arteries.

17α-ethinylestradiol (EE2), a derivative of 17β-estradiol (E2), is a potent estrogenic substance that is used as the estrogenic component of oral contraceptives (OCPs). However, women who take OCPs have an increased risk of cardiovascular events. Since few studies have examined EE2 endothelial effects, we explored the effects of EE2 on endothelial function in ovariectomized and isoflavone-free rats. After ovariectomy, 12-week-old female Sprague-Dawley rats were assigned to EE2, E2 or control groups. After 16 weeks, the EE2 and E2 groups were orally administered EE2 (8.3 μg/day) and E2 (12.6 μg

Ca2+-dependent K+ efflux regulates deoxycholate-induced apoptosis of BHK-21 and Caco-2 cells.

BACKGROUND & AIMS: Deoxycholate (DC) has proapoptotic and tumorigenic effects in different cell types of the gastrointestinal tract. Exposure of BHK-21 (stromal) cells to DC induces Ca(2+) entry at the plasma membrane, which affects intracellular Ca(2+) signaling. We assessed whether DC-induced increases in [Ca(2+)] can impinge on plasma membrane properties (eg, ionic conductances) involved in cell apoptosis. METHODS: Single- and double-barreled microelectrodes were used to measure membrane potential (V(m)) and extracellular [K(+)] in BHK-21 fibroblasts and Caco-2 colon carcinoma cells. Apopt

TRPC1 associates with BK(Ca) channel to form a signal complex in vascular smooth muscle cells.

TRPC1 (transient receptor potential canonical 1) is a Ca(2+)-permeable cation channel involved in diverse physiological function. TRPC1 may associate with other proteins to form a signaling complex, which is crucial for channel function. In the present study, we investigated the interaction between TRPC1 and large conductance Ca(2+)-sensitive K(+) channel (BK(Ca)). With the use of potentiometric fluorescence dye DiBAC(4)(3), we found that store-operated Ca(2+) influx resulted in membrane hyperpolarization of vascular smooth muscle cells (VSMCs). The hyperpolarization was inhibited by an anti-T
