Genome-wide analyses of cassava Pathogenesis-related (PR) gene families reveal core transcriptome responses to whitefly infestation, salicylic acid and jasmonic acid.

BACKGROUND: Whiteflies are a threat to cassava (Manihot esculenta), an important staple food in many tropical/subtropical regions. Understanding the molecular mechanisms regulating cassava's responses against this pest is crucial for developing control strategies. Pathogenesis-related (PR) protein families are an integral part of plant immunity. With the availability of whole genome sequences, the annotation and expression programs of the full complement of PR genes in an organism can now be achieved. An understanding of the responses of the entire complement of PR genes during biotic stress a

Children consuming cassava as a staple food are at risk for inadequate zinc, iron, and vitamin A intake.

Cassava contains little zinc, iron, and beta-carotene, yet it is the primary staple crop of over 250 million Africans. This study used a 24-hour dietary recall to test the hypothesis that among healthy children aged 2-5 years in Nigeria and Kenya, cassava's contribution to the childrens' daily diets is inversely related to intakes of zinc, iron, and vitamin A. Dietary and demographic data and anthropometric measurements were collected from 449 Kenyan and 793 Nigerian children. Among Kenyan children 89% derived at least 25% of their dietary energy from cassava, while among the Nigerian children

New insight into the molecular etiopathogenesis of konzo: Cyanate could be a plausible neurotoxin contributing to konzo, contrary to thiocyanate.

INTRODUCTION: Chronic cassava-derived cyanide poisoning is associated with the appearance of konzo, a tropical spastic paraparesis due to selective upper motor neuron damage. Whether the disease is caused by a direct action of cyanide or its metabolites is still an open question. This preliminary study assessed the neurotoxic effects of thiocyanate (SCN) and cyanate (OCN), two cyanide metabolites hypothesized to be plausible toxic agents in konzo. METHODS: Cultured mouse neuroblastoma (Neuro-2A) and human neuroblastoma (SH-SY5Y) cell lines were incubated (24, 48, and 72 hours) in sodium OCN o

Effect of cyanide ions (CN-) extracted from cassava (Manihotesculenta Crantz) on Alveolar Epithelial Cells (A549 cells).

Cassava (Manihotesculenta Crantz) is one of the most important root crops in tropical countries. It is a major source of cyanogenic glycosides viz. linamarin and lotaustralin, and these on breakdown liberate HCN and ketone. Cassava cyanide extract (CCE) from cassava leaves and tuber rinds were formulated as a biopesticide against certain borer insect pests of horticultural crops. Adenocarcinomic human alveolar basal epithelial cells (A549) were treated with three different concentrations (100, 200, 400 ppm) of CCE. The MTT and NRU assays showed dose-dependent cytotoxicity. The DCFH-DA assay do

Switching to a "turn-on" fluorescent probe for selective monitoring of cyanide in food samples and living systems.

A fluorescent probe (TPA-BTD-MT) was designed to monitor cyanide ions (CN-) with a "turn-on" response, changing from "turn-off" behavior due to the structural change. TPA-BTD-MT exhibited high selectivity for sensing CN- in several food samples and was successfully used for imaging CN- in living cells and animals with strong "turn-on" fluorescence.

Different roughage:concentrate ratios with and without liquid residue of cassava for lambs.

This aim of this study was to evaluate the intake and apparent digestibility of nutrients, ingestive behavior, blood parameters, and performance of lambs fed two roughage:concentrate (R:C) ratios, with or without liquid residue of cassava (LRC). Forty lambs (19.5 ± 1.45 kg body weight) were distributed in a completely randomized 2 × 2 factorial design. The intake of dry matter (1311 g/day) and total digestible nutrients (755 g/day), and the intake (172 g/day) and digestibility (552 g/kg) of crude protein were higher for lambs fed R:C of 40:60. The R:C ratio of 80:20 promoted a higher intake (5

Occupational exposure to hydrogen cyanide during large-scale cassava processing, in Alagoas State, Brazil.

The cassava roots used for flour production contain high amounts of cyanogenic glycosides and are, therefore, potential hydrogen cyanide (HCN) releasers. This fact is the cause of an increasing health concern in the sector of cassava processing. Brazilian workers engaged in the flour production may be chronically exposed to HCN in levels above the safety limits. This hypothesis is based on the drastic reduction in cyanide content of cassava roots during a traditional Brazilian method of processing and in the physical properties of the compound, which makes it very susceptible to volatilization

Bioactivation of cyanide to cyanate in sulfur amino acid deficiency: relevance to neurological disease in humans subsisting on cassava.

Neurological disorders have been reported from parts of Africa with protein-deficient populations and attributed to cyanide (CN-) exposure from prolonged dietary use of cassava, a cyanophoric plant. Cyanide is normally metabolized to thiocyanate (SCN-) by the sulfur-dependent enzyme rhodanese. However, in protein-deficient subjects where sulfur amino acids (SAA) are low, CN may conceivably be converted to cyanate (OCN-), which is known to cause neurodegenerative disease in humans and animals. This study investigates the fate of potassium cyanide administered orally to rats maintained for up to

Overexpression of hydroxynitrile lyase in cassava roots elevates protein and free amino acids while reducing residual cyanogen levels.

Cassava is the major source of calories for more than 250 million Sub-Saharan Africans, however, it has the lowest protein-to-energy ratio of any major staple food crop in the world. A cassava-based diet provides less than 30% of the minimum daily requirement for protein. Moreover, both leaves and roots contain potentially toxic levels of cyanogenic glucosides. The major cyanogen in cassava is linamarin which is stored in the vacuole. Upon tissue disruption linamarin is deglycosylated by the apolplastic enzyme, linamarase, producing acetone cyanohydrin. Acetone cyanohydrin can spontaneously de

Thyroid function in a cassava-eating population affected by epidemic spastic paraparesis.

Thyroid function was studied in a rural population in Mozambique that had been affected by an epidemic of spastic paraparesis attributed to dietary cyanide exposure from cassava. Laboratory investigation on a sample of this population demonstrated very high levels of serum and urinary thiocyanate, indicating a heavy exposure to cyanide. The urinary excretion of iodine was within normal limits, indicating an adequate intake of iodine. The serum levels of FT4I were somewhat decreased and serum FT3I, T3/T4 ratio and TSH were somewhat raised. This hormone pattern suggests an adaptation to the anti

Rational design and application of a fluorogenic chemodosimeter for selective detection of cyanide in an aqueous solution via excimer formation.

A new quinone-benzothiazole imine based chemodosimeter (R) was rationally designed, synthesized and characterized using NMR and mass spectral techniques. The receptor colorimetrically senses cyanide in aq. HEPES buffer: DMF (2:8 v/v) solution with an instantaneous colour change from yellow to bluish green. An enhancement of fluorescence intensity at 429 nm with excimer formation is also observed after addition of cyanide to the receptor, which is accompanied with a colour change from yellow to blue under UV lamp. Nucleophilic addition of cyanide to imine C-atom inhibits intra-molecular charge
