The ultrastructure of the human liver infected with Cryptococcus yeast cells was investigated using transmission electron microscopy (TEM). Infected liver specimens were collected from a 3-year-old female diagnosed with liver cryptococcosis after clinical examination. TEM revealed several types of aberrant structures in different liver cell types. Filled with lipid droplets, stellate cells were evident in the patient's liver, indicating collagen assembly and liver cirrhosis. Kupffer cells, fibroblasts, and stellate cells had multilamellar bodies with multiple ring-like structures, implying the

This study evaluates the transplantation of induced pluripotent stem cell (iPSC)-derived retinal organoids into patients with advanced retinitis pigmentosa using adaptive optics optical coherence tomography (AO-OCT) to monitor retinal changes over two years post transplantation. Our results confirmed successful engraftment and increased retinal thickness, with AO-OCT providing detailed visualization of cellular structures such as an outer plexiform layer-like line and highly reflective particles within rosette-like formations, indicative of photoreceptor development. Low-density lipoprotein particles are taken up by cells and delivered to the lysosome where their cholesterol esters are cleaved off by acid lipase. The released, free cholesterol is then exported from lysosomes for cellular needs or storage. This article summarizes recent advances in our understanding of the molecular basis of cholesterol export from lysosomes. Cholesterol export requires NPC intracellular cholesterol transporter 1 (NPC1) and NPC2, genetic mutations of which can cause Niemann-Pick type C disease, a disorder characterized by massive lysosomal accumulation of cholesterol and gl

The cellular mechanisms that configure the cytoskeleton during migration of dendritic cells (DCs) are poorly understood. Immature DCs assemble specialized adhesion structures known as podosomes at their leading edge; these are associated with the localized recruitment of the Wiskott-Aldrich Syndrome protein (WASp) and the actin organizing actin-related protein 2/3 complex. In immature DCs lacking WASp, podosomes are absent, residual dysmorphic lamellipodia and filopodia are nonpolarized, and migration is severely compromised. Microinjection studies indicate that podosome assembly and polarizat

Stress granules form in response to diverse cellular perturbations to sequester translation components until the stress is resolved. Stress granules are composed of RNA-protein assemblies in membrane delimited structures and must be rapidly disassembled to release components to allow translation to resume. Disassembly of stress granules formed in response to heat stress is dependent on ubiquitiylation of stress granule components such as G3BP1. Ubiquitylation of stress granule proteins recruits the AAA-ATPase p97 (also known as VCP) to enable ubiquitin-dependent disassembly of these structures

The prevalence of antibiotic resistance genes (ARGs) contamination in sludge anaerobic digestion (AD) systems poses a critical challenge to environmental protection. This review elaborates ARGs control from a microbial metabolism perspective after systematically outlining the strategies, performance and mechanisms of ARGs removal in sludge AD systems. External addition strategies appeared to activate multiple mechanisms simultaneously. Although mechanism studies still mainly focus on microbial community structures in current publications, the substantial increase in metabolism-centered investi

Engineering mRNA stability is a promising yet underexplored approach for improving recombinant protein production in bacterial systems. In this study, we evaluated the effect of synthetic 3'-UTR hairpin structures on mRNA stability and protein yield in Escherichia coli using two SUMO-fusion expression systems. Hairpin elements with defined structural features were introduced downstream of the coding sequence. In all constructs, 3'-UTR hairpins increased mRNA half-life, with stabilization ranging from approximately 2-fold to 3-fold (n = 3 biological replicates). In the SUMO-SARS-CoV-2-derived p

Endometrial epithelial organoids (EEOs) represent a valuable tool for investigating endometrial development and establishing in vitro models of related diseases. Traditional organoid cultures, which are dependent on mouse-derived Matrigel, fail to recapitulate species-specific microenvironments. This study optimized the fabrication of porcine uterine decellularized extracellular matrix (DECM) scaffolds using a freeze-thaw cycle combined with low-damage chemical decellularization, significantly enhancing the biomimetic properties. We preserved collagen, glycosaminoglycans (GAG), and key structu

The lamin proteins are classified into A- and B-types, and together with their associated proteins, they form the nuclear lamina, which governs diverse nuclear structures and functions, including nuclear mechanics, chromatin organization, and gene regulation. Mutations of these proteins give rise to a strikingly diverse group of tissue-specific disorders, the laminopathies, including muscular dystrophies, cardiomyopathies, lipodystrophies, neuropathies, and premature aging syndromes, despite their broad expression. Unraveling the basis of this tissue selectivity has revealed that lamins functi

Huntington's disease (HD) is a neurodegenerative disorder in which mutated fragments of the huntingtin protein (Htt) undergo misfolding and aggregation. Since aggregated proteins can cause cellular stress and cytotoxicity, there is an interest in the development of small molecule aggregation inhibitors as potential modulators of HD pathogenesis. Here, we study how a polyphenol modulates the aggregation mechanism of huntingtin exon 1 (HttEx1) even at sub-stoichiometric ratios. Sub-stoichiometric amounts of curcumin impacted the primary and/or secondary nucleation events, extending the pre-aggre

Sequestration of parasite-infected red blood cells (RBCs) in the microvasculature is an important pathological feature of both bovine babesiosis caused by Babesia bovis and human malaria caused by Plasmodium falciparum. Surprisingly, when compared with malaria, the cellular and molecular mechanisms that underlie this abnormal circulatory behaviour for RBCs infected with B. bovis have been relatively ignored. Here, we present some novel insights into the adhesive and mechanical changes that occur in B. bovis-infected bovine RBCs and compare them with the alterations that occur in human RBCs inf

Genomic guanine-rich segments are prone to adopting self-associated noncanonical structures termed G-quadruplex structures. An overabundance of reports reveals their existence prevalently at important genomic locations, including oncogenic promoters, exonic regions, immunoglobulin switch regions, telomeres, etc. Undeniably, these structures have been the subject of curiosity for many decades, leading to mounting evidence suggestive of their involvement in key cellular processes. Herein, we identified a 45-mer G-rich segment containing G5 tracts, present at the intersection of the promoter and

Several proteins, such as polymeric immunoglobulin (Ig) receptor/secretory component (pIgR/SC), immunoglobulins (Igs) and joining (J) chain, and cellular components of the secretory immune system (SIS) of the human embryo and fetus were analyzed and compared with reviewed information concerning SIS organization and function. All organs and structures, including extracorporeal ones, of 18 embryos (4-8 weeks of development) and 45 fetuses (9-38 weeks) were studied using methods of pathomorphology, immunohistochemistry and morphometry. This approach enabled us to analyze the problem in the whole

A weakness in glycobiology research is the insufficient integration of approaches used to connect glycan structure to function. From glycogene knockouts to understanding glycan structures, dynamics, and interactions, to determining whether a single glycan structure underlies a specific cellular function, no single approach provides sufficient information to establish these relationships. We argue that resolving glycan structure-function relationships requires the integration of complementary approaches spanning genetics, analytical glycomics, glycoengineering, structural biology, and computati

Neuronal stimulation is an emerging field in modern medicine to control organ function and reestablish physiological homeostasis during illness. The nervous system innervates most of the peripheral organs and provides a fine tune to control the immune system. Most of these studies have focused on vagus nerve stimulation and the physiological, cellular and molecular mechanisms regulating the immune system. Here, we review the new results revealing afferent vagal signaling pathways, immunomodulatory brain structures, spinal cord-dependent circuits, neural and non-neural cholinergic/catecholamine

Stages of fetal and postnatal development of the normal human chamber angle are reevaluated. Major growth phases can be detected: trabecular anlage formation (around the 15th week), differentiation into definitive structures (around the 24th week), specialisation of the definitive structures (around the 28th week), achievement of final components (birth) and final moulding of the chamber angle and the maturation of its cellular and extracellular constituents (1-8 years). A cleavage or gradual atrophy of chamber angle tissues could not be detected during any of the above phases, nor was a membr

In the slowly progressive neurodegenerative disorders like Parkinson's disease and Alzheimer's disease very different neuronal populations undergo degenerative processes, although the cascades of cellular events leading to death are supposed to be similar. We suggest that the complex pattern of degeneration in Parkinson's disease depends on two processes, a 'primary neurodegeneration' that takes place in the striato-nigral dopamine neurons and a 'secondary degeneration', occurring in distant structures of the basal ganglia network. For the purpose of explaining the regionally different express

The role of primary cilia has recently garnered significant attention in the field of neurodegeneration. This review explores the diversity of primary cilia in the mature brain and their interrelationships with a multitude of cellular structures, including axons and synapses. Importantly, an overview of the growing prominence of ciliary-related dysfunctions in neurodegenerative diseases is summarized, with a special emphasis on Parkinson's disease (PD) and neuropsychiatric disorders.

The cryopreservation of cells and human tissues has generated a great interest from the scientific community since 1949 when the cryoprotective activity of glycerol was discovered. For a homogeneous cellular group or a one-layer cellular tissue it is easy to define the optimal technique conditions of its cryopreservation (cryoprotective agents, speed and steps of freezing, speed of warming). It is considered successful when a high recovery of the cellular structures and tissue components after warming is achieved. The cryopreservation of a whole composite tissue is less easy to obtain. The intercellular communication of mechanotransduction has a significant impact on various cellular processes. Tunneling nanotubes (TNTs) have been documented to possess the capability of transmitting mechanical stimulation between cells, thereby triggering an influx of Ca2+ ions. However, the related kinetic information on the TNT-mediated intercellular mechanotransduction communication is still poorly explored. Herein, we developed a classic and sensitive Pt-functionalized carbon fiber microelectrochemical sensor (Pt/CF) to study the intercellular communication of endothelial mechanotransduc
