Identification and Exploration of Immunity-Related Genes and Natural Products for Alzheimer's Disease Based on Bioinformatics, Molecular Docking, and Molecular Dynamics.

BACKGROUND: Recent research highlights the immune system's role in AD pathogenesis and promising prospects of natural compounds in treatment. This study explores immunity-related biomarkers and potential natural products using bioinformatics, machine learning, molecular docking, and kinetic simulation. METHODS: Differentially expressed genes (DEGs) in AD were analyzed using GSE5281 and GSE132903 datasets. Important AD module genes were identified using a weighted co-expression algorithm (WGCNA), and immune-related genes (IRGs) were obtained from the ImmPortPortal database. Intersecting these

Larvicidal Efficacy of Chloris virgata Extracts Against Aedes aegypti, Anopheles stephensi, and Culex quinquefasciatus with Insights into Mode of Action via Molecular Docking.

The indiscriminate use of synthetic insecticides for mosquito control has resulted in significant challenges, such as environmental pollution and growing insecticide resistance. In response, there is increasing interest in eco-friendly alternatives like plant-based larvicides. This study assesses the larvicidal efficacy of Chloris virgate (Poales: Poaceae) extracts against Aedes aegypti (Diptera: Culicidae), Culex quinquefasciatus (Diptera: Culicidae), and Anopheles stephensi (Diptera: Culicidae), and investigates the bioactive chemical compounds responsible for this activity. Baihe Dihuang Tang Exerts Antidepressant Effects via Modulation of MAOA-Mediated Serotonin Metabolism and Synaptic Plasticity.

Background/Objectives: Baihe Dihuang Tang (BDT), a classical herbal formula from Zhang Zhongjing's Han Dynasty work Jin Gui Yao Lue, is widely used to treat depressive disorder by nourishing Yin, clearing heat, and tonifying the heart and lungs. However, its pharmacological mechanisms remain unclear. This study aims to explore BDT's antidepressant effects via MAOA-regulated serotonin (5-HT) metabolism and synaptic plasticity, supported by experimental validation, while using network pharmacology to predict MAOA-targeting active components. Methods: Active components and targets of BDT were scr

Antiangiogenic Potential of Beneficial Sterols from Parotoid Gland Secretion of Indian Common Toads (Duttaphrynus melanostictus) in the Coastal Region of the Indian Subcontinent: An In Vivo to In Silico Approach.

The toxins of the Bufonid toads have been used formerly as ethnomedicine to treat different diseases, including chronic hepatitis, hypertension, and multiple cancers. Thus, toads' venom has a great impact on traditional health care. However, the main emphasis of this study is to identify natural components present in toad parotoid gland secretion and evaluate their antiangiogenic effects. Sterol-rich extracts of parotoid gland secretions were isolated in an ethyl acetate medium from a natural population of Indian common toads (Duttaphrynus melanostictus) from the coastal region (Purba Medinipu

VEGFR-Mediated Cytotoxic Activity of Pulicaria undulata Isolated Metabolites: A Biological Evaluation and In Silico Study.

Natural products play a remarkable role not only in the synthesis, design, and discovery of new drugs but also as the most prominent source of drugs and bioactive substances. Adding to the search for new sources of safe innovative antitumor drugs, here we reported a phytochemical study on Pulicaria undulata which revealed promising antiangiogenic agents. Six compounds were isolated and identified as xanthoxyline (1), stigmasterol (2), oleanolic acid (3), salvigenin (4), rhamnetin (5) and dihydroquercetin-4'-methyl ether (6) using nuclear magnetic resonance (NMR) spectroscopic techniques.

Identification of Antidiabetic Metabolites from Paederia foetida L. Twigs by Gas Chromatography-Mass Spectrometry-Based Metabolomics and Molecular Docking Study.

Paederia foetida L. (Rubiaceae) is a climber which is widely distributed in Asian countries including Malaysia. The plant is traditionally used to treat various diseases including diabetes. This study is to evaluate the enzymatic inhibition activity of Paederia foetida twigs extracts and to identify the metabolites responsible for the bioactivity by gas chromatography-mass spectrometry (GC-MS) metabolomics profiling. Three different twig extracts, namely, hexane (PFH), chloroform (PFC), and methanol (PFM), were submerged for their α-amylase and α-glucosidase inhibition potential in 5 replicate

Azadirachta indica inhibits key enzyme linked to type 2 diabetes in vitro, abates oxidative hepatic injury and enhances muscle glucose uptake ex vivo.

The progression of secondary complications in type 2 diabetes (T2D) has been linked to oxidative stress caused by hyperglycemia. Therefore, the control of hyperglycemia is the main target in the treatment of diabetes. The present study investigated the scavenging and ameliorative potentials of different fractions of Azadirachta indica (A. indica) ethanol stem bark extract in Fe2+-induced oxidative injury in hepatic tissue as well as their ability to inhibit enzymes linked to diabetes and in enhancing muscle glucose uptake via some in vitro and ex vivo experimental models. The results revealed

Isolation, biological evaluation and validated HPTLC-quantification of the marker constituent of the edible Saudi plant Sisymbrium irio L.

Phytochemical investigation and chromatographic purification of the n-hexane fraction of the aerial parts of the edible Saudi plant Sisymbrium irio led to the isolation of β-sitosterol (1), stigmasterol (2) and β-sitosterol-β-d-glucoside (3). The cytotoxic effects of the n-hexane, dichloromethane, ethyl acetate and n-butanol fractions were tested against three cancer cell lines viz., MCF-7, HCT-116 and HepG2, using the crystal violet staining (CVS) method, while the antibacterial activity against a number of pathogenic bacterial strains, was also estimated using the broth microdilution assay.

Ampelozizyphus amazonicus Ducke is a tree commonly found in the Amazon region and an extract of its stem bark is popularly used as an antimalarial and anti-inflammatory agent and as an antidote to snake venom. Ursolic acid; five lupane type triterpenes: betulin, betulinic acid, lupenone, 3beta-hydroxylup-20(29)-ene-27,28-dioic acid, and 2alpha,3beta-dihydroxylup-20(29)-ene-27,28-dioic acid, and three phytosteroids: stigmasterol, sitosterol and campesterol, have been isolated from stem extracts of A. amazonicus Ducke. Their structures were characterized by spectral data including COSY and HMQC.

Mechanistic insights into Tuoli Xiaozheng Formula for hepatocellular carcinoma: An integrated network pharmacology, molecular docking, and Mendelian randomization analysis.

Hepatocellular carcinoma (HCC) is a malignancy with a poor prognosis and low overall survival. Tuoli Xiaozheng Formula (TLXZF), a traditional Chinese compound prescription, has shown clinical efficacy in treating HCC, but its multi-component and multi-target mechanisms remain unclear. This study aimed to elucidate the molecular mechanisms of TLXZF against HCC by integrating network pharmacology, molecular docking, and Mendelian randomization (MR) analyses. Active compounds and targets were identified from the Traditional Chinese Medicine Systems Pharmacology, high-throughput experiment- and re
