A Longitudinal Operant Assessment of Cognitive and Behavioural Changes in the HdhQ111 Mouse Model of Huntington's Disease.

Huntington's disease (HD) is characterised by motor symptoms which are often preceded by cognitive and behavioural changes, that can significantly contribute to disease burden for people living with HD. Numerous knock-in mouse models of HD are currently available for scientific research. However, before their use, they must be behaviourally characterised to determine their suitability in recapitulating the symptoms of the human condition. Thus, we sought to longitudinally characterise the nature, severity and time course of cognitive and behavioural changes observed in HdhQ111 heterozygous kno

Baseline sensorimotor GABA levels shape neuroplastic processes induced by motor learning in older adults.

Previous research in young adults has demonstrated that both motor learning and transcranial direct current stimulation (tDCS) trigger decreases in the levels of gamma-aminobutyric acid (GABA) in the sensorimotor cortex, and these decreases are linked to greater learning. Less is known about the role of GABA in motor learning in healthy older adults, a knowledge gap that is surprising given the established aging-related reductions in sensorimotor GABA. Here, we examined the effects of motor learning and subsequent tDCS on sensorimotor GABA levels and resting-state functional connectivity in th

Development of a standardized scoring method for the Graphic Sequence Test suitable for use in psychiatric populations.

Although the graphic version of the Alternating Sequences Test which was introduced by Luria exists for years little has been done to standardize it. The aim of the current study was to develop a novel and detailed standardized method of administration and scoring. The study sample included 93 normal control subjects (53 women and 40 men) aged 35.87+/-12.62 and 127 patients suffering from schizophrenia (54 women and 73 men) aged 34.07+/-9.83. The psychometric assessment included the Positive and Negative Symptoms Scale the Young Mania Rating Scale, and the Montgomery-Asberg Depression Rating S

Examining ventral and dorsal prefrontal function in bipolar disorder: a functional magnetic resonance imaging study.

Several lines of research suggest both dorsal and ventral prefrontal cortical dysfunction in bipolar disorder (BD). We used functional magnetic resonance imaging to compare patterns of brain activation in remitted BD patients and controls whilst performing tasks selected for their relative specificity in engaging either the dorsal (n-back sequential-letter working memory task) or ventral (gambling task) PFC. Seven BD patients were selected from participants of the Maudsley Bipolar Disorder Project on the basis of clinical remission, absence of cognitive deficits, and monotherapy with mood stab

Fronto-striatal deficit in Parkinson's disease during semantic event sequencing.

Studies of Parkinson's disease (PD) suggest that cognitive deficits accompany the classically recognized motor symptoms, and that these cognitive deficits may result from damage to frontal-basal ganglia circuits. PD patients are impaired on ordering events and action components into coherent sequences. In this study, we examined early-stage, nondemented, medicated PD subjects and matched control subjects during a semantic event sequencing task using functional MRI (fMRI). The task required subjects to examine four pictures of meaningful events, determine the correct temporal relationship betwe

Administration of eliprodil during ethanol withdrawal in the neonatal rat attenuates ethanol-induced learning deficits.

RATIONALE: Prenatal exposure to alcohol can disrupt brain development, leading to a variety of behavioral alterations, including learning deficits. We have postulated that some central nervous system damage may be due to N-methyl-D-aspartate (NMDA) receptor-mediated excitotoxicity that occurs during ethanol withdrawal. Consistent with this hypothesis, we previously demonstrated that administration of MK-801, an NMDA receptor antagonist, during ethanol withdrawal attenuates ethanol-related learning deficits using an animal model of fetal alcohol effects. However, MK-801 binds to the phencyclidi

Perinatal choline supplementation attenuates behavioral alterations associated with neonatal alcohol exposure in rats.

Children exposed to alcohol prenatally suffer from a variety of behavioral alterations, including hyperactivity and learning deficits. Given that women continue to drink alcohol during pregnancy, it is critical that effective interventions and treatments be identified. Previously, we reported that early postnatal choline supplementation can reduce the severity of learning deficits in rats exposed to alcohol prenatally. The present study examined whether choline supplementation can reduce the severity of behavioral alterations associated with alcohol exposure during the third trimester equivale

Machine Learning for Myocardial Infarction Compared With Guideline-Recommended Diagnostic Pathways.

BACKGROUND: Collaboration for the Diagnosis and Evaluation of Acute Coronary Syndrome (CoDE-ACS) is a validated clinical decision support tool that uses machine learning with or without serial cardiac troponin measurements at a flexible time point to calculate the probability of myocardial infarction (MI). How CoDE-ACS performs at different time points for serial measurement and compares with guideline-recommended diagnostic pathways that rely on fixed thresholds and time points is uncertain. METHODS: Patients with possible MI without ST-segment-elevation were enrolled at 12 sites in 5 countr

Computerized analysis of error patterns in digit span recall.

We analyzed error patterns during digit span (DS) testing in four experiments. In Experiment 1, error patterns analyzed from a community sample of 427 subjects revealed strong primacy and recency effects. Subjects with shorter DSs showed an increased incidence of transposition errors in comparison with other error types and a greater incidence of multiple errors on incorrect trials. Experiment 2 investigated 46 young subjects in three test sessions. The results replicated those of Experiment 1 and demonstrated that error patterns of individual subjects were consistent across repeated test admi

Proactive interference in a semantic short-term memory deficit: role of semantic and phonological relatedness.

Previous research has indicated that patients with semantic short-term memory (STM) deficits demonstrate unusual intrusions of previously presented material during serial recall tasks (Martin and Lesch, 1996). These intrusions suggest excessive proactive interference (PI) from previous lists. Here, we explore one such patient's susceptibility to PI. Experiment 1 demonstrated patient M.L.'s extreme susceptibility to PI using a probe recognition task that manipulates the recency of negative probes (the recent negatives task). When stimuli consisted of letters, M.L. showed greatly exaggerated eff

Investigating the acute effect of low and moderate intensity aerobic exercise on whole-body task learning and cognition in young adults.

Recent studies have shown that a single bout of exercise has acute improvements on various forms of memory, including procedural motor learning, through mechanisms such as the plasticity-promoting effect. This study aimed to examine (1) the acute effects of timing and intensity of aerobic exercise on the acquisition and retention of motor learning in healthy adults, (2) the effect of sleep quality of the night before and after acquisition on motor learning, and (3) the acute effects of low and moderate-intensity aerobic exercise on cognitive functions. Seventy-five healthy adults were divided

Autoimmune gastritis detection from preprocessed endoscopy images using deep transfer learning and moth flame optimization.

Gastric Tract Disease (GTD) constitutes a medical emergency, emphasizing the critical importance of early diagnosis and intervention to lessen its severity. Clinical practices often utilize endoscopy-supported examinations for GTD screening. The images obtained during this procedure are examined to identify the presence of the disease and investigate its severity. Autoimmune Gastritis (AIG) is a chronic inflammatory GTD and timely detection and treatment is crucial to reduce its harshness. This research aims to develop a deep-learning (DL) tool to detect the AIG from clinical-grade endoscopic

Early Use of the Palliative Approach to Improve Patient Outcomes in HIV Disease: Insights and Findings From the Care and Support Access (CASA) Study 2013-2019.

Young men of color who have sex with men (yMSM) living with human immunodeficiency virus (HIV) in syndemic environments have been difficult-to-retain in care resulting in their being at-risk for poor health outcomes despite availability of effective once-daily antiretroviral treatment (ART). Multiple methods have been implemented to improve outcomes for this cohort; none with sustainable results. Outpatient HIV staff themselves may be a contributing factor. We introduced multidisciplinary staff to the concept of using a palliative approach early (ePA) in outpatient HIV care management to enabl

Differential effects of total sleep deprivation on contextual and spatial memory: modulatory effects of modafinil.

The aim of the present work was to investigate in mice the effects of a total 10-hr sleep deprivation on contextual (episodic-like) and spatial (reference) memory tasks. For that purpose, mice learned two consecutive discriminations (D1 and D2) in a 4-hole board involving either identical (Serial Spatial Discrimination, SSD) or distinct (Contextual Serial Discrimination, CSD) internal contextual cues. In a second step, we intended to assess the corrective effect of modafinil on memory impairments generated by sleep deprivation. Sleep deprivation was triggered through an alternative platform ap

Lesions of the medial striatum in monkeys produce perseverative impairments during reversal learning similar to those produced by lesions of the orbitofrontal cortex.

The ability to switch responding between two visual stimuli based on their changing relationship with reward is dependent on the orbitofrontal cortex (OFC). OFC lesions in humans, monkeys, and rats disrupt performance on a common test of this ability, the visual serial discrimination reversal task. This finding is of particular significance to our understanding of psychiatric disorders such as obsessive-compulsive disorder (OCD) and schizophrenia, in which behavioral inflexibility is a prominent symptom. Although OFC dysfunction can occur in these disorders, there is considerable evidence for

Training and transfer of clustering and cumulative rehearsal strategies in retarded individuals.

Forty-four retarded adolescents varying in MA, IQ, and etiology were taught one of three active rehearsal strategies or assigned to a control condition. Immediate, short-term (1 day), and long-term (2 weeks) transfer of the strategy was assessed by presenting new 9-item serial lists on each day. Exposure durations, which indicated the rate of subject-pacing, showed that the two highest MA-IQ (mean IQ = 55) groups, one diagnosed as organically impaired and the other as cultural-familially retarded, successfully transferred their strategies over three sessions, while no transfer occurred in the

Longitudinal evaluation of neurocognitive outcomes in a cohort with persistent post-COVID olfactory dysfunction.

Persistent COVID-19-associated olfactory dysfunction (C19OD), along with other neurologic and cognitive deficits are common features of long COVID. This study aims to evaluate longitudinal trends in neurocognitive performance within a cohort with C19OD. In individuals with perceived C19OD we performed serial psychophysical olfactory and neurocognitive assessments at baseline and follow-up one year later. At baseline evaluation, individuals with C19OD were found to have diminished cognitive functioning compared to normosmic counterparts across several domains, including attention, executive fun

Early detection and progression of sepsis-related myocardial calcification with subsequent left ventricular systolic dysfunction: a case report.

Sepsis-related myocardial calcification (SRMC) is a life-threatening complication. However, it is a rare entity and its clinical course is not well-understood. A 54-year-old man after bone graft surgery presented with septic shock due to surgical site infection. The initial computed tomography (CT) showed no deposit of calcium in the left ventricle (LV), and echocardiography demonstrated preserved left ventricular ejection fraction (LVEF) of 61%. On the 10th day of admission, CT detected new-onset LV myocardial calcification with preserved LVEF of 60% in echocardiography. On the 63rd day, foll

Concordance of flicker comparison versus side-by-side comparison in glaucoma.

BACKGROUND: Flicker comparison superimposes baseline photos of the optic nerve head to subsequent photos, alternating them at a high frequency rate to create the perception of motion showing glaucomatous structural changes over time. The primary purpose of this study was to determine the degree of concordance among examiners in judging glaucomatous progression between serial optic nerve head photos using digital image flicker comparison versus the traditional side-by-side photograph comparison method. The secondary purpose was to determine if flicker comparison was quicker than side-by-side co
