Trace-n-Seq combines retrograde fluorescent tracing with sorting and single-cell sequencing of innervating peripheral neurons.

Neuronal innervation plays a pivotal role in controlling tissue function during homeostasis and regeneration, as well as in pathological contexts such as inflammation, autoimmune disorders, fibrosis and cancer. A major obstacle in understanding neuron-tissue interactions at the molecular level is that neuronal cell bodies reside in peripheral ganglia often far outside the organ of interest and are therefore typically excluded in common single-cell datasets of tissues. Here we developed Trace-n-Seq to address this issue-a method that combines retrograde tracing with fluorescence-activated cell

Posttraumatic growth following culturally grounded intervention among tribal young women with dissociative disorders in Sikkim: an interpretative phenomenological analysis.

BACKGROUND: Trauma recovery among tribal adolescent girls and young women in Northeast India remains poorly understood, particularly for those with dissociative disorders. Existing frameworks rely on Western paradigms that do not account for the cultural, spiritual, and environmental realities of tribal communities. METHODS: Using Interpretative Phenomenological Analysis (IPA), this study examined the lived healing experiences of three young women from Sikkim who had completed a culturally grounded intervention comprising Pranayama, Ana Pana Sati meditation, and raga-based music therapy in 20

Prenatal adversity: a risk factor in borderline personality disorder?

BACKGROUND: Patients with borderline personality disorder (BPD) show a high prevalence of early adversity, such as childhood trauma. It has also been reported that prenatal adverse conditions, such as prenatal maternal stress, drug taking, tobacco smoking or medical complications, may be associated with an increased risk of mental disorders in the offspring. Prenatal adversity is investigated here for the first time as a potential risk factor in the diagnosis of BPD. Method A total of 100 patients with a DSM-IV diagnosis of BPD and 100 matched healthy controls underwent semi-structured intervi

A Hybrid 2D/3D Approach for Neural Differentiation Into Telencephalic Organoids and Efficient Modulation of FGF8 Signaling.

Human brain development relies on a finely tuned balance between the proliferation and differentiation of neural progenitor cells, followed by the migration, differentiation, and connectivity of post-mitotic neurons with region-specific identities. These processes are orchestrated by gradients of morphogens, such as FGF8. Disruption of this developmental balance can lead to brain malformations, which underlie a range of complex neurodevelopmental disorders, including epilepsy, autism, and intellectual disabilities. Studying the early stages of human brain development, whether under normal or p

Sport-related activities as a psychosocial strategy for mental health recovery following natural disasters: a narrative review.

Natural disasters-earthquakes, floods, tsunamis, wildfires, and storms-inflict profound and often enduring psychological trauma on affected populations. Survivors exhibit elevated rates of post-traumatic stress disorder (PTSD), depression, anxiety, peritraumatic dissociation, social dysfunction, and somatic complaints. Despite this burden, mental health services in disaster-affected regions remain under-resourced, particularly in low- and middle-income countries. This narrative review provides a conceptual synthesis of findings from 11 studies that examined the role of sport-related activities

Subtypes of borderline personality features in adolescence: Insights from cross-lagged panel network analysis.

Borderline personality disorder (BPD) is a complex and severe psychiatric condition characterized by emotional, self-image, behavior, and relational instability. While adult BPD heterogeneity has been extensively studied, the phenomenological borderline personality features (BPFs) in adolescence remain uninvestigated. This study aimed to explore the potentially dynamic causal relationships between BPFs in adolescence and identify the subtypes through cross-lagged panel network (CLPN) analysis. Two independent Chinese adolescent samples were followed over 18 months (N₁ = 1,056, M1age = 15.37, S

[123I]Iomazenil SPECT Detects a Reversible Lesion of the Left Medial Temporal Lobe in a Case of Global Autobiographical Amnesia.

Global autobiographical amnesia is a rare disorder that is characterized by a sudden loss of autobiographical memories covering many years of an individual's life. Generally, routine neuroimaging studies such as CT and MRI yield negative findings in individuals with global autobiographical amnesia. However, in recent case reports, functional analyses such as SPECT and fMRI have revealed changes in activity in various areas of the brain when compared with controls. Studies using iomazenil (IMZ) SPECT with individuals with global autobiographical amnesia have not been reported. We report the cas

Reading without words or target detection? A re-analysis and replication fMRI study of the Landolt paradigm.

The Landolt paradigm is a visual scanning task intended to evoke reading-like eye-movements in the absence of orthographic or lexical information, thus allowing the dissociation of (sub-) lexical vs. visual processing. To that end, all letters in real word sentences are exchanged for closed Landolt rings, with 0, 1, or 2 open Landolt rings as targets in each Landolt sentence. A preliminary fMRI block-design study (Hillen et al. in Front Hum Neurosci 7:1-14, 2013) demonstrated that the Landolt paradigm has a special neural signature, recruiting the right IPS and SPL as part of the endogenous at

Psychosocial interventions for conversion and dissociative disorders in adults.

BACKGROUND: Conversion and dissociative disorders are conditions where people experience unusual neurological symptoms or changes in awareness or identity. However, symptoms and clinical signs cannot be explained by a neurological disease or other medical condition. Instead, a psychological stressor or trauma is often present. The symptoms are real and can cause significant distress or problems with functioning in everyday life for the people experiencing them. OBJECTIVES: To assess the beneficial and harmful effects of psychosocial interventions of conversion and dissociative disorders in ad

Comparative Electrophysiological Analysis of Trigeminal and Dorsal Root Ganglion Neurons in Mice.

Cultured dissociated trigeminal ganglion (TG) and dorsal root ganglion (DRG) neurons are widely used to study peripheral sensory function, yet direct electrophysiological comparisons under identical experimental conditions remain limited. We compared intrinsic electrophysiological properties of mouse TG (mTG) and mouse DRG (mDRG) neurons using whole-cell patch-clamp electrophysiology from male mice. In addition to conventional comparisons of membrane properties, action potential waveform characteristics, and firing behavior, neurons were stratified by soma size and analyzed using principal com

Neurabin I haploinsufficiency disrupts ion channel regulation and synaptic maturation in human cortical neurons in neurodevelopmental disorders.

Heterozygous loss-of-function variants in Neurabin I (PPP1R9A), responsible for encoding a cytoskeletal scaffolding protein essential for synaptic plasticity, are recurrently associated with neurodevelopmental and neuropsychiatric disorders, yet their direct effects on human neuronal maturation remain unclear. Here, we establish the first comprehensive human mechanistic model of PPP1R9A haploinsufficiency using an isogenic CRISPR/Cas9-engineered iPSC system differentiated into cortical neurons to define dosage-dependent functional consequences. PPP1R9A+/- neurons exhibited pronounced hyperspin

Deep FLASH-seq profiling of purified canine sensory neurons uncovers species-specific signatures relevant to pain and itch.

Naturally occurring pain and itch disorders in the domestic dog represent an important and underexploited opportunity for translational sensory neuroscience. These conditions largely mirror human disease, highlighting the need for detailed comparative understanding of canine somatosensory neurobiology. Here, we present a single-cell transcriptomic characterisation of the canine dorsal root ganglion (DRG), providing molecular insights into sensory neuron diversity in a species of direct veterinary and biomedical relevance. We develop a novel mechanical dissociation and fluorescence-activated ce

Open Trial of Trauma-Focused Psychodynamic Psychotherapy for Posttraumatic Stress Disorder Among LGBTQ Individuals.

OBJECTIVE: Lesbian, gay, bisexual, transgender, and queer (LGBTQ) individuals report higher rates of exposure to traumatic events and posttraumatic stress disorder (PTSD) compared with heterosexual and cisgender individuals. No treatment outcomes research has focused on PTSD in the LGBTQ population. Trauma-focused psychodynamic psychotherapy (TFPP) is a brief, manualized, attachment- and affect-focused psychotherapy for PTSD. TFPP explicitly incorporates broad identity-related and societal factors into its conceptualization of trauma and its consequences, which may be especially helpful for LG

Pharmacological Effects on Ceroid Lipofuscin and Neuronal Structure in Cln3 ∆ex7/8 Mouse Brain Cultures.

Juvenile Batten disease (JBD) is an inherited disorder that is characterized by the development of blindness, seizures, and progressive motor, psychiatric, and cognitive impairment. A model of JBD expressing the predominant human mutation (Cln3 ∆ex7/8 ) has been explored. Dissociated brain cultures from Cln3 ∆ex7/8 knock-in mice were compared to wild type (WT) for effects on granules of ceroid lipofuscin (CL) and neuronal structure. Utilizing high content image analysis of CL granules identified with antibodies to mitochondrial ATP synthase subunit c or tripeptidyl peptidase-1, significant inc

Complex regional pain syndrome - Autoimmune or functional neurologic syndrome.

Complex regional pain syndrome (CRPS) purports to explain extremity pain accompanied by a variety of subjective complaints, including sensitivity to touch, fatigue, burning sensations, allodynia and signs consistent with voluntary immobilization, including skin changes, edema and trophic changes. By its own definition, CRPS pain is disproportionate to any inciting event or underlying pathology, which means that the syndrome describes non-anatomic and exaggerated symptoms. Although CRPS was coined in the early 1990s, physicians have described unexplained exaggerated pain for centuries. Before a

Sub-second and multi-second dopamine dynamics underlie variability in human time perception.

Aberrant interval timing in the millisecond-to-second range is a consistent feature of neurological and neuropsychiatric disorders involving striatal dysfunction, yet its neurochemical substrates in the human brain remain poorly characterised. Striatal dopamine has long been implicated in temporal processing, but evidence from animal models and human pharmacological studies has proved difficult to reconcile, in part because endogenous dopamine dynamics in humans have been inaccessible at relevant timescales. Patients with Parkinson's disease (N=5, 62-73 years; two women) completed a temporal b

Psychogenic amnesia: syndromes, outcome, and patterns of retrograde amnesia.

There are very few case series of patients with acute psychogenic memory loss (also known as dissociative/functional amnesia), and still fewer studies of outcome, or comparisons with neurological memory-disordered patients. Consequently, the literature on psychogenic amnesia is somewhat fragmented and offers little prognostic value for individual patients. In the present study, we reviewed the case records and neuropsychological findings in 53 psychogenic amnesia cases (ratio of 3:1, males:females), in comparison with 21 consecutively recruited neurological memory-disordered patients and 14 he

[Contribution of animal models in the understanding of AIDS encephalopathy].

The neuropathology associated with HIV (Human Immunodeficiency Virus) infection is one of the major complications of this disease. The virological and cellular mechanisms by which HIV infection induces motor and cognitive disorders remain unknown. This lack of understanding of the pathophysiology is partly due to the difficulty of experimental analysis in man because only post-mortem samples from terminal phases of the disease and cerebrospinal fluid samples are available. Two animal models, very closely resembling human HIV infection, are available: the cat model infected by FIV (Feline Immun
