Muscarinic Inhibition of Hypoglossal Motoneurons: Possible Implications for Upper Airway Muscle Hypotonia during REM Sleep.

Proper function of pharyngeal dilator muscles, including the genioglossus muscle of the tongue, is required to maintain upper airway patency. During sleep, the activity of these muscles is suppressed, and as a result individuals with obstructive sleep apnea experience repeated episodes of upper airway closure when they are asleep, in particular during rapid-eye-movement (REM) sleep. Blocking cholinergic transmission in the hypoglossal motor nucleus (MoXII) restores REM sleep genioglossus activity, highlighting the importance of cholinergic transmission in the inhibition of hypoglossal motor ne

[Various aspects of IL-1 biological activity. II. IL-1 beta in diseases and the Central Nervous System].

Precise understanding of the mechanisms of reciprocal relations between the nervous and the immune systems, has been the subject of numerous studies for the recent two decades. These mechanisms are significant, particularly at the stage of early response to bacterial, parasite, or viral infections. They are also essential from the medical point of view, as they may help in the development of the new methods of treatment of infectious diseases, and also may provide better methods to neutralize possible side effects of the therapy. As it is commonly understood, both forms of IL-1 (alpha and beta

Collicular dysfunction in attention deficit hyperactivity disorder.

Attention deficit hyperactivity disorder (ADHD) is a prevalent neurodevelopmental disorder characterised by (inter alia) an increase in distractibility. The current front-line pharmacotherapies for the treatment of ADHD, namely the psychostimulants methylphenidate and amphetamines, have clear abuse potential, hence there is a strong need to develop new drug treatments for this disorder. Central to this process is the identification of the pathophysiological changes which underlie ADHD. Given the heterogeneity of the disorder, multiple loci are probably involved, providing multiple potential th

Segregation of direction selective neurons and synaptic organization of inhibitory intranuclear connections in the medial terminal nucleus of the rat: an electrophysiological and immunoelectron microscopical study.

A combined electrophysiological and morphological investigation of the medial terminal nucleus (MTN) in the rat was undertaken, aimed at a better understanding of the relationship between structure and function in this nucleus. The locations of upward and downward direction selective units in the MTN were documented with extracellular electrophysiological recording. By means of tracer experiments, with Phaseolus vulgaris-leucoagglutinin, biocytin, and cholera toxin subunit B-horseradish peroxidase, the internal connections of the MTN, its retinal afferents, and the projection neurons to the in

Investigating the efficacy of osteopathic manipulative treatment for intractable hiccups: a pilot study.

CONTEXT: Intractable hiccups is a rare medical condition defined as daily hiccups continuing for more than 30 days and is recognized by the National Organization for Rare Disorders (NORD) and NIH Genetic and Rare Diseases (GARD) Information Center. Hiccups are spasmodic contractions of the respiratory muscles, triggered by activation of the hiccup reflex arc. Intractable hiccups can occur if this arc is chronically or aberrantly activated. The hiccup reflex arc has three components: (1) afferent pathways involving the phrenic, sympathetic, and vagal nerves; (2) central processing centers withi

The cerebellum and cerebellum-like structures of cartilaginous fishes.

The cerebellum is well developed in cartilaginous fishes, with the same cell types (barring basket cells) and organizational features found in other vertebrate groups, including mammals. In particular, the lattice-like organization of cerebellar cortex (with a molecular layer of parallel fibers, interneurons, spiny Purkinje cell dendrites, and climbing fibers) is a defining characteristic. In addition to the cerebellum, cartilaginous fishes have cerebellum-like structures in the dorsolateral wall of the hindbrain. These structures are adjacent to and, in part, contiguous with the cerebellum. T

ENG analysis of 150 cases of posterior fossa disease.

Analysis of the ENG records of 150 patients with well substantiated neurologic disease involving the structure of the posterior fossa revealed a high incidence of diminished optic fixation inhibition (DOFI) of caloric induced nystagmus. DOFI is defined as less than 50 percent decrease of speed of the slow phase when eyes are open and fixated. The whole group was subdivided into seven subgroups according to type and location of disease and the incidence of DOFI was found to vary significantly in the different subgroups. The highest incidence was found in the group of cerebellar mass lesions and

Tumors of the optic nerve.

This is a review of primary and secondary tumors of the optic nerve. The emphasis is an optic nerve gliomas and meningiomas. Optic nerve gliomas are slowly growing astrocytic neoplasms of the anterior visual pathways, the majority of which occur within the first two decades of life, with equal sex incidence in about 1 of 200,000 patients presenting with eye complaints. The incidence is greater in neurofibromatosis. The typical presentation is visual impairment in a verbal pre-school child with optic canal enlargement and optic atrophy. An intraorbital location of the tumor leads to axial, irre

Modulation of pain by affective touch: effects of childhood trauma, depression, chronic pain, and spinal segment.

Chronic pain is highly prevalent, and non-pharmacological tools are a crucial part of a multimodal approach to reducing burden of pain. Among these touch-based interventions such as massage have long been popular, yet their underlying pain-relieving mechanisms are poorly understood. Touch-based therapies often involve gentle stroking associated with the C-tactile afferent pathway and deep pressure. Although chronic pain patients often find touch less pleasant than individuals without pain, the effects of chronic pain on touch-pain modulation have not been directly studied. In the current study

[Deciphering the dynamic characteristics of non-neuronal cells in dorsal root ganglion of rat at different developmental stage based on single cell transcriptome data].

Dorsal root ganglia (DRG) is an essential part of the peripheral nervous system and the hub of the peripheral sensory afferent. The dynamic changes of neuronal cells and their gene expression during the development of dorsal root ganglion have been studied through single-cell RNAseq analysis, while the dynamic changes of non-neuronal cells have not been systematically studied. Using single cell RNA sequencing technology, we conducted a research on the non-neuronal cells in the dorsal root ganglia of rats at different developmental stage. In this study, primary cell suspension was obtained from

Molecular and cellular correlates of human nerve regeneration: ADCYAP1/PACAP enhance nerve outgrowth.

We only have a rudimentary understanding of the molecular and cellular determinants of nerve regeneration and neuropathic pain in humans. This cohort study uses the most common entrapment neuropathy (carpal tunnel syndrome) as a human model system to prospectively evaluate the cellular and molecular correlates of neural regeneration and its relationship with clinical recovery. In 60 patients undergoing carpal tunnel surgery [36 female, mean age 62.5 (standard deviation 12.2) years], we used quantitative sensory testing and nerve conduction studies to evaluate the function of large and small fi

Myostatin inhibition in combination with antisense oligonucleotide therapy improves outcomes in spinal muscular atrophy.

BACKGROUND: Spinal muscular atrophy (SMA) is caused by genetic defects in the survival motor neuron 1 (SMN1) gene that lead to SMN deficiency. Different SMN-restoring therapies substantially prolong survival and function in transgenic mice of SMA. However, these therapies do not entirely prevent muscle atrophy and restore function completely. To further improve the outcome, we explored the potential of a combinatorial therapy by modulating SMN production and muscle-enhancing approach as a novel therapeutic strategy for SMA. METHODS: The experiments were performed in a mouse model of severe SM

Adenosine and dopamine oppositely modulate a hyperpolarization-activated current Ih in chemosensory neurons of the rat carotid body in co-culture.

KEY POINTS: Adenosine and dopamine (DA) are neuromodulators in the carotid body (CB) chemoafferent pathway, but their mechanisms of action are incompletely understood. Using functional co-cultures of rat CB chemoreceptor (type I) cells and sensory petrosal neurons (PNs), we show that adenosine enhanced a hyperpolarization-activated cation current Ih in chemosensory PNs via A2a receptors, whereas DA had the opposite effect via D2 receptors. Adenosine caused a depolarizing shift in the Ih activation curve and increased firing frequency, whereas DA caused a hyperpolarizing shift in the curve and

STUDIES IN RESUSCITATION: IV. THE RETURN OF FUNCTION IN THE CENTRAL NERVOUS SYSTEM AFTER TEMPORARY CEREBRAL ANAEMIA.

The experiments on cerebral anaemia have enabled us to duplicate, by an entirely different method, many of the results obtained by anatomical division or removal of parts of the central nervous system. In some respects the method of anaemia permits of greater precision than the method of division or excision, and avoids, in great measure, the disturbances due to the wound and to the hemorrhage caused by the latter method. The method of general anaemia, as Couty pointed out long ago, leaves something to be desired in the matter of exact localization, but this objection may be met, in some degre

Astrocyte targeted SMN1 gene therapy and forskolin application improves astrocyte filopodia actin defects and motor neuron synaptic dysfunction in human SMA disease pathology.

BACKGROUND: Peri-synaptic astrocyte processes (PAPs) play a fundamental role in synapse formation and function. Central afferent synapse loss and astrocyte dysfunction greatly impede sensory-motor circuitry in spinal muscular atrophy (SMA) disease progression, however mechanisms underpinning tripartite synapse dysfunction remains to be fully elucidated. The aims of this study were to further define PAP and motor neuron synaptic defects in human SMA disease pathology and implement a therapeutic intervention strategy to improve motor neuron function. METHODS: We derived astrocyte monocultures a

A Case Report of Rapid Vision Improvement in a Patient With Optic Nerve Sheath Meningioma Treated Using ZAP-X Radiosurgery.

Optic nerve sheath meningioma (ONSM) is a rare benign tumor of the central nervous system. Although lesions in this region typically grow slowly, their proximity to the optic nerve directly impacts the frontal visual pathway and may lead to severe vision problems. The mainstream treatment for ONSM has shifted from traditional surgery to stereotactic radiosurgery (SRS). Among these, ZAP-X (ZAP Surgical Systems, Inc., San Carlos, California), the latest generation of SRS systems, offers a novel approach to clinical treatment of ONSM with its noncoplanar rotational focus technology, six-dimension

Critical appraisal of pathology transmission in the α-synuclein fibril model of Lewy body disorders.

Lewy body disorders are characterized by the emergence of α-synucleinopathy in many parts of the central and peripheral nervous systems, including in the telencephalon. Dense α-synuclein+ pathology appears in regio inferior of the hippocampus in both Parkinson's disease and dementia with Lewy bodies and may disturb cognitive function. The preformed α-synuclein fibril model of Parkinson's disease is growing in use, given its potential for seeding the self-propagating spread of α-synucleinopathy throughout the mammalian brain. Although it is often assumed that the spread occurs through neuroanat

Vestibular and visual climbing fiber signals evoked in the uvula-nodulus of the rabbit cerebellum by natural stimulation.

1. The cerebellar uvula-nodulus receives vestibular projections from primary and secondary vestibular afferents as well as vestibularly related climbing fibers. It also receives visually related information from climbing fiber pathways. In this experiment we investigated how this information is mapped onto the uvula-nodulus. We studied the specificity, dynamics, and topographic distribution of climbing fiber responses (CFRs), simple spike responses, and mossy fiber terminal responses evoked by vestibular and optokinetic stimulation in rabbits anesthetized with alpha-chloralose. 2. Vestibularly

Central sites involved in lateral hypothalamus conditioned neural responses to acoustic cues in the rat.

1. Unit activity in the lateral hypothalamus (LHA) of the rat was recorded during discrimination learning of cue-tone stimuli (CTS) predicting glucose (CTS1+) or intracranial self-stimulation (ICSS) (CTS2+) as positive reinforcement or electric shock (CTS1-) or tail pinch (CTS2-) as negative reinforcement. The same action, licking, was used as the behavioral response to all stimuli. Procaine hydrochloride, a local anesthetic, was microinjected into the ventral tegmental area (VTA) and the amygdala (AM). LHA neuron responses and licking were analyzed to investigate the afferent input pathway(s)
