New insights regarding Acinetobacter genomic island-related elements.

The objective of this study was to mobilise the Acinetobacter genomic island 1-A (AGI1-A) from Enterobacter hormaechei EclCSP2185 (E. cloacae complex) and to search for the distribution and structure of AGI1-related elements in the NCBI database. AGI1-A was transferred to Escherichia coli. Analysis of the attachment (att) sites could locate the possible recombination crossover in the att sequences at position 10-11 (GG) in the last 18 bp of trmE. In silico detection of AGI backbones in the WGS database identified AGI variants in Salmonella enterica (83 strains), Vibrio cholerae (33), E. hormae

Simultaneous Transformation of Commingled Trichloroethylene, Tetrachloroethylene, and 1,4-Dioxane by a Microbially Driven Fenton Reaction in Batch Liquid Cultures.

UNLABELLED: Improper disposal of 1,4-dioxane and the chlorinated organic solvents trichloroethylene (TCE) and tetrachloroethylene (also known as perchloroethylene [PCE]) has resulted in widespread contamination of soil and groundwater. In the present study, a previously designed microbially driven Fenton reaction system was reconfigured to generate hydroxyl (HO˙) radicals for simultaneous transformation of source zone levels of single, binary, and ternary mixtures of TCE, PCE, and 1,4-dioxane. The reconfigured Fenton reaction system was driven by fed batch cultures of the Fe(III)-reducing facu

A 3D-printed algal-bacterial microreactor for photobiological hydrogen production coupled with actual wastewater treatment.

Microalgae-driven photosynthetic hydrogen production has emerged as a promising approach to tackle the global energy crisis and reduce carbon emissions. Simultaneously, microalgae can remove phosphorus and nitrogen from wastewater while sequestering CO2 during hydrogen production. However, challenges such as the intrinsic oxygen sensitivity of hydrogenases and the relatively low hydrogen productivity remain major hurdles that constrain the process viability of microalgae for combined wastewater treatment and hydrogen production. A 3D-bioprinted algal-bacterial microreactor (ABMR) with a core-s

Living bacterial reactor potently activates tumor immunogenic ferroptosis via cysteine depletion and photothermal therapy.

Bacterial combination therapy offers immense promise for treating aggressive "cold" solid tumors, such as triple-negative breast cancer (TNBC). However, the clinical translatability of traditional genetic engineering is often hampered by operational complexity and genetic instability. Here, we developed A-SPB-a non-genetically engineered, multi-functional living bioreactor based on Shewanella oneidensis MR-1 (SO), surface-modified with Prussian blue (PB) nanoparticles and attenuated via deoxycholic acid (DA) treatment. This platform serves as an in situ metabolic factory that "hijacks" the sul

Exploring the anti-quorum sensing potential of sea anemone bacterial symbionts from Samalona Island (Spermonde Archipelago) in South Sulawesi, Indonesia.

Vannamei shrimp is a promising aquaculture commodity, but presence of Vibrio parahaemolyticus bacteria in cultivation poses a challenge to increasing productivity. Quorum sensing allows bacteria to regulate pathogenicity based on population density. The aim of this study were to screen antiquorum sensing agents that can inhibit bacterial communication, thereby reducing pathogen virulence. The method used in the screening process goes through several stages, such as isolation of anemone symbiont bacteria, screening for inhibition of violacein and antibiofilm production, as well as molecular ide

Study of the key biotic and abiotic parameters influencing ammonium removal from wastewaters by Fe3+-mediated anaerobic ammonium oxidation (Feammox).

The release of ammonia (as NH4+) into water bodies causes serious environmental problems. Therefore, the removal of ammonia from wastewater effluents has become a worldwide concern. New autotrophic biological alternatives for ammonia removal could reduce the limitations of conventional organic carbon-dependent nitrification-denitrification methods. Here, the potential of anaerobic ammonium oxidation coupled to Fe3+ reduction (a process known as Feammox) is studied in wastewater treatment plants of the yeast and beer production industry, not related to ammonium or iron treatment. This process i

Biogenic ferrihydrite-humin coprecipitate as an electron donor for the enhancement of microbial denitrification by Pseudomonas stutzeri.

Nitrate pollution of groundwater has become an increasingly serious environmental problem that poses a great threat to aquatic ecosystems and to human health. Previous studies have shown that solid-phase humin (HM) can act as an additional electron donor to support microbial denitrification in the bioremediation of nitrate-contaminated groundwater where electron donor is deficient. However, the electron-donating capacities of HMs vary widely. In this study, we introduced ferrihydrite and prepared ferrihydrite-humin (Fh-HM) coprecipitates via biotic means to strengthen their electron-donating c

Low Prevalence of Carbapenem-Resistant Bacteria in River Water: Resistance Is Mostly Related to Intrinsic Mechanisms.

Carbapenems are last-resort antibiotics to handle serious infections caused by multiresistant bacteria. The incidence of resistance to these antibiotics has been increasing and new resistance mechanisms have emerged. The dissemination of carbapenem resistance in the environment has been overlooked. The main goal of this research was to assess the prevalence and diversity of carbapenem-resistant bacteria in riverine ecosystems. The presence of frequently reported carbapenemase-encoding genes was inspected. The proportion of imipenem-resistant bacteria was on average 2.24 CFU/ml. Imipenem-resist

Total and pathogenic Vibrio parahaemolyticus in shrimp: fast and reliable quantification by real-time PCR.

Vibrio parahaemolyticus is found in aquatic environments and is the leading cause of gastroenteritis due to seafood consumption worldwide. We evaluated a quantitative real-time PCR (Q-PCR) assay with hydrolysis probes, to determine whether this method could be used for the efficient counting of total, tdh and trh-positive V. parahaemolyticus in shrimps. We assessed the specificity of this assay, using 62 strains from 12 non target bacterial species of the Vibrio, Photobacterium, Shewanella and Aeromonas genera. Only V. parahaemolyticus with the appropriate target gene generated a fluorescent s

Identification of a plasmid-encoded gene from Haemophilus ducreyi which confers NAD independence.

Members of the family Pasteurellaceae are classified in part by whether or not they require an NAD supplement for growth on laboratory media. In this study, we demonstrate that this phenotype can be determined by a single gene, nadV, whose presence allows NAD-independent growth of Haemophilus influenzae and Actinobacillus pleuropneumoniae. This gene was cloned from a 5.2-kb plasmid which was previously shown to be responsible for NAD independence in Haemophilus ducreyi. When transformed into A. pleuropneumoniae, this cloned gene allowed NAD-independent growth on complex media and allowed the u

Rapid Decomposition of Gallic Acid in Anaerobic Conditions Mediated by Bacteria-Iron Mineral Interactions.

It is widely held that polyphenols accumulate in soil due to limited phenol oxidase activity, thereby inhibiting resident microbes and promoting carbon stabilization. Contradicting this paradigm, we demonstrate that bioreduced Fe(II) from iron minerals may be enriched under anaerobic conditions, which accelerates phenol decomposition. Using gallic acid (GA) as a model, the biodegradation rate was 1.6-2.2 times higher under anaerobic conditions (k = 0.021-0.26 h-1) compared to aerobic environments in bacteria (Bacillus megaterium, Lactoccus Lactis, or Shewanella putrefaciens) cocultured with ir

Mariculture Probiotics: A Scoping Review of Research Trends, Knowledge Gaps and Opportunities.

The use of probiotics - live microorganisms - to enhance growth and prevent disease in mariculture species is an emerging alternative to conventional methods including antibiotics, vaccines and chemical intervention. While others have reviewed specific host organisms and probiotic genera, this review synthesises the scientific literature on the use of probiotics across a range of marine organisms to identify knowledge gaps and research opportunities. We identified widespread methodological heterogeneity, making direct inter-study comparisons difficult. Despite this limitation, 86% of in vivo s

A type of multifunctional cellulose nanocrystal composite silicone-based polymer coating for marine antibiofouling.

Nanocomposite polymer coatings are being used as a new generation of marine antibiofouling coatings because of their toxin-free chemical composition and ease of large-scale adoption. Cellulose nanocrystal (CN) exhibits significant potential for composite reinforcement. Herein, CN was surface-modified via α,ω-bis(3-(2-hydroxyl-terminated polydimethylsiloxane (HTPDMS), resulting in dihydroxyl-terminated poly(dimethylsiloxane)-grafted CN (HP-g-CN). The amine-terminated PDMS as the foundational component was sequentially reacted with isophorone diisocyanate, isophthalaldehyde, and carbon disulfide

Antimicrobial Drug-Resistant Gram-Negative Saprophytic Bacteria Isolated from Ambient, Near-Shore Sediments of an Urbanized Estuary: Absence of β-Lactamase Drug-Resistance Genes.

We assessed the prevalence of antimicrobial resistance and screened for clinically relevant β-lactamase resistance determinants in Gram-negative bacteria from a large urbanized estuary. In contrast to the broad literature documenting potentially hazardous resistance determinants near wastewater treatment discharge points and other local sources of aquatic pollution, we employed a probabilistic survey design to examine ambient, near-shore sediments. We plated environmental samples from 40 intertidal and shallow subtidal areas around San Francisco Bay (California, USA) on drug-supplemented MacCo

A multi-heme flavoenzyme as a solar conversion catalyst.

The enzyme flavocytochrome c3 (fcc3), which catalyzes hydrogenation across a C═C double bond (fumarate to succinate), is used to carry out the fuel-forming reaction in an artificial photosynthesis system. When immobilized on dye-sensitized TiO2 nanoparticles, fcc3 catalyzes visible-light-driven succinate production in aqueous suspension. Solar-to-chemical conversion using neutral water as the oxidant is achieved with a photoelectrochemical cell comprising an fcc3-modified indium tin oxide cathode linked to a cobalt phosphate-modified BiVO4 photoanode. The results reinforce new directions in th

Hierarchical scaffolding with Bambus.

The output of a genome assembler generally comprises a collection of contiguous DNA sequences (contigs) whose relative placement along the genome is not defined. A procedure called scaffolding is commonly used to order and orient these contigs using paired read information. This ordering of contigs is an essential step when finishing and analyzing the data from a whole-genome shotgun project. Most recent assemblers include a scaffolding module; however, users have little control over the scaffolding algorithm or the information produced. We thus developed a general-purpose scaffolder, called B
