The sigma-receptor antagonist BD-1063 decreases ethanol intake and reinforcement in animal models of excessive drinking.

Sigma-Receptors (SigRs) have been implicated in behavioral and appetitive effects of psychostimulants and may also modulate the motivating properties of ethanol. This study tested the hypothesis that SigRs modulate ethanol reinforcement and contribute to excessive ethanol intake. The effects of subcutaneous treatment with the potent, selective Sig-1R antagonist BD-1063 on operant ethanol self-administration were studied in two models of excessive drinking-Sardinian alcohol-preferring (sP) rats and acutely withdrawn ethanol-dependent Wistar rats-and compared to ethanol self-administration in no

An Operant Conditioning Task to Assess the Choice between Wheel Running and Palatable Food in Mice.

Wheel running, especially in the homecage, has been widely used to study the neurobiology of exercise because animal tends to use it voluntarily. However, as for each reward, its consumption (in the present case, running performance) does not specifically provide information on its incentive value, i.e., the extent to which animals are motivated to run independently from their consumption of that reward. This is a major drawback, especially when focusing on the neurobiology governing the pathological imbalances between exercise and e.g., feeding (obesity, anorexia nervosa). Yet, few studies ha

Dopamine, motivation, and the evolutionary significance of gambling-like behaviour.

If given a choice between certain and uncertain rewards, animals tend to prefer the uncertain option, even when the net gain is suboptimal. Animals are also more responsive to reward-related cues in uncertain situations. This well-documented phenomenon in many animal species is in opposition to the basic principles of reinforcement as well as the optimal foraging theory, which suggest that animals will prefer the option associated with the highest reward rate. How does the brain code the attractiveness of unreliable/poor reward sources? And how can we interpret this evidence from an adaptive p

Schedule-induced polydipsia in rats with gastric fistulas.

We have investigated the development and maintenance of schedule-induced polydipsia (SIP) when ingested water (and food) was allowed to drain from the stomach. Fourteen male Long-Evans rats were prepared with permanent gastric cannulas and, after recovery, their body weight was reduced to 80%. Water intake was measured, with cannulas open or closed, during 42 daily 1-h sessions in which 45-mg food pellets were delivered one per minute. Allowing ingested material to drain from the stomach impaired the development of SIP and reduced the polydipsia in rats in which SIP had already been establishe

Effects of drugs on the temporal distribution of schedule-induced polydipsia in rats.

Drinking was induced in food-deprived rats by a fixed-time 1-min schedule of food presentation. With three rats, d-amphetamine (0.25, 0.5, 1.0, and 2.0 mg/kg) led to a dose-related increase in licking early in the interfood intervals, the peak of the temporal distribution of licking being shifted to earlier values. These effects were seen even when d-amphetamine had no effect on overall rates of licking and drinking. With another three rats, however, diazepam (0.5, 1.0, 2.0, and 4.0 mg/kg) did not shift the peak of the temporal distribution of licks in interfood intervals, even at doses that p

Two types of bias in psychophysical detection and recognition procedures: nonparametric indices and effects of drugs.

In many conditional discrimination procedures, subjects are required to emit one of two responses in the presence of one stimulus and the other response in its absence. The two responses that occur may be formally differentiated, for example by their positions in the experimental space, and functionally differentiated, by the discriminative stimuli that control their occurrence. For example, the stimuli displayed on two response keys, such as color, may be discriminative stimuli for the particular key on which responses are reinforced. The behavior that occurs under these procedures can be ana

Elimination of rumination by controlled eating and differential reinforcement.

The purpose of the present study was to examine the effectiveness of a controlled eating technique and a procedure which combined controlled eating with differential reinforcement of other behavior (DRO) for the reduction of rumination in an obese, 23-year-old, severely retarded and autistic male. The controlled eating technique resulted in a 75 per cent reduction in the rate of rumination while controlled eating with DRO resulted in an overall 95 per cent reduction. A six month follow-up showed that the participant's ruminative behavior had declined to zero.

Drug-behavior interaction history: modification of the effects of morphine on punished behavior.

Squirrel monkeys were trained to respond under a multiple fixed-interval, fixed-interval schedule in which the first response after 5 min terminated a visual stimulus in the presence of which electric shocks could occur. During one component of the schedule, correlated with one color of stimulus lights, every 30th response also produced electric shock; responding was suppressed during this component to approximately 10 to 12% of that occurring in the alternate component in which responding was not punished. In contrast to previous research, morphine (0.03 to 1.0 mg/kg) increased punished respo

Stereoselective behavioral effects of the isomers of pentobarbital and secobarbital in the pigeon.

Little information is available on the contribution of the isomers of pentobarbital and secobarbital to the behavioral effects of the racemic mixtures. To determine the contribution of each isomer on food-maintained behavior, pigeons were trained to respond under either a multiple fixed-ratio 30, fixed-interval 600 (mult FR30 FI600) schedule, or a multiple fixed-ratio 30, variable-interval 60-sec (mult FR30 VI60) schedule under which every response under both the FR and VI components also produced a brief electrical shock (punishment). Dose-response curves were determined separately for each i

Rat strain differences in the acquisition of conditioned avoidance responses and in the effects of diazepam.

Adult male albino rats of three strains--Wistar, Sprague-Dawley and Holtzman--were trained to press a lever to avoid electric shocks under Sidman-type (R-S interval-20 sec; S-S interval=5 sec) and discriminated avoidance (ITI-15 sec; warning duration=5 sec) schedules, and the acquisition processes of avoidance responses, and the properties of behavioral baselines were investigated. Under both schedules, Wistar strain rats, though showing poorer results than the other two in the beginning, rapidly progressed with the repetitive training, and finally displayed excellent and stable performances.

Effects of central neurokinin-1 receptor antagonism on cocaine- and opiate-induced locomotor activity and self-administration behaviour in rats.

The neuropeptide substance P (SP) and its preferred receptor, the neurokinin-1 (NK-1) receptor, have been implicated in some of the reward-related behavioural effects of abused drugs, including psychostimulants and opiates. The first objective of the present series of experiments was to assess the role of the NK-1 receptor in two reward-related behavioural effects of cocaine: locomotor activity and self-administration. In tests for locomotor activity, rats were given intracerebroventricular (ICV) infusions of the selective NK-1 receptor antagonist, GR82334 (0, 10, 50 pmol), prior to systemic i

Perseverative structuring of responses by schizophrenic and affective disorder patients.

Some theoretical implications from a CNS stimulation model of psychosis were tested by analysing the response patterns of schizophrenic (N = 34), affective disorder patients (N = 18), schizoid personality disorder patients (N = 9), neurotic patients (N = 9) and normal controls (N = 34) on a visual two-choice task with reinforcement. The subjects were required to press a left and right button in order to cause a cross (+) to appear on the monitor screen above one of the buttons. Certain subsequences of right and left presses were required in order for the cross to appear, and a coin-reinforceme

Age-dependent morphine intake and cue-induced reinstatement, but not escalation in intake, by adolescent and adult male rats.

Despite increasing rates of opioid abuse by human adolescents, few laboratory experiments address adolescent vulnerability to opiates. We examined intravenous morphine self-administration after adolescent- vs. adult-onset, followed by extinction and cue-induced reinstatement. Adolescent male Sprague-Dawley rats [postnatal day (P) 35 at start] and adults (P91) acquired lever pressing maintained by 0.375 mg/kg/infusion morphine on a fixed ratio one schedule of reinforcement. Subjects were subsequently divided into short or long daily access conditions (ShAcc, 1-h vs. LgAcc, 8-h; 18 sessions). Af

Development and Validation of an Assessment-Driven Behavioral Intervention for Primary Complex Motor Stereotypies in Young Children.

Complex motor stereotypies are rhythmic, repetitive, fixed, and non-goal directed movements (e.g., bilateral flapping/waving movements of the hands/arms). Movements typically begin in early childhood and can occur in otherwise normally developing ("primary") or autistic ("secondary") children. Stereotypies persist, occur multiple times a day, have prolonged durations, can be socially stigmatizing, and may lead to bullying and isolation. Prior behavioral treatment studies have focused on older children (ages 6-12) and report modest reductions in stereotypy (i.e., between 14% and 33%). The curre

Separation of drug effects on timing and behavioral inhibition by increased stimulus control.

Impulsive behavior may represent, in part, a failure of behavioral inhibition (the ability to delay or inhibit a response). In this study, use of a multiple signaled-unsignaled differential-reinforcement-of-low-rates (DRL) 15-s schedule allowed examination of drug effects in conditions in which level of stimulus control differed. Results showed that whereas diazepam increased premature responding during signaled and unsignaled DRL components, amphetamine and delta9-tetrahydrocannabinol increased premature responding primarily during unsignaled components when timing was necessary for efficient
