Regulation of feeding and defecation in Drosophila by trpγ, piezo, and DH44R2.

Normal gastrointestinal (GI) motility, including defecation, is crucial for nutrient absorption, energy balance, and overall health in species ranging from insects to humans. Disruptions in GI motility can lead to conditions like constipation or severe diseases. Mechanosensors, including TRP channels and Piezo, are known to play key roles in regulating gut physiology in Drosophila melanogaster, but their precise involvement in defecation is not fully understood. Additionally, neuropeptides like DH44 have been implicated in gut regulation. This study explores the roles of Trpγ, Diuretic hormone

Molecular action of larvicidal flavonoids on ecdysteroidogenic glutathione S-transferase Noppera-bo in Aedes aegypti.

BACKGROUND: Mosquito control is a crucial global issue for protecting the human community from mosquito-borne diseases. There is an urgent need for the development of selective and safe reagents for mosquito control. Flavonoids, a group of chemical substances with variable phenolic structures, such as daidzein, have been suggested as potential mosquito larvicides with less risk to the environment. However, the mode of mosquito larvicidal action of flavonoids has not been elucidated. RESULTS: Here, we report that several flavonoids, including daidzein, inhibit the activity of glutathione S-tra

Interfering with cholesterol metabolism impairs tick embryo development and turns eggs susceptible to bacterial colonization.

Cholesterol is a known precursor of arthropod molecules such as the hormone 20-hydroxyecdysone and the antimicrobial boophiline, a component of tick egg wax coat. Because the cholesterol biosynthetic pathway is absent in ticks, it is necessarily obtained from the blood meal, in a still poorly understood process. In contrast, dietary cholesterol absorption is better studied in insects, and many proteins are involved in its metabolism, including Niemann-Pick C (NPC) transporter and acyl-CoA:cholesterol acyltransferase (ACAT), as well as enzymes to convert between free cholesterol and esterified

Azadirachtin acting as a hazardous compound to induce multiple detrimental effects in Drosophila melanogaster.

Azadirachtin, a tetranortriterpenoid botanical insecticide, has varied sub-lethal effects against many insect pests, including antifeedant, repellent, and growth regulatory. Despite extensive studies of the mechanisms that underline these physiological effects, little attention has been given to multiple toxic effects of azadirachtin under a coherent concentration, and there is no definitive overarching consensus on its toxicity. Here, we investigated multiple sub-lethal effects induced by 4 mg L-1 of azadirachtin, which did not elicit antifeedant behavior in Drosophila melanogaster, on metric

Redox Control of Aphid Resistance through Altered Cell Wall Composition and Nutritional Quality.

The mechanisms underpinning plant perception of phloem-feeding insects, particularly aphids, remain poorly characterized. Therefore, the role of apoplastic redox state in controlling aphid infestation was explored using transgenic tobacco (Nicotiana tabacum) plants that have either high (PAO) or low (TAO) ascorbate oxidase (AO) activities relative to the wild type. Only a small number of leaf transcripts and metabolites were changed in response to genotype, and cell wall composition was largely unaffected. Aphid fecundity was decreased significantly in TAO plants compared with other lines. Lea

Synergistic action of E74B and ecdysteroid receptor in activating a 20-hydroxyecdysone effector gene.

A number of insect effector genes activated by the steroid hormone 20-hydroxyecdysone (20E) are dually controlled by the ecdysteroid receptor (EcR/USP) and products of ecdysteroid early responsive genes (E74, E75, and Broad). However, the molecular mechanism of this dual action is poorly understood. Here we examined transcriptional activation of the vitellogenin (Vg) gene in the yellow fever mosquito, Aedes aegypti, by EcR/USP and E74 in response to an elevation of 20E titers. There are two isoforms of the Aedes E74 gene, AaE74A and AaE74B, which have a common C-terminal Ets DNA-binding domain

Biotin-containing proteins of the insect nervous system, a potential source of interference with immunocytochemical localization procedures.

When the biotinylated Manduca sexta adipokinetic hormone gene was used as a probe for in situ hybridization, the intrinsic neurosecretory cells were stained with a biotin detection system that contained streptavidin or avidin. Further experiments showed that the DNA probe was not necessary for staining these cells by streptavidin-alkaline phosphatase, and that they were not stained by alkaline phosphatase alone. Similarly, the intrinsic neurosecretory cells were stained directly by streptavidin conjugated to a fluorescent dye. Other parts of the central nervous system could also be stained wit

Functional properties of the PTH/PTHrP receptor.

The PTH/PTHrP receptor belongs to a novel family of G-protein-coupled receptors which also includes an insect receptor for a diuretic hormone and the protein encoded by a genomic DNA clone from Caenorhabditis elegans. Despite significant structural conservation, rat, opossum, and human PTH/PTHrP receptor homologs display distinct functional characteristics when tested with either [Arg2, Tyr34]hPTH(1-34)amide or [Nle8.18, Tyr34]bPTH(7-34)-amide. These PTH analogs, and chimeras between rat/opossum and between rat/human PTH/PTHrP receptors, led to the identification of receptor residues that appe

Regulation, function and potential origin of the Drosophila gene spalt adjacent, which encodes a secreted protein expressed in the early embryo.

During early embryogenesis of Drosophila the spatial and temporal expression patterns of the region-specific homeotic gene spalt (sal) and the neighbouring gene spalt adjacent (sala) extensively overlap. We show that the initial expression patterns of the two genes in the blastoderm also have identical genetic controls. However, while sal encodes a transcription factor, sala encodes a precursor protein from which a functional signal peptide is cleaved off to generate the secreted sala protein. Ectopic expression or absence of sala protein does not affect embryonic development, adult viability

Biochemical and histopathological investigation to study the impact of pyriproxyfen exposure on ovarian morphology and reproductive function in female rats.

BACKGROUND: Pyriproxyfen (PYR) is a pyridine-based broad-spectrum insect growth regulator and pesticide which works as an analogue of juvenile hormone. Its exposure to aquatic animals and crops is linked with various hazardous effects on biological functions. We aimed to find the possible reprotoxic effects of pyriproxyfen in adult female Sprague-Dawley rats through histological and biochemical approaches. METHODS: Adult female rats were assigned to four groups and were administered 0 mg/kg (Control), 62 mg/kg b.w, 124 mg/kg b.w, and 186 mg/kg b.w., of PYR dissolved in distilled water for 28

The role of the microbiome in the neurobiology of social behaviour.

Microbes colonise all multicellular life, and the gut microbiome has been shown to influence a range of host physiological and behavioural phenotypes. One of the most intriguing and least understood of these influences lies in the domain of the microbiome's interactions with host social behaviour, with new evidence revealing that the gut microbiome makes important contributions to animal sociality. However, little is known about the biological processes through which the microbiome might influence host social behaviour. Here, we synthesise evidence of the gut microbiome's interactions with var

AOP Report: Inhibition of Chitin Synthase 1 Leading to Increased Mortality in Arthropods.

Arthropods (including insects, crustaceans, and arachnids) rely on the synthesis of chitin to complete their life cycles (Merzendorfer 2011). The highly conserved chitin synthetic process and the absence of this process in vertebrates make it an exploitable target for pest management and veterinary medicines (Merzendorfer 2013; Junquera et al. 2019). Susceptible, nontarget organisms, such as insects and aquatic invertebrates, exposed to chitin synthesis inhibitors may suffer population declines, which may have a negative impact on ecosystems and associated services. Hence, it is important to p

Emamectin benzoate induced enzymatic and transcriptional alternation in detoxification mechanism of predatory beetle Paederus fuscipes (Coleoptera: Staphylinidae) at the sublethal concentration.

In this study, the detoxification enzyme activity and the transcriptional profile changes in the second instar through RNA-sequencing technology due to emamectin benzoate (EB) were assessed. The cytochrome P450 monooxygenases (P450) enzyme activity was not altered by EB due to the change in concentration and exposure time in all treatments. The glutathione S-transferase (GST) enzyme was not considerably varying in all treatments, while exposure time significantly changed the enzyme activity. Results showed that the esterase (Ests) activity was elevated with the increasing concentrations and ex

Juvenile hormone regulation of microRNAs is mediated by E75 in the Dengue vector mosquito Aedes aegypti.

MicroRNAs (miRNAs) are small noncoding RNAs that play critical roles in controlling posttranscriptional gene regulation and have a profound effect on mosquito reproduction and metabolism. Juvenile hormone (JH) is critical for achieving reproductive competence in the main vector of human arboviral diseases, Aedes aegypti We report a JH-mediated mechanism governing miRNA expression. Using a transcription factor screen with multiple primary miRNA (pri-miRNA) promoters, we identified that the Ecdysone-induced protein E75 (E75) isoform (E75-RD) induced miRNA gene promoter activity. Regulation of growth-blocking peptide expression during embryogenesis of the cabbage armyworm.

Growth-blocking peptide (GBP) is an insect cytokine with diverse biological functions. Northern blot analysis revealed high heterogeneity in the size distribution of GBP mRNAs as well as in the tissues where they are detected. The spatio-temporal transcription pattern is dynamic, especially during embryogenesis. Gel shift assays demonstrated that the cabbage armyworm embryo nuclear extract specifically binds to a 178-bp element, at position +234 to +411 from the transcription start site of the 1.3 kb GBP transcript, in which two Drosophila Deformed (Dfd) binding sites are repeated in tandem. Evaluating the sterilizing effect of pyriproxyfen treated mosquito nets against Anopheles gambiae at different blood-feeding intervals.

Pyrethroid resistant malaria vectors are widespread throughout sub-Saharan Africa and new insecticides with different modes of action are urgently needed. Pyriproxyfen is a juvenile hormone mimic that reduces fecundity and fertility of adult Anopheles mosquitoes when used as a contact insecticide. A long-lasting insecticidal net incorporating pyriproxyfen is under development. As wild, host-seeking females may succeed in blood-feeding at different intervals after initial contact with mosquito nets the aim of this study was to determine the effect that age and gonotrophic status (nulliparous or

Regulation of a decapentaplegic midgut enhancer by homeotic proteins.

The clustered homeotic genes encode transcription factors that regulate pattern formation in all animals, conferring cell fates by coordinating the activities of downstream 'target' genes. In the Drosophila midgut, the Ultrabithorax (Ubx) protein activates and the abdominalA (abd-A) protein represses transcription of the decapentaplegic (dpp) gene, which encodes a secreted signalling protein of the TGF beta class. We have identified an 813 bp dpp enhancer which is capable of driving expression of a lacZ gene in a correct pattern in the embryonic midgut. The enhancer is activated ectopically in

The andropin gene and its product, a male-specific antibacterial peptide in Drosophila melanogaster.

In our study of the cecropin locus in Drosophila we have found a gene for a new peptide, andropin, with antibacterial properties. Transcripts from this gene, Anp, could be detected in newly eclosed males and reached steady-state levels after 1 day. Transcription was strongly induced in response to mating and is strictly confined to the ejaculatory duct of adult males. The deduced peptide sequence reveals a hydrophobic amino terminus with striking similarity to the signal peptide of the cecropins. The sequence of the predicted mature andropin shows no direct homology with the cecropins, but the
