An update on the ability of St. John's wort to affect the metabolism of other drugs.

INTRODUCTION: Hypericum perforatum (HP), more commonly known as St. John's wort, is a popular medicinal herb used for the treatment of depression. HP affects the pharmacokinetics of many drugs by inducing cytochrome P450 (CYP) isozymes, such as CYP3A4, CYP2C19, CYP2C9, and the P-glycoprotein (P-gp) transporter. AREAS COVERED: This review focuses on drugs that are metabolized by CYP3A4, CYP2C19, CYP2C9 and P-gp as their plasma concentrations show the effects of concomitant use of HP. For the purpose of this review, all electronic databases such as PubMed, Scopus, Google Scholar and Cochrane li

[Herbal drug-drug interaction and adverse drug reactions].

Herbal medicines are used by many patients. Their known or potential adverse events should be taken into account during treatment with herbal medicines. In this article adverse effects of commonly used herbs are presented. St. John's wort is known to be a potent inducer of cytochrome P450 (CYP) 3A4 leading to reduced blood concentrations of a number of CYP3A4 substrates. For many other combinations evidence is sparse but due to a number of case reports of adverse interactions they should only cautiously be combined with certain critical dose drugs until their risk is fully assessed. Pertinent

The Effect of Hesperidin on Inflammatory Response and Oxidant-Antioxidant Systems in Benzo[a]pyrene-Exposed Non-Small Cell Lung Cancer (A549) Cells.

Lung cancer represents a substantial global oncology burden, exhibiting mortality rates that surpass those of prostate, pancreatic, and breast cancers. Extensive research has established a correlation between exposure to polycyclic aromatic hydrocarbon mixtures-particularly those containing benzo(a)pyrene (BaP)-and elevated risks of pulmonary and dermal malignancies across various species, including humans. Hesperidin (HSP), a prominent bioactive flavonoid found in citrus fruits and medicinal herbs such as Hypericum perforatum, is recognized for its diverse pharmacological properties. The pres

Hypericin prolongs action potential duration in hippocampal neurons by acting on K+ channels.

BACKGROUND AND PURPOSE: Synaptic deficiency is generally accepted to be involved in major depression, and accordingly classic antidepressants exert their effects through enhancing synaptic efficiency. Hypericin is one of the major active constituents of extracts of St. John's Wort (Hypericum perforatum L.) with antidepressive actions, but little is known about its therapeutic mechanisms. Our aim was to explore whether hypericin has a modulatory effect on neuronal action potential (AP) duration by acting on voltage-gated ion channels. EXPERIMENTAL APPROACH: We used voltage-clamp and current-cl

[The virtue of that precious balsam...: approach to Don Quixote from the psychopharmacological perspective].

The most outstanding novel of the Spanish literature, Don Quixote, represents the source to which the different specialists who intend to deepen their knowledge of the late Renaissance society usually address. This masterpiece of Miguel de Cervantes has been frequently approached from the psychopathological perspective to obtain a psychiatric diagnosis of its main character, Alonso Quijano. Also, other clinical approaches from the traumatological and general therapeutical view (oils, ointments, balms and other pharmacy preparations) have been frequent. We have tackled Don Quixote from the psyc

A neuroprotective herbal mixture inhibits caspase-3-independent apoptosis in retinal ganglion cells.

It was previously demonstrated that Menta-FX, a mixture of Panax quinquefolius L. (PQE), Ginkgo biloba (GBE), and Hypericum perforatum extracts (HPE), enhances retinal ganglion cell survival after axotomy. However, the mechanisms of neuroprotection remain unknown. The aim of this study is to elucidate the neuroprotective mechanisms of Menta-FX. Since PQE, GBE and HPE have all been observed to display anti-oxidative property, the involvement of anti-oxidation in Menta-FX's neuroprotective effect was investigated. Menta-FX lowered nitric oxide (NO) content in axotomized retinas without affecting

Better tolerability of St. John's wort extract WS 5570 compared to treatment with SSRIs: a reanalysis of data from controlled clinical trials in acute major depression.

Selective serotonin reuptake inhibitors (SSRIs) like paroxetine have replaced older antidepressants mainly because of a more favorable safety profile, but they are still associated with burdensome side effects. We investigate the tolerability of St. John's wort extract WS 5570, a herbal antidepressant with proven efficacy, in comparison to paroxetine and other SSRIs and placebo. A reanalysis was performed based on the original data from four controlled clinical trials during which 1661 outpatients with major depression (Diagnostic and Statistical Manual of Mental Disorders, 4th Edition criteri

The extent of induction of CYP3A by St. John's wort varies among products and is linked to hyperforin dose.

OBJECTIVE: Induction of CYP3A by St. John's wort (SJW) extracts with high hyperforin (HYF) content is well described. Since SJW products vary in the amount of HYF and other main constituents, the aim of the study was to evaluate the effect on CYP3A function of SJW preparations with a range from very low to high HYF content. METHODS: Forty-two male, healthy volunteers were randomized into six parallel SJW medication groups with varying composition especially with regard to HYF content. Midazolam plasma concentration profiles were characterized after a single oral dose of 7.5 mg midazolam on th

Hypericin and hyperforin from St. John's Wort are potent intestinal UGT inhibitors with distinct binding modes: mechanistic and quantitative prediction of clinical drug-drug interaction risk.

St. John's Wort (SJW; Hypericum perforatum) is widely used for mild-to-moderate depression and is known to induce certain cytochrome P450 enzymes (notably CYP3A4, with reported effects on CYP2C9 and CYP2C19) and intestinal P-glycoprotein, but its effects on UDP-glucuronosyltransferases (UGTs), the principal phase-II drug-metabolizing enzymes, remain largely uncharacterized. Using recombinant human UGTs with 4-methylumbelliferone as the probe substrate, we characterized inhibition by the two major SJW constituents, hypericin and hyperforin, across eleven isoforms, and combined AlphaFold2-based

Anthelmintic Activity of Traditional Medicinal Plants Used in Europe.

Synthetic anthelmintic drugs not only contribute to the development of pathogen resistance and environmental pollution but also to the development of pathogen resistance. Therefore, identifying the anthelmintic properties of widely used medicinal plants could be of great practical interest to veterinary and human medicine. In our experiment, we evaluated the in vitro survivability of the noninvasive and invasive (L1-2 and L3, respectively) larvae of Strongyloides papillosus and Haemonchus contortus subject to aqueous solutions of ethanolic tinctures of traditional medicinal plants (46 species)

Hyperforin Attenuates Scopolamine Induced Alzheimer's Pathology in Rat Model: Abrogation of Caspase-1/11 Mediated Pyroptosis via Inhibition of HMGB1-Mediated RAGE Pathway.

Alzheimer's disease (AD) is a neurodegenerative disorder associated with the loss of memory, accumulation of amyloid-beta (Aβ) plaques, and inflammation of the nervous system. Scopolamine, an antagonist of muscarinic receptors, is commonly used to mimic the cognitive and behavioral deficits of AD in laboratory animals. In this study, we aimed to test the neuroprotective properties of hyperforin (HPF), a compound extracted from the St. John's wort plant (Hypericum perforatum), in a scopolamine rat model of AD. Sprague-Dawley rats were divided into four groups: control (saline), scopolamine (10
