Luteolin Is a Potential Immunomodulating Natural Compound against Pulpal Inflammation.

AIM: The present study evaluated the therapeutic effects of luteolin in alleviating pulpitis of dental pulp- (DP-) derived microvesicles (MVs) via the inhibition of protein kinase R- (PKR-) mediated inflammation. Methodology. Proteomic analysis of immortalized human dental pulp (DP-1) cell-derived MVs was performed to identify PKR-associated molecules. The effect of luteolin on PKR phosphorylation in DP-1 cells and the expression of tumor necrosis factor-α (TNF-α) in THP-1 macrophage-like cells were validated. The effect of luteolin on cell proliferation was compared with that of chemical PKR

A VAMS-based LC-MS/MS method for precise cenobamate quantification in epilepsy (patients).

OBJECTIVE: Cenobamate (CNB), a recently approved antiseizure medication by the European Medicines Agency (EMA), serves as an adjunctive therapy for focal-onset seizures in adult patients unresponsive to at least two other treatments. Administered in polytherapy, CNB can potentially interact with co-administered drugs in epilepsy patients, necessitating dose adjustments and the need for effective therapeutic drug monitoring (TDM). METHODS: In this study, we introduce a novel LC-MS/MS method for precise CNB quantification using Volumetric Absorptive Microsampling (VAMS), following validation ac

[Therapy of chronic apical periodontitis in traumatically injuring front teeth with ongoing root growth].

Because of the patient's age and the width of the apical foramen, conventional or surgical root canal fillings are frequently not indicated for the treatment of chronic apical periodontitis in teeth whose roots have not yet fully grown. Therefore resorbable paste fillings are applied either to induce deposition of apical hard substance or to bridge the time until root canal filling is possible. In a comparative clinical and radiological study in 171 young patients, Walkhoff's iodoform paste and calcium hydroxide were examined with regard to their inductive effect on the formation of an apical

Polyethylene glycol functionalized reduced graphene oxide coupled with zinc oxide composite adsorbent for removal of phenolic wastewater.

The development of agricultural activities and industrialization recently has various adverse impacts on living organisms. The ever-increasing problem of organic pollution has been an environmental concern to the community. Among these, phenolic pollutants like 2,4-dichlorophenol (2,4-DCP), phenol, 2-chlorophenol (2-CP), and bisphenol-A (BPA) are priority toxic pollutants that are continuously released into environment from many industries. In this work, a biocompatible zinc oxide incorporated polyethylene glycol functionalized reduced graphene oxide composite (RGO-PEG-ZnO) was synthesized and

Chromate-induced activation of hydrogen peroxide for oxidative degradation of aqueous organic pollutants.

The oxidation of organic compounds in water was investigated with using chromate as an activator of H(2)O(2). 4-chlorophenol (4-CP) was used as a main model substrate, and its degradation was successfully achieved at circumneutral pH. Unlike the traditional Fenton-based activation of H(2)O(2) that is mainly limited to acidic condition, the oxidative capacity of the proposed Cr(VI)/H(2)O(2) system is active over a wide range of pH 3-11. H(2)O(2) substitutes the oxo ligands of chromate by the peroxo ligands and, subsequently, converts chromate(VI) into a tetraperoxochromate(V) complex. The insta

Tetrachlorobisphenol A inhibits steroidogenesis in pubertal rat Leydig cells through disruption of copper homeostasis.

Tetrachlorobisphenol A (TCBPA) is a widely used flame retardant with potential endocrine-disrupting properties. This study investigated the effects of TCBPA on steroidogenesis in pubertal rat Leydig cells, focusing on copper homeostasis and on whether the cuproptosis pathway is engaged. Male Sprague-Dawley rats (35 days old) were administered TCBPA (0, 10, 100, and 200 mg/kg/day) for 21 days. TCBPA exposure (100 and 200 mg/kg) significantly reduced serum and testicular testosterone levels without affecting testis weight or Leydig cell numbers. TCBPA downregulated steroidogenic genes (Lhcgr, Sc

Cenobamate as an Early Adjunctive Treatment in Drug-Resistant Focal-Onset Seizures: An Observational Cohort Study.

BACKGROUND AND OBJECTIVES: Cenobamate (CNB) is a new antiseizure medication (ASM) to treat drug-resistant, focal-onset seizures. Data on its use in early therapy lines are not yet available, and clinicians frequently consider CNB to be a later ASM drug choice. We investigated the efficacy and safety of CNB as an early adjunctive treatment in drug-resistant, focal-onset seizures. METHODS: The study population were patients with drug-resistant, focal-onset seizures who were initiated with CNB after they did not respond to two or three lifetime ASMs, including all prior and concomitant ASMs. The

In vitro characterization of 3-chloro-4-hydroxybenzoic acid building block formation in ambigol biosynthesis.

The cyanobacterium Fischerella ambigua is a natural producer of polychlorinated aromatic compounds, the ambigols A-E. The biosynthetic gene cluster (BGC) of these highly halogenated triphenyls has been recently identified by heterologous expression. It consists of 10 genes named ab1-10. Two of the encoded enzymes, i.e. Ab2 and Ab3, were identified by in vitro and in vivo assays as cytochrome P450 enzymes responsible for biaryl and biaryl ether formation. The key substrate for these P450 enzymes is 2,4-dichlorophenol, which in turn is derived from the precursor 3-chloro-4-hydroxybenzoic acid. H

Cumulative and bolus in vitro contracture testing with 4-chloro-3-ethylphenol in malignant hyperthermia positive and negative human skeletal muscles.

In this study we evaluated the in vitro effects of 4-chloro-3-ethylphenol (CEP) using cumulative (12.5-200 micromol/L) or bolus (75 and 100 micromol/L) administrations, on muscle specimens from malignant hyperthermia (MH) susceptible and MH nonsusceptible patients, respectively. In the cumulative CEP in vitro contracture test, contractures were significantly greater in the MH susceptible compared with the MH nonsusceptible muscles in all concentrations between 25 and 100 micromol/L. There was no overlap between the diagnostic groups at 75 micromol/L of CEP, so this test appears to be feasible

Effects of pentachlorophenol on rat liver changes induced by hexachlorobenzene, with special reference to porphyria, and alterations in mixed function oxygenases.

Hexachlorobenzene (HCB, 1000 ppm) and 500 ppm pentachlorophenol (PCP) were fed separately or in combination to female Wistar rats. A control group was provided with standard food without HCB or PCP. Subgroups of 4 rats were killed after 1, 2, 4, 6 and 8 weeks. No significant difference was found between the amounts of HCB accumulated in the livers of the HCB and HCB + PCP fed rats. Administering HCB together with PCP caused a noticeable accumulation of PCP in the liver, compared to the results after administering HCB and PCP separately. In the HCB and HCB + PCP fed groups liver weight increase

The Pharmacokinetic and Pharmacodynamic Relationship of Clinically Used Antiseizure Medications in the Maximal Electroshock Seizure Model in Rodents.

The assessment of the efficacy of antiseizure medications (ASMs) in animal models of acute seizures has played a critical role in these drugs' success in clinical trials for human epilepsy. One of the most widely used animal models for this purpose is the maximal electroshock seizure (MES) model. While there are numerous published reports on the efficacy of conventional ASMs in MES models, there is a need to expand the understanding on the brain concentrations that are needed to achieve optimal levels of efficacy in this model. We assessed the pharmacokinetic/pharmacodynamic (PK/PD) profiles o
