RNA expression in the early characterization of hepatotoxicants in Wistar rats by high-density DNA microarrays.

High-density microarrays are useful tools to study gene expression for the purpose of characterizing functional tissue changes in response to the action of drugs and chemicals. To test whether high-density expression data can identify mechanisms of toxicity and to identify an unknown sample through its RNA expression pattern, groups of male Wistar rats were administered 6 hepatotoxicants. The compounds chosen for this study were microcystin-LR (MLR), phenobarbital (PB), lipopolysaccharide (LPS), carbon tetrachloride (CT), thioacetamide (THA), and cyproterone acetate (CPA). These hepatotoxicant

Mutation of a major keratin phosphorylation site predisposes to hepatotoxic injury in transgenic mice.

Simple epithelia express keratins 8 (K8) and 18 (K18) as their major intermediate filament (IF) proteins. One important physiologic function of K8/18 is to protect hepatocytes from drug-induced liver injury. Although the mechanism of this protection is unknown, marked K8/18 hyperphosphorylation occurs in association with a variety of cell stresses and during mitosis. This increase in keratin phosphorylation involves multiple sites including human K18 serine-(ser)52, which is a major K18 phosphorylation site. We studied the significance of keratin hyperphosphorylation and focused on K18 ser52 b

Acute toxic effects in mice of an extract from the marine algae Gonyaulax monilata.

Unialgal cultures of Gonyaulax monilata were cultured and harvested. A modified Westphall procedure was used to prepare an extract which did not contain saxitoxin, the gonyautoxins and structurally related toxins. The extract was administered i.p. to young adult, male CD-1 mice and produced: sedation, abdominal constriction, fecal clumping in the perianal area, ataxia, tremors, cyanosis, loss of reflexes, convulsions and death (LD50 = 2.28 mg/kg). Gross and microscopic pathology in the treated mice included: acute active hyperemia of the viscera, multifocal areas of necrosis of the musculature

Is the future blue-green? A review of the current model predictions of how climate change could affect pelagic freshwater cyanobacteria.

There is increasing evidence that recent changes in climate have had an effect on lake phytoplankton communities and it has been suggested that it is likely that Cyanobacteria will increase in relative abundance under the predicted future climate. However, testing such a qualitative prediction is challenging and usually requires some form of numerical computer model. Therefore, the lake modelling literature was reviewed for studies that examined the impact of climate change upon Cyanobacteria. These studies, taken collectively, generally show an increase in relative Cyanobacteria abundance wit

Detection of ciguatoxin-like and paralysing toxins in Trichodesmium spp. from New Caledonia lagoon.

Marine pelagic cyanobacteria Trichodesmium are widespread in the New Caledonia lagoon. Blooms of these Oscillatoriales are suspected to be a potential source of toxins in the ciguatera food chain and were previously reported to contain certain types of paralysing toxins. In the present study, toxicity experiments were conducted on lipid- and water-soluble extracts of freeze-dried samples of these cyanobacteria. Lipid-soluble fractions revealed a ciguatoxin-like activity in both in vivo (mouse bioassay) and in vitro (mouse neuroblastoma cells assay and receptor binding assay using tritiated bre

Toxin mixture in cyanobacterial blooms--a critical comparison of reality with current procedures employed in human health risk assessment.

Cyanobacteria are the oldest life forms on earth known to produce a broad spectrum of secondary metabolites. The functions/advantages of most of these secondary metabolites (peptides and alkaloids) are unknown, however, some of them have adverse effects in humans and wildlife, especially when ingested, inhaled or upon dermal exposure. Surprisingly, some of these cyanobacteria are ingested voluntarily. Indeed, for centuries mankind has used cyanobacteria as a protein source, primarily Spirulina species. However, recently also Aphanizomenon flos-aquae are used for the production of so called blu

A Review of the Evidence of the Toxicity of Chemical Substances Included in the European Union Ambient Air Quality and Drinking Water Directives: Perspectives for Health Impact Assessments.

The European Union (EU) Ambient Air Quality Directive (AAQD) and Drinking Water Directive (DWD) are aimed at maintaining and improving air quality and ensuring high standards for potable water across the EU. Besides several other indicators, the AAQD and DWD consist of chemical parameters (substances or substance groups) that are regulated within this framework. All the substances are associated with various health outcomes, and many of them are classified as carcinogenic or probably/possibly carcinogenic with causal links. To quantify the health burden of the chemical substances included in t

Peroxynitrite is key to Cylindrospermopsin-mediated MASLD to MASH progression via triggering TXNIP binding to NLRP3 and subsequent inflammasome activation.

Harmful algal bloom (HAB) toxins are shown to be associated with Metabolic dysfunction-associated steatohepatitis (MASH) progression. Several studies link the HAB toxin microcystin to hepatic inflammasome activation, but the role of cylindrospermopsin (CYN) in Metabolic dysfunction-associated steatotic liver disease (MASLD) pathology remains unknown. Using a mouse model of MASLD, we show that CYN exposure served as a second hit for MASLD to MASH progression, as shown by histopathology and NAS scoring. The pathology was attenuated by the use of NADPH oxidase 2 (NOX2) inhibitor apocynin and pero

Lake browning counteracts cyanobacteria responses to nutrients: Evidence from phytoplankton dynamics in large enclosure experiments and comprehensive observational data.

Lakes worldwide are affected by multiple stressors, including climate change. This includes massive loading of both nutrients and humic substances to lakes during extreme weather events, which also may disrupt thermal stratification. Since multi-stressor effects vary widely in space and time, their combined ecological impacts remain difficult to predict. Therefore, we combined two consecutive large enclosure experiments with a comprehensive time-series and a broad-scale field survey to unravel the combined effects of storm-induced lake browning, nutrient enrichment and deep mixing on phytoplan

Changes in Carboxy Methylation and Tyrosine Phosphorylation of Protein Phosphatase PP2A Are Associated with Epididymal Sperm Maturation and Motility.

Mammalian sperm contain the serine/threonine phosphatases PP1γ2 and PP2A. The role of sperm PP1γ2 is relatively well studied. Here we confirm the presence of PP2A in sperm and show that it undergoes marked changes in methylation (leucine 309), tyrosine phosphorylation (tyrosine 307) and catalytic activity during epididymal sperm maturation. Spermatozoa isolated from proximal caput, distal caput and caudal regions of the epididymis contain equal immuno-reactive amounts of PP2A. Using demethyl sensitive antibodies we show that PP2A is methylated at its carboxy terminus in sperm from the distal c

An indoor air quality study of an alligator (Alligator mississippiensis) holding facility.

An environmental microbiologic investigation was conducted in an alligator (Alligator mississippiensis) holding facility in a zoo in the southeastern U.S. The facility had housed five alligators between March 1999 and February 2005. In the exhibit, one alligator died and all experienced poor health. It was hypothesized that environmental microbial contamination was associated with these issues. Samples were collected for fungal identification and quantification, microcystin analysis, and airborne mycotoxins. Analyses of air and water were conducted and an examination of the heating, ventilatio

Complementary DNA (cDNA) cloning and expression analysis of selenium glutathione peroxidase (Se-GPx) and glutathione S-transferases (GST) 1 in red claw crayfish (Cherax quadricarinatus).

Selenium glutathione peroxidase (Se-GPx) and Glutathione S-transferases (GST) are crucial parts of cellular detoxification systems that protect cells from reactive oxygen species (ROS). In this study, cDNA sequences of CqGST1 and CqSe-GPx were cloned from red claw crayfish (Cherax quadricarinatus). The molecular masses of the predicted protein are 25.36 and 21.49 kDa, respectively. CqGST1 and CqSe-GPx have open reading frames (ORFs) of 657 bp and 570 bp and encode peptides of 218 and 189 amino acids, respectively. Sequence alignment showed that the amino acid sequence of CqSe-GPx identity with

Ammonium recycling supports toxic Planktothrix blooms in Sandusky Bay, Lake Erie: Evidence from stable isotope and metatranscriptome data.

Sandusky Bay, Lake Erie, receives high nutrient loadings (nitrogen and phosphorus) from the Sandusky River, which drains an agricultural watershed. Eutrophication and cyanobacterial harmful algal blooms (cyanoHABs) persist throughout summer. Planktothrix agardhii is the dominant bloom-forming species and the main producer of microcystins in Sandusky Bay. Non-N2 fixing cyanobacteria, such as Planktothrix and Microcystis, thrive on chemically reduced forms of nitrogen, such as ammonium (NH4+) and urea. Ammonium regeneration and potential uptake rates and total microbial community demand for NH4+

Effects of rainfall patterns on toxic cyanobacterial blooms in a changing climate: between simplistic scenarios and complex dynamics.

Toxic cyanobacterial blooms represent a serious hazard to environmental and human health, and the management and restoration of affected waterbodies can be challenging. While cyanobacterial blooms are already a frequent occurrence, in the future their incidence and severity are predicted to increase due to climate change. Climate change is predicted to lead to increased temperature and changes in rainfall patterns, which will both have a significant impact on inland water resources. While many studies indicate that a higher temperature will favour cyanobacterial bloom occurrences, the impact o

[Swimming in summer].

It happens every summer: 'Swimming prohibited because of cyanobacteria'. Blooming of these toxine producing bacteria in warm summer months is a well known risk for swimming human beings. But how big is this risk for swimming dogs? And how big are other risks? In this article these risks are assessed for dogs that occasionally swim in surface water during the summer in the Netherlands. Based on these findings an advice will be given for dog owners to keep these risks for waterborne infections as small as possible.

Pharmaceutical cocktails in aquatic ecosystems: unveiling the hidden ecotoxicological threats to Microcystis aeruginosa.

Pharmaceutical compounds (PhACs), continuously released into the environment, represent a growing ecotoxicological threat to aquatic ecosystems. While the toxicity of individual pharmaceuticals has been extensively investigated, the effects of complex mixtures, the so-called "cocktail effect", remain poorly understood, particularly for primary producers such as cyanobacteria. This study evaluates the individual and combined effects of four commonly detected pharmaceuticals in aquatic environments: acetaminophen (APAP), atenolol (ATN), carbamazepine (CBZ), and diazepam (DZP), using the model cy

Effect of ultraviolet radiation (type B) and titanium dioxide nanoparticles on the interspecific interaction between Microcystis flos-aquae and Pseudokirchneriella subcapitata.

Anthropogenic activities have led to the depletion of the ultraviolet radiation screening ozone layer, exposing aquatic biota to its harmful effects. Also, the rising applications of nanotechnology are resulting in the release and contamination of aquatic ecosystems with engineered nanometals like titanium dioxide nanoparticles (nTiO2). The rise in ultraviolet radiation interacts with nanometals, increasing their bioactivities to susceptible aquatic organisms such as algae and cyanobacteria. The effect of ultraviolet radiation B (UVB) and nTiO2 on Microcystis flos-aquae and Pseudokirchneriella

Multiclass Cyanotoxins in Aquatic Environments: Occurrence, Impacts and Detection Techniques.

Cyanobacterial harmful algal blooms (cyanoHABs) typically involve the release of a wide spectrum of toxic metabolites produced by different cyanobacteria, which can cause adverse health effects in animals and humans. At the same time, many cyanobacteria are capable of producing more than one toxin simultaneously. As a result, the multiclass toxins released into aquatic environments can exert complex and combined effects on various ecosystem components. This review aims to provide an overview of the occurrence and effects of different cyanotoxins in aquatic systems and current analytical method

Avian high-dose toxicity of cyanobacterial biomass.

OBJECTIVES: Previous studies using oral administration of environmentally relevant doses of cyanobacterial biomass containing microcystins (MCs) induced only sub-lethal effects in experimental birds. Therefore, the objective of this study was to obtain data on avian high-dose toxicity of MCs and compute LD50, if possible, following the natural oral route of administration. DESIGN: Responses of birds to single high-dose exposure to MCs were evaluated in fourteen-day old Japanese quail males (Coturnix coturnix japonica) with average body weight of 50 g which were randomly divided into five grou
