Systematic regional variations in the loss of cortical cholinergic fibers in Alzheimer's disease.

The loss of cortical cholinergic fibers in Alzheimer's disease was investigated using choline acetyltransferase immunohistochemistry and acetylcholinesterase histochemistry. Within both the normal and Alzheimer's cerebral cortex, the two methods revealed an identical pattern of fiber staining. In the normal brain, cholinergic fiber density was highest in limbic and paralimbic cortical zones, intermediate in most sensory-motor and association zones, and lowest within the primary visual and visual association areas of the occipital lobe. In general, supragranular cortical layers contained a high

Visual hemifield mapping using transcranial magnetic stimulation coregistered with cortical surfaces derived from magnetic resonance images.

The perception of a visual stimulus can be inhibited by occipital transcranial magnetic stimulation. This visual suppression effect has been attributed to disruption in the cortical gray matter of primary visual cortex or in the fiber tracts leading to V1 from the thalamus. However, others have suggested that the visual suppression effect is caused by disruption in secondary visual cortex. Here the authors used a figure-eight coil, which produces a focal magnetic field, and a Quadropulse stimulator to produce visual suppression contralateral to the stimulated hemisphere in five normal voluntee

Adult visual cortical plasticity.

The visual cortex has the capacity for experience-dependent change, or cortical plasticity, that is retained throughout life. Plasticity is invoked for encoding information during perceptual learning, by internally representing the regularities of the visual environment, which is useful for facilitating intermediate-level vision--contour integration and surface segmentation. The same mechanisms have adaptive value for functional recovery after CNS damage, such as that associated with stroke or neurodegenerative disease. A common feature to plasticity in primary visual cortex (V1) is an associa

Adults vs. neonates: Differentiation of functional connectivity between the basolateral amygdala and occipitotemporal cortex.

The amygdala, a subcortical structure known for social and emotional processing, consists of multiple subnuclei with unique functions and connectivity patterns. Tracer studies in adult macaques have shown that the basolateral subnuclei differentially connect to parts of visual cortex, with stronger connections to anterior regions and weaker connections to posterior regions; infant macaques show robust connectivity even with posterior visual regions. Do these developmental differences also exist in the human amygdala, and are there specific functional regions that undergo the most pronounced de

Spatial attention triggered by eye gaze enhances and speeds up visual processing in upper and lower visual fields beyond early striate visual processing.

OBJECTIVE: The detection of a lateralized visual target is faster when preceded by a face gazing to the location of this stimulus. Here we aimed to clarify the time-course of the visual processing modulated by these reflexive shifts of attention. METHODS: ERPs were measured on 16 subjects performing a speeded location task on a circular checkerboard. The checkerboard target appeared either on the left or right of the upper or lower visual field, and was preceded by a central face orienting its gaze obliquely to one of the four possible corner locations for the target to appear. RESULTS: Cong

Chemoarchitecture of layer 4 isocortex in the American water shrew (Sorex palustris).

We examined the chemoarchitecture of layer 4 isocortex and the number of myelinated nerve fibers of selected cranial nerves in the American water shrew (Sorex palustris). This study took advantage of the opportunity to examine juvenile brain tissue, which often reveals the most distinctive cortical modules related to different sensory representations. Flattened cortical sections were processed for the metabolic enzyme cytochrome oxidase, revealing a number of modules and septa. Subdivisions related to sensory representations were tentatively identified by performing microelectrode recordings i

The neural basis of spatial vision losses in the dysfunctional visual system.

Human vision relies on correct information processing from the eye to various visual areas. Disturbances in the visual perception of simple features are believed to come from low-level network (e.g., V1) disruptions. In the present study, we modelled monocular losses in spatial vision through plausible multiple network modifications in early visual coding. We investigated perceptual deficits in anisometropic amblyopia and used the monocular tilt illusion as a probe of primary visual cortex orientation coding and inhibitory interactions. The psychophysical results showed that orientation misper

Sequence-dependent effects of iTBS and perceptual learning on binocular balance.

Sensory eye dominance (SED) reflects imbalanced treatment of the two eyes' inputs by the brain. Both V1-targeted intermittent theta-burst stimulation (iTBS) and dichoptic perceptual learning (PL) can reduce SED, implicating primary visual cortex (V1); however, how these interventions interact and whether they interact in an order-dependent manner remains unknown. Forty-four healthy control (HC) adults completed three daily sessions receiving either iTBS followed by PL (iTBS-PL) or PL followed by iTBS (PL-iTBS). We indexed observers' SED and motion discrimination thresholds before and after eac

Visuomotor system uses target features unavailable to conscious awareness.

After lesions to primary visual cortex, patients lack conscious awareness of visual stimuli. Interestingly, however, some retain the ability to make accurate judgments about the visual world (i.e., so-called blindsight). Similarly, damage to inferior occipitotemporal regions of cortex (e.g., lateral occipital cortex) can result in an inability to perceive object properties while retaining the ability to act on them (i.e., visual form agnosia). In the present work, we demonstrate that the ability to interact with objects in the absence of conscious awareness is not isolated to those with restri

Pyramidal cell specialization in the occipitotemporal cortex of the Chacma baboon (Papio ursinus).

Pyramidal cell structure varies systematically in occipitotemporal visual areas in monkeys. The dendritic trees of pyramidal cells, on average, become larger, more branched and more spinous with progression from the primary visual area (V1) to the second visual area (V2), the fourth (V4, or dorsolateral DL visual area) and inferotemporal (IT) cortex. Presently available data reveal that the extent of this increase in complexity parallels the expansion of occipitotemporal cortex. Here we extend the basis for comparison by studying pyramidal cell structure in occipitotemporal cortical areas in t

Pathway-dependent brain stimulation responses indicate motion processing integrity after stroke.

Homonymous hemianopia (HH), a common visual impairment resulting from occipital lobe lesions, affects a significant number of stroke survivors. Intensive perceptual training can foster recovery, possibly by enhancing surviving visual pathways. This study employed cortico-cortical paired associative stimulation (ccPAS) to induce associative plasticity within the residual and bidirectional primary visual cortex (V1)-middle temporal area (MT) pathways in stroke patients. We used ccPAS, which is thought to tap into Hebbian-like spike-timing dependent plasticity, over a motion processing pathway in

Sparsely distributed contours dominate extra-striate responses to complex scenes.

The human visual system exploits redundancy in natural scenes to derive useful information. Such redundancy is frequently associated with either contours or textures within images. In this study we use fMRI to evaluate how the total amount of contrast-energy contained in contours and textures within natural images affect responses in visual cortex. We used both the entire natural image and parts of it containing predominantly contour or texture information. We modified these natural images in order to match other image properties that are known to affect cortical responses as closely as possib

Altered GABAergic markers, increased binocularity and reduced plasticity in the visual cortex of Engrailed-2 knockout mice.

The maturation of the GABAergic system is a crucial determinant of cortical development during early postnatal life, when sensory circuits undergo a process of activity-dependent refinement. An altered excitatory/inhibitory balance has been proposed as a possible pathogenic mechanism of autism spectrum disorders (ASD). The homeobox-containing transcription factor Engrailed-2 (En2) has been associated to ASD, and En2 knockout (En2 (-/-)) mice show ASD-like features accompanied by a partial loss of cortical GABAergic interneurons. Here we studied GABAergic markers and cortical function in En2 (-

Functional magnetic resonance imaging of the visual cortex performed in children under sedation to assist in presurgical planning.

OBJECT: Advances in brain imaging have allowed for more sophisticated mapping of crucial neural structures. Functional MRI (fMRI) measures local changes in blood oxygenation associated with changes in neural activity and is useful in mapping cortical activation. Applications of this imaging modality have generally been restricted to cooperative patients; however, fMRI has proven successful in localizing the motor cortex for neurosurgical planning in uncooperative children under sedation. The authors demonstrate that the use of fMRI to localize the visual cortex in sedated children can be safel

Altered stability of dynamic brain functional architecture in primary open-angle glaucoma: a surface-based resting-state fMRI study.

Delineating the neuropathological characteristics of primary open-angle glaucoma (POAG) is critical for understanding its pathophysiology. While temporal stability represents a crucial aspect of the brain's functional architecture, the specific patterns underlying its contribution to POAG remain unclear. This study aims to analyze the brain functional abnormalities in POAG using functional stability, a dynamic functional connectivity (DFC) approach based on resting-state functional magnetic resonance imaging (rs-fMRI). Seventy patients with POAG and forty-five healthy controls underwent rs-fMR

Gating by induced Α-Γ asynchrony in selective attention.

Visual selective attention operates through top-down mechanisms of signal enhancement and suppression, mediated by α-band oscillations. The effects of such top-down signals on local processing in primary visual cortex (V1) remain poorly understood. In this work, we characterize the interplay between large-scale interactions and local activity changes in V1 that orchestrates selective attention, using Granger-causality and phase-amplitude coupling (PAC) analysis of EEG source signals. The task required participants to either attend to or ignore oriented gratings. Results from time-varying, dire

Visualization of hypermetabolism in the ventral visual pathway by FDG-PET in the Charles Bonnet syndrome.

A 74-year-old man presented with complex visual hallucinations with a left inferior quadrantanopia. The characteristics of the visual hallucinations met the criteria for the Charles Bonnet syndrome. Brain magnetic resonance imaging (MRI) revealed a right occipital falx meningioma. Fusion images of gadolinium-enhanced MRI and 18F-fluorodeoxyglucose (FDG)-positron emission tomography (PET) of the brain demonstrated hypometabolism in the right primary and secondary visual cortices, and an ipsilateral hypermetabolism in a focal area of the medial aspect of the secondary visual cortex as well as th

In vivo visuotopic brain mapping with manganese-enhanced MRI and resting-state functional connectivity MRI.

The rodents are an increasingly important model for understanding the mechanisms of development, plasticity, functional specialization and disease in the visual system. However, limited tools have been available for assessing the structural and functional connectivity of the visual brain network globally, in vivo and longitudinally. There are also ongoing debates on whether functional brain connectivity directly reflects structural brain connectivity. In this study, we explored the feasibility of manganese-enhanced MRI (MEMRI) via 3 different routes of Mn(2+) administration for visuotopic brai

Different Excitation-Inhibition Correlations Between Spontaneous and Tone-evoked Activity in Primary Auditory Cortex Neurons.

Tone-evoked synaptic excitation and inhibition are highly correlated in many neurons with V-shaped tuning curves in the primary auditory cortex of pentobarbital-anesthetized rats. In contrast, there is less correlation between spontaneous excitation and inhibition in visual cortex neurons under the same anesthetic conditions. However, it was not known whether the primary auditory cortex resembles visual cortex in having spontaneous excitation and inhibition that is less correlated than tone-evoked excitation and inhibition. Here we report whole-cell voltage-clamp measurements of spontaneous ex
