Cyclase-associated protein 1 facilitates actin-dependent mitochondrial fission in podocytes associated with diabetic nephropathy.

Aberrant mitochondrial dynamics in podocytes are closely associated with the progression of diabetic nephropathy (DN); however, the underlying mechanisms remain incompletely understood. Recent research has demonstrated that cyclase-associated protein 1 (CAP1), an actin-binding protein, mediates mitochondrial fission. Therefore, we aimed to explore the regulatory role and mechanisms of CAP1 in high glucose (HG)-induced mitochondrial dysfunction in podocytes. CAP1 knockout reduced the severity of glomerular injuries and curtailed mitochondrial fission in podocytes in streptozotocin (STZ)-induced

Loss of Cyclin G-Associated Kinase Leads to Lysosome Dysfunction and Immune Modulation in Podocytes.

KEY POINTS: Loss of podocyte-associated cyclin G-associated kinase resulted in mistrafficking of lysosomal hydrolases. Loss of podocyte-associated cyclin G-associated kinase induced immune modulation. BACKGROUND: Given the postmitotic nature of podocytes, adapting to both physiologic and pathologic stress is crucial to prevent podocyte loss. An important component of maintaining cellular homeostasis are lysosomes, which are membrane-bound organelles responsible for degradation and recycling of damaged organelles and other macromolecules. Lysosome impairment has been shown to cause cellular an

Dual blockade of DPP-4 and CXCL12/CXCR4 axes synergistically protects podocytes in lupus nephritis.

BACKGROUND: Lupus nephritis (LN), a severe complication of systemic lupus erythematosus (SLE), is characterized by podocyte injury that contributes to disease progression. Dipeptidyl peptidase-4 (DPP-4) inhibitors, though developed for diabetes, have shown renoprotective potential. However, DPP-4 inhibition may elevate CXCL12/CXCR4 signaling, a pathway implicated in LN pathogenesis. This study aimed to determine whether dual DPP-4 and CXCL12/CXCR4 blockade confers enhanced renal protection in LN. METHODS: MRL/lpr lupus-prone mice were treated with the DPP-4 inhibitor linagliptin, either alone

ANGPTL3 in Podocytes Contributes to the Pathogenesis of Lupus Nephritis by Activating MSR1 in Macrophages.

INTRODUCTION: Lupus nephritis (LN), a severe complication of systemic lupus erythematosus (SLE), is characterized by persistent immune dysregulation and glomerular injury predominantly driven by macrophage infiltration and type I interferon (IFN-I) signaling. Crosstalk between injured podocytes and infiltrating macrophages is increasingly recognized as a key contributor to LN progression; however, the molecular mediators and their corresponding receptors remain poorly defined. This study investigates whether ANGPTL3 upregulation in podocytes activates macrophages through MSR1 and drives interf

Defective autophagy in diabetic nephropathy sensitizes podocytes to hyperuricemia induced injury by amplifying inflammasome activation.

Hyperuricemia was associated with faster progression of proteinuria in diabetic nephropathy. However, the underlying mechanisms remained largely unknown. Therefore, this study aimed to explore whether there was a tightly regulated process involved in susceptibility to hyperuricemia under diabetic conditions. We established a diabetes model with hyperuricemia superimposed both in vivo and in vitro. The urinary albumin: creatinine ratio, biomarkers of podocyte damage, markers of inflammasome activation, and autophagosome markers were assessed. In vivo, we found that hyperuricemia aggravated podo

Podocytes as novel sources of neutrophil serine proteases: expression and regulation by inflammatory molecular patterns.

Podocytes are essential components of the glomerular filtration barrier and are increasingly recognized as immunologically active cells. Here, we demonstrate that human and rat podocytes express enzymatically active neutrophil serine proteases (NSPs), including neutrophil elastase, proteinase 3, and cathepsin G, as well as their endogenous inhibitors, serpins. We show that the expression and activity of these proteases are regulated by pathogen- and damage-associated molecular patterns. Notably, podocytes release NSPs in extracellular vesicles and secrete mitochondrial DNA in response to infla

Activation of the inflammasome and pyroptosis cascade in podocytes of patients with minimal change disease.

BACKGROUND: In contrast to childhood minimal change disease (MCD), adult-onset MCD frequently recurs and requires prolonged immunosuppressive therapy. Accordingly, an investigation of the pathogenesis of adult MCD is required. MCD is usually accompanied by severe dyslipidaemia. Oxidized low-density lipoprotein (ox-LDL) is known to function in a damage-associated molecular pattern (DAMP) through CD36, triggering the NOD-like receptor thermal protein domain-associated protein 3 (NLRP3) inflammasome and programmed cell death called pyroptosis. However, the relationship between MCD pathogenesis an

Effects of a novel ANLN E841K mutation associated with SRNS on podocytes and its mechanism.

BACKGROUND: Steroid-resistant nephrotic syndrome (SRNS) is characterized by unrelieved proteinuria after an initial 4-8 weeks of glucocorticoid therapy. Genes in podocytes play an important role in causing SRNS. OBJECTIVE: This study aimed to report a pathogenic mutation in SRNS patients and investigate its effects on podocytes, as well as the pathogenic mechanism. METHODS: We screened out a novel mutation by using whole-exon sequencing in the SRNS cohort and verified it via Sanger sequencing. Conservative analysis and bioinformatic analysis were used to predict the pathogenicity of the muta

Urolithins Modulate the Viability, Autophagy, Apoptosis, and Nephrin Turnover in Podocytes Exposed to High Glucose.

Urolithins are bioactive compounds generated in human and animal intestines because of the bacterial metabolism of dietary ellagitannins (and their constituent, ellagic acid). Due to their multidirectional effects, including anti-inflammatory, antioxidant, anti-cancer, neuroprotective, and antiglycative properties, urolithins are potential novel therapeutic agents. In this study, while considering the future possibility of using urolithins to improve podocyte function in diabetes, we assessed the results of exposing mouse podocytes cultured in normal (NG, 5.5 mM) and high (HG, 25 mM) glucose c

AKAP1 contributes to impaired mtDNA replication and mitochondrial dysfunction in podocytes of diabetic kidney disease.

Podocyte injury is involved in the onset and progression of diabetic kidney disease (DKD) and is associated with mitochondrial abnormalities. Defective mitochondrial DNA (mtDNA) replication results in mitochondrial dysfunction. However, whether podocyte mtDNA replication is impaired in DKD is still unclear. A-kinase anchoring protein 1 (AKAP1) is localized in the outer mitochondrial membrane (OMM) and acts as a regulator and conductor of mitochondrial signals. Herein, we investigated the role of AKAP1 in high glucose-induced mtDNA replication. Decreased mtDNA replication and mitochondrial dysf

The role of TRPC6 in α1-AR activation-induced calcium signal changes in human podocytes.

BACKGROUND: An accumulating amount of evidence has suggested that there is a contributive role of sympathetic nervous hyperactivity in the pathogenesis of chronic kidney disease (CKD). α1-AR promotes an increase in calcium levels in podocytes and adjusts podocyte contraction. Changes in TRPC6 expression and function can directly affect the podocyte cytoskeleton, which is a key component in podocyte injury. This study proposed to clarify the correlation between α1-AR activation-induced signal cascade reaction and TRPC6 in human podocytes. METHODS: Human podocytes were incubated with the calciu

Plasma Soluble Urokinase Receptor Level Is Correlated with Podocytes Damage in Patients with IgA Nephropathy.

BACKGROUND: Focal segmental glomerulosclerosis (FSGS) lesions are similar in characteristics to S lesions of the Oxford classification of IgA nephropathy (IgAN) and may predict poor prognosis. In the present study, we aimed to explore the association between plasma soluble urokinase receptor (suPAR) levels and S lesions and podocytes damage in IgAN patients. METHODS: We enrolled 569 IgAN patients with follow-up data and detected plasma suPAR levels at renal biopsy by enzyme-linked immunosorbent assay. RESULTS: Plasma suPAR levels in IgAN patients with or without S lesions did not differ sign

Nicorandil, a K(atp) channel opener, alleviates chronic renal injury by targeting podocytes and macrophages.

Nicorandil exhibits a protective effect in the vascular system, which is thought to be due to vasodilatation from opening ATP-dependent potassium channels and donation of nitric oxide. Recently, nicorandil was shown to be renoprotective in models of acute kidney injury and glomerulonephritis. However, the specific mechanisms of renoprotection are unclear. We evaluated the effect of nicorandil on the rat remnant kidney model of chronic kidney disease. Blood pressure was unchanged by a 10-wk course of nicorandil, while albuminuria was significantly reduced. Glomerular injury and tubulointerstiti

Planar cell polarity pathway regulates actin rearrangement, cell shape, motility, and nephrin distribution in podocytes.

Glomerular podocytes are highly polarized cells characterized by dynamic actin-based foot processes (FPs). Neighboring FPs form specialized junctions, slit diaphragms (SDs), which prevent passage of proteins into the ultrafiltrate. The SD protein complex is linked to cytoskeletal actin filaments and mutations in SD proteins lead to a dramatic change in cell morphology; proteinuria is accompanied by FP retraction and loss of SD structure. Thus, organization of the podocyte cytoskeleton is tightly linked to filtration barrier function. In a variety of cell systems, cytoskeleton arrangement is re

Is vacuolization of podocytes and glomerular endothelial cells of prognostic value with respect to FSGS?

Focal segmental glomerulosclerosis (FSGS) poses a major problem both from the clinical and pathomorphological viewpoint. The diagnosis of FSGS in its early stage is vital mainly because of rapidly developing therapeutic modalities. In the literature various changes are discussed which may be of prognostic value (may predict the development of FSGS). One the these changes in vacuolization, mainly of podocytes and less frequently of endothelial cells. The purpose of the present study was to analyse biopsy specimens to find out to what extent vacuolization of podocytes and endothelial cells is as

RENAL-CHIP: Rejection Evaluation via Non-Invasive Analysis of Circulating Podocytes With Herringbone-Chip Isolation Platform.

Immune rejection limits long-term renal allograft survival, yet current diagnostics lack non-invasive, and precise detection. Here, donor-derived circulating podocytes (CPCs) are identified as a blood-based surrogate of acute rejection. A magnetically reversible herringbone microfluidic chip equipped with epithelial cell adhesion molecule (EpCAM) antibody-functionalized magnetic beads efficiently isolates CPCs from 1 mL of peripheral blood (capture efficiency 84.4%, release 95.5%, viability 96.0%). Among 65 participants, only transplant recipients with biopsy-confirmed rejection exhibited≥35 C

Targeting LRH-1 alleviates diabetes-induced lipotoxicity in podocytes: role of PLIN5-mediated lipid droplet turnover.

Diabetic kidney disease (DKD) is characterized by podocyte injury driven by intracellular lipid accumulation. Liver receptor homolog-1 (LRH-1) is a key nuclear receptor regulating lipid metabolism, yet its role in podocyte lipotoxicity remains unclear. This study identifies the LRH-1/perilipin 5 (PLIN5) axis as a critical pathway for maintaining lipid homeostasis in podocytes and a promising pharmacological target for DKD. We demonstrated that hyperglycemia suppressed the LRH-1/PLIN5 axis, leading to lipid droplet accumulation, oxidative stress, and podocyte injury in db/db mice and cultured p
