N6-methyladenosine-mediated feedback regulation of abscisic acid perception via phase-separated ECT8 condensates in Arabidopsis.

N6-methyladenosine (m6A) is the most abundant internal modification in eukaryotic mRNAs, yet how plants recognize this chemical modification to swiftly adjust developmental plasticity under environmental stresses remains unclear. Here we show that m6A mRNA modification and its reader protein EVOLUTIONARILY CONSERVED C-TERMINAL REGION 8 (ECT8) act together as a key checkpoint for negative feedback regulation of abscisic acid (ABA) signalling by sequestering the m6A-modified ABA receptor gene PYRABACTIN RESISTANCE 1-LIKE 7 (PYL7) via phase-separated ECT8 condensates in stress granules in respons

Turning Up the Heat: More Persistent Precipitation Regimes Weaken the Micro-Climate Buffering Capacity of Forage Grasses During a Hot Summer.

Developing climate-proof forage grasslands does not only require developing plant communities that are soil drought resistant, but also adept at buffering elevated atmospheric temperatures to minimize heat stress for plant and soil. Previous studies indicate that the emerging trend towards rainfall regimes with longer dry and wet spells negatively affects forage grass performance (i.e., greater physiological plant stress and yield loss) in Western Europe. We conducted a 120-day open-air experiment testing whether a hot summer (+3°C for the first 60 days) exacerbates the negative effects of inc

Alleviation of osmotic stress effects by exogenous application of salicylic or abscisic acid on wheat seedlings.

The aim of the study was to assess the role of salicylic acid (SA) and abscisic acid (ABA) in osmotic stress tolerance of wheat seedlings. This was accomplished by determining the impact of the acids applied exogenously on seedlings grown under osmotic stress in hydroponics. The investigation was unique in its comprehensiveness, examining changes under osmotic stress and other conditions, and testing a number of parameters simultaneously. In both drought susceptible (SQ1) and drought resistant (CS) wheat cultivars, significant physiological and biochemical changes were observed upon the additi

Functional validation of EXLB1 in leaf and stomatal development in tobacco.

Leaf morphological traits are the key determinants of tobacco yield and stress responses, and cell wall synthesis and metabolism play an important role in leaf development. However, the underlying regulatory mechanism of cell wall on leaf morphology remains largely unknown in tobacco. Here, we compared two pairs of tobacco cultivars (DMC vs. DX3 and DMC vs. WM3) with distinct leaf traits, and identified a cell wall-related expansin gene, NtEXLB1, based on transcriptome data analysis. The homolog NbEXLB1 from N. benthamiana is highly expressed in the mature leaf, and the NbEXLB1 protein is loca

The Trihelix transcription factor GT2-like 1 (GTL1) promotes salicylic acid metabolism, and regulates bacterial-triggered immunity.

The Trihelix Transcription factor GT2-like 1 (GTL1) was previously shown to be a key regulator of ploidy-dependent trichome growth and drought tolerance. Here, we report that GTL1 plays an important role in coordinating plant immunity. We show that gtl1 mutants are compromised in the regulation of basal immunity, microbial pattern-triggered immunity (PTI) and effector-triggered RIN4-mediated immunity. Transcriptome analysis revealed that GTL1 positively regulates defense genes and inhibits factors that mediate growth and development. By performing hormonal measurements and chromatin-immunoprec

Secreted proteins of tobacco cultured BY2 cells: identification of a new member of pathogenesis-related proteins.

Cultured cells of tobacco BY2 secrete more than 100 proteins into culture medium. Six major proteins were purified, and partial protein sequences were determined. Five of them were found to be similar to an ascorbic acid oxidase, three peroxidase isozymes and a beta-1,3-exoglucanase, respectively. A cDNA clone encoding the remaining polypeptide, whose amino acid sequence showed no similarity with earlier reported proteins, was isolated. It encoded a putative 27 kDa protein of 242 amino acids with resemblance to WCI-5, a wheat protein induced by benzo(1,2,3)thiadiazole-7-carbothioic acid S-meth

E3 Ubiquitin Ligase PdSINA2D in Populus deltoides Positively Regulates ABA-Dependent Stress Tolerance through Ubiquitination of FREE1 in Poplar.

Populus deltoides "Shalinyang" (PdS) displays reduced tolerance to abiotic stresses. SINA (seven in absentia) proteins, a subtype of E3 ubiquitin ligases, play crucial roles in regulating plant growth and responses to abiotic stresses. However, the characteristics and potential functions of SINA proteins in PdS remain unclear. We identified 12 SINA genes via comprehensive bioinformatic characterization. Overexpression of PdSINA2D improved drought and saline-alkali tolerance in PdS, whereas gene silencing exacerbated plant sensitivity to these stresses. We also found that the abscisic acid (ABA

Impact of environmental DDT concentrations on gill adaptation to increased salinity in the tilapia Sarotherodon melanotheron.

Estuaries of tropical developing countries suffering from severe droughts induced by climate change are habitats to fish, which face drastic salinity variations and the contact with pollutants. The Western Africa tilapia Sarotherodon melanotheron is highly resistant to hypersalinity, but the effect of human-released xenobiotics on its adaptation is barely known. Controlled experiments were conducted to observe S. melanotheron gill adaptation to abrupt salinity variations in the presence of waterborne DDT, at concentrations detected in their natural habitat. The gills appeared as an important s

Ecophysiological Responses of the Lesser Mealworm Alphitobius diaperinus Exposed to Desiccating Conditions.

In order to improve predictions of the impacts of climate change on insects, this study aimed to uncover how exposure to dry conditions can affect the biology of the invasive pest beetle Alphitobius diaperinus in terms of longevity, activity, water content, metabolic profiles, and fecundity. We measured desiccation resistance in adults of A. diaperinus by recording the time the beetles could survive desiccation stress. We found that the species was highly desiccation resistant, with about 50% of the insects exposed to desiccation being able to survive for 30 days, and some individuals even sur

[The use of L-phenylalanine ammonia lyase inhibitors in plant ecophysiological studies].

Phenylalanine ammonia lyase (PAL) is a key enzyme controlling the biosynthesis of phenolic compounds in plants. PAL catalyzes ammonia elimination from L-phenylalanine in a reaction that yields cinnamic acid, a precursor of a large group of phenylpropanoid compounds. Phenylpropanoids and their derivatives play an important role in regulating plant resistance mechanisms under environmental stresses. By reducing the level of phenolic compounds, PAL inhibitors can induce changes in plant metabolism. This paper presents the current state of knowledge on the use of PAL inhibitors in plant biology, a

H2S-modulated alternative splicing of the protein phosphatase gene HAB2 drives divergent stomatal dynamics in Arabidopsis.

Abscisic acid (ABA) serves as a critical regulator of plant adaptive responses to environmental stresses. Currently, the function of HOMOLOGY TO ABI2 (HAB2) of Arabidopsis, a member of the ABA signaling component the protein phosphatase 2C family (PP2C), remains uncharacterized. Hydrogen sulfide (H2S) plays a vital role throughout the plant life cycle. We found that exogenous H2S induced stomatal closure in Arabidopsis hab2 mutants. Detailed characterization then showed that HAB2 underwent alternative splicing (AS), generating three transcripts that we designated as HAB2.1-2.3, with HAB2.2 tar

Role of plant peroxisomal catalase in temperature and drought stress: Physio-biochemical and molecular perspectives.

Climate change-driven abiotic stresses, mainly drought and extreme temperatures, significantly hinder global crop productivity. These stresses lead to excessive accumulation of reactive oxygen species (ROS), which trigger oxidative damage and impair physiological and molecular mechanisms. Plants counter oxidative stress through ROS-metabolizing enzymatic, e.g., catalase (CAT), superoxide dismutase, ascorbate peroxidase, among others and non-enzymatic (e.g., glutathione, ascorbic acid, etc.) antioxidant systems. Among these, peroxisomal CAT is one of the key enzymes for ROS detoxification; stil

Transcription Factor McHB7 Improves Ice Plant Drought Tolerance through ABA Signaling Pathway.

As global climate change continues, drought episodes have become increasingly frequent. Studying plant stress tolerance is urgently needed to ensure food security. The common ice plant is one of the model halophyte plants for plant stress biology research. This study aimed to investigate the functions of a newly discovered transcription factor, Homeobox 7 (HB7), from the ice plant in response to drought stress. An efficient Agrobacterium-mediated transformation method was established in the ice plant, where ectopic McHB7 expression may be sustained for four weeks. The McHB7 overexpression (OE)

Recurrent heatwaves reduce production but reveal species mixture‑mediated resistance and recovery in grasslands.

Current and future heatwaves threaten temperate grasslands as they impair plant temperature regulation, photosynthesis, and eventually lead to production losses. Species mixture could mitigate the negative effects of heatwaves as documented for drought. Our main objective was to test the effect of species mixture on grassland responses to heatwaves by comparing mixtures with monocultures. We conducted a greenhouse experiment to simulate four heatwave treatments: i) control ii) two mild heatwaves, iii) a severe heatwave, and iv) recurrent scenario combining the mild and severe heatwaves. We ana

Persistence of Resident and Transplanted Genotypes of the Undomesticated Yeast Saccharomyces paradoxus in Forest Soil.

One might expect yeasts in soil to be highly dispersed via water or insects, forming ephemeral, genetically heterogeneous populations subject to competition and environmental stochasticity. Here, we report persistence of genotypes of the yeast Saccharomyces paradoxus in space and time. Within 1 km2 in a mixed hardwood forest on scales from centimeters to tens of meters, we detected persistence over 3 years of native genotypes, identified by single nucleotide polymorphisms (SNPs) genome-wide, of the wild yeast Saccharomyces paradoxus growing around Quercus rubra and Quercus alba Yeasts were rec

Isolation and functional validation of the CmLOX08 promoter associated with signalling molecule and abiotic stress responses in oriental melon, Cucumis melo var. makuwa Makino.

BACKGROUND: Lipoxygenases (LOXs) play significant roles in abiotic stress responses, and identification of LOX gene promoter function can make an important contribution to elucidating resistance mechanisms. Here, we cloned the CmLOX08 promoter of melon (Cucumis melo) and identified the main promoter regions regulating transcription in response to signalling molecules and abiotic stresses. RESULTS: The 2054-bp promoter region of CmLOX08 from melon leaves was cloned, and bioinformatic analysis revealed that it harbours numerous cis-regulatory elements associated with signalling molecules and ab

Metabolic engineering of Saccharomyces cerevisiae to enhance resistance toward microbial toxicity in agave extracts.

Agave, which thrives in arid regions of North Americas, is a promising alternative feedstock for biofuel production due to its high sugar content and drought tolerance. A major challenge to using agave as a feedstock for bioprocessing is its toxicity. Saponins, natively produced glycosides, disrupt cell membranes and are highly toxic to Saccharomyces cerevisiae even at low concentrations. To enable agave as a feedstock, we combined rational engineering with adaptive laboratory evolution (ALE) to create new S. cerevisiae strains with high tolerance to agave extracts. Heterologous overexpression

The exodermis: a variable apoplastic barrier.

The exodermis (hypodermis with Casparian bands) of plant roots represents a barrier of variable resistance to the radial flow of both water and solutes and may contribute substantially to the overall resistance. The variability is a result largely of changes in structure and anatomy of developing roots. The extent and rate at which apoplastic exodermal barriers (Casparian bands and suberin lamellae) are laid down in radial transverse and tangential walls depends on the response to conditions in a given habitat such as drought, anoxia, salinity, heavy metal or nutrient stresses. As Casparian ba

Specific Rhizobacteria Responsible in the Rhizosheath System of Kengyilia hirsuta.

The rhizosheath is a critical interface supporting the exchange of resources between plants and their associated environment of soil. Favorable microenvironment of rhizosphere soil provides the rhizosheath formed and then promotes desert plant survival. However, it remains unclear how rhizosheath benefits the colonization of pioneer plants in alpine desert under changing environment. In this study, we investigated the effect of different soil moisture and sterilization treatments (three moisture levels and unsterilized or sterilized soil) on rhizosheath forming process of Kengyilia hirsuta (K.
