Duration-aware unsupervised segmentation reveals collective movement modes in homing flights of pigeon flocks.

BACKGROUND: Homing flights of pigeon flocks require rapid adjustments in individual behaviour while maintaining group cohesion. However, how heading adjustments and coordinated movement changes are temporally organised during continuous flight, and whether they form repeatable collective movement modes, remain poorly characterised. METHODS: During homing flights of domestic pigeon (Columba livia) flocks, we synchronously recorded individual GPS trajectories and inertial measurement data. After standardised preprocessing, candidate flock-level kinematic features were constructed to characteris

Controlled and Real-Life Investigation of Optical Tracking Sensors in Smart Glasses for Monitoring Eating Behavior Using Deep Learning: Cross-Sectional Study.

BACKGROUND: The increasing prevalence of obesity necessitates innovative approaches to better understand this health crisis, particularly given its strong connection to chronic diseases such as diabetes, cancer, and cardiovascular conditions. Monitoring dietary behavior is crucial for designing effective interventions that help decrease obesity prevalence and promote healthy lifestyles. However, traditional dietary tracking methods are limited by participant burden and recall bias. Exploring microlevel eating activities, such as meal duration and chewing frequency, in addition to eating episod

Prediabetes transitions to normoglycaemia or type 2 diabetes and associated risk factors in the Obesity, Diabetes and Cardiovascular Disease Collaboration: an individual-level pooled analysis of 19 prospective cohort studies.

BACKGROUND: With the increasing global burden of type 2 diabetes, prevention strategies that target prediabetes, a state of hyperglycaemia that puts individuals at high risk of type 2 diabetes, are required. We aimed to estimate global rates of transition from prediabetes to normoglycaemia or type 2 diabetes, stratified by age, sex, and race and ethnicity. We also aimed to quantify the effect of modifiable and non-modifiable risk factors on these transitions. METHODS: In this pooled analysis of individual-level data, we included original data from 19 prospective cohort studies conducted in As

Comprehensive genomic identification and functional analysis of bHLH transcription factors in Ganoderma lucidum.

OBJECTIVE: The basic helix-loop-helix (bHLH) transcription factors (TFs) are pivotal in regulating fungal growth, development, and secondary metabolism. However, the knowledge about the Ganoderma lucidum bHLHs (GlbHLHs) in ganoderic acid (GA) biosynthesis of G. lucidum was limited. This study aimed to explore the functions of bHLH genes in ganoderic acid biosynthesis during G. lucidum growth development. METHODS: First, the genome-wide identification of GlbHLHs was performed through Hidden Markov model searches and Two-way blast. Furthermore, through physicochemical properties, gene structure

Genomic copy number variants: evidence for association with antibody response to anthrax vaccine adsorbed.

BACKGROUND: Anthrax and its etiologic agent remain a biological threat. Anthrax vaccine is highly effective, but vaccine-induced IgG antibody responses vary widely following required doses of vaccinations. Such variation can be related to genetic factors, especially genomic copy number variants (CNVs) that are known to be enriched among genes with immunologic function. We have tested this hypothesis in two study populations from a clinical trial of anthrax vaccination. METHODS: We performed CNV-based genome-wide association analyses separately on 794 European Americans and 200 African-America

Enhanced microbiota-derived mucinases in colorectal cancer patients revealed by gut metagenome probing coupled with functional validation.

Mucinases produced by the gut microbiota play a dual role in regulating the integrity and renewal of the mucus layer, which is essential for maintaining gut homeostasis and human health. In this study, we constructed protein hidden Markov models based on 11 known mucinases and used them to systematically identify mucinase sequences from gut metagenome-assembled genomes derived from 80 colorectal cancer (CRC) patients and 86 healthy (Healthy) subjects. A total of 1,869 mucinases were detected, widely distributed across the studied cohorts, with the majority originating from Bacteroides, Phocaei

Eight Unexpected Selenoprotein Families in Organometallic Biochemistry in Clostridium difficile, in ABC Transport, and in Methylmercury Biosynthesis.

The bioinformatics of a nine-gene locus, designated selenocysteine-assisted organometallic (SAO), was investigated after identifying six new selenoprotein families and constructing hidden Markov models (HMMs) that find and annotate members of those families. Four are selenoproteins in most SAO loci, including Clostridium difficile. They include two ABC transporter subunits, namely, permease SaoP, with selenocysteine (U) at the channel-gating position, and substrate-binding subunit SaoB. Cytosolic selenoproteins include SaoL, homologous to MerB organomercurial lyases from mercury resistance loc

Transcriptomic decoding of dynamic brain activity alterations in chronic non-specific low back pain.

BACKGROUND: Investigating dynamic brain fluctuations helps reveal changes in neural activity patterns associated with disease. Chronic non-specific low back pain (CNLBP) is characterized by altered brain function, yet less is known about the dynamic changes in brain activity and their relationship with regional gene expression profiles. METHODS: Based on resting-state functional MRI data, a hidden Markov model (HMM) was applied to identify recurrent whole-brain activity patterns, referred to as HMM states, in 80 patients with CNLBP and 80 healthy controls (HCs). Additionally, partial least sq

Disrupted Dynamic Functional Network Connectivity Among Cognitive Control Networks in the Progression of Alzheimer's Disease.

Background: Alzheimer's disease (AD) is the most common age-related dementia that promotes a decline in memory, thinking, and social skills. The initial stages of dementia can be associated with mild symptoms, and symptom progression to a more severe state is heterogeneous across patients. Recent work has demonstrated the potential for functional network mapping to assist in the prediction of symptomatic progression. However, this work has primarily used static functional connectivity (sFC) from resting-state functional magnetic resonance imaging. Recently, dynamic functional connectivity (dFC

Resting state dynamic functional connectivity in children with attention deficit/hyperactivity disorder.

Attention deficit/hyperactivity disorder (ADHD) is characterized by inattention, hyperactivity and impulsivity. In this study, we investigated group differences in dynamic functional connectivity (dFC) between 113 children with inattentive (46 ADHDI) and combined (67 ADHDC) ADHD and 76 typically developing (TD) children using resting-state functional MRI data. For dynamic connectivity analysis, the data were first decomposed into 100 independent components, among which 88 were classified into eight well-known resting-state networks (RSNs). Three discrete FC states were then identified using k-

Epilepsy as a dynamic disease: A Bayesian model for differentiating seizure risk from natural variability.

OBJECTIVE: A fundamental challenge in treating epilepsy is that changes in observed seizure frequencies do not necessarily reflect changes in underlying seizure risk. Rather, changes in seizure frequency may occur due to probabilistic variation around an underlying seizure risk state caused by normal fluctuations from natural history, leading to seizure unpredictability and potentially suboptimal medication adjustments in epilepsy management. However, no rigorous statistical approach exists to systematically distinguish expected changes in seizure frequency due to natural variability from chan

Autosome-wide copy number variation association analysis for rheumatoid arthritis using the WTCCC high-density SNP genotype data.

OBJECTIVE: Rheumatoid arthritis (RA) is a complex autoimmune rheumatic disease that is strongly influenced by genetic factors. Numerous genes are convincingly associated with RA, including genes in tumor necrosis factor signaling (TNF) and the nuclear factor-κB pathway. To date, except for genes within the HLA region, no data exist regarding potential copy number variations (CNV) involving RA-associated genes. We set out to identify genes affected by CNV that are associated with RA at a genome-wide level. METHODS: Data from the Wellcome Trust Case Control Consortium (WTCCC) were used in our a

Optical computer recognition of facial expressions associated with stress induced by performance demands.

Application of computer vision to track changes in human facial expressions during long-duration spaceflight may be a useful way to unobtrusively detect the presence of stress during critical operations. To develop such an approach, we applied optical computer recognition (OCR) algorithms for detecting facial changes during performance while people experienced both low- and high-stressor performance demands. Workload and social feedback were used to vary performance stress in 60 healthy adults (29 men, 31 women; mean age 30 yr). High-stressor scenarios involved more difficult performance tasks

RC-Mamba: regime-conditioned selective state-space models for probabilistic time-series forecasting.

Selective state-space models (SSMs) such as Mamba achieve linear-time sequence modeling through input-dependent recurrence, yet they apply the same dynamics regardless of the latent regime generating the data-a structural mismatch for non-stationary signals whose statistics shift abruptly between regimes. We introduce RC-Mamba, a probabilistic forecaster whose selective-SSM backbone is dynamically conditioned on the posterior over a latent regime variable. Regime posteriors from a Student-t emission Hidden Markov Model with a sticky Dirichlet prior modulate the SSM discretisation step Δ t toge

High Functioning Autism with Missense Mutations in Synaptotagmin-Like Protein 4 (SYTL4) and Transmembrane Protein 187 (TMEM187) Genes: SYTL4- Protein Modeling, Protein-Protein Interaction, Expression Profiling and MicroRNA Studies.

We describe a 7-year-old male with high functioning autism spectrum disorder (ASD) and maternally-inherited rare missense variant of Synaptotagmin-like protein 4 (SYTL4) gene (Xq22.1; c.835C>T; p.Arg279Cys) and an unknown missense variant of Transmembrane protein 187 (TMEM187) gene (Xq28; c.708G>T; p. Gln236His). Multiple in-silico predictions described in our study indicate a potentially damaging status for both X-linked genes. Analysis of predicted atomic threading models of the mutant and the native SYTL4 proteins suggest a potential structural change induced by the R279C variant which elim

Semantic segmentation of cerebrospinal fluid and brain volume with a convolutional neural network in pediatric hydrocephalus-transfer learning from existing algorithms.

BACKGROUND: For the segmentation of medical imaging data, a multitude of precise but very specific algorithms exist. In previous studies, we investigated the possibility of segmenting MRI data to determine cerebrospinal fluid and brain volume using a classical machine learning algorithm. It demonstrated good clinical usability and a very accurate correlation of the volumes to the single area determination in a reproducible axial layer. This study aims to investigate whether these established segmentation algorithms can be transferred to new, more generalizable deep learning algorithms employin

Emotion Forecasting: A Transformer-Based Approach.

BACKGROUND: Monitoring the emotional states of patients with psychiatric problems has always been challenging due to the noncontinuous nature of clinical assessments, the effect of the health care environment, and the inherent subjectivity of evaluation instruments. However, mental states in psychiatric disorders exhibit substantial variability over time, making real-time monitoring crucial for preventing risky situations and ensuring appropriate treatment. OBJECTIVE: This study aimed to leverage new technologies and deep learning techniques to enable more objective, real-time monitoring of p

Neurophysiological effects of transcranial alternating current stimulation combined with multidisciplinary rehabilitation in Parkinson's disease.

INTRODUCTION: Motor impairment in Parkinson's disease (PD) often becomes inadequately controlled as the disease progresses. Rehabilitation therapy combined with noninvasive neuromodulation has shown benefits, yet the neural mechanisms underlying this remain unclear. This study aimed to identify neurophysiological and brain network features associated with motor improvement following high-intensity transcranial alternating current stimulation (Hi-tACS) combined with multidisciplinary intensive rehabilitation therapy (MIRT). METHODS: This secondary analysis was based on a randomized controlled

Distribution and Classification of Serine β-Lactamases in Brazilian Hospital Sewage and Other Environmental Metagenomes Deposited in Public Databases.

β-lactam is the most used antibiotic class in the clinical area and it acts on blocking the bacteria cell wall synthesis, causing cell death. However, some bacteria have evolved resistance to these antibiotics mainly due the production of enzymes known as β-lactamases. Hospital sewage is an important source of dispersion of multidrug-resistant bacteria in rivers and oceans. In this work, we used next-generation DNA sequencing to explore the diversity and dissemination of serine β-lactamases in two hospital sewage from Rio de Janeiro, Brazil (South Zone, SZ and North Zone, NZ), presenting diffe

Characterizing Rare DNA Copy-Number Variants in Pediatric Obsessive-Compulsive Disorder.

OBJECTIVE: Pediatric obsessive-compulsive disorder (OCD) is a common neuropsychiatric disorder in which genetic factors play an important role. Recent studies have demonstrated an enrichment of rare de novo DNA single-nucleotide variants in persons with OCD compared to controls, and larger studies have examined copy-number variants (CNVs) using microarray data. Our study examines rare de novo CNVs using whole-exome sequencing (WES) data to provide additional insight into genetic factors and biological processes underlying OCD. METHOD: We detected CNVs using whole-exome DNA sequencing (WES) da
