Loudness perception affected by early age hearing loss.

Tinnitus and hyperacusis, commonly seen in adults, are also reported in children. Although clinical studies found children with tinnitus and hyperacusis often suffered from recurrent otitis media, there is no direct study on how temporary hearing loss in the early age affects the sound loudness perception. In this study, sound loudness changes in rats affected by perforation of the tympanic membranes (TM) have been studied using an operant conditioning based behavioral task. We detected significant increases of sound loudness and susceptibility to audiogenic seizures (AGS) in rats with bilater

An anatomical assessment of the supracerebellar midline and paramedian approaches to the inferior colliculus for auditory midbrain implants using a neuronavigation model on cadaveric specimens.

The inferior colliculus (IC) is an alternative site for electrode placement in neural deafness due to its surgical accessibility and its well-known tonotopic stratification. In patients where tumor surgery has already occurred and the cerebellopontine angle contains scar tissue or tumor-remnants, midline and paramedian supracerebellar approaches are alternative routes. They are often avoided due to concerns regarding the venous drainage of the cerebellum, the electrode trajectory and the course of the electrode cable. We studied these surgical routes in five neuronavigated fixed cadaveric spec

Causality of parenchymal and vascular changes in rats with experimental thiamine deficiency encephalopathy.

The causality of vascular and parenchymal damage to the central nervous system (CNS) was examined in rats with thiamine deficiency. Male Sprague-Dawley rats were divided into two groups; one was given a thiamine-deficient diet (TDD) and injected intraperitoneally with 10 micrograms/100 g bodyweight pyrithiamine (PT) in order to analyze morphometrically the topographical and sequential relationship between vascular and parenchymal changes and vasodilatation, and the other was given a TDD and 50 micrograms/100 g bodyweight PT in order to determine hemorrhagic sites using serial sections. Temporal dynamics of contextual processing in the inferior colliculus of a rat model of autism: Sex- and age-dependent trajectories.

In autism, sensory differences have been linked to altered contextual processing and to how temporal structure supports the formation of regularities and predictions. Here, we examined how temporal and contextual auditory processing are shaped by sex, development, and prenatal valproic acid (VPA) exposure in the non-lemniscal inferior colliculus at low sound levels. Single-unit recordings from control and VPA-exposed rats were obtained using oddball and cascade paradigms, allowing dissociation of mismatch (iMM), repetition suppression (iRS), and prediction-error (iPE) components in both respon

Pairing tones with vagus nerve stimulation improves brain stem responses to speech in the valproic acid model of autism.

Receptive language deficits and aberrant auditory processing are often observed in individuals with autism spectrum disorders (ASD). Symptoms associated with ASD are observed in rodents prenatally exposed to valproic acid (VPA), including deficits in speech sound discrimination ability. These perceptual difficulties are accompanied by changes in neural activity patterns. In both cortical and subcortical levels of the auditory pathway, VPA-exposed rats have impaired responses to speech sounds. Developing a method to improve these neural deficits throughout the auditory pathway is necessary. The

Neural representations of concurrent sounds with overlapping spectra in rat inferior colliculus: Comparisons between temporal-fine structure and envelope.

Perceptual segregation of multiple sounds, which overlap in both time and spectra, into individual auditory streams is critical for hearing in natural environments. Some cues such as interaural time disparities (ITDs) play an important role in the segregation, especially when sounds are separated in space. In this study, we investigated the neural representation of two uncorrelated narrowband noises that shared the identical spectrum in the rat inferior colliculus (IC) using frequency-following-response (FFR) recordings, when the ITD for each noise stimulus was manipulated. The results of this

Cholinergic Modulation of Stimulus-Specific Adaptation in the Inferior Colliculus.

UNLABELLED: Neural encoding of an ever-changing acoustic environment is a complex and demanding process that depends on modulation by neuroactive substances. Some neurons of the inferior colliculus (IC) exhibit "stimulus-specific adaptation" (SSA), i.e., a decrease in their response to a repetitive sound, but not to a rare one. Previous studies have demonstrated that acetylcholine (ACh) alters the frequency response areas of auditory neurons and therefore is important in the encoding of spectral information. Here, we address how microiontophoretic application of ACh modulates SSA in the IC of

Expression of tumor necrosis factor-α and interleukin-1β genes in the cochlea and inferior colliculus in salicylate-induced tinnitus.

BACKGROUND: Changes in the gene expressions for tumor necrosis factor-α (TNF-α) and/or interleukin-1β (IL-1β) during tinnitus have not been previously reported. We evaluated tinnitus and mRNA expression levels of TNF-α, IL-1β, and N-methyl D-aspartate receptor subunit 2B (NR2B) genes in cochlea and inferior colliculus (IC) of mice after intraperitoneal injections of salicylate. METHODS: Forty-eight 3-month-old male SAMP8 mice were randomly and equally divided into two groups: salicylate-treated and saline-treated. All mice were trained to perform an active avoidance task for 5 days. Once cond

Effects of salicylate on transient outward and delayed rectifier potassium channels in rat inferior colliculus neurons.

The effects of salicylate (a tinnitus inducer) were studied on the transient outward potassium current (I(K(A))) and the delayed rectifier potassium current (I(K(DR))) in acutely dissociated rat inferior colliculus neurons by the whole-cell voltage-clamp method. Salicylate's inhibition of the amplitude of I(K(A)) and I(K(DR)) was concentration-dependent. The IC(50) values for the blocking action of salicylate on I(K(A)) and I(K(DR)) were 2.27 and 0.80 mM, respectively. At a concentration of 1 mM, salicylate significantly shifted the activation and inactivation curves of I(K(DR)) negatively by

Phase spectral analysis of auditory brainstem response in cats.

In order to apply the phase spectral analysis of auditory brainstem response (ABR) to the clinical diagnostic test, the phase spectra of ABRs were investigated under adequate stimulus intensity in normal cats and after the destruction of acoustic nerve and brainstem auditory pathways in cats. The results were as follows; i) In normal ABRs, the phase spectra were mainly composed of three frequency components at 0-300 Hz (component A), 300-900 Hz (component B) and 900-1500 Hz (component C). ii) A greater decrease of the component synchrony measure (CSM) occurred if ipsilateral destruction was pe

Acoustic trauma induced the alteration of the activity balance of excitatory and inhibitory neurons in the inferior colliculus of mice.

We examined the effect of acoustic trauma on the spontaneous activities of the glutamatergic and GABAergic neurons in the inferior colliculus (IC) of mice. Optogenetics was used to identify the neuron type. In control animals, the spontaneous firing rate was higher in GABAergic neurons than in glutamatergic neurons. However, in the animals with acoustic trauma, the balance of spontaneous activities between glutamatergic and GABAergic neurons was inverted. The spontaneous firing rate was enhanced in glutamatergic neurons only, with bursting episodes occurring frequently. Moreover, the spike sha

The relation between perception and brain activity in gaze-evoked tinnitus.

Tinnitus is a phantom sound percept that can be severely disabling. Its pathophysiology is poorly understood, partly due to the inability to objectively measure neural correlates of tinnitus. Gaze-evoked tinnitus (GET) is a rare form of tinnitus that may arise after vestibular schwannoma removal. Subjects typically describe tinnitus in the deaf ear on the side of the surgery that can be modulated by peripheral eye gaze. This phenomenon offers a unique opportunity to study the relation between tinnitus and brain activity. We used functional magnetic resonance imaging in humans to show that in n

L-type calcium channel modulation reveals the relationship between neuronal synchrony and hyperactivity in the inferior colliculus following noise-induced hearing loss.

Previous studies have established the protective effects of calcium channel inhibition on the peripheral auditory system in response to noise exposure. While these studies implicate L-type calcium channels (LTCCs) in noise-generated dysfunction in the auditory periphery, contributions of LTCCs to noise-induced central dysfunction remains unclear. To begin to elucidate the roles of LTCCs in hearing, peripheral and central auditory function were assessed longitudinally after LTCC inhibition. Neuronal synchrony and activity were assessed by analyzing wave I (peripheral) and wave V (central) audit

Underlying mechanisms of tinnitus: review and clinical implications.

BACKGROUND: The study of tinnitus mechanisms has increased tenfold in the last decade. The common denominator for all of these studies is the goal of elucidating the underlying neural mechanisms of tinnitus with the ultimate purpose of finding a cure. While these basic science findings may not be immediately applicable to the clinician who works directly with patients to assist them in managing their reactions to tinnitus, a clear understanding of these findings is needed to develop the most effective procedures for alleviating tinnitus. PURPOSE: The goal of this review is to provide audiolog

Spontaneous activity in the inferior colliculus of CBA/J mice after manipulations that induce tinnitus.

Several physiological studies have linked experimentally induced tinnitus to increases in the spontaneous activity of auditory neurons. These results have led to the proposal of hyperactivity models of tinnitus in which elevated neural activity in the absence of auditory stimulation is perceived as phantom sound. Such models are appealing in their simplicity but remain controversial because a generalized elevation of spontaneous rates may not be observed after treatments that induce tinnitus in humans and experimental animals. Our study addressed these issues by characterizing the effects of c

[Symmetrical necrosis in the gray matter of the brainstem and spinal cord. Two adult cases of anoxic encephalopathy].

We describe distinctive necrosis in the brainstem of two adult patients of anoxic encephalopathy. The patients were a 75-year-old man and an 82-year-old woman, who were resuscitated after cardiac arrest that lasted about 20 minutes and artificially ventilated. The first patient remained comatose without recovery of brainstem reflexes and had persistent hypotension requiring continuous infusion of catecholamine until death that occurred two weeks after the onset of illness. In the second patient, her general circulation recovered on the next day resuscitation, and traces of brainstem reflexes r

Overexpression of Isl1 under the Pax2 Promoter, Leads to Impaired Sound Processing and Increased Inhibition in the Inferior Colliculus.

The LIM homeodomain transcription factor ISL1 is essential for the different aspects of neuronal development and maintenance. In order to study the role of ISL1 in the auditory system, we generated a transgenic mouse (Tg) expressing Isl1 under the Pax2 promoter control. We previously reported a progressive age-related decline in hearing and abnormalities in the inner ear, medial olivocochlear system, and auditory midbrain of these Tg mice. In this study, we investigated how Isl1 overexpression affects sound processing by the neurons of the inferior colliculus (IC). We recorded extracellular ne

The immediate effects of acoustic trauma on excitation and inhibition in the inferior colliculus: A Wiener-kernel analysis.

Noise-induced tinnitus and hyperacusis are thought to correspond to a disrupted balance between excitation and inhibition in the central auditory system. Excitation and inhibition are often studied using pure tones; however, these responses do not reveal inhibition within the excitatory pass band. Therefore, we used a Wiener-kernel analysis, complemented with singular value decomposition (SVD), to investigate the immediate effects of acoustic trauma on excitation and inhibition in the inferior colliculus (IC). Neural responses were recorded from the IC of three anesthetized albino guinea pigs

[The significance of stress: its role in the auditory system and the pathogenesis of tinnitus].

Emotional stress is often associated with auditory phenomena such as hyperacusis, tinnitus, Ménière's disease and vertigo. Stress develops as a result of a person's attempts to come to terms with the increased or unexpected demands of his or her environment. Stress serves to protect one from physical danger and to temporarily increase one's performance in order to increase the probability of survival. Sleep and appetite are particularly reduced, while anxiety increases. The mental changes induced by stress may contribute to the onset or exacerbation of tinnitus. The following links exist betwe

Temporal processing in inferior colliculus and auditory cortex affected by high doses of salicylate.

Speech recognition and language learning can be affected by both peripheral and central auditory system impairment. However, whether sensorineural hearing loss would affect central auditory processing is not clear. Recent studies found that salicylate not only affects outer hair cell motility in the cochlea, but also blocks GABAergic neuron activities in central nervous systems. This provides a good animal model to evaluate the role of sensorineural hearing loss and central inhibition in auditory temporal processing. In this study, gap prepulse inhibition (gap-PPI) of the acoustic startle refl
