Design of a microneedle-based enzyme biosensor using a simple and cost-effective electrochemical strategy to monitor superoxide anion released from cancer cells.

Early detection of Reactive oxygen species (ROS) concentration is very important in cancer diagnosis, pathological examinations, and health screening. Studies show that changes in ROS concentration occurs in a short time, causing irreparable damage to living cells and organs. Miniaturized sensors and microelectrodes are capable of online monitoring of electrochemical reactions both in vitro and in vivo. In this study, an enzymatic biosensor based on an electrochemically roughened gold microneedle electrode (RAuME) has been developed to measure superoxide anion released from prostate cancer cel

Ultrastructural Analysis of Neuroimplant-Parenchyma Interfaces Uncover Remarkable Neuroregeneration Along-With Barriers That Limit the Implant Electrophysiological Functions.

Despite increasing use of in vivo multielectrode array (MEA) implants for basic research and medical applications, the critical structural interfaces formed between the implants and the brain parenchyma, remain elusive. Prevailing view assumes that formation of multicellular inflammatory encapsulating-scar around the implants [the foreign body response (FBR)] degrades the implant electrophysiological functions. Using gold mushroom shaped microelectrodes (gMμEs) based perforated polyimide MEA platforms (PPMPs) that in contrast to standard probes can be thin sectioned along with the interfacing

Dominant and recessive congenital myasthenic syndromes caused by SYT2 mutations.

INTRODUCTION/AIMS: We studied a patient with a congenital myasthenic syndrome (CMS) caused by a dominant mutation in the synaptotagmin 2 gene (SYT2) and compared the clinical features of this patient with those of a previously described patient with a recessive mutation in the same gene. METHODS: We performed electrodiagnostic (EDX) studies, genetic studies, muscle biopsy, microelectrode recordings and electron microscopy (EM). RESULTS: Both patients presented with muscle weakness and bulbar deficits, which were worse in the recessive form. EDX studies showed presynaptic failure, which was m

Omeprazole induces high intraglandular pressure in the rat gastric mucosa.

BACKGROUND & AIMS: In a few patients receiving high doses of omeprazole for a prolonged period, cystic changes have developed in the gastric mucosa, possibly consisting of enlarged gland lumina. To investigate this question, the luminal pressure in gastric glands was studied after omeprazole treatment in Inactin-anesthetized rats. METHODS: The gland luminal pressure and mucus thickness were measured with microelectrodes. For pressure measurements (servo-null technique) the microelectrode was inserted into a gland at an angle of approximately 30 degrees to the mucosal surface. RESULTS: Animal

The effects of neuron morphology and spatial distribution on the selectivity of dorsal root ganglion stimulation.

Objective. For prosthesis users, sensory feedback that appears to come from the missing limb can improve function, confidence, and phantom limb pain. Numerous pre-clinical studies have considered stimulation via penetrating microelectrodes at the dorsal root ganglion (DRG) as a potential approach for somatosensory neuroprostheses. However, to develop clinically translatable neuroprosthetic devices, a less invasive approach, such as stimulation via epineural macroelectrodes, would be preferable. This work explores the feasibility of using such electrodes to deliver focal sensory feedback by exam

The Long-Term Stability of Intracortical Microstimulation and the Foreign Body Response Are Layer Dependent.

Intracortical microstimulation (ICMS) of the somatosensory cortex (S1) can restore sensory function in patients with paralysis. Studies assessing the stability of ICMS have reported heterogeneous responses across electrodes and over time, potentially hindering the implementation and translatability of these technologies. The foreign body response (FBR) and the encapsulating glial scar have been associated with a decay in chronic performance of implanted electrodes. Moreover, the morphology, intrinsic properties, and function of cells vary across cortical layers, each potentially affecting the

Validation of quantitative BOLD MRI measurements in kidney: application to unilateral ureteral obstruction.

BACKGROUND: Blood oxygenation level dependent (BOLD) magnetic resonance imaging (MRI) provides a measure of deoxyhemoglobin content and therefore an indirect measure of the partial oxygen pressure (pO(2)). The main purpose of this study was to examine the relationship between the apparent relaxation rate (R2*) in the pig kidney by BOLD imaging and renal tissue pO(2) levels measured directly by oxygen-sensitive microelectrodes. Second, BOLD imaging was applied to kidneys in pigs subjected to acute unilateral ureteral obstruction (UUO) to examine whether this condition is associated with changes

Gold/vertical-graphene-based dual-channel electrochemical immunosensor for point-of-care testing of tetanus toxin in blood.

The tetanus spasm toxin has been identified as the main pathogenic substance responsible for tetanus, and its trace detection is expected to provide reliable diagnostic biomarkers. In this study, we developed the first electrochemical tetanus-toxin immunosensor capable of rapid on-site detection and real-time analysis. This electrochemical sensor was developed by combining a mutually verifiable dual-channel (oxidation and reduction channels) strategy and a gold-nanoparticle-embedded vertical graphene (Au-VG) microelectrode with a high antibody-loading capacity, which led to a sensor with super

Microscale maps of bursting dynamics across human hippocampal slices from patients with epilepsy.

Neuronal firing patterning in the dentate gyrus of patients with epilepsy remains unknown at the microcircuit level. Advancements in high-density CMOS-based microelectrode arrays can be harnessed to study network activity with unprecedented spatial and temporal resolution. We use novel computational methods with high-density electrophysiology recordings to spatially map network activity of human hippocampal brain slices from six patients with mesial temporal lobe epilepsy. Two slices from the dentate gyrus exhibited synchronous bursting activity in the presence of low magnesium media with kain

µSpikeHunter: An advanced computational tool for the analysis of neuronal communication and action potential propagation in microfluidic platforms.

Understanding neuronal communication is fundamental in neuroscience, but there are few methodologies offering detailed analysis for well-controlled conditions. By interfacing microElectrode arrays with microFluidics (μEF devices), it is possible to compartmentalize neuronal cultures with a specified alignment of axons and microelectrodes. This setup allows the extracellular recording of spike propagation with a high signal-to-noise ratio over the course of several weeks. Addressing these μEF devices, we developed an advanced yet easy-to-use publically available computational tool, μSpikeHunter

What visual perception tells us about mind and brain.

Recent studies of visual perception have begun to reveal the connection between neuronal activity in the brain and conscious visual experience. Transcranial magnetic stimulation of the human occipital lobe disrupts the normal perception of objects in ways suggesting that important aspects of visual perception are based on activity in early visual cortical areas. Recordings made with microelectrodes in animals suggest that the perception of the lightness and depth of visual surfaces develops through computations performed across multiple brain areas. Activity in earlier areas is more tightly co

[Sodium channel blocking effect of scorpion venom on cultured mouse myocardiocytes].

Myocardiocytes of mice were cultured. Action potentials were recorded with microelectrodes inside the cells. Scorpion venom from Buthus martensii Karsch 3.75 or 7.5 micrograms.ml-1 decreased the duration of action potential and all of the depolarization concerned parameters of myocardiocytes. The Vmax, TP, APA behaved apparently in a dosage-dependent way. Restoration happened after washing out. Tetrodotoxin 2.5 micrograms.ml-1 acted in a similar way. Nimodipine 3.0 micrograms.ml-1 led to a decrease in action potential duration. BaCl2 0.1 mmol.L-1 elongated the action potential duration, while

Two cytochrome oxygen consumption model and mechanism for carotid body chemoreception.

We have measured sinus nerve discharge, tissue PO2 and oxygen consumption (VO2) in cat carotid bodies under different experimental conditions using our recessed oxygen microelectrode. Our results indicate that the change in chemoreceptor activity with oxygen disappearance following blood flow occlusion can be related to a two cytochrome model for oxygen consumption as previously proposed by Mills and Jöbsis (1972).

Alkaline biodegradable implants for osteoporotic bone defects--importance of microenvironment pH.

UNLABELLED: Change of microenvironment pH by biodegradable implants may ameliorate unbalanced osteoporotic bone remodeling. The present work demonstrated that a weak alkaline condition stimulated osteoblasts differentiation while suppressed osteoclast generation. In vivo, implants with an alkaline microenvironment pH (monitored by a pH microelectrode) exhibited a promising healing effect for the repair of osteoporotic bone defects. INTRODUCTION: Under osteoporotic conditions, the response of the bone microenvironment to an endosseous implant is significantly impaired, and this substantially i

[Effect of furosemide on the ampullar endolymphatic potential and endolymphatic Ca2+ activity].

The ampullar endolymphatic potential (AEP) and the endolymphatic Ca2+ activity were measured with double-barred Ca2+ selective microelectrodes in the experimental hydrops of guinea pigs. They showed 3.16 +/- 0.42 mV (n = 9) and (4.10 +/- 1.22) x 10(-4) M, respectively. The AEP decreased significantly at about 6 min after intravenous administration of 10 mg/kg furosemide. Its minimal value was 3.04 +/- 0.53 mV at about 40 min. But there was no significant difference between that and the value before injection. The Ca2+ activity increased significantly and reached (7.10 +/- 1.32) x 10(-4) M at a

Axonal conduction velocity and input conductance in petrosal ganglion primary sensory neurones of the cat.

Petrosal ganglion chemoreceptor neurones were impaled with glass microelectrodes in an 'in vitro' preparation maintained at 36.5 degrees C. For each cell, the input conductance and the axonal conduction velocity were calculated from the voltage change produced by a hyperpolarizing pulse, and from the latency of the spike evoked by carotid nerve stimulation. In 34 neurones, the average values for these variables were 3.7 X 10(-8) Siemens and 10 m/s. The two parameters were related by a power function with an exponent of 0.9, which is lower than the value of 2 expected on the basis of simple lin

Needle-integrated ultrathin bioimpedance microsensor array for early detection of extravasation.

Extravasation is a common complication during intravenous therapy in which infused fluids leak into the surrounding tissues. Timely intervention can prevent severe adverse consequences, but early detection remains an unmet clinical need because existing sensors are not sensitive to leakage occurring in small volumes (< 200 μL) or at deep venipuncture sites. Here, an ultrathin bioimpedance microsensor array that can be integrated on intravenous needles for early and sensitive detection of extravasation is reported. The array comprises eight microelectrodes fabricated on an ultrathin and flexibl

An alternating current electrokinetics biosensor for rapid on-site serological screening of Taenia solium cysticercosis infection.

Cysticercosis, caused by Taenia solium infection, is a leading cause of acquired epilepsy in many developing countries. Several types of immunoassays have been developed for the detection of Taenia solium infection in both infected humans and livestock animals. However, these methods require central laboratory facilities and are both time- and labor-consuming with longer than desired turnaround time. In this work, we demonstrated that AC electrokinetics (ACEK) capacitive sensing can be used to realize point-of-care immunosensor in general, with the on-site screening of Taenia solium infection

The use of digital image correlation for measurement of strain fields in a novel wireless intraspinal microstimulation interface.

BACKGROUND: Intraspinal microstimulation (ISMS) has emerged as a promising neuromodulation technique for restoring standing and overground walking in individuals with spinal cord injury. Current and emerging ISMS implant designs connect the electrodes to the stimulator through lead wires that cross the dura mater. To reduce possible complications associated with transdural leads such as tethering and leakage of cerebrospinal fluid, we aim to develop a wireless, fully intradural ISMS implant based on our prior work in the cortex with the Wireless Floating Microelectrode Array (WFMA). [Features of the spontaneous unit activity in the human thalamic parafascicular (CM-Pf) complex and its modification in functional cerebral changes].

New data on neural organization of the human thalamic parafascicular (CM-Pf) complex were revealed with microelectrode technique during 11 stereotaxic operations in alert diskinetic patients suffering from the tonic forms of spasmodic torticollis. The data were obtained as follows: the functional heterogeneity in cellular organization of the human CM-Pf thalamic nuclei and the existence of three (A, B and C) different types of neurons in these thalamic nuclei: with the irregular discharges (A-type, 18%); with short (10-20 ms) bursts characterised by unstable rhythmic 2-5 Hz pattern and by the
