To examine the connection between caecal size and urea concentration in the caecal contents urease inhibition was tested in conventional animals and urea and urease were administered to germ-free rats and mice. Administration of alloxan and barbituric acid and immunization with urease led to slightly larger caeca in conventional animals. Neomycin treatment caused a clear enlargement of the caecum. In germ-free animals urease administration led to a reduction in the caecal size, whereas urea in the drinking water caused enlargement. The urea concentration and the urease activity in the caecal c

The correlation between urease activity of Campylobacter pylori and atrophic gastritis was studied. On the basis of fundamental study on the optimal pH of C. pylori urease activity, urease activity of 38 biopsied specimens were measured under pH 5 condition, and compared with the positive ratio of C. pylori. In this study, sensitivity was 86.7%, and specificity was 87.0%, respectively. Mean urease activity of C. pylori positive specimens was 3.69 mIU/mg protein, and under this condition, C. pylori was likely to produce ammonia of 0.0218 mumole per minute, enough to damage the gastric mucosa. Urea degradation by urease represents a promising green strategy for reducing ethyl carbamate in alcoholic beverages. This study characterized the urea metabolism capacity of indigenous Schizosaccharomyces japonicus, with strains FBKL2.9SZJ3 and FBKL2.9792 exhibiting robust urease activities (1.20 and 1.26 U/mL). They degraded over 95% of urea during alcoholic fermentation with initial levels of 1.5-6.0 g/L. Although the highest urease activity was induced in the simulated fermentation with initial urea level of 20 g/L, the formation of ethanol was restrained. Interestingly, the restrain was r

UNLABELLED: In this study, our objective was to determine the presence of Helicobacter pylori and other rapid urease-positive (RUP) organisms in gastric biopsies in 70 patients, 30 of whom had ulcers or erosions. A retrospective chart review was undertaken to correlate diagnostic tests (culture and direct urease) with results of endoscopic examination and the patient's clinical information. Eleven of 70 (15.7%) patients' biopsies were positive by both culture and direct urease for H. pylori, seven (10%) patients' biopsies were positive by culture only, and eight (11.4%) biopsies by direct urea

Herein, the zwitterionic material poly (carboxybetaine acrylamide) was grafted onto iron oxide to obtain biocompatible magnetic nanoparticles Fe3O4-pCBAA which were employed to immobilize enzymes. The nanocomplxes Fe3O4-pCBAA were characterized using scanning electron microscopy (SEM), dynamic light scattering (DLS), zeta potential, Fourier transform-infrared (FT-IR) spectra and energy dispersive X-ray spectrometry (EDX). The urease as a model enzyme was immobilized with the novel supports and the properties of immobilized urease were further investigated in comparison with the free urease cou

BACKGROUND: Urease is a virulence factor that plays a role in the resistance of Brucella to low pH conditions, both in vivo and in vitro. Brucella contains two separate urease gene clusters, ure1 and ure2. Although only ure1 codes for an active urease, ure2 is also transcribed, but its contribution to Brucella biology is unknown.
RESULTS: Re-examination of the ure2 locus showed that the operon includes five genes downstream of ureABCEFGDT that are orthologs to a nikKMLQO cluster encoding an ECF-type transport system for nickel. ureT and nikO mutants were constructed and analyzed for urease ac

In this work, urea detection techniques based on the pH sensitivity of CdSe/ZnS QDs were developed using three types of sol-gel membranes: a QD-entrapped membrane, urease-immobilized membrane and double layer consisting of a QD-entrapped membrane and urease-immobilized membrane. The surface morphology of the sol-gel membranes deposited on the wells in a 24-well microtiter plate was investigated. The linear detection range of urea was in the range of 0-10mM with the three types of sol-gel membranes. The urea detection technique based on the double layer consisting of the QD-entrapped membrane a

Helicobacter pylori (H. pylori) is a small Gram negative bacillus, recently discovered, found in the stomach of patients with active chronic gastritis and duodenal ulcers. Production of a potent urease has been described as a trait common to all H. pylori so far isolated. To clarify the role of urease in the pathogenic process, as well as to engineer genetic tools useful for the diagnosis of H. pylori, we cloned the genes responsible for urease activity. A genomic library was constructed in Escherichia coli (E. coli) from the chromosomal DNA of the H. pylori stain 85P using a shuttle cosmid ve

Staphylococcus aureus (S. aureus) is a major opportunistic pathogen, causing acute and chronic infections including urinary tract infection (UTI). S. aureus relies on the Accessory gene regulator (Agr), a central quorum sensing (QS) system and the urease genes ureABCEFGD, for the regulation of urease expression and its pathogenicity. Urease is a key virulence factor for S. aureus, modulating immune responses by altering the local urinary pH and impairing immune defenses in the hostile urinary environment. QS is regulated by the external stimuli and in urological disease, the role of different

Urease has become an important therapeutic target because it stimulates the pathogenesis of many human health conditions, such as pyelonephritis, the development of urolithiasis, hepatic encephalopathy, peptic ulcers, gastritis and gastric cancer. A series of alkyl chain-linked thiourea derivatives were synthesized to screen for urease inhibition activity. Structure elucidation of these compounds was done by spectral studies, such as IR, 1H NMR and 13C NMR, and MS analysis. In vitro urease enzyme inhibition assay revealed that compound 3c was the most potent thiourea derivative among the serie

Elevated urease activity in tuberculosis-infected individuals is often suggestive of bacterial virulence by promoting survivability and establishment of the causative pathogen (Mycobacterium tuberculosis) in the host. Thus, heightened urease activity could be a promising biomarker for screening pathogenic mycobacterial strains. In this proof-of-concept study, we evaluate the suitability of electrochemical techniques for direct screening of mycobacterial strains based on urease activity. Mycolicibacterium smegmatis wildtype (WT), a derived knockout mutant of the LCP protein family (∆0107) with

The development of novel scaffolds that can increase the effectiveness, safety, and convenience of medication therapy using drug conjugates is a promising strategy. As a result, drug conjugates are an active area of research and development in medicinal chemistry. This research demonstrates acetamide-sulfonamide scaffold preparation after conjugation of ibuprofen and flurbiprofen with sulfa drugs, and these scaffolds were then screened for urease inhibition. The newly designed conjugates were confirmed by spectroscopic techniques such as IR, 1HNMR, 13CNMR, and elemental analysis. Ibuprofen con

Epitope vaccine is a promising option for prophylactic and therapeutic vaccination against Helicobacter pylori infection. Urease is an essential virulence factor and colonization factor for H. pylori. In this study, we constructed a multi-epitope vaccine named CTB-UE with mucosal adjuvant cholera toxin B subunit (CTB) and tandem copies of Th and B cell epitopes from H. pylori urease A and B subunits. The immunogenicity, specificity, ability to induce neutralizing antibodies against H. pylori urease, and prophylactic and therapeutic efficacy of the CTB-UE vaccine were evaluated in BALB/c mice m

The nickel-containing enzymes of Helicobacter pylori, urease and hydrogenase, are essential for efficient colonization in the human stomach. The insertion of nickel into urease and hydrogenase is mediated by the accessory protein HypA. HypA contains an N-terminal nickel-binding site and a dynamic structural zinc-binding site. The coordination of nickel and zinc within HypA is known to be critical for urease maturation and activity. Herein, we test the hydrogenase activity of a panel of H. pylori mutant strains containing point mutations within the nickel- and zinc-binding sites. We found that

The mechanism by which Helicobacter pylori, a noninvasive bacterium, initiates chronic antral gastritis in humans is unknown. We now show that H. pylori releases products with chemotactic activity for monocytes and neutrophils. This chemotactic activity was inhibited by antisera to either H. pylori whole bacteria or H. pylori-derived urease. Moreover, surface proteins extracted from H. pylori and purified H. pylori urease (a major component of the surface proteins) exhibited dose-dependent, antibody-inhibitable chemotactic activity. In addition, a synthetic 20-amino acid peptide from the NH2-t
