Development of a Nucleoside Photosensitizer Efficiently Activated by One- or Two-Photon Absorption in the Optical Therapeutic Window.

Developing DNA/RNA nucleoside prodrugs that absorb in the red-to-near-infrared is crucial for applications requiring deep-tissue penetration. This study reports the rational design and synthesis of a novel thiocarbonyl analog, 5-(5-(4-(dimethylamino)phenyl)thiophen-2-yl)-2',3',5'-tri-O-benzoyl-6-aza-2,4-dithiouridine (DATU), efficiently activated through one- and two-photon absorption in the red-to-near-infrared spectrum. We report DATU's electronic structure and its photophysical and triplet-state properties under both excitation, comparing these results to its carbonyl counterpart, 5-(5-(4-(

Encapsulation of Glucose Oxidase in Porous Cu(II)-Doped Zinc Phosphate@polydopamine Nanoparticles for Triple-Combination Therapy of Cancer.

Designing multifunctional nanotherapeutic platforms that integrate multiple therapeutic capabilities with enhanced tumor specificity and low systemic toxicity has emerged as a promising strategy for cancer therapy. Herein, we put forward a simple and clear route to construct multifunctional nanoparticles (NPs) integrating chemodynamic therapy (CDT), starvation therapy (ST), and photothermal therapy (PTT) by using polydopamine as a protective layer to coat Cu2+-doped zinc phosphate loaded with glucose oxidase (designated as Cu-ZnP@GOx/PDA/PEG NPs) for optimizing therapeutic efficacy. The caprices of a trace element: selenium's considerable effects on Hashimoto's thyroiditis, though few on Graves' disease.

A simplified figure displaying the main events leading to Hashimoto's thyroiditis and Grave's disease via Th1 and Th2 activation, and the potential sites of selenium (Se) action. (A) Activated by the dendritic cell, naïve helper T cells (CD4+ T cells) can mainly differentiate into two subsets, Th1 and Th2, which are crucial in orchestrating immune responses. Th1 cells produce tumor necrosis factor (TNF-a) along with interferon-γ (IFN-γ), exacerbating inflammation, in synergy with interleukin-6 (ΙL-6), leading to apoptosis (death of follicular cells). In parallel, oxidative stress induces activ

Turning Waste into Treasure: Radiation Byproduct-Induced Fe(III)/Fe(II) Conversion for Efficient Ferroptosis to Improve Iodine-131-Based Transarterial Radioembolization for Liver Tumors.

Transarterial radioembolization (TARE) is a primary palliative treatment for advanced liver cancer. Nonetheless, its therapeutic efficacy is frequently hindered by resistance to tumor cell apoptosis induced by inter-radiotherapy. Induction of multiple cell death modalities provides a potential solution to this challenge. Ferroptosis, a distinct form of cell death from apoptosis, is dependent on the intracellular Fe2+-mediated Fenton reaction for the production of hydroxyl radicals (·OH) and is gaining recognition as a promising approach for cancer treatment. In this study, we synthesized a the

Synergistic multimodal therapy using bimetallic-doped graphene quantum dot nanozyme for the eradication of MRSA biofilms and enhancement of infected wound healing.

Bacterial biofilm-associated infections, particularly those caused by methicillin-resistant Staphylococcus aureus (MRSA), present a major challenge in clinical wound management due to their intrinsic resistance to conventional antibiotics and their capacity to severely impair tissue repair and wound healing. To address the limitations of current therapeutic approaches in combating biofilm-associated infections, we developed a multifunctional bimetal-doped nanozyme system, Cu,Fe,S,N-GQDs@Ru-NO, which synergistically integrates chemodynamic therapy (CDT), photothermal therapy (PTT), photodynamic

Comparative assessment of hepatoprotective properties of Artesunate and flavonoids from Artemisia annua on acetaminophen and carbon tetrachloride-induced cytotoxicity in primary mice hepatocytes.

BACKGROUND: Artesunate (ART) is a semi-synthetized molecule from Artemisinin, an active compound isolated from the medicinal plant Artemisia annua, widely used for the treatment of malaria. Previous studies reported that ART may exert a dual effect on the liver. Accordingly, this study investigated the potential protective action of ART against Acetaminophen (APAP) and Carbon tetrachloride (CCl4)-induced hepatotoxicity in primary mice hepatocytes, in comparison to that of flavonoid extracted from A. annua (FAA). In addition, the antioxidant properties of FAA were also assessed. METHODS: The a

Ozonation of lake water and wastewater: Identification of carbonous and nitrogenous carbonyl-containing oxidation byproducts by non-target screening.

Ozonation of drinking water and wastewater is accompanied by the formation of disinfection byproducts (DBPs) such as low molecular weight aldehydes and ketones from the reactions of ozone with dissolved organic matter (DOM). By applying a recently developed non-target workflow, 178 carbonous and nitrogenous carbonyl compounds were detected during bench-scale ozonation of two lake waters and three secondary wastewater effluent samples and full-scale ozonation of secondary treated wastewater effluent. An overlapping subset of carbonyl compounds (20%) was detected in all water types. Moreover, wa

Anti-inflammatory drugs degradation during LED-UV365 photolysis of free chlorine: roles of reactive oxidative species and formation of disinfection by-products.

Light-emitting diode (LED) is environmentally friendly with longer life compared with traditionally mercury lamps. This study investigated the non-steroidal anti-inflammatory drugs (NSAIDs)- phenacetin (PNT) and acetaminophen (ACT)- removal during LED-UV (365 nm) photolysis of free available chlorine (FAC). Degradation of PNT and ACT during LED-UV365/FAC treatment at pH 5.5-8.5 followed the pseudo-first order kinetics. The presence of hydroxyl radicals (·OH), reactive chlorine species (RCS), and ozone (O3, transformed from O (3P)) were screened by using scavengers of ethanol (EtOH), tert-Butan

Phenobarbital at low dose exerts hormesis in rat hepatocarcinogenesis by reducing oxidative DNA damage, altering cell proliferation, apoptosis and gene expression.

Our recent research indicated that phenobarbital (PB) may inhibit the development of N-diethylnitrosamine (DEN)-initiated pre-neoplastic lesions at low doses in a rat liver medium-term bioassay (Ito test), while high doses exhibit promoting activity. This raises the question of whether treatment with low doses of PB might reduce cancer risk. For clarification, male 6-week-old F344 rats were treated with PB at doses of 0, 2, 15 and 500 p.p.m. in the diet for 10 or 33 weeks after initiation of hepatocarcinogenesis with DEN. In a second, short-term experiment, animals were given PB at doses of 2,

Urinary thymidine glycol as a biomarker for oxidative stress after kidney transplantation.

Reactive oxygen species are generated during ischemia-reperfusion tissue injury. Oxidation of thymidine by hydroxyl radicals (HO*) causes formation of 5,6-dihydroxy-5,6-dihydrothymidine (thymidine glycol). Thymidine glycol excreted in urine can be used as a biomarker of oxidative DNA damage. The aim of this study was to investigate the oxidative DNA damage in patients showing immediate allograft function after kidney transplantation, and to find out whether this damage correlates with glomerular and tubular lesions. Time dependent changes in urinary excretion rates of thymidine glycol, but als

Ethnopharmacological potential and mechanistic study of Platycodon grandiflorum stems and leaves and Lonicera japonica stems and leaves against acute lung injury.

ETHNOPHARMACOLOGICAL RELEVANCE: Platycodon grandiflorum and Lonicera japonica are two plants that have been widely used in traditional ethnic medicine for both medicinal and edible purposes. They are commonly applied in the prevention and treatment of lung-heat-related and inflammatory diseases. However, the stems and leaves of these plants have historically been regarded as by-products, and their therapeutic potential has not been thoroughly explored. Recent studies have shown that the Platycodon grandiflorum stems and leaves, and Lonicera japonica stems and leaves are rich in bioactive compo

Ferrocene-Cystamine Conjugate Encapsulated Hyaluronic Acid Nanomicelles with Sequential pH/GSH Responsiveness for Synergistic Chemodynamic-Immunotherapy.

Metastasis and postoperative recurrence of malignant tumors remain critical obstacles that undermine clinical therapeutic efficacy. Traditional chemodynamic therapy (CDT) is plagued by three inherent, hard-to-resolve drawbacks: abundant intratumoral glutathione (GSH) quenches reactive oxygen species; robust systemic antitumor immunity fails to be activated; and unbound iron catalysts initiate off-target Fenton cascades within healthy tissues, inducing systemic oxidative injury and toxic side effects. To address these critical barriers, we rationally designed a bifunctional ferrocene-cystamine

Impact of hydrogen peroxide photolysis on viable bacterial count and composition of in vivo dental biofilm-an ex vivo study.

BACKGROUND: A bactericidal technique utilizing hydrogen peroxide (H₂O₂) photolysis, which generates hydroxyl radicals, was developed. Since it has demonstrated the potential to kill in vitro Streptococcus mutans biofilms, this technique has the possibility to be applied to dental caries. However, its efficacy on in vivo cariogenic dental biofilms remains unclear. This study aimed to evaluate the bactericidal effect of H₂O₂ photolysis on in vivo biofilms obtained from volunteers, focusing on its potential application in dental caries treatment. METHODS: Sixteen participants, prescreened for the

Long-range transport and transformation of volcanic sulfur emissions: A modeling study of the 2020 Nishinoshima event.

Volcanic emissions can undergo substantial chemical transformations during long-range atmospheric transport, potentially influencing air quality in downwind regions. This study quantitatively analyzes a sulfate (SO42-) pollution event in Jeju, South Korea, following the 2020 Nishinoshima volcanic eruption. The transport and transformation of volcanic emissions were simulated using the Community Multiscale Air Quality (CMAQ) model, which revealed three distinct stages in the transport of the volcanic plume: an initial northwestward transport away from the volcano, chemical transformation under

Metabolites augment oxidative stress to sensitize antibiotic-tolerant Staphylococcus aureus to fluoroquinolones.

If left unchecked, infections involving antibiotic-refractory bacteria are expected to cause millions of deaths per year in the coming decades. Beyond genetically resistant bacteria, persisters, which are genetically susceptible cells that survive antibiotic doses that kill the rest of the clonal population, can potentially contribute to treatment failure and infection relapse. Stationary-phase bacterial cultures are enriched with persisters, and it has been shown that stimulating these populations with exogenous nutrients can reduce persistence to different classes of antibiotics, including t

Tracking the reaction networks of acetaldehyde oxide and glyoxal oxide Criegee intermediates in the ozone-assisted oxidation reaction of crotonaldehyde.

The reaction of unsaturated compounds with ozone (O3) is recognized to lead to the formation of Criegee intermediates (CIs), which play a key role in controlling the atmospheric budget of hydroxyl radicals and secondary organic aerosols. The reaction network of two CIs with different functionality, i.e. acetaldehyde oxide (CH3CHOO) and glyoxal oxide (CHOCHOO) formed in the ozone-assisted oxidation reaction of crotanaldehyde (CA), is investigated over a temperature range between 390 K and 840 K in an atmospheric pressure jet-stirred reactor (JSR) at a residence time of 1.3 s, stoichiometry of 0

Synergy of DNA intercalation and catalytic activity of a copper complex towards improved polymerase inhibition and cancer cell cytotoxicity.

Improving the binding of metal complexes to DNA to boost cancer cell cytotoxicity requires fine tuning of their structural and chemical properties. Copper has been used as a metal center in compounds containing intercalating ligands due to its ability to catalytically generate reactive oxygen species (ROS), such as hydroxyl radicals (OH˙). We envision the synergy of DNA binding and ROS generation in proximity to target DNA as a powerful chemotherapy treatment. Here, we explore the use of [Cu(2CP-Bz-SMe)]2+ (2CP-Bz-SMe = 1,3-bis(1,10-phenanthrolin-2-yloxy)-N-(4-(methylthio)benzylidene)propan-2-
