Absence of endo-1,4-β-glucanase KOR1 alters the jasmonate-dependent defence response to Pseudomonas syringae in Arabidopsis.

During plant-pathogen interactions, the plant cell wall forms part of active defence against invaders. In recent years, cell wall-editing enzymes, associated with growth and development, have been related to plant susceptibility or resistance. Our previous work identified a role for several tomato and Arabidopsis endo-1,4-β-glucanases (EGs) in plant-pathogen interactions. Here we studied the response of the Arabidopsis thaliana T-DNA insertion mutant lacking EG Korrigan1 (KOR1) infected with Pseudomonas syringae. KOR1 is predicted to be an EG which is thought to participate in cellulose biosyn

Chitosan-based nanoparticles for topical genetic immunization.

Numerous studies have reported the prophylactic and therapeutic use of genetic vaccines for combating a variety of infectious diseases in animal models. Recent human clinical studies with the gene gun have validated the concept of direct targeting of dendritic cells (Langerhan's cells) in the viable epidermis of the skin. However, it is unclear whether the gene gun technology or other needle-free devices will become commercially viable. The objective of our studies was to investigate the topical application of chitosan-based nanoparticles containing plasmid DNA (pDNA) as a potential approach t

High-throughput droplet screening of filamentous fungi on solid substrates.

Droplet-based microfluidics has been frequently used for high-throughput single-cell microbial screening, where individual cells are encapsulated into nanoliter or picoliter-sized droplets and sorted based on measurable phenotypes such as fluorescent proteins or enzyme activity using fluorescence or absorbance-based substrates. However, using droplet microfluidics with filamentous fungi poses a unique challenge: fungal strains tend to escape from droplets as their hyphae grow, causing individual droplets to merge and eventually burst which prevents measuring individual phenotypes. In our work,

PGPR-Treated Spent Mushroom Substrate Enhances Lignocellulose Degradation, Enzyme Activities, and Microbial Restructuring to Sustain Blueberry Rhizosphere Fertility.

Spent mushroom substrate (SMS) is a major agricultural byproduct whose complex lignocellulosic matrix hinders direct reuse and poses environmental risks when stockpiled. This study evaluated whether pretreatment with plant growth-promoting rhizobacteria (PGPR) could enhance SMS as a soil amendment for blueberry cultivation. Two PGPR-treated SMS formulations, along with raw SMS and a blank control, were applied to blueberry seedlings in a 10-month greenhouse experiment. Plant height, rhizosphere soil nutrients, enzyme activities, lignocellulose fractions, and the microbial communities were moni

Fe2O3@D201 Enhanced Efficiency of Food Waste Degradation by Microbial Inoculum Under Aerobic Condition.

The global quantity of food waste (FW) is increasing at an alarming rate, making safe disposal a pressing issue in urban management. The inappropriate disposal of FW will put risks on health and environment. Aerobic degradation equipment has emerged as a promising solution for FW disposal by adding microbial agents. However, current equipment faces challenges such as long processing duration and low efficiency. Therefore, we investigated the impact of combining microbial agents with iron oxide nano-resin (Fe2O3@D201) on the aerobic degradation of FW. We conducted experiments using 10% microbia

Simultaneous stabilization of high-concentration multi-heavy metal(loid)s co-contaminated soil using Fe/Mn (hydr)oxide-humic acid composites:A novel approach towards sustainable environmental remediation.

In this work, a green and highly efficient functional material (FMHs) was developed through the precise optimization of Fe2+, Fe3+, Mn2+, and humic acid (HA) ratios. The stabilization efficacy of FMHs on the high-concentration Cd-Pb-Cu-Zn-As-Sb co-contaminated soil and its potential ecological impacts were comprehensively evaluated from multiple perspectives. After 60 days of soil remediation with 5 wt% FMHs, the extractable Cd, Pb, Cu, Zn, As, and Sb were reduced by 59.06 %, 99.83 %, 92.60 %, 92.43 %, 98.06 %, and 77.04 %, respectively. These reductions were primarily attributed to the FMHs-d

[Isolation of protoplasts from vegetable tissues using extracellular lytic enzymes from fusarium oxysporum f.sp. melonis].

Fusarium oxysporum f.sp. melonis, a pathogen of melon (Cucumis melo L.), was grown in shaken cultures at 26 degrees C in a mineral salts medium containing glucose, xylan and apple pectin as carbon sources. The extracellular enzymic complex obtained from these cultures showed lytic activity on plant tissues, causing maceration of melon fruits, potato tubers and carrot roots. Protoplasts were isolated from melon fruits when the maceration was carried out under appropriate osmotic conditions. This fact suggest a possible relationship between the enzymes produced by Fusarium oxysporum f.sp. meloni

Hologenomics of xylotrophic bivalves reveals a minimalist, remote-acting evolutionary strategy of wood digestion.

Wood constitutes the largest reservoir of biogenic carbon on Earth, yet remarkably few animals can exploit it. While terrestrial wood-feeders like termites rely on highly diverse gut microbiomes, xylotrophic marine bivalves have evolved a fundamentally different approach: a spatially segregated system where intracellular gill symbionts produce enzymes that act remotely within a nearly sterile cecum. However, the genetic and evolutionary basis of this unique symbiosis remains largely elusive. Here, we integrate hologenomics, transcriptomics, and biochemistry of a shallow-water shipworm ( Teredo

A multifunctional Streptomyces murinus bioinoculant integrates immune priming and microbiome restructuring for sustainable control of Sclerotinia minor in Brassica rapa.

BACKGROUND: Harnessing beneficial soil microbes provides an eco-friendly alternative to pesticides for sustainable crop protection. Members of the genus Streptomyces combine broad antagonistic activity with plant immune priming and growth promotion, yet their ecological roles and field reliability remain insufficiently understood. We investigated the soil actinobacterium Streptomyces murinus JS029 in the Brassica rapa-Sclerotinia minor pathosystem to elucidate its mechanisms of pathogen suppression, plant immune activation and rhizosphere microbiome modulation, and to develop a field-deployabl
