Intestinal absorption of lithocholic acid sulfates in the rat: inhibitory effects of calcium.

Sulfation of lithocholic acid has been proposed as a mechanism for elimination of this hepatotoxic bile acid from the body by accelerating its fecal excretion. However, quantitative data on the absorption characteristics of sulfated lithocholic acid conjugates in vivo are scarce. We studied the intestinal absorption of 14C-labeled glycolithocholic acid (GLC), taurolithocholic acid (TLC), and their 3 alpha-sulfate esters, SGLC and STLC, respectively. Studies were performed in unanesthetized rats with a permanent biliary drainage. At an intestinal infusion rate of 125 nmol/min, which is comparab

Long-term supplementation of taurine induces hepatic steatosis and disrupts bile acid homeostasis in male mice.

Taurine, a sulfur-containing amino acid, is widely used in energy drinks and nutraceuticals. However, the long-term effects of taurine supplementation on liver metabolism remain incompletely understood. Here, male C57BL/6 mice were administered 3 % taurine in drinking water for 32 weeks. Taurine treatment reduced body weight and abdominal fat, suggesting potential anti-obesity effects. However, hepatic lipid accumulation and steatosis were evident in taurine-treated mice. Serum levels of alanine aminotransferase, alkaline phosphatase, cholesterol, non-esterified fatty acids (NEFA), total bile

Cholestyramine treatment of healthy humans rapidly induces transient hypertriglyceridemia when treatment is initiated.

Bile acid (BA) production in mice is regulated by hepatic farnesoid X receptors and by intestinal fibroblast growth factor (FGF)-15 (in humans, FGF-19), a suppressor of BA synthesis that also reduces serum triglycerides and glucose. Cholestyramine treatment reduces FGF-19 and induces BA synthesis, whereas plasma triglycerides may increase from unclear reasons. We explored whether FGF-19 may suppress BA synthesis and plasma triglycerides in humans by modulation of FGF-19 levels through long-term cholestyramine treatment at increasing doses. In a second acute experiment, metabolic responses from

[Simultaneous determination of gastric emptying and bile and pancreatic enzyme secretion].

Intestinal perfusion methods with a nonabsorbable marker allow an exact quantitative determination of intestinal absorption and secretion provided that methodological pitfalls are avoided. A modified technique is applied to the simultaneous measurement of biliary and pancreatic secretion during and depending on emptying of a mixed test meal. A duodenal segment was perfused with an isotonic polyethyleneglycol solution (PEG). Reinjection of duodenal aspirates maintained a normal enterohepatic circulation of bile acids (interruption less than 10%). The perfusion was performed in healthy volonteer

Vitamin B12 absorption and malabsorption.

Vitamin B12 is assimilated and transported by complex mechanisms that involve three transport proteins, intrinsic factor (IF), haptocorrin (HC) and transcobalamin (TC) and their respective membrane receptors. Vitamin deficiency is mainly due to inadequate dietary intake in vegans, and B12 malabsorption is related to digestive diseases. This review explores the physiology of vitamin B12 absorption and the mechanisms and diseases that produce malabsorption. In the stomach, B12 is released from food carrier proteins and binds to HC. The degradation of HC by pancreatic proteases and the pH change

Gallstone Disease and Microbiome.

Gallstone disease (GSD) has, for many years, remained a high-cost, socially significant public health problem. Over the past decade, a number of studies have been carried out-both in humans and in animal models-confirming the role of the microbiota in various sections of the gastrointestinal tract as a new link in the etiopathogenesis of GSD. The microbiome of bile correlates with the bacterial composition of saliva, and the microbiome of the biliary tract has a high similarity with the microbiota of the duodenum. Pathogenic microflora of the oral cavity, through mechanisms of immunomodulation

Bile acid signaling in health and disease.

Bile acids (BAs), cholesterol-derived amphipathic metabolites which are traditionally regarded as digestive surfactants, are now recognized as pleiotropic signaling molecules that act through ligand-, receptor-, and tissue-specific mechanisms to maintain systemic homeostasis. They exert biological functions via nuclear receptors (FXR, PXR, VDR, CAR) and membrane receptors (TGR5, S1PR2, MRGPRX4). These receptors activate transcriptional and membrane-initiated programs, including FXR-SHP/FGF19 feedback, TGR5-cAMP-PKA signaling, S1PR2-ERK/AKT/RhoA pathways, MRGPRX4-dependent Ca2⁺ signaling, PXR/C

Tamoxifen induced hepatotoxicity via gut microbiota-mediated hyodeoxycholic acid depletion and Farnesoid X receptor signaling disruption.

Tamoxifen (TAM) is a widely used estrogen receptor modulator for breast cancer treatment. However, TAM exhibits significant hepatotoxicity in the clinic, affecting nearly 50% of patients and thereby limiting its clinical utility. The specific mechanisms underlying TAM-induced liver injury remain poorly understood. In this study, we elucidated the mechanistic role of the gut microbiota in the hepatotoxicity associated with TAM. TAM administration induced substantial liver injury and gut microbiota dysbiosis in mice, characterized by an increased abundance of Escherichia and a reduction in Lachn

Zinc supplementation for prevention of neonatal hyperbilirubinemia: A systematic review and meta-analysis.

Given the potential of enteral zinc to reduce enterohepatic circulation of unconjugated bilirubin, we aimed to synthesize evidence from randomized controlled trials (RCTs) on zinc supplementation for prevention of neonatal hyperbilirubinemia (NNH). We searched PubMed, Embase, Scopus, and CENTRAL for RCTs evaluating zinc supplementation in healthy or at-risk neonates for prevention of NNH. Primary outcomes were incidence of significant NNH and requirement for phototherapy. Secondary outcomes included duration of phototherapy, bilirubin levels, and adverse effects. Two reviewers independently sc

Circulating levels of PYY are increased in individuals with bile acid diarrhoea.

BACKGROUND: Bile acid diarrhoea (BAD) is a chronic disease caused by a disturbance of the enterohepatic circulation resulting in an abundance of bile acids in the colon, which in turn causes diarrhoea. Since bile acids stimulate the secretion of the hormone peptide YY (PYY), we investigated PYY levels in plasma samples from earlier studies enrolling patients with BAD and healthy volunteers. METHODS: We analysed total PYY in 1) a case-control study comparing individuals with BAD with matched healthy controls during acute administration of the bile acid sequestrant colesevelam and placebo; 2) a

Liupao tea alleviates high-fat diet-induced obesity by re-establishing metabolic homeostasis of short-chain fatty acids and chenodeoxycholic acid.

Metabolic dyshomeostasis of bile acid (BA) and microbiota-derived short-chain fatty acid (SCFA) is a critical driver of obesity pathogenesis. Liupao tea (LPT) has garnered attention for its anti-obesity potential, owing to potent lipid-lowering, microbiota- and BA metabolism-regulatory properties. However, whether LPT coordinately modulates BA and SCFA homeostasis remains unclear. This study investigated the therapeutic potential of LPT extract (LPTe) in alleviating obesity by focusing on the host-microbial re-establishment of SCFA and BA homeostasis. Results showed that five potential bioacti

Antiplasmodial Activity and Pharmacokinetic Profiling of Cryptolepine and 2,7-Dibromocryptolepine With a View to Informing the Design of Novel Antimalarial Cryptolepine Analogues.

The roots of the climbing shrub Cryptolepis sanguinolenta are traditionally used in West Africa for the treatment of malaria. The principal constituent, cryptolepine (1), has been shown to have antimalarial activity but there are concerns regarding its toxicity on account of its DNA-intercalating property. The synthetic analogue, 2,7-dibromocryptolepine, (2) does not intercalate into DNA and is markedly more active than the parent against Plasmodium sp. in vitro and in vivo. The aim of this study was to carry out a pre-clinical assessment of 1 and 2, and if appropriate, carry out in vivo pharm

Neonatal Hyperbilirubinemia in Uganda: Mechanisms, Clinical Consequences, and Health-System Challenges.

OBJECTIVE: Neonatal hyperbilirubinemia is a prevalent condition characterized by elevated serum bilirubin levels, affecting approximately 50% of term and 80% of preterm infants. While mild cases often resolve without intervention, severe hyperbilirubinemia can lead to life-threatening complications such as bilirubin-induced neurological dysfunction (BIND) and kernicterus, resulting in permanent brain damage, developmental delays, and hearing impairment. MATERIALS AND METHODS: A literature review was conducted by searching PubMed, Google Scholar, and UpToDate for English-language articles publ

Zinc Supplementation in Neonatal Hyperbilirubinemia: An Updated Systematic Review and Meta-Analysis of Randomized Controlled Trials.

BACKGROUND AND OBJECTIVES: In vitro studies suggest that oral zinc supplementation reduces enterohepatic circulation of unconjugated bilirubin and serum bilirubin levels. Multiple randomized controlled trials (RCTs) have assessed the efficacy of oral zinc supplementation in reducing bilirubin levels and the need for phototherapy. However, the evidence remains conflicting. Therefore, we aimed to evaluate the effects of oral zinc supplementation in neonatal hyperbilirubinemia. METHODS: We searched PubMed, Embase, Web of Science, and various clinical registries for RCTs published until May 25, 2

Nisin lantibiotic prevents NAFLD liver steatosis and mitochondrial oxidative stress following periodontal disease by abrogating oral, gut and liver dysbiosis.

Oral microbiome dysbiosis mediates chronic periodontal disease, gut microbial dysbiosis, and mucosal barrier disfunction that leads to steatohepatitis via the enterohepatic circulation. Improving this dysbiosis towards health may improve liver disease. Treatment with antibiotics and probiotics have been used to modulate the microbial, immunological, and clinical landscape of periodontal disease with some success. The aim of the present investigation was to evaluate the potential for nisin, an antimicrobial peptide produced by Lactococcus lactis, to counteract the periodontitis-associated gut d

High-grain diet feeding alters ileal microbiota and disrupts bile acid metabolism in lactating dairy cows.

Bile acids (BAs) play an important role in the regulation of lipid metabolic homeostasis, but little is known about their metabolism in dairy cows fed a high-grain (HG) diet. In the present study, we examined the bacterial community, BA profile, and the FXR/FGF19 signaling pathway in the ileum and liver to investigate the gut microbe-BA metabolism interactions response to HG diet and the changes in the subsequent enterohepatic circulation of dairy cows. The results showed that the ileal bacterial community was altered, with an increase of Paraclostridium, Anaerobutyricum, Shuttleworthia, and S

Featuring molecular regulation of bile acid homeostasis in pediatric short bowel syndrome.

BACKGROUND: Disturbed bile acid homeostasis may foster development of short bowel syndrome (SBS) associated liver disease during and after weaning off parenteral nutrition (PN). Our aim was to study hepatic molecular regulation of bile acid homeostasis in relation to serum and fecal bile acid profiles in pediatric SBS. METHODS: Liver histopathology and mRNA expression of genes regulating synthesis, uptake and export of bile acids, and cellular receptors involved in bile acid signaling were measured in SBS patients (n = 33, median age 3.2 years). Simultaneously, serum (n = 24) and fecal (n = 1

Bile Acids, Gut Microbiome and the Road to Fatty Liver Disease.

This article describes the complex interactions occurring between diet, the gut microbiome, and bile acids in the etiology of fatty liver disease. Perhaps 25% of the world's population may have nonalcoholic fatty liver disease (NAFLD) and a significant percentage (∼20%) of these individuals will progress to nonalcoholic steatohepatitis (NASH). Currently, the only recommended treatment for NAFLD and NASH is a change in diet and exercise. A Western-type diet containing high fructose corn syrup, fats, and cholesterol creates gut dysbiosis, increases intestinal permeability and uptake of LPS causi
