Constrained S-estimators for linear mixed effects models with covariance components.

Linear mixed effects (LME) models are increasingly used for analyses of biological and biomedical data. When the multivariate normal assumption is not adequate for an LME model, then a robust estimation approach is preferable to the maximum likelihood one. M-estimators were considered before for robust estimation of the LME models, and recently a constrained S-estimator was proposed. This S-estimator cannot be applied directly to LME models with correlated error terms and vector random effects with correlated dimensions. Therefore, a modification is proposed, which extends application of the c

Race and emotion in computer-based HIV prevention videos for emergency department patients.

Computer-based video provides a valuable tool for HIV prevention in hospital emergency departments. However, the type of video content and protocol that will be most effective remain underexplored and the subject of debate. This study employs a new and highly replicable methodology that enables comparisons of multiple video segments, each based on conflicting theories of multimedia learning. Patients in the main treatment areas of a large urban hospital's emergency department used handheld computers running custom-designed software to view video segments and respond to pre-intervention and pos

How Well Do Commonly Used Co-contraction Indices Approximate Lower Limb Joint Stiffness Trends During Gait for Individuals Post-stroke?

Muscle co-contraction generates joint stiffness to improve stability and accuracy during limb movement but at the expense of higher energetic cost. However, quantification of joint stiffness is difficult using either experimental or computational means. In contrast, quantification of muscle co-contraction using an EMG-based Co-Contraction Index (CCI) is easier and may offer an alternative for estimating joint stiffness. This study investigated the feasibility of using two common CCIs to approximate lower limb joint stiffness trends during gait. Calibrated EMG-driven lower extremity musculoskel

Methodology for Estimation of Annual Risk of Rupture for Abdominal Aortic Aneurysm.

BACKGROUND AND OBJECTIVE: Estimating patient specific annual risk of rupture of abdominal aortic aneurysm (AAA) is currently based only on population. More accurate knowledge based on patient specific data would allow surgical treatment of only those AAAs with significant risk of rupture. This would be beneficial for both patients and health care system. METHODS: A methodology for estimating annual risk of rupture (EARR) of abdominal aortic aneurysms (AAA) that utilizes Bayesian statistics, mechanics and patient-specific blood pressure monitoring data is proposed. EARR estimation takes into c

The Effect of Cranial Sutures Should Be Considered in Transcranial Electrical Stimulation.

BACKGROUND: Computational modeling is used to optimize transcranial electrical stimulation (tES) approaches, and the precision of these models is dependent on their anatomical accuracy. We are unaware of any computational modeling of tES that has included cranial sutures. OBJECTIVES: The aims of the study were to review the literature on the timing of closure of the coronal and squamous sutures, which are situated under electrode placements used in tES; to review the literature regarding differences in skull and suture conductivity and to determine a more accurate conductivity for sutures; an

Comparison of hemodynamic endpoints between normal subject and tetralogy patient using Womersley velocity profile and MR based flow measurements.

Right ventricular (RV) enlargement and pulmonary valve insufficiency (PI) are well-known, unavoidable long term sequelae encountered by patients who undergo tetralogy of Fallot (TOF) surgery. Despite their lifelong need for cardiac surveillance and occasional re-intervention, there is a paucity of numerical data characterizing blood flows in their pulmonary arteries (PA). Specifically, although PA regurgitation is well-known to be ubiquitously present in adult repaired TOF (rTOF) patients yet, there have been only limited numerical studies to fully characterize this process. The few studies av

Craniofacial Superimposition: Historical Review and Current Issues.

Methodology of craniofacial superimposition has evolved dramatically since its inception in the early 20th century. Contemporary approaches involve advanced procedures of digitization, use of video cameras, warping algorithms, skull positioning devices and data derived from computed tomography, radiography, and ultrasound technology. Applications have proven most useful in exclusion and casework frequency has declined in some regions in recent years as molecular approaches to identification have become more widely available. Additional research is needed to clarify the probabilities involved a

Radiologic diagnosis of osteoporosis. Current methods and perspectives.

Osteoporosis is defined as a decrease in bone mass accompanied by structural changes, leading to an increase in fracture propensity. Early diagnosis of osteoporosis, fracture risk prediction, and assessment of efficacy of therapy therefore are of great interest. A number of noninvasive techniques are available for measuring bone mass at multiple sites of the skeleton. This article reviews basic methodology and developments in radiology such as x-ray and photon absorptiometry and quantitative computed tomography, which are routinely used in clinical practice. Recent techniques for assessment of

Immunoscore and beyond: evolution and clinical integration of immune contexture-based biomarkers across solid tumours.

The tumour immune microenvironment is increasingly recognised as a critical determinant of cancer progression, prognosis, and therapeutic response, challenging the sufficiency of anatomy-based staging systems such as TNM. The Immunoscore (IS), originally validated in colorectal cancer, provides a standardised, spatially resolved assessment of CD3+ and CD8+ T cells within the tumour core (TC) and invasive margin (IM). Expanding upon this framework, adapted and modified IS models, as well as computational and imaging-based surrogates, have been investigated across multiple solid malignancies, in

Variation in center of mass estimates for extant sauropsids and its importance for reconstructing inertial properties of extinct archosaurs.

Inertial properties of animal bodies and segments are critical input parameters for biomechanical analysis of standing and moving, and thus are important for paleobiological inquiries into the broader behaviors, ecology and evolution of extinct taxa such as dinosaurs. But how accurately can these be estimated? Computational modeling was used to estimate the inertial properties including mass, density, and center of mass (COM) for extant crocodiles (adult and juvenile Crocodylus johnstoni) and birds (Gallus gallus; junglefowl and broiler chickens), to identify the chief sources of variation and

Comparison of quantitative methodologies to define chronic pressure ulcer measurements.

The objective of this study was to evaluate and compare various methodologies of measuring the characteristics of pressure ulcers. This prospective, four-week, follow-up study consisted of 20 patients, of whom 17 completed the study. Each patient had at least one full-thickness pressure ulcer (surface area between 1.2 and 61.6 cm2) that had been present for at least four weeks. The ulcers were assessed weekly for four weeks using the following techniques: direct measurement (length, width, and depth), tracing of the ulcer outline onto transparent material, standard photography, and volume meas

Chromatin Immunoprecipitation Followed by Next-Generation Sequencing in Ewing Sarcoma.

Chromatin immunoprecipitation followed by sequencing (ChIP-seq) is a widely used technique to study the distribution of transcription factors, histones, posttranslational modifications, or other DNA-associated proteins across the genome. This methodology is based on the isolation of DNA fragments bound by the protein of interest using specific antibodies. Computational analysis of sequencing libraries derived from the isolated DNA fragments will delineate genome-wide maps of protein-DNA interactions. ChIP-seq profiling of EWSR1::FLI1 oncogene has enormously contributed to the understanding of

An Anomaly Self-Supervised Representation To Classify Malignant Lung Nodules.

Nodules are the primary biomarker for diagnosing and stratifying lung cancer malignancy. From CT scans, it is possible to characterize these abnormal masses, but analysis remains expert-dependent with remarkable challenges due to the typical morphological and textural variability. Computer-aided disease methodologies have recently shown capability support to classify lung nodules, yet they remain limited due to the following of classical supervised and balanced schemes, which results unrealistic, considering that less than 10% of the nodules are malignant. This proposed method takes advantage

Estimating the reliability of repeatedly measured endpoints based on linear mixed-effects models. A tutorial.

There are various settings in which researchers are interested in the assessment of the correlation between repeated measurements that are taken within the same subject (i.e., reliability). For example, the same rating scale may be used to assess the symptom severity of the same patients by multiple physicians, or the same outcome may be measured repeatedly over time in the same patients. Reliability can be estimated in various ways, for example, using the classical Pearson correlation or the intra-class correlation in clustered data. However, contemporary data often have a complex structure t

A protocol for computational design of mRNA vaccines with high functionality and specificity.

The design of effective mRNA vaccines requires rigorous target selection, optimization, and validation to ensure stability, immunogenicity, specificity, and overall efficacy. A standardized protocol is needed to guide the process from gene selection to in silico optimization of the final vaccine construct. This protocol provides a comprehensive computational framework integrating all stages of mRNA vaccine design by defining the design space, essential criteria, and a step-by-step methodology. To demonstrate its applicability, the protocol is applied to the SARS-CoV-2 spike protein, targeting

Diagnostic accuracy of positive oral contrast in abdominal computed tomography for non-traumatic pathologies: a systematic review protocol.

OBJECTIVE: The objective of this review is to determine the diagnostic accuracy of positive oral contrast versus no oral contrast for common, non-traumatic computed tomography abdominal indications in adults. INTRODUCTION: Positive oral contrast is regularly administered to patients to drink prior to computed tomography scans of the abdomen and pelvis. The purpose of the preparation is to improve visualization of the bowel, however, technological advances in scanner design may mean positive oral contrast is no longer required. INCLUSION CRITERIA: Eligible studies will consider the use of pos
