Feeding Byproduct-Based Concentrates Instead of Human-Edible Feed Ingredients Increases Net Food Production and Improves Performance of High-Producing Holstein Cows.

The effect of feeding greater amounts of byproducts (BP) as a replacement for human-edible (HE) feed ingredients on nutrient intake, chewing activity, rumen fermentation, production performance, human-edible feed conversion efficiency (HeFCE) and net food production (NFP) of high-producing Holstein cows was evaluated. Twelve multiparous Holstein cows (BW = 673 ± 44, DIM = 112 ± 8 d; 48 ± 2.25 kg/d of milk; mean ± SE) were used in a replicated 3 × 3 Latin square design with 28-d periods. Each period consisted of 21 d of adaptation followed by 7 d of data collection. Treatments diets were (DM ba

Characterization of raft microdomains in bovine mammary tissue during lactation: How they are modulated by fatty acid treatments.

The objective of the current study was first to characterize lipid raft microdomains isolated as detergent-resistant membranes (DRM) from mammary gland tissue, and second to determine how dietary fatty acids (FA) such as conjugated linoleic acid (CLA), 19:1 cyclo, and long-chain n-3 polyunsaturated FA affect lipid raft markers of mammary cells, and to finally establish relationships between these markers and lactation performance in dairy cows. Eight Holstein cows were used in a replicated 4 × 4 Latin square design with periods of 28 d. For the first 14 d, cows received daily an abomasal infus

Risk of transmission of carbapenem-resistant Enterobacteriaceae and related "superbugs" during gastrointestinal endoscopy.

To evaluate the risk of transmission of carbapenem-resistant Enterobacteriaceae (CRE) and their related superbugs during gastrointestinal (GI) endoscopy. Reports of outbreaks linked to GI endoscopes contaminated with different types of infectious agents, including CRE and their related superbugs, were reviewed. Published during the past 30 years, both prior to and since CRE's emergence, these reports were obtained by searching the peer-reviewed medical literature (via the United States National Library of Medicine's "MEDLINE" database); the Food and Drug Administration's Manufacturer and User

Analytical aspects of thyroid antibodies estimation.

The search for antibodies that will reliably diagnose, predict and monitor the autoimmune thyroid diseases is a quest that is beset with difficulties. This review will describe the various antigens involved in autoimmune thyroid disease, the development of a humoral response to these antigens and some of the technical difficulties involved in trying to detect and then measure their concentrations. Multiple antigen configurations of thyroglobulin (TG) are produced when it is iodinated resulting in functionally active but immunologically distinct molecules. Thyroid peroxidase (TPO), originally d

Time-dependent transition probabilities and the assessment of seasonal effects on within-day variations in chewing behaviour of housed sheep.

State transitions in the chewing behaviour of six half-breed (Île de FrancexTexel) yearling female sheep (Ovis aries L.) were studied by using jaw movements recorded continuously over 5 days at the end of a number of experimental periods from 21 September 1992 to 4 April 1993. The sheep were housed in individual pens. Each of them received the same diet, that is, 250 g/day of concentrate mix (15.5% crude protein (CP), 36.5% neutral detergent fibre (NDF)) fed at 0900 h and natural grass hay (6.7% CP, 69.1% NDF) fed ad libitum at 0915 and 1600 h. Mineral salt blocks and water were continuously a

Matrix glycosaminoglycans in the growth phase of fibroblasts: more of the story in wound healing.

BACKGROUND: Understanding wound healing and ways to accelerate the healing process includes understanding the factors that influence the synthesis of granulation tissue, which fills the wound before epithelialization. An important phase of early wound healing involves secretion of glycosaminoglycans (GAGs) by fibroblasts which form a hydrophilic matrix suitable for remodeling during healing. The complexity of GAG structure and function in the extracellular matrix (ECM) remains poorly studied in wound healing. There is no established model for cutaneous wound healing due to variations in donor

Calreticulin-independent regulation of the platelet integrin alphaIIbbeta3 by the KVGFFKR alphaIIb-cytoplasmic motif.

The platelet integrin alphaIIbbeta3 alters conformation in response to platelet activation and ligand binding, although the molecular mechanisms involved are not known. We previously showed that a lipid modified peptide, corresponding to the membrane proximal 989KVGFFKR995 portion of the alphaIIb cytoplasmic tail, independently activates platelet alphaIIbbeta3. Calreticulin (CRT) is a potential integrin regulatory protein based on its interaction with the highly conserved alpha-integrin sequence KxGFFKR. We therefore examined the possible interaction of calreticulin and alphaIIbbeta3 in human

Tyrosine-to-cysteine modification of human alpha-synuclein enhances protein aggregation and cellular toxicity.

The deposition of alpha-synuclein and other cellular proteins in Lewy bodies in midbrain dopamine neurons is a pathological hallmark of Parkinson's disease. Nitrative and oxidative stress can induce alpha-synuclein protein aggregation, possibly initiated by the formation of stable cross-linking dimers. To determine whether enhanced dimer formation can accelerate protein aggregation and increase cellular toxicity, we have substituted cysteine for tyrosine at positions 39, 125, 133, and 136 in human wild-type (WT) alpha-synuclein, and in A53T and A30P mutant alpha-synuclein. To reduce the likeli

Characterization of rat HSA/CD24 protein bearing the B lineage-restricted epitope recognized by MAb HIS50.

HIS50 MAb recognizes a GPI-linked molecule on the surface of rat cells committed to the B lineage but is absent from cells of other hematopoietic lineages. Eukaryotic expression cloning of the cDNA has revealed that HIS50 recognizes a rat homologue of human CD24 and murine HSA. CD24/HSA exists in multiple glycoforms and plays an important role in hematopoietic and neural cell development. HIS50 is significant in that it is the only, presently available, anti-rat CD24/HSA MAb. Furthermore it is the only reported MAb with specificity restricted to B cell forms of CD24. Here we report Western blo

Differential extraction and enrichment of human sperm surface proteins in a proteome: identification of immunocontraceptive candidates.

The objective of this study was to discover previously unknown human sperm surface proteins that may be candidate contraceptive vaccinogens. To this end, methods of concentrating human sperm proteins for microsequencing by mass spectrometry were used, which increased the likelihood of identifying surface proteins. Vectorial labeling, differential extraction and two-dimensional (2-D) gel electrophoresis were employed to identify and isolate proteins accessible at the cell surface. Percoll harvested or swim-up sperm were either solubilized directly or solubilized after surface labeling with sulf

Enzymatic syntheses of T antigen-containing glycolipid mimicry using the transglycosylation activity of endo-alpha-N-acetylgalactosaminidase.

Thomsen-Friedenreich antigen (T antigen) disaccharide, beta-D-galactose-(1-->3)-alpha-N-acetyl-D-galactosamine (beta-D-Gal-(1-->3)-alpha-D-GalNAc), containing glycolipid mimicry was synthesized using the transglycosylation activity of endo-alpha-N-acetylgalactosaminidase from Bacillus sp. This enzyme could transfer the disaccharide from a p-nitrophenyl substrate to water-soluble 1-alkanols and other alcohols at a transfer ratio of 70% or more. Although the transfer ratios were lower for water-insoluble than water-soluble alcohols, they were shown to increase by adding sodium cholate to the rea

Impacts of grain source and sodium bicarbonate supplementation level on calf performance, nutrient digestion, and rumen fermentation.

The aim of this experiment was to investigate the main and interactive effects of sodium bicarbonate (SB) supplementation level (1% vs. 3% of dietary DM) and the grain source (barley vs. corn grain) in calf starter feed containing 5% chopped alfalfa hay. Our hypothesis was that a higher SB level would improve calf performance by buffering rumen pH and that the grain source of the calf starter feed would differentially influence rumen fermentation. Further, we hypothesized an interaction effect where a higher SB supplementation level would provide greater benefits in barley-based starter diets

Amended final report of the safety assessment of Drometrizole as used in cosmetics.

Drometrizole is used in cosmetics as an ultraviolet (UV) light absorber and stabilizer. In an earlier safety assessment, the available data were found insufficient to support the safety of this ingredient, but new data have been provided and assessed. In voluntary industry reports to the Food and Drug Administration, this ingredient is reported to be used in noncoloring hair care products, and in an industry use concentration survey, uses in nail care products at 0.07% were reported. Drometrizole has absorbance maxima at 243, 298, and 340 nm. Drometrizole is used widely as a UV absorber and st

Structural requirements for potent anti-human immunodeficiency virus (HIV) and sperm-immobilizing activities of cyclohexenyl thiourea and urea non-nucleoside inhibitors of HIV-1 reverse transcriptase.

The current pandemic of sexually transmitted human immunodeficiency virus (HIV)/acquired immunodeficiency syndrome (AIDS) has created an urgent need for a new type of microbicide, one that is both a spermicide and a virucide. In a systematic effort to identify a non-detergent-type antiviral spermicide, we have rationally designed and synthesized a series of cyclohexenyl thiourea (CHET) nonnucleoside inhibitors (NNIs) of HIV-1 reverse transcriptase (RT) with sperm-immobilizing activity (SIA). To gain further insight into the structural requirements for the optimal activity of these dual-functio

Reconstituted versus dry alfalfa hay in starter feed diets of Holstein dairy calves: Effects on feed intake, feeding and chewing behavior, feed preference, and health criteria.

We investigated the effect of reconstitution of alfalfa hay on feeding and chewing behavior, sorting activity, and health status using 20 neonate Holstein male calves (3 d of age; 40.3 ± 1.30 kg of body weight) that were assigned randomly to 2 treatments, a starter feed containing either 10% dry (AH) or reconstituted alfalfa hay (RAH), each consisting of 10 calves. Alfalfa hay was reconstituted with water 24 h before feeding to achieve a theoretical dry mater (DM) content of 20%. Both starter diets had the same ingredients and nutrient compositions but differed in their DM content (90.9 vs. 66
