Genetic transformation in Streptococcus sanguis. Identification, surface spreading and competence of invasive strains of Streptococcus sanguis Lancefield groups H and W and other invasive viridans streptococci.

Of 46 viridans streptococci isolated from cases of probably true bacteremia, 26 strains or 56% could be grouped serologically in the Lancefield system. Eight strains (17%) were identified as serogroup H (characteristic of Streptococcus sanguis), ten strains (22%) as serogroup W (a new group antigen found in S. sanguis) and eight strains (17%) as serogroup K (related to S. mitis (mitior)). The high number of serogroupable viridans streptococci was specially due to the introduction of serogroup W and indicates the importance of this serogroup in cases of bacteremia. A set of six diagnostic tests

Multi-doped glasses with Mg2+ and Zn2+ ions in glass-ionomer cements.

OBJECTIVE: To develop a new, biocompatible glass-ionomer cement (GIC) with improved antibacterial activity and physical properties through multiple ionic substitutions of strontium with magnesium and zinc. METHODS: A fluoro-alumino-silicate glass (LG99Sr) with composition 4.5SiO₂-3Al₂O₃-1.5 P₂O₅-3SrF₂-2SrO was synthesized using the melt-quench technique. LG99Sr was modified by partially substituting SrO with MgO and ZnO, yielding a new composition 4.5SiO₂-3Al₂O₃-1.5 P₂O₅-3SrF₂-0.5SrO-1MgO-0.5ZnO (LG99Sr-Mg-Zn). Following glass characterization, GICs were prepared using Fuji IX (GC Corporation

The Role of Indoor Microbiome and Metabolites in Shaping Children's Nasal and Oral Microbiota: A Pilot Multi-Omic Analysis.

Maintaining a diverse and well-balanced nasal and oral microbiota is vital for human health. However, the impact of indoor microbiome and metabolites on nasal and oral microbiota remains largely unknown. Fifty-six children in Shanghai were surveyed to complete a questionnaire about their personal and environmental characteristics. The indoor microbiome and metabolites from vacuumed indoor dust were profiled via shotgun metagenomics and untargeted liquid chromatography-mass spectrometry (LC-MS). The nasal and oral microbiota in children was characterized using full-length 16S rRNA sequencing fr

Evaluation of swirling, pH, and glucose tests for the detection of bacterial contamination in platelet concentrates.

BACKGROUND: Although infrequent, episodes of transfusion-associated bacterial sepsis may lead to serious outcomes or death and therefore are of concern. This study evaluates the sensitivity of three surrogate tests for the presence of bacteria in platelet concentrates: cessation of swirling, low extracellular pH, and low plasma glucose levels. STUDY DESIGN AND METHODS: Day 0 platelet concentrates were inoculated with low levels of one of seven bacterial strains and stored with agitation at 20 to 24 degrees C for up to 5 days. In the morning and afternoon of each day of storage, bacterial leve

Treatment of infection caused by animal commensal pathogen Staphylococcus simulans.

INTRODUCTION: Staphylococcus simulans is an animal pathogen belonging to the coagulase-negative staphylococci (CoNS) group that was occasionally identified in the skin of healthy people. Although it was suggested that S. simulans share some virulence factors with S. aureus, little is known about human infections, and clinical relevance is limited to a few reports. Herein, we report a case of bacteremia caused by S. simulans. CASE PRESENTATION: A 39-year-old man was brought to the hospital due to heart attack. During the course of admission, his procalcitonin, C-reactive protein, and erythrocy

A Rare Case of Ruptured Distal Posterior Cerebral Artery Aneurysm Followed by Middle Cerebral Artery Occlusion due to Delayed Diagnosis of Infective Endocarditis.

A 31-year-old female presented to our hospital with sudden headache and altered consciousness. Computed tomography showed left acute subdural hematoma, and digital subtraction angiography revealed a small aneurysm on the left distal posterior cerebral artery. Coil embolization was conducted, and the patient was discharged with no neurological deficits. However, two weeks later, she presented with complete left hemiplegia and with the National Institutes of Health Stroke Scale of 20. Magnetic resonance angiography showed the occlusion of right middle cerebral artery, and the Diffusion-Weighted

Mediastinitis and subcutaneous abscess complicated after EBUS-TBNA of 2R mediastinal lymph node.

Endobronchial ultrasound-guided transbronchial needle aspiration (EBUSTBNA) is a minimally invasive diagnostic tool used for the evaluation of mediastinal lymphadenopathy. It is a safe procedure, but complications such as bleeding and infection may occur. We report a case of a patient who developed a subcutaneous abscess abscess and mediastinitis after EBUSTBNA. A 75-year-old male with a history of right nephrectomy due to renal cell carcinoma and lung adenocarcinoma history underwent EBUS-TBNA for the evaluation of a right upper paratracheal lymph node. Two weeks after the procedure, the pati

Secondary streptococcal infection following influenza.

Secondary bacterial infection following influenza type A virus (IAV) infection is a major cause of morbidity and mortality during influenza epidemics. Streptococcus pneumoniae has been identified as a predominant pathogen in secondary pneumonia cases that develop following influenza. Although IAV has been shown to enhance susceptibility to the secondary bacterial infection, the underlying mechanism of the viral-bacterial synergy leading to disease progression is complex and remains elusive. In this review, cooperative interactions of viruses and streptococci during co- or secondary infection w

Wound Botulism Caused by Botulinum Neurotoxin Type A in a Chronic Parenteral Drug Abuser.

Botulism is an acute paralytic disease caused by botulinum neurotoxin (BoNT)-mediated inhibition of neurosignaling at the neuromuscular junction. BoNTs are produced by gram positive, anaerobic, spore-forming bacteria from the genus Clostridium, most commonly Clostridium botulinum. Over the last decade, a previously uncommon form of botulism, wound botulism, has increased in prevalence possibly due to the rise in parenteral drug abuse. A 53-year-old patient with a history of drug abuse presents to a rural emergency department with rapidly progressing lower extremity weakness over the past few da

Using Selective Agar Containing Ciprofloxacin and Tetracycline Reveals Resistant Oral Microbiota in Healthy and Periodontitis Patients.

The oral cavity may act as a reservoir for antibiotic resistance. This study aimed to directly isolate and identify phenotypically resistant bacteria from the oral biofilm of healthy individuals and patients with periodontitis, using tetracycline, and ciprofloxacin containing selective agar. Furthermore, resistance of selected bacteria towards ampicillin was also evaluated. Plaque samples were collected from 12 patients (six healthy, six with periodontitis). Bacteria were cultured on selective agar containg defined antibiotic concentration and non-selective media under aerobic and anaerobic co

Elective endoscopic variceal ligation is not a risk factor for bacterial infection in patients with liver cirrhosis.

BACKGROUND: Patients with cirrhosis are at high risk of bacterial infections. Invasive procedures are generally believed to increase this susceptibility. AIMS: We investigated the incidence of bacterial infections in cirrhotic patients undergoing elective endoscopic variceal ligation (EVL). METHODS: We enrolled 60 consecutive cirrhotic patients who underwent a total number of 112 elective EVL procedures. One to seven bands were applied at each session until variceal eradication. Markers of inflammation/infection and blood cultures were obtained before and 24 h after EVL. RESULTS: Aetiology

New structural insights into the PI-2 pilus from Streptococcus oralis, an early dental plaque colonizer.

Streptococcus oralis is a member of the mitis group of oral streptococci and an early colonizer in dental plaque biofilm, a major cause of periodontal disease, dental caries, and other oral infections. S. oralis promotes biofilm growth by coaggregating in a mutualistic partnership with other early colonizers such as Actinomyces oris. For this cell-to-cell interaction, A. oris is known to use its sortase-dependent pilus (type 2), but whether S. oralis uses its PI-2 (pilus islet 2) pilus is still to be determined. The PI-2 pilus is predicted to have a heterodimeric structure consisting of two di

Shotgun Metagenomics of Biofilm Microbiome in Oral Lichen Planus With Desquamative Gingivitis.

INTRODUCTION: Oral lichen planus (OLP) is a chronic inflammatory condition often associated with desquamative gingivitis (DG). The oral microbiome's role in OLP and DG (OLP-DG) is gaining recognition, but prior 16S rRNA studies lacked taxonomic resolution. This study introduced shotgun metagenomic sequencing to thoroughly compare the supragingival and subgingival plaque microbiomes of individuals with and without OLP-DG. METHODS: Twenty-seven participants (9 OLP-DG, 18 non-OLP) were recruited. Supra- and subgingival plaque samples were collected separately. Genomic DNA was analyzed using shot

Lymphedema in patients with X-linked severe combined immunodeficiency.

BACKGROUND: IL2RG and JAK3 mutations cause T-cell-negative, B-cell-positive, natural killer cell-negative severe combined immunodeficiency (SCID). Cell therapies such as hematopoietic stem cell transplantation (or gene therapy for IL2RG deficiency) can effectively restore T-cell immunity but-when performed without myeloablation-may leave residual immune defects. These include persistent B-cell dysfunction, poor innate lymphoid cell recovery, and poor reconstitution of secondary lymphoid organs. Severe/recurrent warts may also occur in the long term. OBJECTIVE: We present 3 IL2RG SCID cases co

Microbial contamination and composition of oral samples subjected to clinical whole genome sequencing.

Biological material from the oral cavity is an excellent source of samples for genetic diagnostics. This is because collection is quick, easy-to-access, and non-invasive. We have set-up clinical whole genome sequence testing for patients with suspected hereditary disease. Beside the excellent quality of human DNA that can be isolated from such samples, we observed the presence of non-human DNA sequences at varying percentages. We investigated the proportion of non-human mapped reads (NHMR) sequenced from buccal swabs and saliva, the type of microbial genomes from which they were derived, and i

Identification of PblB mediating galactose-specific adhesion in a successful Streptococcus pneumoniae clone.

The pneumococcal genome is variable and there are minimal data on the influence of the accessory genome on phenotype. Pneumococcal serotype 14 sequence type (ST) 46 had been the most prevalent clone causing pneumonia in children in Taiwan. A microarray was constructed using the genomic DNA of a clinical strain (NTUH-P15) of serotype 14 ST46. Using DNA hybridization, genomic variations in NTUH-P15 were compared to those of 3 control strains. Microarray analysis identified 7 genomic regions that had significant increases in hybridization signals in the NTUH-P15 strain compared to control strains
