Inulin and Lycium barbarum polysaccharides mitigate the diabetic inflammatory response by modulating bile acid metabolism to enhance regulatory T cell (Treg) activation in a rat model of diabetes.

BACKGROUND: Type 2 diabetes mellitus (T2DM) is a chronic metabolic disease that poses a serious threat to health. Currently, there are no completely effective treatment options. Modulating intestinal flora and its metabolites may represent a promising new approach for diabetes therapy. Regulating intestinal microbiota and its metabolites through prebiotics, mediated by regulatory T (Treg) cells, could offer a novel strategy to improve the chronic inflammatory state associated with diabetes. OBJECTIVE: This study aimed to investigate the improvement of diabetes-related chronic inflammation by

Lysosomal Dysfunction Is Associated With Intervertebral Disc Degeneration: Multiomics and Machine Learning Identify Molecular Subtypes and Hub Genes.

Intervertebral disc degeneration (IVDD) is closely associated with cellular senescence and defective autophagic degradation, but the molecular heterogeneity and functional significance of lysosome-related alterations remain incompletely understood. We integrated bulk transcriptomic datasets (GSE56081 and GSE70362) and single-cell RNA-sequencing data (GSE153066) using consensus clustering, weighted gene coexpression network analysis, machine-learning algorithms, immune-signature analysis, and covariate-adjusted correlation testing. Candidate genes were evaluated in human and rat disc tissues, a

Functional and metabolic dysbiosis of the gut microbiome in the tumoral immune microenvironment of hepatocellular carcinoma.

With its asymptomatic and heterogeneous nature, hepatocellular carcinoma (HCC) ranks as one of the most fatal malignancies globally, contributing to a deteriorating five-year overall survival. The gut microbiome, as the resident microorganism of the digestive tract, has been linked to a variety of hepatic disorders based on the internal "gut-liver" axis, accompanied by alterations in microbial components and derived metabolites. We found that patients with HCC had a greater abundance of Streptococcus salivarius (S. salivarius) and lithocholic acid (LCA), but its role in the tumor immune microe

CAR, the continuously advancing receptor, in drug metabolism and disease.

The detoxification and elimination of potentially toxic foreign and endogenous compounds depends on the concerted action of xenobiotic metabolizing enzymes. Nuclear hormone receptors (NHRs) have emerged as key regulators of the expression of these enzymes and his review focuses on the xenosenor CAR (Constitutive Androstane Receptor, NR1I3). CAR is highly expressed in the liver and the small intestine, two key tissues expressing xenobiotic metabolizing enzymes, and mediates the induction of their expression by the widely used antiepileptic drug, phenobarbital (PB) and the potent synthetic induc

Ultrastructural evidence of intrahepatic cholestasis before and after chenodeoxycholic acid therapy in patients with cholelithiasis: the national cooperative gallstone study.

Electron microscopy was performed to assess potential hepatotoxicity before and after 9 and 24 months of chenodeoxycholic acid (CDCA) therapy (375 or 750 mg, daily) in 103 patients with cholelithiasis. Prior to treatment, 64% of the biopsies demonstrated ultrastructural evidence of intrahepatic cholestasis, manifested by abnormal bile canaliculi, thickened pericanalicular ectoplasm, and retention of biliary material within liver cells or pericellular spaces. After 9 months of CDCA therapy, several changes (including increased free cytoplasmic "biliary pigment," decreased canalicular microvilli

Decreased C-reactive protein induces abnormal vascular structure in a rat model of liver dysfunction induced by bile duct ligation.

BACKGROUND/AIMS: Chronic liver disease leads to liver fibrosis, and although the liver does have a certain regenerative capacity, this disease is associated with dysfunction of the liver vessels. C-reactive protein (CRP) is produced in the liver and circulated from there for metabolism. CRP was recently shown to inhibit angiogenesis by inducing endothelial cell dysfunction. The objective of this study was to determine the effect of CRP levels on angiogenesis in a rat model of liver dysfunction induced by bile duct ligation (BDL). METHODS: The diameter of the hepatic vein was analyzed in rat l

Enhanced glucose homeostasis via Clostridium symbiosum-mediated glucagon-like peptide 1 inhibition of hepatic gluconeogenesis in mid-intestinal bypass surgery.

BACKGROUND: The small intestine is known to play a crucial role in the development and remission of diabetes mellitus (DM). However, the exact mechanism by which mid-small intestinal bypass improves glucose metabolism in diabetic rats is not fully understood. AIM: To elucidate the mechanisms by which mid-small intestinal bypass improves glucose metabolism. METHODS: Streptozotocin (STZ) was used to induce DM in Sprague-Dawley (SD) rats at a dose of 60 mg/kg. The rats were then randomly divided into two groups: The mid-small intestine bypass (MSIB) group and the sham group (underwent switch la

Total flavonoids from Abrus cantoniensis alleviate fatty liver hemorrhagic syndrome in laying hens by regulating inflammation, oxidative stress, and cecal metabolites and microbiota.

Fatty liver hemorrhagic syndrome (FLHS) is a metabolic disease in laying hens. Total flavonoids from Abrus cantoniensis (TFAC) comprise multiple bioactive compounds with potential benefits against FLHS. This study aimed to explore the effects and mechanism of TFAC in improving FLHS. Firstly, analysis by liquid chromatography-tandem mass spectrometry identified 20 flavonoid compounds in the TFAC, including vicenin-3 and acacetin. Subsequently, 144 laying hens at 28 weeks of age with similar body weight (1.45 ± 0.03 kg) were randomly divided into six groups (eight replicates per group and three

Effects of Composite Antimicrobial Peptides on Colonic Microbiota and Barrier Integrity in Holstein Steers.

Antimicrobial peptides (APs) are increasingly used in livestock production because of their effective antibacterial properties and low risk of causing drug resistance. However, their effects on intestinal barrier function in ruminants remain relatively unexplored. This study explored the effect of composite APs on colonic microbiota and barrier integrity in Holstein steers. Twelve steers were randomly assigned to two groups (n = 6): a control (CK) group that received a basal diet and an AP group that received the same diet supplemented with 8 g/ (d· head) of APs over a 270-day feeding period.

Activation of pregnane X receptor protects against cholestatic liver injury by inhibiting hepatocyte pyroptosis.

Our previous study shows that activation of pregnane X receptor (PXR) exerts hepatoprotection against lithocholic acid (LCA)-induced cholestatic liver injury. In this study we investigated whether PXR activation could inhibit hepatocyte pyroptosis, as well as the underlying mechanisms. Male mice were treated with mouse PXR agonist pregnenolone 16α-carbonitrile (PCN, 50 mg·kg-1·d-1, i.p.) for 7 days, and received LCA (125 mg/kg, i.p., bid) from D4, then sacrificed 12 h after the last LCA injection. We showed that LCA injection resulted in severe cholestatic liver injury characterized by signifi

Impact of brewer's spent grain (BSG) and bio-transformed BSG biscuits on metabolic and gut health in adults with metabolic impairments: a randomized controlled trial.

BACKGROUND: Brewers’ spent grain (BSG), a fiber-rich by-product of the brewing industry, has high nutritional value. Fermentation with Rhizopus oligosporus further enhances its composition by increasing soluble dietary fiber and releasing antioxidants, potentially conferring superior metabolic regulatory and prebiotic effects. This study evaluated the effect of long-term consumption of BSG or fermented BSG-containing biscuits on metabolic and gut health in Singapore adults with metabolic impairments. METHODS: Thirty-one metabolically impaired subjects were randomized to consume 90 g/day of con

A universal plasma metabolites-derived signature predicts cardiovascular disease risk in MAFLD.

BACKGROUND: Metabolic associated fatty liver disease (MAFLD) is a novel concept proposed in 2020, which is more practical for identifying patients with fatty liver disease with high risk of disease progression. Fatty liver is a driver for extrahepatic complications, particularly cardiovascular diseases (CVD). Although the risk of CVD in MAFLD could be predicted by carotid ultrasound test, a very early stage prediction method before the formation of pathological damage is still lacking. METHODS: Stool microbiomes and plasma metabolites were compared across 196 well-characterized participants e

Targeted therapy of colorectal neoplasia with rapamycin in peptide-labeled pegylated octadecyl lithocholate micelles.

Many powerful drugs have limited clinical utility because of poor water solubility and high systemic toxicity. Here, we formulated a targeted nanomedicine, rapamycin encapsulated in pegylated octadecyl lithocholate micelles labeled with a new ligand for colorectal neoplasia, LTTHYKL peptide. CPC;Apc mice that spontaneously develop colonic adenomas were treated with free rapamycin, plain rapamycin micelles, and peptide-labeled rapamycin micelles via intraperitoneal injection for 35days. Endoscopy was performed to monitor adenoma regression in vivo. We observed complete adenoma regression at the

Drug metabolism in end-stage liver disease. In vitro activities of some phase I and phase II enzymes.

The activities of a number of drug metabolising enzymes were measured in liver samples obtained from three groups of subjects: normal donors, patients with primary biliary cirrhosis (PBC), and patients with other types of liver disease. In the latter group, all the enzyme activities determined were impaired relative to the normal group. In the PBC group, however, enzyme activities were altered more selectively. (a) Activities of the methyl cholanthrene-inducible forms of cytochrome P-450 were decreased compared to normal controls, whereas the activities of the phenobarbitone-inducible isozymes

Acute myocardial infarction induces sex-specific, time-dependent remodeling of the gut microbiome and intestinal immune compartment in retired breeder C57BL/6N mice.

INTRODUCTION: The gut microbiome influences cardiovascular health through metabolite production and immune modulation. Gut microbial dynamics and cardiovascular outcomes are also shaped by biological sex. However, sex-specific responses to myocardial infarction (MI) that involve the gut microbiome and intestinal milieu remain poorly defined, particularly in older hosts. Here, we characterize gut microbiome structure and function alongside physiological and immune responses to MI across multiple tissues in aging male and female mice. METHODS: MI was induced by permanent LAD ligation and confir
