In vitro determinants of asbestos fiber toxicity: effect on the relative toxicity of Libby amphibole in primary human airway epithelial cells.

BACKGROUND: An abnormally high incidence of lung disease has been observed in the residents of Libby, Montana, which has been attributed to occupational and environmental exposure to fibrous amphiboles originating from a nearby contaminated vermiculite mine. The composition of Libby amphibole (LA) is complex and minimal toxicity data are available. In this study, we conduct a comparative particle toxicity analysis of LA compared with standard reference asbestiform amphibole samples. METHODS: Primary human airway epithelial cells (HAEC) were exposed to two different LA samples as well as stand

[A role for reactive oxygen species in the pathogenesis of focal glomerulosclerosis].

Previous studies have indicated that reactive oxygen species play a key role in puromycin aminonucleoside (PAN)-induced proteinuria in the acute nephrotic phase, and glomerular infiltration of macrophages (M phi) in the acute phase has a critical impact on the subsequent progression to glomerular sclerosis. In the present study, we examined the effect of deferoxamine (DFO), which is known to inhibit the generation of hydroxyl radical on the proteinuria, renal function and morphological changes in PAN-induced chronic glomerulopathy. Rats given a single dose of PAN (150 mg/kg BW, i.p.) were not

Activation of C5 by cobra venom factor is required in neutrophil-mediated lung injury in the rat.

Cobra venom factor (CVF)-induced systemic activation of the complement system in the rat has been shown to result in the development of acute lung microvascular injury and appearance in lungs and plasma of lipid peroxidation products. The pathogenesis of these events is dependent on complement and neutrophils and is sensitive to pretreatment of experimental animals with iron chelators or scavengers of hydroxyl radical. In order to further analyze the role of complement in the pathogenesis of acute lung injury in rats after systemic complement activation, two different CVFs have been employed i

Zika virus infections of human stem cell-derived cerebral organoids reveal viral lineage-specific pathogenesis responses.

UNLABELLED: Zika virus (ZIKV) has multiple lineages and strains that cause a range of disease severity, underscoring the need to elucidate differential neuropathogenesis mechanisms. Here, we performed systematic, side-by-side comparisons of African, Asian, and American ZIKV lineage infections using cerebral organoids derived from human embryonic stem cells, a relevant human model experimental system. African lineage ZIKV strains, as well as the ancestral Asian Malaysia strain, persistently infected neural progenitor cells, causing apoptosis and severe disruption of ventricular cytoarchitecture

Eugenol-derived carbamates as sub-nanomolar dual cholinesterase inhibitors with neuroprotective activity against Alzheimer's disease: Structure-activity relationships and molecular dynamics insights.

Carbamate-based cholinesterase inhibitors represent a clinically validated therapeutic strategy for Alzheimer's disease (AD); however, sub-nanomolar AChE inhibitory activity within natural phenol-derived scaffolds has not been previously reported. To address this gap, a series of eleven eugenol-based carbamate derivatives (E1-E11) was rationally designed, synthesized, and evaluated as multifunctional anti-AD agents. Systematic variation of the carbamate N-substituent enabled fine modulation of inhibitory potency and isoform selectivity, revealing well-defined structure-activity relationships.

Fingerprint profiling for quality evaluation and the related biological activity analysis of polysaccharides from Liuweizhiji Gegen-Sangshen beverage.

INTRODUCTION: Liuweizhiji Gegen-Sangshen beverage (LGS) is popular in China, which has been used for alleviating alcohol-mediated discomfort and preventing alcoholic liver disease (ALD). This beverage is consisted of six herbal components that are known as functional foods and fruits. LGS is rich in polysaccharides, however, the activity and quality evaluation of LGS-derived polysaccharides remain unexplored. The purpose of this study is thus to establish a comprehensive quality control methodology for the assessment of LGS polysaccharides (LGSP) and to further explore the anti-oxidant, anti-i

NIR-II triggered Cu(I) phosphide for chemodynamic and photothermal periodontitis treatment: Efficient reduction of bacterial co-aggregation.

The synergy between chemodynamic therapy (CDT) and photothermal therapy (PTT) offers a promising antimicrobial strategy for periodontitis, yet faces challenges like complex material structure and limited NIR-I light penetration. Additionally, low endogenous H2O2 levels in biofilm and a focus on bacterial eradication over colonization prevention limit current treatments. To address these issues, we newly introduce a single-material system (Cu3P@PAH@Lox) that integrates dual functionalities to synergistically enhance antimicrobial effects and significantly reduce pathogen co-aggregation. Iron-chelating agents attenuate NMDA-Induced neuronal injury via reduction of oxidative stress in the rat retina.

Excitoneurotoxicity is regarded as one of the mechanisms of the death of retinal ganglion cells induced by retinal central artery occlusion and glaucoma. Oxidative stress is at least in part involved in excitoneurotoxicity. Fenton reaction, which is catalyzed by Fe2+, is known to cause formation of hydroxyl radical, one of reactive oxygen species, suggesting that chelation of iron may be protective against excitoneurotoxicity. In the present study, we histologically evaluated whether zinc-deferoxamine (Zn-DFO) and deferasirox (DFX), common iron-chelating agents, were protective against N-methy

Biochemical activity of reactive oxygen species scavengers do not predict retinal ganglion cell survival.

PURPOSE: Retinal ganglion cells (RGCs) die as a result of axonal injury in a variety of optic neuropathies, including glaucoma. Reactive oxygen species (ROS) act as intracellular signaling molecules and initiate apoptosis in nerve growth factor-deprived sympathetic neurons and axotomized RGCs. Determination of the role of specific ROS relies on the use of small molecule or protein scavengers with various degrees of specificity. The pro- or anti-cell-death effect of several ROS generating and scavenging systems in cultured RGCs was correlated with their activity in cell-free assays. METHODS: N

Activation of JNK, p38 and ERK mitogen-activated protein kinases by chromium(VI) is mediated through oxidative stress but does not affect cytotoxicity.

In this study we have explored the involvement of oxidative stress in Cr(VI)-induced JNK, p38 and ERK signaling pathways and their effects on Cr(VI) cytotoxicity in human non-small cell lung carcinoma CL3 cells. Exposure to K(2)Cr(2)O(7) markedly activated JNK and p38 and moderately activated ERK in a dose- (10-80 microM) and time-dependent (1-12 h) manner. The activated p38 decreased markedly and rapidly and the activated JNK decreased gradually when Cr(VI) was removed from the medium. Post-incubation of Cr(VI)-treated cells with H(2)O(2) increased the activities of JNK and p38, but not ERK.

Characteristics of the active oxygen in covalent binding of the pesticide methoxychlor to hepatic microsomal proteins.

This study examined the characteristics of the active oxygen species involved in generation of the reactive intermediate of methoxychlor which covalently binds to liver microsomal proteins. The possibility that the active oxygen participating in the above reaction is the superoxide anion (O2-) or a species generated from O2- was examined with the help of superoxide dismutase (SOD) and with an SOD-mimetic agent, CuDIPS [Cu2+(3,5-diisopropylsalicylic acid)2]. It was observed that, whereas CuDIPS inhibited covalent binding of methoxychlor metabolite(s), SOD did not. However, ZnDIPS [Zn2+(3,5-diis

Ultrasound-Assisted Flash Extraction of Juglans cathayensis Polysaccharides and Their Phosphorylation Treatment: Structural Characterization, Antioxidant, and Hypoglycemic Activities.

Traditional methods for extracting Juglans cathayensis polysaccharides (JCP) suffer from low yield and poor efficiency; moreover, the biological activity of these natural polysaccharides is often limited, which severely hampers their functional development and application. Juglans cathayensis (JC) is a plant with medicinal and edible values. This study adopted the ultrasound-assisted flash extraction (UAFE) method to isolate polysaccharides from the fruits of JC. Response surface methodology (RSM) was employed to optimize the extraction conditions. Meanwhile, the purified polysaccharides (JCP)

Redox-modulating metallic nanoparticles: mechanistic insights into pharmacological activity and cellular therapeutics.

Redox imbalance refers to abnormal production of reactive oxygen species (ROS) and reactive nitrogen species (RNS), which causes cellular dysfunction and damage, playing a critical role in the onset and progression of many diseases including cancer, cardiovascular disorders, and neurodegenerative conditions such as Alzheimer's and Parkinson's disease. While many previous reviews have covered individual systems of nanoparticles (NP) or single disease applications separately, there is a missing link that has not been adequately summarized regarding the ability of the same metallic core to act as

Co-Cu Bimetallic ZIF-67@PDA-Based Fabrics: Trimodal Synergistic Antibacterial Performance for Reusable Protective Textiles.

Protective textiles are essential for public health security, yet conventional textiles only physically filter microorganisms without rapid surface sterilization, easily becoming potential cross-infection sources. Commercial antibacterial variants also suffer from slow sterilization, poor reusability, and insufficient multifunctional integration, thus failing to meet the demand for efficient and long-term protection. Herein, a facile and scalable two-step strategy (vacuum impregnation followed by PDMS spray coating) was developed to fabricate Co-Cu bimetallic ZIF-67@polydopamine (PDA)-based fa

Hyaluronic acid-mediated artemisinin/ferrocene co-delivery Nanoplatform enhances immune checkpoint blockade response by triggering tumor cell immunogenic cell death via H₂O₂-independent Chemodynamic therapy.

Immune checkpoint blockade (ICB) therapy faces major challenges including low clinical response rates, while traditional chemodynamic therapy (CDT) is highly dependent on intratumoral hydrogen peroxide (H₂O₂) levels. To address these limitations, we constructed a pH-responsive hyaluronic acid (HA)-based nanosystem (HA-Fc@ART) for targeted cancer therapy. This nanoplatform achieves tumor-specific delivery through the specific interaction between HA and CD44 receptors overexpressed on tumor cells. Upon accumulation in the acidic tumor microenvironment (TME), the nanosystem undergoes Schiff base
