Minimal residual disease assessment in childhood acute lymphoblastic leukaemia: a Swedish multi-centre study comparing real-time polymerase chain reaction and multicolour flow cytometry.

Minimal residual disease (MRD) assessment is a powerful prognostic factor for determining the risk of relapse in childhood acute lymphoblastic leukaemia (ALL). In this Swedish multi-centre study of childhood ALL diagnosed between 2002 and 2006, the MRD levels were analysed in 726 follow-up samples in 228 children using real-time quantitative polymerase chain reaction (RQ-PCR) of rearranged immunoglobulin/T-cell receptor genes and multicolour flow cytometry (FCM). Using an MRD threshold of 0·1%, which was the sensitivity level reached in all analyses, the concordance between RQ-PCR and FCM MRD

Oligonucleotide microarray analysis of gene expression profiles followed by real-time reverse-transcriptase polymerase chain reaction assay in chronic active Epstein-Barr virus infection.

Chronic active Epstein-Barr virus infection (CAEBV) is characterized by recurrent infectious mononucleosis-like symptoms and has high mortality and morbidity. To clarify the mechanisms of CAEBV, the gene-expression profiles of peripheral blood obtained from patients with CAEBV were investigated. Twenty genes were differentially expressed in 4 patients with CAEBV. This microarray result was verified using a real-time reverse-transcriptase polymerase chain reaction assay in a larger group of patients with CAEBV. Eventually, 3 genes were found to be significantly upregulated: guanylate binding pr

INK4A/ARF deletions are acquired at relapse in childhood acute lymphoblastic leukaemia: a paired study on 25 patients using real-time polymerase chain reaction.

Current risk-adjusted intensive therapies for childhood acute lymphoblastic leukaemia (ALL) are expected to result in an event-free survival of greater than 75%. In sharp contrast, relapsed paediatric ALL is a difficult disease to treat. In this study, 25 paediatric patients with ALL were analysed at diagnosis and relapse for their p16 (exon 2) status using the most accurate method of detection, real-time polymerase chain reaction (PCR). The median time to relapse for the group was 27 months. At diagnosis, the incidence of p16 homozygous and hemizygous deletion in this group was 32% and 20% re

Comparison of lower and upper respiratory tract specimens for the detection of respiratory viruses and atypical pneumonia pathogens by multiplex real-time polymerase chain reaction.

Multiplex respiratory polymerase chain reaction (PCR) testing allows for the rapid and reliable identification of viral, bacterial and fungal causative agents of respiratory tract infections. Traditionally, PCR testing is performed on nose and throat swabs which are easy to collect and are accessible to most patients; however, many commercial kits have several additional validated specimen types including sputum and bronchial washings. These specimens may be more representative of the lower respiratory tract (LRT) environment. The current study aimed to investigate the performance of PCR when

Melting curve analyses in the quantitative real-time polymerase chain reaction of methylated/non-methylated DNA toward the detection of oral cancer using gargle fluid.

The early detection and early treatment of oral cancer are of paramount importance for improving the patients' prognoses. Our group previously reported a noninvasive test for oral cancer that uses a commercially available methylation-specific multiple ligation-dependent probe amplification (MS-MLPA) method, focusing on the DNA methylation of cells in gargle fluid. However, the conventional MS-MLPA method is complicated, costly, and thus difficult to use for screening and other health examinations. We therefore sought to develop a simple screening method for the diagnosis of oral cancer by cond

In situ collected small intestinal mucosal swabs as alternative analytes to diagnose Echinococcus multilocularis infection in its definitive hosts by real-time polymerase chain reaction.

The cestode Echinococcus multilocularis parasitizes small intestines of carnivores such as foxes (Vulpes vulpes) and raccoon dogs (Nyctereutes procyonoides) as definitive hosts and represents an important zoonotic threat. This study aimed to determine whether E. multilocularis DNA detection methods by real-time PCR involving easy-to-collect sample types, such as intestinal swabs could represent alternatives to traditional parasitological techniques. Three analytes from foxes and raccoon dogs were tested. i. Swabs taken immediately during necropsy (in situ) from the small intestinal mucosa (Fas

Diagnostic performance of an in-house real-time polymerase chain reaction (PCR) assay and antigen detection from respiratory samples for the diagnosis of pulmonary cryptococcosis.

INTRODUCTION: The currently available diagnostic tests lack sensitivity to diagnose pulmonary cryptococcosis. In the current study, we developed and standardized an in-house real-time PCR assay and evaluated the antigen detection in respiratory samples for the diagnosis of pulmonary cryptococcosis. MATERIALS AND METHODS: We standardized an in-house real-time PCR assay (using URA5 and STR1 primers; index test 1) and cryptococcal antigen detection (BIOSYNEX® CryptoPS, France) from the respiratory samples (index test 2). We considered a sample positive for PCR assay when both gene targets (URA5

Comparison between Two Molecular Techniques: Nested and Real-Time Polymerase Chain Reaction Targeting 100-kDa Hc Protein for Detection of Histoplasma capsulatum in Environmental Samples.

Histoplasmosis, one of the most frequent endemic mycoses in the Americas, is caused by the inhalation of airborne conidia of Histoplasma capsulatum. Better understanding of the distribution of this fungus in the environment is important for the development of appropriate public health measures to prevent human infections. Previously, we used Hc100 nested polymerase chain reaction (PCR) to identify H. capsulatum DNA in 10% of environmental samples in Colombia. Here, we validate a 100-kDa real-time PCR assay for the detection of this fungus in the environment. Using this method, we identified H.

Evaluation of diagnostic performance of comprehensive respiratory virus panel: comparison of next-generation-sequencing to real-time polymerase chain reaction for detection of respiratory viruses.

BACKGROUND: Respiratory viral infections remain a global health concern, particularly among children, the elderly, and immunocompromised individuals. Although real-time polymerase chain reaction (RT-PCR) is the diagnostic gold standard, its limitations in strain-level typing and mutation tracking highlight the need for complementary approaches such as next-generation sequencing (NGS). METHODS: We compared a hybridization-based NGS respiratory virus panel (RVP) in comparison with RT-PCR using 81 nasopharyngeal swab (NPS) specimens. The performance metrics included concordance rates, cycle thre
