Age-related macular degeneration affects the optic radiation white matter projecting to locations of retinal damage.

We investigated the impact of age-related macular degeneration (AMD) on visual acuity and the visual white matter. We combined an adaptive cortical atlas and diffusion-weighted magnetic resonance imaging (dMRI) and tractography to separate optic radiation (OR) projections to different retinal eccentricities in human primary visual cortex. We exploited the known anatomical organization of the OR and clinically relevant data to segment the OR into three primary components projecting to fovea, mid- and far-periphery. We measured white matter tissue properties-fractional anisotropy, linearity, pla

Plasticity in the adult visual cortex: implications for the diagnosis of visual field defects and visual rehabilitation.

It has recently been shown that, contrary to long-held beliefs, sensory and motor maps are not stable in the adult cerebral cortex. Alteration of input from the periphery results in changes in topography in the cortex, including the primary visual cortex. Mechanisms involved consist mainly of reshaping the receptive field of cortical cells and increasing the sensitivity of deprived cells in the visual cortex. Cortical plasticity allows the brain to adapt to background modifications or to damage of the nervous system. It also underlies learning and attention processes. Cortical changes occurrin

Impact of blindness onset on the functional organization and the connectivity of the occipital cortex.

Contrasting the impact of congenital versus late-onset acquired blindness provides a unique model to probe how experience at different developmental periods shapes the functional organization of the occipital cortex. We used functional magnetic resonance imaging to characterize brain activations of congenitally blind, late-onset blind and two groups of sighted control individuals while they processed either the pitch or the spatial attributes of sounds. Whereas both blind groups recruited occipital regions for sound processing, activity in bilateral cuneus was only apparent in the congenitally

Approaches to a cortical vision prosthesis: implications of electrode size and placement.

OBJECTIVE: In order to move forward with the development of a cortical vision prosthesis, the critical issues in the field must be identified. APPROACH: To begin this process, we performed a brief review of several different cortical and retinal stimulation techniques that can be used to restore vision. MAIN RESULTS: Intracortical microelectrodes and epicortical macroelectrodes have been evaluated as the basis of a vision prosthesis. We concluded that an important knowledge gap necessitates an experimental in vivo performance evaluation of microelectrodes placed on the surface of the visual

Optic nerve transection affects development and use-dependent plasticity in neocortex of the rat: Quantitative acetylcholinesterase imaging.

We investigated the effects of neonatal optic nerve transection on cortical acetylcholinesterase (AChE) activity in hooded rats during postnatal development and following behavioral manipulation after weaning. AChE reaction product was quantified on digitized images of histochemically stained sections in layer IV of primary somatic sensory, primary visual and visual association cortex. Rats with optic nerve transection were compared to sham-operated littermates. In all cortical regions of both types of animal, AChE reaction product was increased to peak 2 weeks after birth and decreased therea

Somatosensory cortex and body representation: Updating the motor system during a visuo-proprioceptive cue conflict.

The brain's representation of hand position is critical for voluntary movement. Representation is multisensory, combining visual and proprioceptive cues. When these cues conflict, the brain recalibrates its unimodal estimates, shifting them closer together to compensate. Converging evidence from research in perception, behavior, and neurophysiology suggest that such updates to body representation are communicated to the motor system to keep hand movements accurate. We hypothesized that primary somatosensory cortex (SI) is crucial in this updating process due to its role in proprioception and c

Cortical hyperexcitability is cortical under-inhibition: evidence from a novel functional test of migraine patients.

Recent studies of the visual cortex in patients with migraine have generally concluded that migraine (particularly migraine with aura) is associated with a state of functional cortical hyperexcitability. The mechanisms giving rise to this hyperexcitability have hitherto been unclear. This paper reports two studies that used a novel investigative technique, derived from basic research in vision science, to examine specific deficits of inhibitory processing in primary visual cortex. The technique is termed the metacontrast test, and it examines visual masking under highly specified conditions. I

Thalamocortical architecture shapes structural and functional plasticity after early sensory loss.

UNLABELLED: Early sensory loss transforms the functional organization of affected sensory systems, with regions that normally support vision or audition participating in noncanonical perceptual and cognitive functions. Such reorganization requires coordination across distributed networks. Higher-order thalamic nuclei are positioned to support these interactions through their widespread reciprocal cortical connectivity, yet studies of sensory-loss plasticity have focused primarily on cortex and first-order sensory pathways. Here, we combined structural and functional MRI in early blind and earl

A functional MRI study of motor dysfunction in Friedreich's ataxia.

Friedreich's ataxia (FRDA) is the most common form of hereditary ataxia. In addition to proximal spinal cord and brain stem atrophy, mild to moderate atrophy of the cerebellum has been reported in advanced FRDA. The aim of this study was to examine dysfunction in motor-related areas involved in the execution of finger tapping tasks in individuals with FRDA, and to investigate functional re-organization of cortico-cerebellar, cortico-striatal and parieto-frontal loops as a result of the cerebellar pathology. Thirteen right-handed individuals with FRDA and fourteen right-handed controls particip

Visual Receptive Field Heterogeneity and Functional Connectivity of Adjacent Neurons in Primate Frontoparietal Association Cortices.

The basic organization principles of the primary visual cortex (V1) are commonly assumed to also hold in the association cortex such that neurons within a cortical column share functional connectivity patterns and represent the same region of the visual field. We mapped the visual receptive fields (RFs) of neurons recorded at the same electrode in the ventral intraparietal area (VIP) and the lateral prefrontal cortex (PFC) of rhesus monkeys. We report that the spatial characteristics of visual RFs between adjacent neurons differed considerably, with increasing heterogeneity from VIP to PFC. In

Molecular signatures of resilience to Alzheimer's disease in neocortical layer 4 neurons.

Selective neuronal vulnerability is a hallmark of Alzheimer's disease (AD), yet the molecular basis of resilience remains poorly understood. Using single-nucleus and spatial transcriptomics to compare neocortical regions affected early (prefrontal cortex, precuneus) or late (primary visual cortex) in AD, we identified a resilient excitatory population in layer 4 of the primary visual cortex expressing RORB, CUX2, and EYA4. Layer 4 neurons in association neocortex shared molecular signatures of resilience. Early-stage resilient neurons upregulated genes associated with synapse maintenance, syna

Slow Theta tACS of the Right Parietal Cortex Enhances Contralateral Visual Working Memory Capacity.

Recent research suggests alteration of visual working memory capacity by modulation of parietal theta frequency via transcranial alternating current stimulation (tACS). However, it remains to be clarified whether this effect is partly driven by co-stimulation of prefrontal cortex and subcortical structures. It was hypothesized that focal tACS over the parietal lobe without additional prefrontal or subcortical stimulation should lead to similar effects as reported in the literature. Healthy, young participants were tested on a visual working memory paradigm while receiving either focal parietal

On the evolution of polysensory superior temporal sulcus and middle temporal gyrus: A key component of the semantic system in the human brain.

The primary auditory cortex and other early auditory cortical areas lie on Heschl's gyrus within the Sylvian fissure. On the adjacent lateral surface of the superior temporal gyrus, the cortex processes higher order auditory information leading to auditory perception. On the ventral surface of the temporal lobe in the primate brain, there are areas that process higher order visual information leading to visual perception. These sensory-specific auditory and visual processing regions are separated by areas that integrate multisensory information within the deep superior temporal sulcus in both

Early-onset hearing loss reorganizes the visual and auditory network in children without cochlear implantation.

The present study investigates the effect of early-onset hearing loss on the reorganization of visual and auditory networks in children without cochlear implants. Eleven congenitally deaf children and 12 age-matched hearing children were included in the study. Bilateral transverse temporal cortices and bilateral lateral occipital cortices were defined as auditory and visual seeds, respectively (as verified using an independent component analysis). The four seed-based connectivity maps were computed for each participant. As a result, group analysis showed that the primary auditory cortex was le

Running towards amblyopia recovery.

Amblyopia is a neurodevelopmental disorder of the visual cortex arising from abnormal visual experience early in life which is a major cause of impaired vision in infants and young children (prevalence around 3.5%). Current treatments such as eye patching are ineffective in a large number of patients, especially when applied after the juvenile critical period. Physical exercise has been recently shown to enhance adult visual cortical plasticity and to promote visual acuity recovery. With the aim to understand the potentialities for translational applications, we investigated the effects of vol

Visual Field Defect Before and After Endovascular Treatment of Occipital Arteriovenous Malformations.

BACKGROUND: Occipital arteriovenous malformations (AVMs) carry a high risk of postoperative morbidity because of their anatomic relation to the visual cortex and optic radiations. Data regarding endovascular management of these lesions are scant. OBJECTIVE: To report our single-center experience with occipital AVMs, most of which were treated endovascularly, with a special interest for postoperative visual impairment. METHODS: From a prospective database, we assessed the clinical and radiological data of all patients with an occipital AVM managed between 1997 and 2018. The extension of the n

Does dominance of crossing retinal ganglion cells make the eyes cross? The temporal retina in the origin of infantile esotropia – a neuroanatomical and evolutionary analysis.

A closer look at the evolution of the eye and the brain provides a possible explanation for both the origin of infantile esotropia and its motor characteristics. In the course of evolution, the eyes have moved from a lateral to a frontal position. Consequently, the monocular visual fields started to overlap resulting in a binocular visual field. In lateral-eyed animals, the retinae project to the contralateral visual cortices only. These projections are also found in binocular mammals and birds with binocular visual fields but in addition there are uncrossed projections from the temporal retin

Mirror therapy in lower limb amputees--a look beyond primary motor cortex reorganization.

PURPOSE: Phantom pain in upper limb amputees is associated with the extent of reorganization in the primary sensorimotor cortex. Mirror visual feedback therapy has been shown to improve phantom pain. We investigated the extent of cortical reorganization in lower limb amputees and changes in neural activity induced by mirror therapy. MATERIALS AND METHODS: Eight lower limb amputees underwent 12 sessions of MVFT and functional magnetic resonance imaging (fMRI) of the brain before the first and after the last MVFT session. FMRI sessions consisted of two runs in which subjects were instructed to

Spinal Cord Injury Disrupts Resting-State Networks in the Human Brain.

Despite 253,000 spinal cord injury (SCI) patients in the United States, little is known about how SCI affects brain networks. Spinal MRI provides only structural information with no insight into functional connectivity. Resting-state functional MRI (RS-fMRI) quantifies network connectivity through the identification of resting-state networks (RSNs) and allows detection of functionally relevant changes during disease. Given the robust network of spinal cord afferents to the brain, we hypothesized that SCI produces meaningful changes in brain RSNs. RS-fMRIs and functional assessments were perfor

Restoration of FMRP expression in adult V1 neurons rescues visual deficits in a mouse model of fragile X syndrome.

Many people affected by fragile X syndrome (FXS) and autism spectrum disorders have sensory processing deficits, such as hypersensitivity to auditory, tactile, and visual stimuli. Like FXS in humans, loss of Fmr1 in rodents also cause sensory, behavioral, and cognitive deficits. However, the neural mechanisms underlying sensory impairment, especially vision impairment, remain unclear. It remains elusive whether the visual processing deficits originate from corrupted inputs, impaired perception in the primary sensory cortex, or altered integration in the higher cortex, and there is no effective
