Remote ischemic postconditioning promotes the survival of retinal ganglion cells after optic nerve injury.

Ischemic conditioning, the application of a mild ischemic stimulus to an ischemia-sensitive structure like the heart or brain either before (preconditioning) or after (postconditioning) its exposure to a lethal ischemic insult, is known to switch on endogenous protective mechanisms. However, most studies of its neuroprotective effect in the central nervous system (CNS) have focused on ischemic damage or related conditions like hypoxia, while its potential in treating other neural diseases remains uncertain. In particular, the recent discovery of remote ischemic postconditioning whereby mild is

Axotomy results in delayed death and apoptosis of retinal ganglion cells in adult rats.

Using quantitative anatomical techniques, we show that after intraorbital optic nerve transection in adult rats, virtually all retinal ganglion cells (RGCs) survive for 5 d and then die abruptly in large numbers, reducing the RGC population to approximately 50% of normal by day 7 and to less than 10% on day 14. During this period of rapid cell loss, some RGCs show cytochemical alterations indicative of apoptosis ("programmed cell death"), a change not previously categorized after axotomy in adult mammals. With intracranial lesions 8-9 mm from the eye, the onset of cell death is delayed until d

Blockade of electrical activity promotes the death of mammalian retinal ganglion cells in culture.

During the first 2 postnatal weeks, up to 50% of the ganglion cells in the mammalian retina normally die. Natural cell death may result from several factors, and electrical activity has been proposed as one critical element. Recent experiments in vivo using intraocular injection of tetrodotoxin (TTX) have suggested that competition for survival between ganglion cells from the two eyes is mediated by their degree of neuronal activity. In addition, the level of activity of afferents to the ganglion cells has been postulated to be an important variable in determining their survival. To investigat

Influence of GABA on the spectral and spatial coding of goldfish retinal ganglion cells.

The responses of center red ON and center red OFF ganglion cells of the isolated goldfish retina to a flickering stimulus light diminish by GABA. Without stimulation GABA blocks the maintained activity of the OFF cells. With a green flickering spot in the receptive field center, the green ON-response of the OFF cells becomes an OFF-response during GABA administration. It is concluded that, besides the common antagonistic green input, the receptive field center of these cells also has a synergistic green input, which is mediated by a pathway other than the center opponent red and green inputs.

Sustained and transient discharges of retinal ganglion cells during spontaneous eye movements of cat.

Discharges of 223 retinal ganglion cells during spontaneous eye movements (saccades) across a stationary grating pattern were studied in chronically prepared cats. Of these 83 showed sustained responses to local differences in luminance of the grating stripes (S-units); 84 showed transient responses to saccades and did not register local differences in luminance (T-units); and 56 showed mixed responses, i.e., transient responses to saccades and sustained firings in response to local luminance (M-units). When tested with diffuse light, 93.9% of the S-units showed either ON-sustained or OFF-sust

Dopamine Alleviated Diabetic Retinal Neurodegeneration through Protecting Retinal Ganglion Cells from Ferroptosis via the Nrf2/HO‑1 Signaling Pathway.

Diabetic retinopathy (DR) is a major cause of vision impairment, with diabetic retinal neurodegeneration (DRN) identified as an early pathological feature. Although dopamine (DA) has demonstrated protective effects in various neurodegenerative diseases, its role in DRN is inadequately understood. This study provides evidence that DA can alleviate DRN impairment and elucidates the underlying mechanisms. Diabetes was induced in C57BL/6 mice through a single large dose injection of streptozotocin, and the dopamine precursor l-DOPA was administered via intraperitoneal injection daily for 2 weeks t

TBK1 Knockdown Alleviates Axonal Transport Deficits in Retinal Ganglion Cells Via mTORC1 Activation in a Retinal Damage Mouse Model.

PURPOSE: Glaucoma is the leading cause of irreversible blindness worldwide and is characterized by progressive retinal ganglion cell (RGC) death and optic nerve degeneration. Axonal transport deficits are the earliest crucial pathophysiological changes in glaucoma. Genetic variation in the TANK-binding kinase 1 gene (TBK1) plays a role in the pathogenesis of glaucoma. This study was designed to investigate intrinsic factors underlying RGCs' damage and to explore the molecular mechanism of TBK1 involvement in glaucomatous pathogenesis. METHODS: We established a mouse model of acute ocular hype

Retinal Ganglion Cells With a Glaucoma OPTN(E50K) Mutation Exhibit Neurodegenerative Phenotypes when Derived from Three-Dimensional Retinal Organoids.

Retinal ganglion cells (RGCs) serve as the connection between the eye and the brain, with this connection disrupted in glaucoma. Numerous cellular mechanisms have been associated with glaucomatous neurodegeneration, and useful cellular models of glaucoma allow for the precise analysis of degenerative phenotypes. Human pluripotent stem cells (hPSCs) serve as powerful tools for studying human disease, particularly cellular mechanisms underlying neurodegeneration. Thus, efforts focused upon hPSCs with an E50K mutation in the Optineurin (OPTN) gene, a leading cause of inherited forms of glaucoma.

Modeling spatial contrast sensitivity in responses of primate retinal ganglion cells to natural movies.

Retinal ganglion cells, the output neurons of the vertebrate retina, often display nonlinear summation of visual signals over their receptive fields. This creates sensitivity to spatial contrast, letting the cells respond to spatially structured visual stimuli even when no net change in overall illumination of the receptive field occurs. Yet, computational models of ganglion cell responses are often based on linear receptive fields, and typical nonlinear extensions, which separate receptive fields into nonlinearly combined subunits, are often cumbersome to fit to experimental data. Repetitive hypoxic preconditioning protects retinal ganglion cells against damage caused by exposure to blast.

Visual system damage and dysfunction caused by exposure to a blast wave has been described in both clinical studies and in pre-clinical models. Within the retina, retinal ganglion cells (RGC) exhibit sensitivity to mild blast-mediated traumatic brain injury (bTBI), which can result in progressive neurodegeneration. The purpose of this study was to determine if repetitive hypoxic preconditioning (HPC) can prevent bTBI-mediated RGC damage and death. This study utilized clinically relevant outcomes of RGC structure and function, supported by histological analysis of the surviving RGCs. Mice were

The rod bipolar cell pathway contributes to surround responses in OFF retinal ganglion cells.

Sensory neurons can be influenced by stimuli beyond their receptive field center, yet the mechanisms underlying this surround modulation remain poorly understood. In the retina, many OFF ganglion cells exhibit responses to ON stimulation outside their receptive field center. However, disentangling the pathways and cell types contributing to these responses has been challenging with traditional experimental approaches. Here, we combined optogenetics, two-photon holographic stimulation, and multielectrode array recordings to identify the intermediate retinal cell types involved in this circuit.

Retinal degeneration increases inter-trial variabilities of light-evoked spiking activities in ganglion cells.

Retinal ganglion cells (RGCs) transmit visual information to the brain in the form of spike trains, which form visual perception. The reliabilities of spike timing and count are thought to play a crucial role in generating stable percepts. However, the effect of retinal degeneration on spike reproducibility remains underexplored. In this study, we examined longitudinal changes of both spike timing and count across different RGC types in response to repeated presentations of an identical light stimulus in retinal degeneration 10 (rd10) mice (B6.CXBl-Pde6brd10/J), a well-established model of ret

Genetic and pharmacological correction of impaired mitophagy in retinal ganglion cells rescues glaucomatous neurodegeneration.

Progressive loss of retinal ganglion cells (RGCs) and degeneration of optic nerve axons are the pathological hallmarks of glaucoma. Ocular hypertension (OHT) and mitochondrial dysfunction are linked to neurodegeneration and vision loss in glaucoma. However, the exact mechanism of mitochondrial dysfunction leading to glaucomatous neurodegeneration is poorly understood. Using multiple mouse models of OHT and human eyes from normal and glaucoma donors, we show that OHT induces impaired mitophagy in RGCs, resulting in the accumulation of dysfunctional mitochondria and contributing to glaucomatous

Region-specific Brain Targets Drive Circuit Formation and Maturation of Human Retinal Ganglion Cells.

UNLABELLED: Human vision relies on retinal ganglion cells (RGCs), and their connectivity with distinct brain regions enables higher order visual processing. RGCs vary considerably between species, and small model organisms display distinct RGC innervation patterns from that in humans. There is thus a need for robust models of human RGC circuit formation that preserves innervation specificity. Here, we developed an in vitro microfluidics eye-to-brain connectivity model using human pluripotent stem cell (hPSC)-derived RGCs to assess brain region-specific connectivity features. We find that cultu
