The clinical use of P300 event related potentials for the evaluation of cholinesterase inhibitors treatment in demented patients.

Centrally acting cholinesterase inhibitors (ChEIs) improve cognitive functions in Alzheimer's disease (AD) and other forms of dementia. Evaluation of treatment efficacy is based mainly on subjective assessment methods such as standardized neuropsychological tests. Therefore, an additional objective tool for the evaluation of drug response would be most helpful.Thirty-two patients suffering from dementia of several etiologies were treated with ChEIs (tacrine 19, donepezil 5, rivastigmine 8). Cognitive response was assessed pre ChEIs initiation (baseline) and after 26 weeks, as optimal tolerated

Pharmacodynamics of cholinesterase inhibitors suggests add-on therapy with a low-dose carbamylating inhibitor in patients on long-term treatment with rapidly reversible inhibitors.

Despite three decades of intensive research in the field of Alzheimer's disease (AD) and numerous clinical trials of new therapeutic agents, cholinesterase inhibitors (ChEIs) are still the mainstay of therapeutics for AD and dementia with Lewy bodies. Pharmacodynamic analyses of ChEIs provide paradoxical observations. Treatment with the rapidly reversible, noncarbamylating ChEIs (donepezil, galantamine, and tacrine) increases acetylcholinesterase (AChE) protein expression, whereas the carbamylating agent, rivastigmine, produces sustained inhibition with no significant change in AChE protein ex

Apolipoprotein E4 allele as a predictor of cholinergic deficits and treatment outcome in Alzheimer disease.

Apolipoprotein E (apoE) is critical in the modulation of cholesterol and phospholipid transport between cells of different types. Human apoE is a polymorphic protein with three common alleles, APO epsilon 2, APO epsilon 3, and APO epsilon 4. ApoE4 is associated with sporadic and late-onset familial Alzheimer disease (AD). Gene dose was shown to have an effect on risk of developing AD, age of onset, accumulation of senile plaques in the brain, and reduction of choline acetyltransferase (ChAT) activity in the hippocampus of AD subjects. To characterize the possible impact of the apoE4 allele on

Synthesis, characterization, crystal structure of novel bis-thiomethylcyclohexanone derivatives and their inhibitory properties against some metabolic enzymes.

In this study, a series of novel bis-thiomethylcyclohexanone compounds (3a-3j) were synthesized by the addition of thio-Michael to the bis-chalcones under mild reaction conditions. The bis-thiomethylcyclohexanone derivatives (bis-sulfides) were characterized by 1H NMR, 13C NMR, FTIR and elemental analysis techniques. Furthermore, the molecular and crystal structures of 3h, 3i and 3j compounds were determined by single crystal X-ray diffraction studies. In this study, X-ray crystallography provided an alternative and often-complementary means for elucidating functional groups at the enzyme inhi

Recent trends in anti-Alzheimer's potential of novel biologically active isatin analogues: synthetic strategies, structural activity relationship studies and molecular docking insights.

Alzheimer's disease is a progressive neurodegenerative illness i.e., characterized by cognitive decline, memory impairment, cholinergic dysfunction, oxidative stress, Aβ aggregation, tau hyperphosphorylation and neuroinflammation. Due to multifactorial nature of AD, conventional single-target therapeutic approaches exhibit limited clinical success. Hence, development of multifunctional small molecules emerged as a promising strategy for management of Alzheimer's diseases i.e., capable to modulate multiple pathological pathways simultaneously. Among various heterocyclic pharmacophores, isatin (

Rivastigmine-centered molecular hybridization strategies for multi-targeted intervention in Alzheimer's disease: Advances (2020-2026).

Alzheimer's disease (AD) is a progressive and multifactorial neurodegenerative disorder characterized by cholinergic dysfunction, amyloid-β (Aβ) aggregation, tau hyperphosphorylation, oxidative stress, mitochondrial impairment, metal dyshomeostasis, and chronic neuroinflammation, collectively leading to irreversible cognitive decline and neuronal degeneration. Despite decades of intensive research, currently approved therapeutic agents, including cholinesterase inhibitors (ChEI) and NMDA receptor antagonists, provide only symptomatic relief and do not substantially alter disease progression. Novel pyrimidine-based N-Acylhydrazones as dual AChE inhibitors and antioxidants targeting Alzheimer's disease.

Alzheimer's disease (AD) is a multifactorial neurodegenerative disorder characterized by cholinergic dysfunction and oxidative stress, making the development of multi-target-directed ligands (MTDLs) an attractive therapeutic strategy. Herein, we designed and synthesized a novel series of pyrimidinone-based acyl hydrazones 6a-g bearing substituted aryl or heteroaryl (furan) moieties. The synthesized compounds were evaluated for their in vitro acetylcholinesterase (AChE) inhibitory and antioxidant activities. The synthesized pyrimidinone-based acyl hydrazones (6a-g) exhibited promising acetylcho

The anti-amnesic and neuroprotective effects of donepezil against amyloid beta25-35 peptide-induced toxicity in mice involve an interaction with the sigma1 receptor.

BACKGROUND AND PURPOSE: The acetylcholinesterase inhibitor, donepezil, is also a high affinity sigma(1) receptor agonist. We examined the involvement of sigma(1) receptors in its anti-amnesic and neuroprotective properties against amyloid beta(25-35) peptide-induced toxicity in mice. EXPERIMENTAL APPROACH: Mice were given an intracerebroventricular (i.c.v.) injection of Abeta(25-35) peptide (9 nmol) 7-9 days before being tested for spontaneous alternation and passive avoidance. Hippocampal lipid peroxidation was measured 7 days after Abeta(25-35) injection to evaluate oxidative stress. Donepe

Effects of intracranial infusions of chlordiazepoxide on spatial learning in the Morris water maze. II. Neuropharmacological specificity.

In the preceding paper it was found that infusions of chlordiazepoxide (CDP) into the medial septal region, but not several other regions possessing a high density of benzodiazepine receptors, impaired spatial learning, but not cue learning or swim speed, in the Morris water maze. The present investigation sought to further characterize the neuropharmacological profile of this effect. Initially, it was reconfirmed that systemically administered CDP impaired spatial learning, but not cue learning or swim speed, in the water maze. Additionally, it was found that systemically administered scopola

THA increases action potential duration of central histamine neurons in vitro.

Long-term administration of 9-amino-1,2,3,4-tetrahydroacridine (THA) has been reported to produce marked clinical improvement in patients suffering from Alzheimer's disease. The dramatic enhancement of cognitive function seen with THA contrasts sharply with the modest improvements seen with other forms of anticholinesterase therapy, suggesting that additional mechanisms might play a role in its therapeutic efficacy. When applied to hypothalamic histamine neurons maintained in vitro, THA produced a dose-dependent increase in action potential duration. By its effect upon action potential duratio

Disentangling the formation, mechanism, and evolvement of the covalent methanesulfonyl fluoride acetylcholinesterase adduct: Insights into an aged-like inactive complex susceptible to reactivation by a combination of nucleophiles.

Chemical warfare nerve agents and pesticides, known as organophosphorus compounds inactivate cholinesterases (ChEs) by phosphorylating the serine hydroxyl group located at the active site of ChEs. Over the course of time, phosphorylation is followed by loss of an organophosphate-leaving group and the bond with ChEs becomes irreversible, a process known as aging. Differently, structurally related irreversible catalytic poisons bearing sulfur instead of phosphorus convert ChEs in its aged form only by covalently binding to the key catalytic serine. Kinetic and crystallographic studies of the int

Non-dopaminergic Alterations in Depression-Like FSL Rats in Experimental Parkinsonism and L-DOPA Responses.

Depression is a common comorbid condition in Parkinson's disease (PD). Patients with depression have a two-fold increased risk to develop PD. Further, depression symptoms often precede motor symptoms in PD and are frequent at all stages of the disease. However, the influence of a depressive state on the responses to antiparkinson treatments is largely unknown. In this study, the genetically inbred depression-like flinders sensitive line (FSL) rats and control flinders resistant line (FRL) rats were studied in models of experimental parkinsonism. FSL rats showed a potentiated tremorgenic respon

Drug interactions with neuromuscular blockers.

Drugs administered to patients undergoing anaesthesia may complicate the use of the neuromuscular blockers that are given to provide good surgical conditions. The various sites of interaction include actions on motor nerve conduction and spinal reflexes, acetylcholine (ACh) synthesis, mobilisation and release, sensitivity of the motor end plate to ACh and the ease of propagation of the motor action potential. In addition, many drugs affect the pharmacokinetics of neuromuscular blockers, especially as most drugs depend to a greater or lesser extent upon renal excretion. The clinically significa

Effects of tetrahydroaminoacridine and nicotine in nucleus basalis and serotonin-lesioned rats.

The present study was designed to investigate the hypothesis that concurrent degeneration of serotonin and acetylcholine cells may decrease the therapeutic effects of cholinergic drugs on cognitive functioning in Alzheimer dementia. Therefore, we compared the effects of pretraining injections of a cholinesterase inhibitor, tetrahydroaminoacridine (1, 3 and 5 mg/kg i.p.), and nicotine (0.03, 0.1 and 0.3 mg/kg i.p.) on spatial navigation (water maze) and passive avoidance in nucleus basalis- and nucleus basalis+p-chlorophenylalanine-lesioned rats. Design, synthesis, biological evaluation, and in silico studies of novel pyrrolidine based hybrids bearing indole, chromone, and benzoylthiourea pharmacophores as multitarget hCA and AChE inhibitors.

In this study, novel pyrrolidine-based compounds bearing indole, chromone, and benzoylthiourea pharmacophores were rationally designed, synthesized, and evaluated as dual inhibitors of carbonic anhydrase I/II (hCA I, hCA II) and acetylcholinesterase (AChE). The target compounds were synthesized via imine azomethine ylide 1,3-dipolar cycloaddition and in selected cases, benzoyl isothiocyanate condensation reactions. Their structures were fully characterized using FTIR, NMR, microanalysis, and single-crystal X-ray diffraction (for 1ba). Compound 2ad (hCA I, Ki = 35.56 ± 5.10 nM, hCA II, Ki = 31.

Traditional herbal medicine Ga-Mi Gongjindan improves muscarinic cholinergic dysfunction through regulation of BDNF/CREB signaling pathway using a scopolamine-induced cognitive impairment of mouse model.

BACKGROUND: Neurodegenerative diseases, such as Alzheimer's disease (AD), are characterized by progressive memory loss and cognitive dysfunction, often linked to cholinergic system deterioration and hippocampal oxidative stress. Current pharmacological treatments offer only modest symptomatic relief and are often accompanied by adverse effects. In traditional Korean medicine, Ga-Mi Gongjindan (GJD), a modified formulation of Gongjindan, has long been used for enhancing cognitive function, but its neuropharmacological basis remains largely unexplored. OBJECTIVE: This study aimed to investigate
