Mitochondrial respiration and redox coupling in articular chondrocytes.

INTRODUCTION: Chondrocytes rely primarily on glycolysis to meet cellular energy needs, but recent studies implicate impaired mitochondrial function in osteoarthritis (OA) pathogenesis. Our objectives were to investigate the ability of chondrocytes to upregulate mitochondrial respiration when challenged with a nutrient stress and determine the effect on mediators of chondrocyte oxidative homeostasis. METHODS: Primary bovine chondrocytes were isolated and cultured in alginate beads. Mitochondrial respiration was stimulated by culturing cells with galactose-supplemented media for a period of 1 o

Effects of in vitro endochondral priming and pre-vascularisation of human MSC cellular aggregates in vivo.

INTRODUCTION: During endochondral ossification, both the production of a cartilage template and the subsequent vascularisation of that template are essential precursors to bone tissue formation. Recent studies have found the application of both chondrogenic and vascular priming of mesenchymal stem cells (MSCs) enhanced the mineralisation potential of MSCs in vitro whilst also allowing for immature vessel formation. However, the in vivo viability, vascularisation and mineralisation potential of MSC aggregates that have been pre-conditioned in vitro by a combination of chondrogenic and vascular

Benefit of supplements in functional dyspepsia after treatment of Helicobacter pylori.

AIM: Functional dyspepsia, though benign, leads to deterioration of the quality of life and high costs for healthcare systems. The optimal therapy for functional dyspepsia is still to be defined because of its multifactorial pathogenesis. In an open multicentric study of patients with functional dyspepsia, we prospectively evaluated the benefit of treatment with a food supplement composed of sodium alginate, carbonate calcium, pineapple, papaya, ginger, α-galactosidase and fennel (Perdiges, Bioten Snc, Turin, Italy). METHODS: Ninety-one consecutive patients were included, suffering from funct

Encapsulated living choroid plexus cells: potential long-term treatments for central nervous system disease and trauma.

In neurodegenerative disease and in acute brain injury, there is often local up-regulation of neurotrophin production close to the site of the lesion. Treatment by direct injection of neurotrophins and growth factors close to these lesion sites has repeatedly been demonstrated to improve recovery. It has therefore been proposed that transplanting viable neurotrophin-producing cells close to the trauma lesion, or site of degenerative disease, might provide a novel means for continuous delivery of these molecules directly to the site of injury or to a degenerative region. The aim of this paper i

Evaluation of the efficacy of a redox agent in the treatment of chronic periodontitis.

A redox dye, methylene blue, was compared with subgingival root surface debridement and sterile water in the treatment of adult periodontitis. Plaque and gingival indices, bleeding on probing, and microbiological samples were obtained at baseline, and at 1, 4, 8 and 12 weeks following treatment. All subjects had matched pockets in each of the 4 quadrants, of 5 mm or more. One treatment consisted of 0.1% methylene blue gel irrigated professionally at 0, 1 and 4 weeks, and by subjects at days in between up to 4 weeks, at chosen sites within a randomly selected quadrant (split-mouth design). In vivo experience with natural scaffolds for myocardial infarction: the times they are a-changin'.

Treating a myocardial infarction (MI), the most frequent cause of death worldwide, remains one of the most exciting medical challenges in the 21st century. Cardiac tissue engineering, a novel emerging treatment, involves the use of therapeutic cells supported by a scaffold for regenerating the infarcted area. It is essential to select the appropriate scaffold material; the ideal one should provide a suitable cellular microenvironment, mimic the native myocardium, and allow mechanical and electrical coupling with host tissues. Among available scaffold materials, natural scaffolds are preferable

[Update on gastroesophageal reflux disease].

Gastroesophageal reflux disease is a highly frequent disorder classically characterized by the presence of heartburn and/or acid regurgitation that improves with drug therapy that reduces acid content in the stomach. However, especially in patients with non-erosive disease, response to proton pump inhibitors is unsatisfactory in approximately 1 out of 3 patients, and consequently, in these patients, it is important to establish a definitive diagnosis and an alternative therapeutic strategy. In the last few years, advances have been made in knowledge of the physiopathology of reflux, such as id

Association of Midgut Bacteria and Their Metabolic Pathways with Zika Infection and Insecticide Resistance in Colombian Aedes aegypti Populations.

INTRODUCTION: Aedes aegypti is the vector of several arboviruses such as dengue, Zika, and chikungunya. In 2015-16, Zika virus (ZIKV) had an outbreak in South America associated with prenatal microcephaly and Guillain-Barré syndrome. This mosquito's viral transmission is influenced by microbiota abundance and diversity and its interactions with the vector. The conditions of cocirculation of these three arboviruses, failure in vector control due to insecticide resistance, limitations in dengue management during the COVID-19 pandemic, and lack of effective treatment or vaccines make it necessary

Establishment and application of a vesicle extraction method for clinical strains of Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a versatile pathogen capable of causing illnesses that range from mild infections to life-threatening conditions. Its virulence is driven by a wide array of factors, among which extracellular vesicles (EVs) have gained recognition as important contributors to pathogenicity. Despite this, the full scope of their roles remains unclear. A major barrier to EV characterization is the difficulty of vesicle isolation-procedures are often lengthy, yield is low, and specialized equipment is required. In this study, we assessed the effectiveness of a rapid vesicle extraction me

Guidelines on diagnosis and management of gastroesophageal reflux disease in infants, children and adolescents: a joint consensus from Italian pediatric societies (SIP and SIGENP) -Part II: management.

BACKGROUND: Suspected gastroesophageal reflux disease (GERD) is one of the most common reasons for referral to pediatric and gastroenterology clinics, and proton pump inhibitors (PPIs) are frequently prescribed. Currently, there is still wide heterogeneity in treatment, especially in neonates and extraesophageal presentations. This article aimed to provide guidelines and a consensus of Italian experts on the management of GERD in infants, children, and adolescents, in order to improve the care and treatment of these patients. METHODS: A multidisciplinary pediatric panel identified four key cli

Pre-Conditioned Bone Marrow Mesenchymal Stromal Cell-Derived Secretome Exerts an Anti-Inflammatory Effect on Degenerative Nucleus Pulposus Cells In Vitro.

BACKGROUND: Intervertebral disc degeneration (IDD) is frequently associated with chronic low back pain (LBP), which contributes significantly to disability, psychological distress, and reduced work capacity. Mesenchymal stromal cell (MSC)-based treatments may offer a promising, less invasive alternative to conventional treatments for IDD. The MSC secretome has shown regenerative potential through paracrine and anti-inflammatory mechanisms. In this study, we investigated the effects of the secretome isolated from interleukin-1β (IL-1β)-preconditioned bone marrow-derived MSCs (BM-MSCs) on human

Proteolysis-Targeting Chimera-Loaded Hydrogel Dressings Orchestrate Immunoregulation and Angiogenesis to Promote Tissue Regeneration in Large Diabetic Wounds.

The delayed healing of diabetic wounds is primarily attributed to a persistent excessive inflammatory microenvironment and impaired angiogenesis. Here, we identify the STimulator of INterferon Genes (STING) as a key regulator driving this pathological state within the wound milieu. To address this, we developed SD-Gel, a thermosensitive protein-degrading hydrogel loaded with a PROteolysis-TArgeting Chimera (PROTAC) designed for targeted STING degradation. This SD-Gel is characterized by its reactive oxygen species (ROS)-responsive degradation, which enables simultaneous ROS scavenging and cont

Use of a Slow-Release Phenylalanine-Free Microtablet Protein Substitute in Children and Adolescents with Phenylketonuria: An Observational Pilot Study.

Background/Objectives: In phenylketonuria (PKU), adherence to protein substitute (PS) is frequently suboptimal due to poor palatability, high volume, and sensory fatigue. A novel phenylalanine (Phe)-free L-amino acid microtablet PS has been developed to address these barriers. The microtablets incorporate a cellulose-based taste-masking coating and a sodium-alginate inner matrix intended to enable controlled amino acid release over approximately three hours. This study evaluated their short-term tolerability, acceptability, and adherence in children with PKU, with extended follow-up in a subgr

Current Challenges and Potential Strategies to Enhance Efficacy of Oral Phage Therapy in Food Animals: A Systematic Review with Quantitative Analysis.

Phage therapy has enormous potential in combating bacterial resistance in food animals. However, its application via the oral route remains limited due to challenges associated with the gastrointestinal tract (GIT) environment and a lack of rigorous clinical trial evidence. Therefore, we systematically searched in Google Scholar, PubMed, Scopus, and Web of Science databases following PRISMA guidelines and finally identified 111 articles on oral phage therapy in food animals from where we summarized the key physiological and chemical factors of the gut environment hindering the effectiveness of

The conserved Pss exopolysaccharide is a common component of Listeria biofilms.

Listeria monocytogenes (Lm) is a major foodborne pathogen capable of persisting on fresh produce and in food-processing environments. Although exopolysaccharides (EPSs) are recognized as central components of resilient bacterial biofilms, Lm biofilms have long been considered to be EPS-less or EPS-poor. We previously identified a listerial EPS, Pss, whose biosynthesis is encoded by the highly conserved pss operon and is stimulated by elevated c-di-GMP levels. However, the prevalence of Pss in wild-type strains remained unclear, in part, due to the lack of a specific detection probe. Here, we e

Ultramicroporous Al(III) MOFs with selective CO2 adsorption, acid resistance, and efficient Cr(VI) sorption properties.

In the present work, we report the hydrothermal synthesis and detailed characterization of a series of ultramicroporous, luminescent, multivariate metal-organic frameworks (MOFs) with the general formula [Al(OH)(IATP)1-x(NH2-BDC)x], constructed from two different dicarboxylate ligands, H2IATP = 2-(((1H-imidazol-4-yl)methyl)amino) terephthalic acid and NH2-H2BDC = 2-aminoterephthalic acid. Structural analysis, using both powder X-ray diffraction (PXRD) and microcrystal electron diffraction (MicroED), confirms that the reported MTV-Al-MOFs are topological analogues of the related NH2-MIL-53(Al)

Decorporation dilemma: Interplay of prussian blue and potassium iodide in radioactive contamination.

The expansion of the nuclear industry has led to various radioactive effluents, originating from routine operations or catastrophic incidents such as those at Three Mile Island (USA), Chernobyl (Ukraine), and Fukushima (Japan). Research conducted after these events emphasizes Cesium-137 (137Cs) and iodine 131 (131I) as major contributors to harmful airborne dispersion and fallout. These isotopes infiltrate the human body via inhalation, ingestion, or wounds, posing significant health risks. Understanding contamination mechanisms and devising effective countermeasures are crucial in mitigating
