Normal cytogenetic values for bone marrow based on studies of bone marrow transplant donors.

For individuals suspected of having hematologic neoplasms, interpretation of the clinical significance of sporadic cells with chromosome breakage, structural anomalies, aneuploidy, or polyploidy is often difficult. To help resolve this problem, we established normal cytogenetic values for bone marrow (BM) by investigating 219 BM transplant (BMT) donors using standard techniques for chromosome analysis. The donors ranged in age from 2 to 58 years and were studied for 7 years. The constitutional karyotype for two individuals was 47,XXY; one was mos45,X/46,XX, one was mos46,XX/47,XX, + mar, and 2

Genetic screening and evaluation for chromosomal abnormalities of infertile males in Jilin Province, China.

Chromosomal abnormality is the most common genetic cause of male infertility, particularly in cases of azoospermia, oligozoospermia, and recurrent spontaneous abortion. Chromosomal rearrangement may interrupt an important gene or exert position effects. The functionality of genes at specific breakpoints, perhaps with a specific role in spermatogenesis, may be altered by such rearrangements. Structural chromosome abnormalities are furthermore known to increase the risk of pregnancy loss. In this study, we aimed to assess chromosomal defects in infertile men from Jilin Province, China, by geneti

Three new patients with congenital unilateral facial nerve palsy due to chromosome 22q11 deletion.

We report three unrelated patients with congenital facial nerve palsy and chromosome 22q11 deletion, a condition hitherto poorly recognized. In the first case, facial palsy was associated with aortic coarctation, ductus arteriosus, and ostium secundum atrial septal defect. In the second case, facial palsy was associated with ostium secundum atrial septal defect, obstruction of the ureteropelvic junction, double ureteropelvic-calicial system, and distal metaphyseal widening of the forearm and leg bones. In both cases, facial palsy was the presenting feature. In the third case, an ostium secundu

Specific chromosomal defects associated with metastatic potential in K-1735 melanoma clones. Involvement of chromosomes 4 and 14.

In this study we sought to identify specific cytogenetic defects associated with the metastatic phenotypes in clones isolated from the parental K-1735 murine melanoma. All nonmetastatic clones (C-3, C-10, and C-19) exhibited trisomy of chromosomes 1, 3, 12, and 15. The only structural defect present in these clones was an interstitial deletion in a chromosome 4. In contrast, the highly metastatic clones (C-4, M-2, BB1, and X-21) exhibited trisomy of chromosomes 1, 3, 12, and 15, plus structural abnormalities of chromosomes 4 and 14, with the net result of a deletion in both. Parental K-1735 ce

MDM2 amplification in rod-shaped chromosomes provides clues to early stages of circularized gene amplification in liposarcoma.

Well-differentiated liposarcoma (WDLS) displays amplification of genes on chromosome 12 (Chr12) in supernumerary ring or giant marker chromosomes. These structures have been suggested to develop through chromothripsis, followed by circularization and breakage-fusion-bridge (BFB) cycles. To test this hypothesis, we compared WDLSs with Chr12 amplification in rod-shaped chromosomes with WDLSs with rings. Both types of amplicons share the same spectrum of structural variants (SVs), show higher SV frequencies in Chr12 than in co-amplified segments, have SVs that fuse the telomeric ends of co-amplif

Normal Fetus in Chromosomal Abnormality Carriers.

BACKGROUND: Both balanced translocation and inversion belong to chromosome structural aberration, in which balanced translocation only changes the corresponding position of the translocation fragment on the chromosome. Inversion refers to the phenomenon that two breaks occur on one chromosome at the same time, and the fragments are reversed 180° and then reconnected. Chromosome balanced translocation and inversion usually have normal phenotype and intelligence because they retain the total amount of original genes. However, due to the reduction of the probability of producing normal gametes, p

ASAR15, A cis-acting locus that controls chromosome-wide replication timing and stability of human chromosome 15.

DNA replication initiates at multiple sites along each mammalian chromosome at different times during each S phase, following a temporal replication program. We have used a Cre/loxP-based strategy to identify cis-acting elements that control this replication-timing program on individual human chromosomes. In this report, we show that rearrangements at a complex locus at chromosome 15q24.3 result in delayed replication and structural instability of human chromosome 15. Characterization of this locus identified long, RNA transcripts that are retained in the nucleus and form a "cloud" on one homo

A cytogenetic survey of an institution for the mentally retarded: I. Chromosome abnormalities.

A cytogenetic survey of 475 patients in an institution for the mentally retarded is reported. The chromosomes of all patients were studied using both a non-banding and a G-banding technique in order to estimate the relative efficiency of the two technique in detecting structural rearrangements of the chromosomes. A total of 57 patients was found to have a chromosome abnormality, including five with a balanced structural rearrangement. The contribution of chromosome aberrations to the etiology of mental retardation is discussed with special emphasis on the contribution of balanced structural re

Testis-Specific PDHA2 Is Required for Proper Meiotic Recombination and Chromosome Organisation During Spermatogenesis.

Proper segregation of homologous chromosomes during meiosis requires crossovers that are tightly regulated by the chromosome structure. PDHA2 is the testis-specific paralog of PDHA1, a core subunit of pyruvate dehydrogenase. However, its role during spermatogenesis is unclear. We show that PDHA2 knockout results in male infertility in mice, but meiotic DSBs in spermatocytes occur normally and are efficiently repaired. Detailed analysis reveals that mid/late recombination intermediates are moderately reduced, resulting in fewer crossovers and many chromosomes without a crossover. Furthermore, d

Cytogenetic studies in patients with reproductive failure.

BACKGROUND: Cytogenetic studies in patients with reproductive failure AIM: To investigate the contribution of chromosomal abnormalities in sub fertility and in couples with repeated abortions. METHODS: Hundred and 13 couples who had at least two or more spontaneous abortions and 65 women and 63 men with infertility were analyzed cytogenetically. RESULTS: Major chromosomal rearrangements were found in 8% and minor variants in 6% in the study population. Major chromosomal aberrations were judged to explain 4.9% of recurrent abortions and 13% of infertility. Chromosomal abnormalities in infert

Transcriptional control thrown for a loop.

The relationships among in vivo chromatin structures, chromosome organization and genome function must be understood in order to reveal the hidden regulatory information in our genomes. Rather than being stable architectural features, it appears that chromatin and chromosome conformations at all levels are highly dynamic, which is the key to their function. Studies in recent years have elucidated long-range interactions or folded chromatin conformations that play significant roles in gene regulation. Most recently, intrachromosomal associations and co-associations with shared nuclear transcrip

A CRISPR-engineered isogenic model of the 22q11.2 A-B syndromic deletion.

22q11.2 deletion syndrome, associated with congenital and neuropsychiatric anomalies, is the most common copy number variant (CNV)-associated syndrome. Patient-derived, induced pluripotent stem cell (iPS) models have provided insight into this condition. However, patient-derived iPS cells may harbor underlying genetic heterogeneity that can confound analysis. Furthermore, almost all available models reflect the commonly-found ~ 3 Mb "A-D" deletion at this locus. The ~ 1.5 Mb "A-B" deletion, a variant of the 22q11.2 deletion which may lead to different syndromic features, and is much more frequ

Involvement of der(12)t(12;21)(p13;q22) and as well as additional rearrangements of chromosome 12 homolog in ETV6/RUNX1-positive acute lymphoblastic leukemia.

The t(12;21) translocation is the most common translocation in childhood B-precursor ALL. Because G-banding cannot detect the cryptic t(12;21), the full characterization of additional aberrations involving chromosome 12 is often difficult. Spectral karyotyping (SKY) can distinguish a der(12)t(12;21) chromosome from a nonrearranged chromosome 12. We therefore applied SKY on an unselected series of 14 patients after G-banding analysis for the detection of additional chromosomal rearrangements and especially for aberrations involving chromosome 12. Chromosome 12 was the most frequently affected b

[Chromosome study on giant cell tumor of bone].

Chromosome study was made in 30 specimens of giant cell tumor of bone (GCT) and osteosarcoma (OS). In 15 tumors, karyotypes were satisfactorily made for chromosome analysis, including 13 GCTs, one OS and one cell-line of same. The karyotypes of all cells were recognized and described according to the international system for human cytogenetic nomenclature. Karyotype analysis showed that GCT differs from OS both in numerical chromosomal aberrations and in structural chromosomal aberrations. The number of chromosome of GCT concentrated on diploid or subdiploid, with the exception of some superdi

Double minutes and other chromosomal aberrations in malignant cell line of Shionogi carcinoma 115.

G-banded karyotypes and structural abnormalities of chromosomes are described in two clones of the mouse mammary carcinoma SC 115 cell line. The cells showed multiple numerical and structural abnormalities. Structural anomalies, such as double minutes, acentric fragments, gaps or breaks, radials, ring chromosomes, endoreduplications, dicentrics, and elongated chromosomes were observed in varying proportions. Unstable chromosomal aberrations, such as breaks, gaps, fragments, and radials, frequently appeared without any particular pretreatment. Increased numbers of double minutes per cell correl

Two-color hybridization with high complexity chromosome-specific probes and a degenerate alpha satellite probe DNA allows unambiguous discrimination between symmetrical and asymmetrical translocations.

This report describes a fluorescence in situ hybridization approach to chromosome staining that facilitates detection of structural aberrations and allows discrimination between dicentric chromosomes and symmetrically translocated chromosomes. In this approach, selected whole chromosomes are stained in one color by hybridization with composite probes whose elements have DNA sequence homology along the length of the target chromosomes. In addition, all chromosomes are counterstained with a DNA specific dye so that structural aberrations between target and non-target chromosomes are clearly visi

The complete sequence of a human Y chromosome.

The human Y chromosome has been notoriously difficult to sequence and assemble because of its complex repeat structure that includes long palindromes, tandem repeats and segmental duplications1-3. As a result, more than half of the Y chromosome is missing from the GRCh38 reference sequence and it remains the last human chromosome to be finished4,5. Here, the Telomere-to-Telomere (T2T) consortium presents the complete 62,460,029-base-pair sequence of a human Y chromosome from the HG002 genome (T2T-Y) that corrects multiple errors in GRCh38-Y and adds over 30 million base pairs of sequence to th
