Commercial coconut palm as an ecotope of Chagas disease vectors in north-eastern Venezuela.

BACKGROUND & OBJECTIVE: There are few reports of Coccus nucifera (Palmae) infestation by triatomines (Hemiptera, Reduviidae, Triatominae), vectors of Trypanosoma cruzi (Kinetoplastida: Trypanosomatidae), the causal agent of American Trypanosomiasis. The aim of this study was to determine if this palm is an appropriate ecotope for Rhodnius prolixus and Triatoma maculata, the main vectors in Venezuela. METHODS: Dry and green leaves, humid debris, interfoliaceus meshes and bracts from C. nucifera from north-eastern Venezuela were examined for the presence of triatomines. Samples of the intestina

Trypanosoma cruzi I genotype among isolates from patients with chronic Chagas disease followed at the Evandro Chagas National Institute of Infectious Diseases (FIOCRUZ, Brazil).

INTRODUCTION:: Trypanosoma cruzi is the etiologic agent of Chagas disease in humans, mainly in Latin America. Trypanosome stocks were isolated by hemoculture from patients followed at Evandro Chagas National Institute of Infectious Diseases (FIOCRUZ) and studied using different approaches. METHODS:: For species and genotype identification, the stocks were analyzed by parasitological techniques, polymerase chain reaction assays targeted to specific DNA sequences, isoenzyme patterns, besides sequencing of a polymorphic locus of TcSC5D gene (one stock). RESULTS:: The isolates presented typical

Characterisation of Trypanosoma cruzi populations by DNA polymorphism of the cruzipain gene detected by single-stranded DNA conformation polymorphism (SSCP) and direct sequencing.

Fifty fresh isolates of Trypanosoma cruzi from Triatoma dimidiata vectors and 31 from patients with Chagas' disease were analysed for DNA polymorphisms within the 432-bp core region of the cruzipain gene which encodes the active site of cathepsin L-like cystein proteinase. The cruzipain gene showed signs of polymorphism consisting of four different DNA sequences in Central and South American isolates of T. cruzi. The PCR fragments of Guatemalan isolates could be divided into three groups, Groups 1, 2 and 3, based on different patterns of single-stranded DNA conformation polymorphism. All of th

Effect of habitat fragmentation on rural house invasion by sylvatic triatomines: A multiple landscape-scale approach.

After the decrease of the relative importance of Triatoma infestans, a number of studies reported the occurrence of sylvatic triatomines dispersing actively to domestic environments in the dry western Chaco Region of Argentina. Anthropic modification of the landscape is mentioned as one of the main causes of the increase in domicile invasion. The aim of this study was to describe the occurrence and frequency of sylvatic triatomines invading rural houses, and to evaluate the effect of habitat fragmentation and other ecological factors on the invasion of rural houses in central Argentina. We hyp

Trypanosoma cruzi transmission in a Colombian Caribbean region suggests that secondary vectors play an important epidemiological role.

BACKGROUND: Colombia, as part of The Andean Countries Initiative has given priority to triatomine control programs to eliminate primary (domiciliated) vector species such as Rhodnius prolixus and Triatoma dimidiata. However, recent events of Trypanosoma cruzi transmission in localities where R. prolixus and T. dimidiata are not present suggest that other species are involved in the T. cruzi transmission cycle. METHODS: We studied T. cruzi transmission on Margarita Island, located on the Magdalena River in the Colombian Caribbean region, where a high number of non-domiciliated triatomines infe

[Entomological survey (1975-1983)].

After the systematization and re-dimension of the vectorial control in all the Country by 1975, it was considered necessary to have an up-to-date information on the distribution of vectors in Brazil, and differentiate precisely the role of each of the different species on the intra-domiciliary transmission of Chagas disease. For this purpose, sampling regional surveys for regions with non reliable information were performed, as well as, a house by house search for vectors on those areas considered at risk. For this last, 1,942 municipalities from 19 states were searched, as by the political di

Importance of species of Triatominae (Heteroptera: Reduviidae) in risk of transmission of Trypanosoma cruzi in western Mexico.

The epidemiological risk of infection by Trypanosoma cruzi Chagas in human populations of western Mexico is still under study. Although most vectors in this region and their vector capability are already known, new studies estimating the risk and the importance of individual Triatominae species (Hemiptera: Reduviidae) for T. cruzi transmission are necessary. For 1 yr, every month, > 400 human dwellings and their surroundings in eight communities of two western Mexico states were searched for triatomines. More than 1,000 specimens representing four species were collected and checked for T. cruz

Chagas disease in a naturally-infected dog from Northeast Brazil: a case report.

BACKGROUND: Dogs are the main domestic reservoir host of the parasite Trypanosoma cruzi, the causative agent of Chagas disease, and they are considered sentinel animals for the detection of new cases of human infection. Canis familiaris is also a well-established experimental T. cruzi infection model, but little is known about the progression of Chagas disease in naturally-infected dogs, especially during the acute phase in these hosts. Triatomine species infected with discrete typing units (DTUs) I, II and III of T. cruzi have been previously found in many of the municipalities of the state o

Beating the odds: Sustained Chagas disease vector control in remote indigenous communities of the Argentine Chaco over a seven-year period.

BACKGROUND: Rapid reinfestation of insecticide-treated dwellings hamper the sustained elimination of Triatoma infestans, the main vector of Chagas disease in the Gran Chaco region. We conducted a seven-year longitudinal study including community-wide spraying with pyrethroid insecticides combined with periodic vector surveillance to investigate the house reinfestation process in connection with baseline pyrethroid resistance, housing quality and household mobility in a rural section of Pampa del Indio mainly inhabited by deprived indigenous people (Qom). American trypanosomiasis, or Chagas disease, in Panama: a chronological synopsis of ecological and epidemiological research.

American trypanosomiasis, or Chagas disease, is a growing public health problem in Panama, and further forest degradation due to human population growth is expected to worsen the situation. Most people infected with the parasite Trypanosoma cruzi are silently ill, and their life expectancy is severely compromised, which contributes to further deterioration of living conditions in endemic regions. Here, we review the outcomes of nearly 100 years of ecological and epidemiological investigation about Chagas disease in Panama, in an attempt to highlight progress, identify needs, and re-orient futu

Vectorial transmission of Trypanosoma cruzi: an experimental field study with susceptible and immunized hosts.

The dynamics of vectorial transmission of Trypanosoma cruzi and the level of host (guinea pigs) protection after immunization with attenuated parasites (TCC strain) was studied under natural climatic conditions in an endemic region of northern Argentina. The experimental design included two guinea pig corrals isolated by mosquito netting. One (controls) had 17 healthy and susceptible adult guinea pigs. The other had 19 guinea pigs immunized with attenuated T. cruzi TCC strain. Each corral was colonized in April 1988 with equal-sized populations of Triatoma infestans naturally infected by T. cr

Chagas Disease Infection Prevalence and Vector Exposure in a High-Risk Population of Texas Hunters.

Chagas disease, caused by the vector-borne parasite Trypanosoma cruzi, remains one of the most significant neglected tropical diseases affecting the Americas. Identifying high-risk populations is important for understanding Chagas disease transmission and directing public health resources. We recently hypothesized that Texas hunters may be at an elevated risk for contracting Chagas disease because of opportunities for vector exposure and contact with blood of infected reservoirs. To assess their unique exposure risks, we conducted a statewide screening program of Texas hunters. A total of 885

New sylvatic hosts of Trypanosoma cruzi and their reservoir competence in the humid Chaco of Argentina: a longitudinal study.

A four-year longitudinal study of the structure of sylvatic transmission cycles of Trypanosoma cruzi, reservoir host competence and parasite discrete typing units was conducted in a disturbed rural area of the humid Chaco in Argentina. Among 190 mammals examined by xenodiagnosis and polymerase chain reaction amplification, the composite prevalence of infection was substantially higher in Dasypus novemcinctus armadillos (57.7%) and Didelphis albiventris opossums (38.1%) than in Euphractus sexcinctus (20.0%), Tolypeutes matacus (12.5%), and Chaetophractus vellerosus (6.3%) armadillos. Metabolic signatures of triatomine vectors of Trypanosoma cruzi unveiled by metabolomics.

Chagas disease is a trypanosomiasis whose causative agent is the protozoan parasite Trypanosoma cruzi, which is transmitted to humans by hematophagous insects known as triatomines and affects a large proportion of South America. The digestive tract of the insect vectors in which T. cruzi develops constitutes a dynamic environment that affects the development of the parasite. Thus, we set out to investigate the chemical composition of the triatomine intestinal tract through a metabolomics approach. We performed Direct Infusion Fourier Transform Ion Cyclotron Resonance Mass Spectrometry on fecal

Chronic Chagas disease: PCR-xenodiagnosis without previous microscopic observation is a useful tool to detect viable Trypanosoma cruzi.

UNLABELLED: We evaluate the elimination of the microscopic stage of conventional xenodiagnosis (XD) to optimize the parasitological diagnosis of Trypanosoma cruzi in chronic Chagas disease. To this purpose we applied under informed consent two XD cages to 150 Chilean chronic chagasic patients. The fecal samples (FS) of the triatomines at 30, 60 and 90 days post feeding were divided into two parts: in one a microscopic search for mobile trypomastigote and/or epimastigote forms was performed. In the other part, DNA extraction-purification for PCR directed to the conserved region of kDNA minicirc

Differential antioxidant enzyme profiles reveal early molecular signatures of virulence in Trypanosoma cruzi DTU-TcI and DTU-TcVI strains.

BACKGROUND: Trypanosoma cruzi, the causative agent of Chagas disease, exhibits significant genetic and phenotypic diversity that influences clinical outcomes. Antioxidant enzymes are crucial for parasite survival under host-induced oxidative stress, yet their role as determinants of virulence remains underexplored. OBJECTIVE: This study aimed to characterize and compare early antioxidant responses between the virulent T. cruzi CL Brener clone and a non-virulent strain isolated from Arequipa, Peru, to test the hypothesis that elevated antioxidant enzyme expression is associated with parasite v

Characterization of Trypanosoma cruzi from Argentina by electrophoretic zymograms.

Polyacrylamide gel electrophoretic patterns for six enzymes in 73 isolates and 38 clones of Trypanosoma cruzi from different areas of Argentina were classified into 12 zymodemes. The isolates were obtained from human patients with acute, chronic or congenital Chagas' disease, vector insects, domestic and sylvatic animals. Two out of 8 isolates cloned were shown to be heterogeneous. Zymodemes 1 and 12 exhibit widespread geographic distribution; isolates belonging to both zymodemes account for 55% of the total analyzed. The other zymodemes are not widely geographically dispersed. Although there

[Trypanosoma cruzi strains resistant to benznidazole occurring in Colombia].

Introduction. Chagas disease caused by Trypanosoma cruzi is one of the most serious public health problems in the Americas. Benznidazole is one of two drugs used to treat Chagas' disease. However, the variation in susceptibility of the parasite to this drug is one of the main causes of treatment failure. Objective. The in vitro susceptibility to benznidazole was assessed in Colombian strains of T. cruzi from several sources and geographical regions. Materials and methods. Thirty-three Colombian T. cruzi strains were isolated from humans, vectors and mammals. These were analyzed in vitro by the

An insight into the salivary gland and fat body transcriptome of Panstrongylus lignarius (Hemiptera: Heteroptera), the main vector of Chagas disease in Peru.

Triatomines are hematophagous arthropod vectors of Trypanosoma cruzi, the causative agent of Chagas Disease. Panstrongylus lignarius, also known as Panstrongylus herreri, is considered one of the most versatile triatomines because it can parasitize different hosts, it is found in different habitats and countries, it has sylvatic, peridomestic and domestic behavior and it is a very important vector of Chagas disease, especially in Peru. Molecules produced and secreted by salivary glands and fat body are considered of important adaptational value for triatomines because, among other functions, t
