Transcriptomic analysis reveals Toxoplasma gondii strain-specific differences in host cell response to dense granule protein GRA15.

Growth and replication of the protozoan parasite Toxoplasma gondii within host cell entail the production of several effector proteins, which the parasite exploits for counteracting the host's immune response. Despite considerable research to define the host signaling pathways manipulated by T. gondii and their effectors, there has been limited progress into understanding how individual members of the dense granule proteins (GRAs) modulate gene expression within host cells. The aim of this study was to evaluate whether T. gondii GRA15 protein plays any role in regulating host gene expression.

Absence of Specific Chlamydia trachomatis Inclusion Membrane Proteins Triggers Premature Inclusion Membrane Lysis and Host Cell Death.

Chlamydia trachomatis is a human pathogen associated with significant morbidity worldwide. As obligate intracellular parasites, chlamydiae must survive within eukaryotic cells for sufficient time to complete their developmental cycle. To promote host cell survival, chlamydiae express poorly understood anti-apoptotic factors. Using recently developed genetic tools, we show that three inclusion membrane proteins (Incs) out of eleven examined are required for inclusion membrane stability and avoidance of host cell death pathways. In the absence of specific Incs, premature inclusion lysis results

Does DNA replication direct locus-specific recombination during host immune evasion by antigenic variation in the African trypanosome?

All pathogens must survive host immune attack and, amongst the survival strategies that have evolved, antigenic variation is a particularly widespread reaction to thwart adaptive immunity. Though the reactions that underlie antigenic variation are highly varied, recombination by gene conversion is a widespread approach to immune survival in bacterial and eukaryotic pathogens. In the African trypanosome, antigenic variation involves gene conversion-catalysed movement of a huge number of variant surface glycoprotein (VSG) genes into a few telomeric sites for VSG expression, amongst which only a

Brucella TIR-like protein TcpB/Btp1 specifically targets the host adaptor protein MAL/TIRAP to promote infection.

Brucella spp. are known to avoid host immune recognition and weaken the immune response to infection. Brucella like accomplish this by employing two clever strategies, called the stealth strategy and hijacking strategy. The TIR domain-containing protein (TcpB/Btp1) of Brucella melitensis is thought to be involved in inhibiting host NF-κB activation by binding to adaptors downstream of Toll-like receptors. However, of the five TIR domain-containing adaptors conserved in mammals, whether MyD88 or MAL, even other three adaptors, are specifically targeted by TcpB has not been identified. Here, we

Experimental viral evolution to specific host MHC genotypes reveals fitness and virulence trade-offs in alternative MHC types.

The unprecedented genetic diversity found at vertebrate MHC (major histocompatibility complex) loci influences susceptibility to most infectious and autoimmune diseases. The evolutionary explanation for how these polymorphisms are maintained has been controversial. One leading explanation, antagonistic coevolution (also known as the Red Queen), postulates a never-ending molecular arms race where pathogens evolve to evade immune recognition by common MHC alleles, which in turn provides a selective advantage to hosts carrying rare MHC alleles. This cyclical process leads to negative frequency-de

Outer membrane protein A of bovine and ovine isolates of Mannheimia haemolytica is surface exposed and contains host species-specific epitopes.

Mannheimia haemolytica is the etiological agent of pneumonic pasteurellosis of cattle and sheep; two different OmpA subclasses, OmpA1 and OmpA2, are associated with bovine and ovine isolates, respectively. These proteins differ at the distal ends of four external loops, are involved in adherence, and are likely to play important roles in host adaptation. M. haemolytica is surrounded by a polysaccharide capsule, and the degree of OmpA surface exposure is unknown. To investigate surface exposure and immune specificity of OmpA among bovine and ovine M. haemolytica isolates, recombinant proteins r

Specific and genotypic identification of Cryptosporidium from a broad range of host species by nonisotopic SSCP analysis of nuclear ribosomal DNA.

The accurate identification of Cryptosporidium (Protozoa: Apicomplexa) species and genotypes is central to the understanding of the transmission and to the diagnosis and control of cryptosporidiosis. In this study, we demonstrate the effectiveness of nonisotopic SSCP analysis of a approximately 300 bp region of the small subunit (pSSU) of ribosomal DNA for the specific identification of and delineation among 18 different Cryptosporidium species and genotypes from a wide range of hosts. This mutation scanning approach allowed the rapid and reliable differentiation between species/genotypes diff

Filarial-specific antibody response in East African bancroftian filariasis: effects of host infection, clinical disease, and filarial endemicity.

The effect of host infection, chronic clinical disease, and transmission intensity on the patterns of specific antibody responses in Bancroftian filariasis was assessed by analyzing specific IgG1, IgG2, IgG3, IgG4, and IgE profiles among adults from two communities with high and low Wuchereria bancrofti endemicity. In the high endemicity community, intensities of the measured antibodies were significantly associated with infection status. IgG1, IgG2, and IgE were negatively associated with microfilaria (MF) status, IgG3 was negatively associated with circulating filarial antigen (CFA) status,

Regulation of OspE-related, OspF-related, and Elp lipoproteins of Borrelia burgdorferi strain 297 by mammalian host-specific signals.

In previous studies we have characterized the cp32/18 loci in Borrelia burgdorferi 297 which encode OspE and OspF orthologs and a third group of lipoproteins which possess OspE/F-like leader peptides (Elps). To further these studies, we have comprehensively analyzed their patterns of expression throughout the borrelial enzootic cycle. Serial dilution reverse transcription-PCR analysis indicated that although a shift in temperature from 23 to 37 degrees C induced transcription for all nine genes analyzed, this effect was often markedly enhanced in mammalian host-adapted organisms cultivated wit

Platform-Specific Immunoassay Development and Orthogonal Characterization for Improved Host Cell Protein Control in Vero Cell-Derived Vaccines.

Vero host cell proteins (HCPs) are critical process-related impurities in Vero cell-derived vaccines and require robust analytical control to support product quality, safety, and manufacturing consistency. During development of the V590 vaccine candidate, discontinuation of the original commercial Vero HCP ELISA prompted evaluation of alternative approaches for HCP monitoring and risk control. We compared first- and second-generation commercial ELISA kits, assessed the impact of process-representative HCP standards and anti-HCP antibodies on HCP quantitation, and characterized antibody coverag

Parents don't dictate the menu: Lack of strong host-specific parental effects may help maintain dietary generalism.

The environment or experiences of a parent generation can impact the fitness of the next generation, a phenomenon known as parental effects. While a high-quality diet can directly benefit an individual and a low-quality diet can reduce fitness, a parental effect occurs only when there is a transgenerational response to these stimuli that manifests in the individual's offspring. In dietary generalists, a parental effect could play an important role for offspring performance if the species experiences considerable variation in quality across host plants; positive parental effects may result from

BSMV-mediated genome editing exhibits host-specific heritability: germline transmission in barley and somatic edits in Nicotiana benthamiana.

BACKGROUND: Plant RNA virus-mediated guide RNA (gRNA) delivery represents a transformative advance in genome editing technologies. Unlike conventional transformation methods that rely on labor-intensive tissue culture and regeneration for each individual gRNA delivery, viral vectors can rapidly and systemically transmit gRNAs into pre-established Cas-expressing plants, providing an accelerated route for functional genomics and trait discovery directly in planta. However, key design parameters, including subgenomic promoter choice, transcript architecture, and their effects on viral fitness and

Impacts of artificial light at night on interactions between four trematode parasites and their marine invertebrate hosts: species-specific effects?

Parasitism is influenced by anthropogenic stressors, among which artificial light at night (ALAN) remains understudied. With the expansion of urban areas and the development of lighting technology, the range and intensity of ALAN's effects on host-parasite interactions are increasing. Trematodes, with their complex life cycles, are particularly sensitive to anthropogenic stressors, especially during the transmission of motile cercariae (infective stages) from the first to the second intermediate host while exposed to external conditions. We examined how three types of ALAN (warm white LED, col

Comparative pangenomics of Pasteurella multocida uncovers adhesins specific to capsular serogroup B strains associated with bovine host tropism.

Haemorrhagic septicaemia (HS), an acute and highly fatal disease of bovines in tropical countries of Africa and Asia, including India, is primarily caused by Pasteurella multocida serogroup B:2 strains. Although P. multocida strains are ubiquitous and known to infect a wide range of host species with varied clinical manifestations hitherto, the molecular basis of host tropism remains unknown. In this study, we present an integrated genomic, structural, and molecular dynamics framework to investigate how P. multocida serogroup B strains selectively adhere to bovine tissues. Development of a Specific Primer-Based Method for Detecting Bithynia siamensis siamensis, an Intermediate Host of the Liver Fluke Opisthorchis viverrini.

BACKGROUND: The liver fluke Opisthorchis viverrini is a major public health concern in Southeast Asia and a leading cause of cholangiocarcinoma. Its transmission relies on freshwater snails of the genus Bithynia as the first intermediate hosts. However, accurately identifying these morphologically similar snails remains challenging. OBJECTIVES: This study aimed to develop species-specific primers for the accurate molecular identification of Bithynia siamensis siamensis, which serves as an important first intermediate host of O. viverrini. METHODS: Random amplified polymorphic DNA polymerase

Stage-specific gene expression profiling of Toxocara canis in canine and human hosts: Implications for the pathogenesis and transmission of zoonotic diseases.

BACKGROUND: Toxocara canis is a zoonotic nematode that mainly infects dogs but can produce visceral, ocular, and neurological toxocariasis in humans. The parasite completes its life cycle within dogs. The molecular processes that enable T. canis to adapt to its host remain largely unknown despite the significance of the parasite to public health. AIM: To understand the molecular basis of immune evasion, tissue migration, and survival strategies, researchers analyzed T. canis gene expression during different life stages within canine and human hosts. METHODS: Third-stage larvae (L3) and adult

Temporal transcriptomics identifies early-response and infection-condition-specific modules guiding host-directed anti-EBOV therapeutics.

Ebola virus (EBOV) is among the most lethal human pathogens, yet effective treatment options remain limited. While extensive efforts have elucidated the functions of viral proteins, the temporal orchestration of host transcriptional responses-and their exploitation by EBOV during infection-remains poorly defined. Here, we performed integrated time-series transcriptomic profiling using both RNA-seq and microarray analyses to characterize dynamic host and viral gene expression. Through the combination of co-expression network construction and causal inference modeling, we reconstructed the seque

Identification of specific host cell potassium ion channels that mediate escape of bunyamwera virus from late endosome compartments.

The Orthobunyavirus genus within the Peribunyaviridae family of enveloped arthropod-borne negative-sense RNA viruses includes species associated with serious or fatal disease in both animals and humans such as Schmallenberg, La Crosse and Oropouche viruses. Orthobunyaviruses (OBVs) are internalized into cells by endocytosis and release their genomes following fusion with late endosome membranes, triggered by low pH of the luminal milieu. There is mounting evidence to suggest OBV endosomal escape is also influenced by potassium ions (K+), which increase in concentration as endosomes mature. End

Reframing Antimicrobial Peptides beyond Direct Antimicrobial Activity toward Host-Directed Functions and Disease-Specific Therapeutic Applications.

Antimicrobial peptides (AMPs) have been extensively investigated as alternatives to conventional antibiotics to combat multidrug-resistant bacteria. This narrow antibiotic-replacement framework underestimates the biological and translational potential of AMPs. The present review reframes them as disease-context-dependent therapeutic platforms rather than universal antibiotic substitutes. In particular, this review critically examines their microbiological basis, mechanisms of action, emerging applications, engineering strategies, formulation approaches, translational limitations, and criteria
