Rational design and synthesis of a safe sNPF-mimetic peptide with cross-species insecticidal activity.

Short neuropeptide F (sNPF) is an insect-specific neuropeptide composed of 6-19 amino acids and features a conserved RLRFa motif, playing a pivotal role in regulating insect growth, development, feeding behavior, and energy homeostasis. Disruption of these processes can impair insect development and survival, highlighting sNPF as a promising scaffold for the development of environmentally safe insecticides in line with green chemistry principles. In the present study, ten novel sNPF analogs were rationally designed and synthesized by incorporating the conformationally rigid amino acid Aib into

Comparison of absorption and excretion of test compounds in sucking versus chewing pests.

A critical understanding of how pests interact with active ingredients is essential for the development of new insect control solutions to maintain crop quality and quantity by reducing insect damage. Absorption of insecticides into insect bodies of targeted pest species is the first critical step that confounds the efficacy of insecticides. This study investigated how different feeding behaviour of two pests, Myzus persicae and Spodoptera littoralis, affects the absorption, metabolism, and excretion (AME) of seven insecticidally inactive test compounds. A feeding contact assay for the chewing

Economic Injury Levels and Dynamic Action Thresholds for Diuraphis noxia (Hemiptera: Aphididae) in Australian Cereal Crops.

The Russian wheat aphid (Diuraphis noxia [Kurdjumov, Hemiptera: Aphididae], RWA) was first detected in Australia in 2016 and is threatening an annual cereal industry valued at nearly 10 billion AUD per annum. Considerable uncertainty surrounds the economic risk of D. noxia to Australian cereals, which limits cost-effective farm management decisions. Through a series of inoculated and non-inoculated field trials in 2018 and 2019 in south-eastern Australia, we generated a range of D. noxia pressure metrics under different growing conditions for barley, wheat, and durum wheat. Relative yield loss

Development and validation of a binomial sequential sampling plan for the greenbug (Homoptera: Aphididae) infesting winter wheat in the southern plains.

From 1997 to 1999, Schizaphis graminum (Rondani), intensity (number per tiller) was estimated on 115 occasions from hard red winter wheat fields located throughout the major wheat growing regions of Oklahoma. A total of 32 and 83 fields was sampled during the fall and spring, respectively. The parameters of linear regressions relating the mean number of greenbugs per tiller (m) and the proportion of infested tillers (PT) differed significantly between fall and spring infestations. The PT-m linear model provided a good fit for data on S. graminum for fall and spring infestations at tally thresh

EcR-JHAMT3 axis regulates reproductive plasticity in the predatory beetle Harmonia axyridis.

Ecdysone (20-hydroxyecdysone, 20E) is a master regulator of insect development, but its function in adult reproduction-particularly in predatory species-remains poorly understood. We employed Harmonia axyridis to explore 20E signaling in reproduction regulation through RNAi, transcriptomic analysis, and hormone rescue assays. The results demonstrated that both JH and 20E biosynthesis were activated in aphid-fed females, whereas injection of 1.0 μg 20E effectively promoted ovarian development in artificial diet-fed individuals. Expression of the 20E receptor EcR was significantly upregulated in

Nano-biointerface interlocking strategy based on bacteriomorphic silica nanocarriers for high-performance foliar delivery of imidacloprid.

Conventional pesticide application suffers from low utilization efficiency due to droplet rebound and rain wash-off. Inspired by gut-colonizing bacteria's adhesion, pilus-like spherical rough hollow silica (SRHS) and rod-shaped rough hollow silica (RRHS) were engineered to improve imidacloprid delivery via leaf-surface mechanical interlocking. Using imidacloprid as active ingredient and sucrose both as an adhesion enhancer, SRHS- and RRHS-based nanopesticides (SRHSIS/RRHSIS) were fabricated via vacuum impregnation. Under simulated rainfall conditions, retention rates were 4.8- and 5.2-fold gre

Spatio-temporal patterns of mortality in Pemphigus populicaulis and P. populitransversus on cottonwoods.

Galls of the aphid species Pemphigus populitransversus and P. populicaulis from a single cottonwood in Clinton, New York were censused for mortality approximately every ten days during the summer of 1978. P. populitransversus had significantly higher survival than P. populicaulis. Both species of aphid showed considerably elevated mortality later in the season. Mortality was not spatially random, but neighboring galls had a tendency to die at close to the same time. This tendency to spatially autocorrelated mortality varied from time to time, being more pronounced earlier in the season than la

A genomic perspective on nutrient provisioning by bacterial symbionts of insects.

Many animals show intimate interactions with bacterial symbionts that provision hosts with limiting nutrients. The best studied such association is that between aphids and Buchnera aphidicola, which produces essential amino acids that are rare in the phloem sap diet. Genomic studies of Buchnera have provided a new means for inferring metabolic capabilities of the symbionts and their likely contributions to hosts. Despite evolutionary reduction of genome size, involving loss of most ancestral genes, Buchnera retains capabilities for biosynthesis of all essential amino acids. In contrast, most g

Multiple phenotypes conferred by a single insect symbiont are independent.

Many microbial symbionts have multiple phenotypic consequences for their animal hosts. However, the ways in which different symbiont-mediated phenotypes combine to affect fitness are not well understood. We investigated whether there are correlations between different symbiont-mediated phenotypes. We used the symbiont Spiroplasma, a striking example of a bacterial symbiont conferring diverse phenotypes on insect hosts. We took 11 strains of Spiroplasma infecting pea aphids (Acyrthosiphon pisum) and assessed their ability to provide protection against the fungal pathogen Pandora neoaphidis and

Toxicity and Sublethal Effects of Flupyradifurone, a Novel Butenolide Insecticide, on the Development and Fecundity of Aphis gossypii (Hemiptera: Aphididae).

The cosmopolitan pest Aphis gossypii (Glover) causes considerable economic losses on various crops by its feeding damage and transmitting diseases around the world. Flupyradifurone is a novel butenolide pesticide; its toxicity on A. gossypii parent generation (F0) was estimated following treatment with LC25 concentration for 48 h. The adult longevity and fecundity of the F0 individuals treated by flupyradifurone showed no significant decrease in comparison with the control. Life table method was used to evaluate the sublethal effects on progeny population (F1). Results showed that the developm

Phytophagy of omnivorous predator Macrolophus pygmaeus affects performance of herbivores through induced plant defences.

Plants possess various inducible defences that result in synthesis of specialized metabolites in response to herbivory, which can interfere with the performance of herbivores of the same and other species. Much less is known of the effects of plant feeding by omnivores. We found that previous feeding of the omnivorous predator Macrolophus pygmaeus on sweet pepper plants significantly reduced reproduction of the two-spotted spider mite Tetranychus urticae and western flower thrips Frankliniella occidentalis on the same plants, also on leaves that had not been exposed to the omnivore. In contras

Biology and biotype determination of greenbug, Schizaphis graminum (Hemiptera: Aphididae), on seashore paspalum turfgrass (Paspalum vaginatum).

Greenbug, Schizaphis graminum (Rondani) (Hemiptera: Aphididae), was first discovered damaging seashore paspalum (Paspalum vaginatum Swartz) turfgrass in November 2003 at Belle Glade, FL. Inquiries to several golf courses with seashore paspalum turf across southern Florida indicated infestation was wide spread by April 2004. Damage symptoms progress from water soaked lesions surrounding feeding sites within 24 h to chlorosis and necrosis of leaf tips within 96 h. Problems caused by greenbug feeding were initially misdiagnosed as fertilizer, disease, other insects, or water management problems b

Aphidius ervi venom regulates Buchnera contribution to host nutritional suitability.

The association between the pea aphid, Acyrthosiphon pisum (Harris) (Homoptera: Aphididae), and the endophagous parasitoid wasp Aphidius ervi Haliday (Hymenoptera: Braconidae) offers a unique model system for studying the molecular mechanisms underlying the complex interactions between the parasitoid, its host and the associated primary symbiont. Here, we investigate in vivo the functional role of the most abundant component of A. ervi venom, Ae-γ-glutamyl transpeptidase (Ae-γ-GT), which is known to induce host castration. [Ecological effects of wheat-oilseed rape intercropping combined with methyl salicylate release on Sitobion avenae and its main natural enemies].

In order to explore the effects of wheat-oilseed rape intercropping in combining with methyl salicylate (MeSA) release on Sitobion avenae and its main natural enemies, a field experiment was conducted at the Tai'an Experimental Station of Shandong Agricultural University in East China from October 2008 to June 2010 to study the temporal dynamics of S. avenae and its main natural enemies as well as the ecological control effect on the aphid. In the plots of intercropping combined with MeSA release, the S. avenae apterae population reached a peak about 12 d in advance of the control, but the pea

Potential of Lecanicillium spp. for management of insects, nematodes and plant diseases.

Fungi in the genus Lecanicillium (formerly classified as the single species Verticillium lecanii) are important pathogens of insects and some have been developed as commercial biopesticides. Some isolates are also active against phytoparasitic nematodes or fungi. Lecanicillium spp. use both mechanical forces and hydrolytic enzymes to directly penetrate the insect integument and the cell wall of the fungal plant pathogen. In addition to mycoparasitism of the plant pathogen, the mode of action is linked to colonization of host plant tissues, triggering an induced systemic resistance. Recently it

Bi-Parental Mapping and Validation of Barley Yellow Dwarf Virus Resistance Locus in Barley Cultivar 'Lightning'.

Barley Yellow Dwarf Virus (BYDV), caused by multiple viruses within the Luteoviridae family and primarily transmitted by aphids, is among the most economically significant viral diseases impacting barley (Hordeum vulgare subsp. vulgare) production worldwide, resulting in stunted growth, leaf discoloration, and considerable reductions in grain yield. To validate previously reported BYDV resistance quantitative trait loci (QTL) in the barley cultivar 'Lightning', a doubled haploid (DH) population (n = 131) derived from a cross between 'Lightning' and the susceptible line 'DH141222' was evaluated
