Copper-ligand clusters dictate size of cyclized peptide formed during alkyne-azide cycloaddition on solid support.

Peptide and peptidomimetic cyclization by copper-catalyzed alkyne-azide cycloaddition (CuAAC) reaction have been used to mimic disulfide bonds, alpha helices, amide bonds, and for one-bead-one-compound (OBOC) library development. A limited number of solid-supported CuAAC cyclization methods resulting in monomeric cyclic peptide formation have been reported for specific peptide sequences, but there exists no general study on monocyclic peptide formation using CuAAC cyclization. Since several cyclic peptides identified from an OBOC CuAAC cyclized library has been shown to have important biologic

Cu/CuxOy NPs architectured COF: a recyclable catalyst for the synthesis of oxazolidinedione via atmospheric cyclizative CO2 utilization.

The present study describes the favourable construction of a crystalline covalent organic framework (COF) with exceptional surface area, tunable pore size and huge CO2 capture efficiency to facilitate a novel multicomponent cyclization by introducing CO2 into extremely reactive organic skeletons. In the presence of a catalytic Cu/CuxOy NP-loaded COF, several 2-bromo-3-alkylacrylic acids combined with several amine derivatives and CO2 (0.1 MPa) are converted to the desired oxazolidinediones in excellent yields (up to 96%) under alkali-free conditions and ambient temperature.

A diaminomaleonitrile-appended BODIPY chemosensor for the selective detection of Cu2+ via oxidative cyclization and imaging in SiHa cells and zebrafish.

A specific Cu2+ sensor, 2-amino-3-(BODIPYmethyleneamino)maleonitrile (BDM), was established by a simple dehydration between BODIPY and diaminomaleonitrile. Cu2+ could be recognized by BDM over other competing metal ions in acetonitrile with distinct fluorescence emission signal response. Upon the addition of Cu2+ to BDM in acetonitrile, the maximum absorption at approximately 530 nm on the longer wavelength side was quenched, and the emission at 530 nm was ignited simultaneously. The fluorescence intensity enhancement could reach a maximum of 204 times the intensity of the BDM blank solution.

Biocompatible and Rapid Cyclization of Peptides with 2,4-Difluoro-6-hydroxy-1,3,5-benzenetricarbonitrile for the Development of Cyclic Peptide Libraries.

We report a biocompatible and rapid reaction between cysteine thiols and 2,4-difluoro-6-hydroxy-1,3,5-benzenetricarbonitrile (DFB), which enables the efficient cyclization of peptides in neutral aqueous solutions. The reaction was further applied to cyclize peptides displayed on the phage surface without reducing phage infectivity, thus affording high-quality cyclic peptide libraries useful for screening of cyclic peptide ligands. Using the DFB-cyclic peptide library, we identified ligands that can distinguish the pro-survival protein Bcl-xl from its close relative Bcl-2. Therefore, this study

AUF1 p45 promotes West Nile virus replication by an RNA chaperone activity that supports cyclization of the viral genome.

UNLABELLED: A central aspect of current virology is to define the function of cellular proteins (host factors) that support the viral multiplication process. This study aimed at characterizing cellular proteins that assist the RNA replication process of the prevalent human pathogen West Nile virus (WNV). Using in vitro and cell-based approaches, we defined the p45 isoform of AU-rich element RNA-binding protein 1 (AUF1) as a host factor that enables efficient WNV replication. It was demonstrated that AUF1 p45 has an RNA chaperone activity, which aids the structural rearrangement and cyclization

Fine mapping of a cis-acting sequence element in yellow fever virus RNA that is required for RNA replication and cyclization.

We present fine mapping of a cis-acting nucleotide sequence found in the 5' region of yellow fever virus genomic RNA that is required for RNA replication. There is evidence that this sequence interacts with a complementary sequence in the 3' region of the genome to cyclize the RNA. Replicons were constructed that had various deletions in the 5' region encoding the capsid protein and were tested for their ability to replicate. We found that a sequence of 18 nucleotides (residues 146 to 163 of the yellow fever virus genome, which encode amino acids 9 to 14 of the capsid protein) is essential for

Cu(II)-Catalyzed Cascade Cyclization and Oxidative Annulation to Access Functionalized 1H-Furo[3,2-b]pyrazolo[4,3-e]pyridines.

A novel Cu(II)-catalyzed [3 + 3]/oxidative cyclization strategy is developed for the construction of highly functionalized 1H-furo[3,2-b]pyrazolo[4,3-e]pyridine derivatives (FPPDs). This transformation employs readily accessible 5-aminopyrazole and (E)-6-phenylhex-3-en-5-yn-2-one as coupling partners and proceeds through a sequential [3 + 3] cyclization/oxidative C-O bond-forming cascade. In 1,4-dioxane at 100 °C, using Cu(OAc)2 as the catalyst, Ag2CO3 and O2 as co-oxidants, and PhCO2H as an additive, the reaction proceeds via sequential 1,6-conjugate addition and aza-Michael addition to const

β-glucuronidase-triggered ortho-hydroxyl-mediated cyclization strategy for the efficient delivery of phenolic payloads in antibody-drug conjugates.

While the payload landscape of antibody-drug conjugates (ADCs) has diversified beyond amine-containing compounds, the application of potent cytotoxic agents possessing phenolic hydroxyl groups remains restricted. This is primarily due to the scarcity of linker technologies capable of balancing systemic stability with efficient release kinetics, as conventional strategies for phenolic conjugation often suffer from premature hydrolysis or sluggish activation. Herein, we report the design and evaluation of a novel self-immolative linker strategy based on an ortho-hydroxyaniline scaffold, which fa

A chemo-selective deprotection-cyclization strategy enables fluorescence imaging of hydroxylamine and reveals its pathological role in Parkinson's disease.

Hydroxylamine (HA), a reactive nitrogen species generated by neuronal nitric oxide synthase (nNOS), has been largely overlooked in neurodegenerative disorders. Herein, we report the first identification of HA overexpression in Parkinson's disease (PD) and elucidate its pathological role using chemo-selective fluorescence imaging combined with proteomic analysis. A general HA-responsive probe platform was developed by introducing a 1-(4,4-dimethyl-2,6-dioxacyclohexylidene)ethyl (Dde) unit as a highly specific HA-recognition motif. Among the resulting probes, DCI-HA-2 exhibited remarkable sensit

PIDA/BF3·OEt2-Mediated Cyclization of Bis(indoylmethyl)benzene Sulfonamides: Assembly of Di-indoloazepines, Spiro Carbolines, and Heterotriquinane Frameworks.

This study presents a metal-free, hypervalent iodine-(III)-promoted oxidative cyclization of N,N-bis(indol-2-ylmethyl)benzenesulfonamide to access isomeric di-indoloazepine under mild conditions. During the synthetic exploration of di-indoloazepine, we observed several unexpected transformations, including the rearrangement and oxidation of the diindoloazepine framework, that generate isomeric spiro β/γ-carbolines and unusual bi-indole-2-carboxaldehydes. In contrast, the oxidative cyclization of an isomeric N,N-bis(indol-3-ylmethyl)benzenesulfonamides yielded typical di-indoloazepines and high

Unconventional β-Hydride Elimination in Pd-Catalyzed Migratory Cyclization: A Theoretical Study of Outer-Sphere C(sp2)-H Activation.

The density functional theory (DFT) study reveals a novel mechanism for Pd-catalyzed C(sp2)-H activation through an unconventional outer-sphere deprotonation pathway. The canonical β-hydride elimination in alkenyl-Pd systems has been unequivocally demonstrated to be both kinetically and thermodynamically unfavorable. Our computational analysis reveals this energetic penalty stems from three key factors: (1) increased ring strain in the four-membered transition state, (2) stronger Pd-C bonding due to π-backdonation in alkenyl-Pd species, and (3) thermodynamic instability of the resulting Pd-H i

Regioselective Multicomponent Sulfonylation/Cyclization of Unactivated Alkenes to Access Sulfonylated Six- and Seven-Membered Cyclic Enamines via SO2 Insertion.

Radical cascade cyclization of alkenes involving the insertion of sulfur dioxide has proven to be a promising tool to access sulfonylnated heterocycle compounds, whereas cyclization of unactivated alkenes has been much less explored. Here, we developed a three-component cascade of unactive alkenes with sulfur dioxide and aryldiazonium tetrafluoroborates to generate sulfonylated tetrahydropyridines and azepines via the cleavage of alkenyl C-H bonds. Moreover, this protocol exhibited excellent chemical and regioselectivity and compatibility with broad functional groups.

Structural and Energetic Evidence Supports the Non-Covalent Phosphate Cyclization by the Class II Phospholipase D from Loxosceles intermedia.

Phospholipase D (PLD) enzymes from Loxosceles spider venom mediate envenomation pathology by cleaving phospholipid headgroups. We revisited the crystal structure of Loxosceles intermedia PLD (PDB: 3RLH) to evaluate two alternative mechanisms-covalent and non-covalent-for headgroup cleavage. The covalent mechanism involves a nucleophilic attack on the substrate's P atom by catalytic histidine, forming a phosphohistidine intermediate. It was originally suggested that this intermediate hydrolyzes, leading to linear phosphates. The non-covalent mechanism relies on the substrate's hydroxyl group pe

Pd(II)-catalyzed cyclization of 2-methyl aromatic ketones with maleimides through weak chelation assisted dual C-H activation.

A palladium-catalyzed dual C-H functionalization of substituted aromatic ketones and ester with maleimides leading to tricyclic heterocyclic molecules with good to excellent yield is reported. In this protocol, weak chelation of the carbonyl groups has been successfully utilized for the selective activation of the ortho-methyl C(sp3)-H bond instead of the ortho-C(sp2)-H bond in the presence of an external bidentate ligand Ac-Ile-OH. The reaction proceeds through two-fold C-H activation to generate a five-membered cyclic ring. The first C-H activation takes place selectively at the benzylic pos

Isocyanide-Based Multicomponent Reaction: Cascade α-Acyloxylation/Carboxamidation and [3 + 1+1] Cyclization of I(III)/S(VI)-Ylides.

A metal-free cascade of α-acyloxylation/carboxamidation of I(III)/S(VI)-ylides, carboxylic acids, and isonitriles via a Passerini-like multicomponent reaction is reported. Unexpectedly, [3 + 1+1] cyclization involving I(III)/S(VI)-ylides and two molecules of ethyl isocyanoacetate was observed. The strategy allows for the synthesis of unsymmetrical α,α-disubstituted ketones and functionalized pyrroles with up to 99% yield and wide substrate compatibility. Notably, the procedure has been extended to the late-stage modification of drugs and natural products, offering an elegant complement to the
