Comparative plastome analysis of the Vandenboschia radicans complex in Korea: insights into its evolutionary history.

BACKGROUND: Vandenboschia radicans and its close relatives are difficult to distinguish based on morphological characteristics because of reticulate evolution within the complex. Therefore, cytological and molecular approaches have been proposed for accurate identification of taxa within this complex. However, conventional plastid markers lack sufficient discriminatory power at the species level. In Korea, the V. radicans complex comprises nine genotypes associated with varying ploidy levels and hybrid origins. The maternal genotypes are classified into three groups: α, β, and γ. In this study

Landscape genomics: natural selection drives the evolution of mitogenome in penguins.

BACKGROUND: Mitochondria play a key role in the balance of energy and heat production, and therefore the mitochondrial genome is under natural selection by environmental temperature and food availability, since starvation can generate more efficient coupling of energy production. However, selection over mitochondrial DNA (mtDNA) genes has usually been evaluated at the population level. We sequenced by NGS 12 mitogenomes and with four published genomes, assessed genetic variation in ten penguin species distributed from the equator to Antarctica. Signatures of selection of 13 mitochondrial prote

Climate-extreme soil exposome: how drought converts contaminated agricultural soils into one-health risk hotspots.

Contaminated agricultural soils are conventionally treated as static pollutant repositories whose hazard state is adequately captured by total contaminant concentration, an assumption that intensifying drought regimes are rendering untenable. This review advances the concept of a climate-sensitive soil exposome, in which drought functions as a hydro-biogeochemical driver that reorganizes the bioavailability, speciation, and biological accessibility of metals, antibiotics, pesticides, per- and polyfluoroalkyl substances, and microplastics without altering their total soil inventories. By reduci

Insights Into Divergent Mechanisms of Pyrite Oxidation by Indigenous Fungi From Mine Tailings.

Fungi can oxidise pyrite, a process which has potential implications for metal bioleaching and acid mine drainage formation. However, this capacity has been demonstrated primarily using only a few model fungal strains. Diverse fungal communities are known to inhabit pyrite-rich mine tailings but how these indigenous fungi may mediate pyrite oxidation and by what mechanisms remain largely unexplored. Here, we isolated 51 pyrite-oxidizing fungal strains spanning 28 genera from three mine tailings areas in South China. From this native library, we selected three potent pyrite oxidisers, Pithomyce

A One Health Molecular Investigation and Exploratory Epidemiological Analysis of Cryptosporidium spp. in A Shared Human-Animal Ecosystem.

INTRODUCTION: Cryptosporidium spp. are enteric protozoan parasites of public health importance, particularly in low-resource settings where humans, animals and environmental water sources frequently interact. This study investigated the prevalence, molecular diversity and environmental exposure-related variables in relation to Cryptosporidium infection in a rural community. METHODS: A cross-sectional study was conducted using 180 faecal samples collected from humans, cattle, chickens, fish, cats, dogs, and ducks. Speciation and subtyping were determined through PCR amplification and sequencin

Copper speciation and total essential trace element levels in Wilson's disease.

Wilson's disease (WD) is a rare genetic disorder characterized by impaired Cu metabolism in the body. This study aimed to compare the total levels of essential trace elements (Cr, Mn, Fe, Co, Cu, Zn, Se, and Mo) in plasma and urine samples of WD-treated patients (cases) and healthy controls, and to conduct Cu speciation in plasma samples. Inductively coupled plasma mass spectrometry (ICP-MS) was used for total element concentration, while high-performance liquid chromatography (HPLC) coupled to ICP-MS (HPLC-ICP-MS) was used to determine the following Cu species: Cu-ceruloplasmin (Cu-Cp), Cu-hu

Molecular phylogeny of Boninia (Platyhelminthes: Polycladida), with description of a new species from the Pacific coasts of Panama.

Mesopsammic polyclad members in the family Boniniidae have attracted attention in terms of their evolutionary shifts of microhabitat and their unique morphology such as a pair of pointed tentacles extending from the anterolateral margins and prostatoid organs harbouring stylets. Here, we establish a new species of this family as Boninia panamensis sp. nov. from the Pacific coasts of Panama, based on its morphological characteristics of (i) four cerebral and 61-80 marginal eyespots, (ii) two prostatoid organs located anterior and posterior to the penis papilla, and (iii) two uterine canals depa

Ecological and geographical speciation in Lucilia bufonivora: The evolution of amphibian obligate parasitism.

Lucilia (Diptera: Calliphoridae) is a genus of blowflies comprised largely of saprophagous and facultative parasites of livestock. Lucilia bufonivora, however, exhibits a unique form of obligate parasitism of amphibians, typically affecting wild hosts. The evolutionary route by which amphibian myiasis arose, however, is not well understood due to the low phylogenetic resolution in existing nuclear DNA phylogenies. Furthermore, the timing of when specificity for amphibian hosts arose in L. bufonivora is also unknown. In addition, this species was recently reported for the first time in North Am

Phenotypic plasticity in haptoral structures of Ligophorus cephali (Monogenea: Dactylogyridae) on the flathead mullet (Mugil cephalus): a geometric morphometric approach.

Evaluating phenotypic plasticity in attachment organs of parasites can provide information on the capacity to colonise new hosts and illuminate evolutionary processes driving host specificity. We analysed the variability in shape and size of the dorsal and ventral anchors of Ligophorus cephali from Mugil cephalus by means of geometric morphometrics and multivariate statistics. We also assessed the morphological integration between anchors and between the roots and points in order to gain insight into their functional morphology. Dorsal and ventral anchors showed a similar gradient of overall s

Description of the first cryptic avian malaria parasite, Plasmodium homocircumflexum n. sp., with experimental data on its virulence and development in avian hosts and mosquitoes.

For over 100 years studies on avian haemosporidian parasite species have relied on similarities in their morphology to establish a species concept. Some exceptional cases have also included information about the life cycle and sporogonic development. More than 50 avian Plasmodium spp. have now been described. However, PCR-based studies show a much broader diversity of haemosporidian parasites, indicating the possible existence of a diverse group of cryptic species. In the present study, using both similarity and phylogenetic species definition concepts, we believe that we report the first char

Contrasting patterns in mammal-bacteria coevolution: bartonella and leptospira in bats and rodents.

BACKGROUND: Emerging bacterial zoonoses in bats and rodents remain relatively understudied. We conduct the first comparative host-pathogen coevolutionary analyses of bacterial pathogens in these hosts, using Bartonella spp. and Leptospira spp. as a model. METHODOLOGY/PRINCIPAL FINDINGS: We used published genetic data for 51 Bartonella genotypes from 24 bat species, 129 Bartonella from 38 rodents, and 26 Leptospira from 20 bats. We generated maximum likelihood and Bayesian phylogenies for hosts and bacteria, and tested for coevoutionary congruence using programs ParaFit, PACO, and Jane. Barton

Adaptation to aridity in the malaria mosquito Anopheles gambiae: chromosomal inversion polymorphism and body size influence resistance to desiccation.

Chromosomal inversions are thought to confer a selective advantage in alternative habitats by protecting co-adapted alleles from recombination. The frequencies of two inversions (2La and 2Rb) of the afro-tropical malaria mosquito Anopheles gambiae change gradually along geographical clines, increasing in frequency with degree of aridity. Such clines can result from gene flow and local selection acting upon alternative karyotypes along the cline, suggesting that these inversions may be associated with tolerance to xeric conditions. Since water loss represents a major challenge in xeric habitats

Mitogenomic Insights Into the Ancient Split and Cryptic Diversity of the Poecilochirus carabi Complex (Acari: Mesostigmata).

Phoretic associations with insects are widespread in the Mesostigmata. These temporary symbiotic associations have been important in driving the evolutionary development of this taxon. The phoretic mite genus Poecilochirus, which is considered obligately associated with burying beetles, is of great value in forensic entomology. However, due to the morphological conservatism and the difficulty of obtaining adult specimens, the taxonomy of this genus has long been based on deutonymphal characters, and the true species diversity is far from resolved. Here we used next-generation sequencing to det

Postnatal Abrogation of VEGFR2 Blocks Terminal Cap2 Differentiation by Preventing the Developmental Progression from a Capillary Intermediate Cell State.

After birth, the alveolar capillary network expands to increase gas exchange surface area and endothelial-derived signals promote alveolarization. Lung capillaries are comprised of two distinct subsets, one with proliferative potential to facilitate growth and repair (Cap1), and the other serving a specialized role in gas exchange (Cap2). However, the molecular mechanisms directing capillary speciation, developmental plasticity, and fate transitions during development and repair are not well understood. Here, we show that Cap2 are absent in late embryonic life but rapidly appear and expand imm

Adaptation of CD4 in gorillas and chimpanzees conveyed resistance to simian immunodeficiency viruses.

Simian immunodeficiency viruses (SIVs) comprise a large group of primate lentiviruses that endemically infect African monkeys. HIV-1 spilled over to humans from this viral reservoir, but the spillover did not occur directly from monkeys to humans. Instead, a key event was the introduction of SIVs into great apes, which then set the stage for infection of humans. Here, we investigate the role of the lentiviral entry receptor, CD4, in this key and fateful event in the history of SIV/HIV emergence. First, we reconstructed and tested ancient forms of CD4 at two important nodes in ape speciation, b

Up-regulated Pro-inflammatory MicroRNAs (miRNAs) in Alzheimer's disease (AD) and Age-Related Macular Degeneration (AMD).

Alzheimer's disease (AD) of the brain neocortex and age-related macular degeneration (AMD) of the retina are two complex neurodegenerative disorders, which (i) involve the progressive dysregulation and deterioration of multiple neurobiological signaling pathways, (ii) exhibit the temporal accumulation of pro-inflammatory lesions including the amyloid beta (Aβ) peptide-containing senile plaques of AD and the drusen of AMD, and (iii) culminate in an insidious inflammatory neurodegeneration ending, respectively, in neural cell atrophy and death and progressive loss of cognition and central visual

Follicle-stimulating hormone receptor (FSHR) alternative skipping of exon 2 or 3 affects ovarian response to FSH.

Genes critical for fertility are highly conserved in mammals. Interspecies DNA sequence variation, resulting in amino acid substitutions and post-transcriptional modifications, including alternative splicing, are a result of evolution and speciation. The mammalian follicle-stimulating hormone receptor (FSHR) gene encodes distinct species-specific forms by alternative splicing. Skipping of exon 2 of the human FSHR was reported in women of North American origin and correlated with low response to ovarian stimulation with exogenous follicle-stimulating hormone (FSH). To determine whether this var

Methods for Individualized Determination of Methylmercury Elimination Rate and De-Methylation Status in Humans Following Fish Consumption.

Methylmercury (MeHg) exposure via fish in the diet remains a priority public health concern. Individual variation in response to a given MeHg exposure and the biotransformation of MeHg that follows complicate our understanding of this issue. MeHg elimination from the human body occurs slowly (elimination rate (kel) approximately 0.01 day(-1) or approximately 70 days half-life [t1/2]) and is a major determinant of the Hg body burden resulting from fish consumption. The underlying mechanisms that control MeHg elimination from the human body remain poorly understood. We describe here improved met

[On the origin of Yersinia pestis, a causative agent of the plague: A concept of population-genetic macroevolution in transitive environment].

An ecological scenario is proposed for the origin of causative agent of the plague (the bacterium Yersenia pestis) from the clone of pseudotuberculous microbe of the first serotype Y. pseudotuberculosis O:1b. Disclosed are the conditions of gradual intrusion of psychrophile saprozoonosis ancestor into the blood of the primary host, Mongolian tarbagan marmot Marmota sibirica. As an inductor of speciation acted the Sartan cooling that occurred in the end of late Pleistocene under conditions of arid ultra-continental climate in Central Asia.
