Assessment of the cardiovascular adverse effects of drug-drug interactions through a combined analysis of spontaneous reports and predicted drug-target interactions.

Adverse drug effects (ADEs) are one of the leading causes of death in developed countries and are the main reason for drug recalls from the market, whereas the ADEs that are associated with action on the cardiovascular system are the most dangerous and widespread. The treatment of human diseases often requires the intake of several drugs, which can lead to undesirable drug-drug interactions (DDIs), thus causing an increase in the frequency and severity of ADEs. An evaluation of DDI-induced ADEs is a nontrivial task and requires numerous experimental and clinical studies. Therefore, we develope

Extravasation of vesicant drugs in combined regimens: A case with a fatal outcome.

IntroductionExtravasation of vesicant chemotherapy is an oncological emergency that may result in severe tissue injury, functional impairment, and long-term morbidity. Management becomes particularly challenging in combination regimens containing agents with different vesicant properties and opposing treatment recommendations.Case ReportWe report a fatal case associated with administration of a combination chemotherapy regimen through difficult peripheral venous access. Mild symptoms during infusion of a non-vesicant agent led to suspected extravasation; consequently, antidotes were not admini

Optimal Designs for Identifying Effective Doses in Drug Combination Studies.

We consider the optimal design problem for identifying effective dose combinations within drug combination studies where the effect of the combination of two drugs is investigated. Drug combination studies are becoming increasingly important as they investigate potential interaction effects rather than the individual impacts of the drugs. In this situation, identifying effective dose combinations that yield a prespecified effect is of special interest. If nonlinear surface models are used to describe the dose combination-response relationship, these effective dose combinations, also called iso

Drug combination treatment as a strategy for inhibiting human adenovirus replication.

Human adenoviruses (HAdVs) are ubiquitous human pathogens that infect the respiratory, ocular, and gastrointestinal tissues. Despite the severity of these infections in immunocompromised patients, there are no clinically approved antiviral medications to treat HAdV infections. Over the past decade, many compounds have been found to interfere with different parts of the HAdV replication cycle. One critical barrier to developing a successful HAdV therapy arises if the drug concentration required for antiviral efficacy is clinically unachievable or too toxic for patient use. This problem can be d

Drug combination assays using Caenorhabditis elegans as a model system.

The C. elegans drug combination assay evaluates the effects of drug combinations in the nematode Caenorhabditis elegans, serving as a valuable tool to assess the efficacy of pharmaceutical agents and natural compounds. Using C. elegans as a model organism, this method allows for the efficient screening of the combined effects of different drugs and evaluation of synergistic effects in drug combinations, which reduces the risk of developing drug resistance. Combination therapy, involving commercial drugs, new agents, or natural products, broadens treatment effectiveness by targeting multiple pa

Combining Fluorescence and Magnetic Resonance Imaging in Drug Discovery-A Review.

Drug discovery is a complex and multi-stage process that requires advanced analytical technologies capable of accelerating preclinical evaluation and improving the precision of therapeutic design. The combination of fluorescence and magnetic resonance imaging (MRI) within multimodal imaging plays an increasingly important role in modern pharmacokinetics, integrating the high molecular sensitivity of fluorescence with the non-invasive anatomical visualization offered by MRI. Fluorescence enables real-time monitoring of cellular processes, including drug-target interactions and molecular dynamic

Combining drugs that bypass p53 to treat TP53-mutated leukemias.

Acute myeloid leukemias (AML) containing TP53 (p53) mutations are routinely treated with decitabine or 5-azacytidine, which deplete DNA methyltransferase 1 (DNMT1; ie, hypomethylating agents [HMA]). Unfortunately, resistance/relapse, characterized by preserved DNMT1, is rapid. HMA are pyrimidine analogs, and to deplete DNMT1, must compete with endogenous pyrimidines. These were substantially increased in HMA-resistant vs parental AML cells, together with upregulation of CAD (carbamoyl-phosphate-synthetase-2/aspartate transcarbamylase/dihydroorotase) that rate limits de novo pyrimidine synthesi

Chinese expert consensus on the combined use of antiviral drugs for influenza.

Influenza is an acute respiratory infectious disease caused by influenza viruses, and it poses a serious threat to global public health. High-risk groups include the elderly, infants and young children, pregnant women, and patients with chronic underlying diseases. These groups are prone to developing severe illness after infection, which can lead to serious complications and even death. Early antiviral treatment is key to reducing the rate of severe illness and death. Currently, authoritative guidelines at home and abroad recommend early, single-agent antiviral therapy as the standard regimen

[Combination drug therapy with individualized bronchoscopic intervention for pulmonary mucormycosis].

This article reported a 46-year-old male patient with pulmonary mucormycosis and a history of poorly controlled diabetes. Chest imaging revealed rapidly progressive patchy opacities and multiple cavities in both lungs. Bronchoscopy showed mucor plug obstructing multiple segmental and subsegmental bronchi. The patient underwent sequential treatment, including local nebulization, oral/intravenous antifungals, and bronchoscopic removal of mucor plugs combined with intrabronchial instillation of amphotericin B deoxycholate. However, this yielded no significant improvement. During the procedure, ma

Electrospun patches to deliver combination drug therapy for fungal infections.

Fungal infections, though affecting healthcare globally, receive insufficient attention in clinical and academic settings. Invasive fungal infections, particularly caused by combat wounds, have been identified as a critical threat by the US Department of Defense. Monotherapy with traditional antifungals is often insufficient, and so combination therapies are explored to enhance treatment efficacy. However, systemic combination treatments can result in severe adverse effects, suggesting the need for localised delivery systems, such as drug-loaded electrospun patches, to administer antifungals d

Combining nanotechnology with monoclonal antibody drugs for rheumatoid arthritis treatments.

Rheumatoid arthritis (RA) is a systemic immune disease characterized by synovial inflammation. Patients with RA commonly experience significant damage to their hand and foot joints, which can lead to joint deformities and even disability. Traditional treatments have several clinical drawbacks, including unclear pharmacological mechanisms and serious side effects. However, the emergence of antibody drugs offers a promising approach to overcome these limitations by specifically targeting interleukin-1 (IL-1), interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), and other cytokines that are

An effective nano drug delivery and combination therapy for the treatment of Tuberculosis.

Drug resistance in tuberculosis is exacerbating the threat this disease is posing to human beings. Antibiotics that were once effective against the causative agent, Mycobacterium tuberculosis (Mtb), are now no longer usable against multi- and extensively drug-resistant strains of this pathogen. To address this issue, new drug combinations and novel methods for targeted drug delivery could be of considerable value. In addition, studies have shown that the use of the antidepressant drug fluoxetine, a serotonin reuptake inhibitor, can be useful in the treatment of infectious diseases, including b

Efficacy of moxifloxacin combined with levofloxacin in the treatment of drug-resistant tuberculosis.

Drug-resistant tuberculosis is a clinically common respiratory-borne chronic infectious disease. Fluoroquinolone drugs can inhibit the replication and transcription of bacterial DNA and cause bacteria to die, and the antibacterial spectrum of such drugs is broad, especially for Mycobacterium tuberculosis-induced diseases. This article observes and compares the clinical efficacy of levofloxacin and moxifloxacin in the treatment of multidrug-resistant tuberculosis (MDR-TB). At the end of the course of treatment, the treatment success rate was 76.4% in the control group and 68.2% in the treatment

Combination drug therapy via nanocarriers against infectious diseases.

Current drug therapy against infections is threatening to become obsolete due to the poor physical, chemical, biological and pharmacokinetic properties of drugs, followed by high risk of acquiring resistance. Taking into account the significant benefits of nanotechnology, nano-based delivery of anti-infectious agents is emerging as a potential approach to combat several lethal infections. Co-delivery of multiple anti-infectious agents in a single nano-based system is beginning to show significant advantages over mono-therapy, such as synergism, enhanced anti-microbial activity, broad anti-micr

Prediction of HIV Drug Resistance by Combining Sequence and Structural Properties.

Drug resistance is a major obstacle faced by therapist in treating HIV infected patients. The reason behind these phenomena is either protein mutation or the changes in gene expression level that induces resistance to drug treatments. These mutations affect the drug binding activity, hence resulting in failure of treatment. Therefore, it is necessary to conduct resistance testing in order to carry out HIV effective therapy. This study combines both sequence and structural features for predicting HIV resistance by applying SVM and Random Forests classifiers. The model was tested on the mutants
