Distinct systemic metabolic signatures in premenopausal women with lipedema revealed by composite indices.

BACKGROUND: Lipedema is a chronic adipose tissue disorder with disproportionate fat accumulation in the extremities and is often misdiagnosed as obesity. Although women with lipedema appear to be metabolically distinct from body mass index (BMI)-matched controls, their fasting metabolism remains insufficiently characterized. We therefore aimed to define the metabolic signature of lipedema using serum NMR metabolomics and anthropometric profiling. METHODS: We conducted a study with 24 premenopausal women with lipedema and 21 BMI-matched controls. Fasting serum samples were analyzed using NMR s

Targeting chaperone-mediated autophagy by nutritional interventions mitigates age-related cardiac fibrosis and diastolic dysfunction.

Cardiac fibrosis, a major pathological hallmark of aging that leads to heart failure, is characterized by excessive collagen deposition. Our knowledge of what sustains collagen synthesis in the aging heart is still very preliminary. Here, we uncover a central role for chaperone-mediated autophagy (CMA), a selective lysosomal degradation pathway, in this process. We demonstrate that CMA is suppressed in the aging heart, which promotes collagen overproduction in fibroblasts, whereas enhancing CMA activity ameliorates fibrosis and diastolic dysfunction. Mechanistically, we identify SHMT2 (serine

Impact of sublethal concentrations of monosodium methylarsonate (MSMA) on zebrafish: Insights from embryotoxicity assessment and NMR-based metabolomics.

Organic arsenicals such as monosodium methylarsonate (MSMA) remain widely used, yet their mechanistic toxicity in aquatic systems is poorly understood. Here, we integrated fish embryo toxicity (FET) assays with 1H NMR-based metabolomics to characterize the effects of acute MSMA exposure in zebrafish (Danio rerio) embryos. The 96-h LC50 was 74 mg L-1, and embryos were exposed to sublethal concentrations (10, 30, and 60 mg L-1). Exposure induced concentration-dependent embryotoxicity, with severe phenotypic alterations at 60 mg L-1, including reduced survival (p = 0.0316), delayed hatching (p =

Dapagliflozin ameliorates pulmonary arterial hypertension-induced right heart failure via ketone body metabolism and mitochondrial function enhancement.

BACKGROUND: Pulmonary arterial hypertension (PAH)-induced right ventricular (RV) failure increases reliance on ketones. The efficacy of SGLT2 inhibitors, known to modulate ketone metabolism, in this context remains unclear. METHODS: A PAH model was established in Sprague-Dawley rats via a single intraperitoneal injection of monocrotaline (MCT), followed by daily oral administration of dapagliflozin (DAPA). Pulmonary hemodynamics and RV function were evaluated using echocardiography and right heart catheterization. Myocardial injury, fibrosis, and mitochondrial ultrastructure were assessed via

Design, Synthesis, Molecular Modeling, and Biological Evaluation of Novel Pyrimidine Derivatives as Potential Calcium Channel Blockers.

Pyrimidines play an important role in modern medical fields. They have a wide spectrum of biological activities such as antimicrobial, anticancer, anti-allergic, anti-leishmanial, antioxidant agents and others. Moreover, in recent years, 3,4-dihydropyrimidin-2(1H)ones have attracted researchers to synthesize them via Biginelli reaction and evaluate their antihypertensive activities as bioisosters of Nifedipine, which is a famous calcium channel blocker. Our new target compounds were prepared through one-pot reaction of thiourea 1, ethyl acetoacetate 2 and/or 1H-indole-2-carbaldehyde, 2-chloroq

Diminished ketone interconversion, hepatic TCA cycle flux, and glucose production in D-β-hydroxybutyrate dehydrogenase hepatocyte-deficient mice.

OBJECTIVE: Throughout the last decade, interest has intensified in intermittent fasting, ketogenic diets, and exogenous ketone therapies as prospective health-promoting, therapeutic, and performance-enhancing agents. However, the regulatory roles of ketogenesis and ketone metabolism on liver homeostasis remain unclear. Therefore, we sought to develop a better understanding of the metabolic consequences of hepatic ketone body metabolism by focusing on the redox-dependent interconversion of acetoacetate (AcAc) and D-β-hydroxybutyrate (D-βOHB). METHODS: Using targeted and isotope tracing high-re

Barth's syndrome-like disorder: a new phenotype with a maternally inherited A3243G substitution of mitochondrial DNA (MELAS mutation).

An Argentine male child died at 4.5 years of age of a lethal mitochondrial disease associated with a MELAS mutation and a Barth syndrome-like presentation. The child had severe failure to thrive from the early months and for approximately two years thereafter. In addition, the patient had severely delayed gross motor milestones, marked muscle weakness, and dilated cardiomyopathy that progressed to congestive heart failure. He also had persistently elevated urinary levels of 3-methylglutaconic and 2-ethylhydracrylic acids and low blood levels of cholesterol. Detailed histopathologic evaluation

Cerebrospinal fluid and plasma metabolites in Parkinson's disease: a Mendelian randomization study.

Metabolites in cerebrospinal fluid (CSF) and plasma have been implicated in Parkinson's disease (PD), but their causal roles remain unclear. This study aims to investigate the causal relationships between specific CSF and plasma metabolites and PD risk, using Mendelian Randomization (MR) analysis. We utilized MR with inverse-variance weighting (IVW) as the primary analytical method, supplemented by four additional MR models to validate robustness. Data included 1,400 plasma metabolites from the Canadian Longitudinal Study on Aging, and 338 CSF metabolites from the Wisconsin Alzheimer's Disease

Causal association between cerebrospinal fluid metabolites and Parkinson's disease: A two-sample bidirectional mendelian randomization study.

OBJECTIVE: Observational studies suggest CSF metabolites may be linked to Parkinson's disease (PD) onset, but causality is uncertain. This study uses a two-sample bidirectional Mendelian randomization approach to investigate the causal relationship between CSF metabolites and PD. METHODS: Data on 338 CSF metabolites and PD-related traits were obtained from genome-wide association studies (GWAS). Causal relationships between CSF metabolites and PD were assessed using inverse variance-weighted (IVW), MR-Egger regression, weighted median method, simple mode, and weighted mode. Pre-analytical considerations in the simultaneous quantification of ketone bodies, lactate, pyruvate and TCA cycle intermediates.

BACKGROUND: Accurate quantification of small metabolites such as ketone bodies (KB: β-hydroxybutyrate (BHB), acetoacetate (AcAc)), pyruvate (Pyr), lactate (Lac) and tricarboxylic acid (TCA) cycle intermediates is essential for diagnostics, therapy monitoring and metabolic research. These metabolites serve as energy substrates and signaling molecules, with their interpretation often relying on physiologically meaningful ratios (Lac/Pyr, BHB/AcAc). However, their chemical instability and susceptibility to rapid post-collection metabolism pose significant pre-analytical challenges. METHOD: We de

SGLT2 Inhibition by Dapagliflozin Attenuates Diabetic Ketoacidosis in Mice with Type-1 Diabetes.

BACKGROUND: SGLT2 inhibitors increase plasma ketone concentrations. It has been suggested that insulinopenia, along with an increase in the counter-regulatory hormones epinephrine, corticosterone, glucagon and growth hormone, can induce ketoacidosis, especially in type-1 diabetes (T1DM). Dehydration precipitates SGLT2 inhibitor-induced ketoacidosis in type-2 diabetes. We studied the effects of dapagliflozin and water deprivation on the development of ketoacidosis and the associated signaling pathways in T1DM mice. METHODS: C57BL/6 mice were fed a high-fat diet. After 7 days, some mice receive
