Effect of dietary supplementation with antioxidants and flavouring prototypes based on acorn and olive on the volatile profile of pork.

This study evaluated the impact of dietary supplementation with antioxidants (AO) and flavouring prototypes based on capsicum (CA), acorn (AC), and olive (OL) on the volatile profile of pork. Two trials were conducted. Trial 1 served as a preliminary screening to evaluate samples of longissimus thoracis et lumborum (LTL) of 32 barrows fed with diets containing different levels of CA, AC, and OL. Results showed that dietary treatments significantly affected the abundance of seven volatile compounds, particularly long-chain aldehydes, while the fatty acid profile remained unaffected. Trial 2 con

Antifungal and Therapeutic Evaluation of l- and d‑Enantiomers of a Plant Defensin-Inspired Peptide against Sporothrix schenckii and Sporothrix brasiliensis.

Sporotrichosis is an emerging subcutaneous mycosis and an important public and veterinary health concern in Brazil. The ongoing expansion of feline-associated transmission, particularly involving Sporothrix brasiliensis, has reinforced the need for new antifungal strategies. In this study, we evaluated the antifungal potential of the bioinspired peptide L-CaDef2.1G27‑K44 (L-CDF-GK) and its enantiomer D-CaDef2.1G27‑K44 (D-CDF-GK), derived from a defensin identified in Capsicum annuum fruits. Antifungal activity was assessed through in vitro and in vivo assays, together with the investigation of

H2O2-generating polyamine oxidases (PAOs) are modulated during sweet pepper ripening: Spermine oxidase (SpmOX) as a case study of post-translational modification regulation.

Polyamine oxidases (PAOs) are flavin-containing enzymes involved in the catabolism of polyamines through the oxidative deamination of spermine (Spm) and spermidine (Spd). Transcriptome analysis of pepper (Capsicum annuum) fruit revealed six PAO genes (CaPAO1 to CaPAO6) expressed during ripening. Time-course expression profiling across three developmental stages, green immature (G), breaking point (BP), and red ripe (R), showed differential regulation: CaPAO5 and CaPAO6 were upregulated during ripening, while CaPAO1 and CaPAO3 were downregulated. CaPAO4 expression remained relatively stable. Ex

New generation capsaicin-diclofenac containing, silicon-based transdermal patch provides prolonged analgesic effect in acute and chronic pain models.

OBJECTIVE: Pain is one of the major public health burdens worldwide, however, conventional analgesics are often ineffective. Capsaicin-the active compound of Capsicum species, being responsible for their pungency-has been part of traditional medicine long ago. Capsaicin is a natural agonist of the Transient Receptor Potential Vanilloid 1 receptor-localized on capsaicin-sensitive sensory neurons and strongly involved in pain transmission-, and has been in focus of analgesic drug research for many years. In this study, we aimed to develop a sustained release transdermal patch (transdermal therap

Self-reported food intolerances in an Indian population: Need for individualization rather than a universal low-FODMAP diet.

BACKGROUND AND AIM: Low-fermentable oligo-, di-, and monosaccharides and polyol (FODMAP) diets have been recommended for individuals with food intolerance and irritable bowel syndrome (IBS). Individual food intolerances may, however, not correspond to the FODMAP content alone. METHODS: We conducted a survey on self-reported intolerance to articles of food commonly identified as high FODMAP in 400 healthy Indian subjects (median age 40 years; 69% men) and 204 consecutive consenting patients with IBS (median age 36 years; 58% men). RESULTS: One-hundred seventy-nine (44.8%) healthy subjects and

CaWRKY30 Positively Regulates Pepper Immunity by Targeting CaWRKY40 against Ralstonia solanacearum Inoculation through Modulating Defense-Related Genes.

The WRKY transcription factors (TFs) network is composed of WRKY TFs' subset, which performs a critical role in immunity regulation of plants. However, functions of WRKY TFs' network remain unclear, particularly in non-model plants such as pepper (Capsicum annuum L.). This study functionally characterized CaWRKY30-a member of group III Pepper WRKY protein-for immunity of pepper against Ralstonia solanacearum infection. The CaWRKY30 was detected in nucleus, and its transcriptional expression levels were significantly upregulated by R. solanacearum inoculation (RSI), and foliar application ethyl

Genome-Wide Identification and Functional Characterization of CaGRAS43 in Enhancing Cold Stress Tolerance in Pepper.

Cold stress severely limits plant growth and crop productivity, necessitating the identification of key regulatory genes that enhance tolerance. In this study, we performed a genome-wide identification and expression analysis of the GRAS transcription factor family in pepper under cold stress. In total, 64 GRAS genes (CaGRAS1-CaGRAS64) were identified in the pepper genome and distributed across 10 different chromosomes. Additional in silico analysis provided key insights on the identified CaGRAS genes, including their conserved domains, exon-intron organization, evolutionary relationships, gen

Impact of plant growth-promoting rhizobacteria and natural enemies on Myzus persicae (Hemiptera: Aphididae) infestations in pepper.

Management of green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae), in bell pepper, Capsicum annuum L., was explored through a combination of plant growth-promoting rhizobacteria (PGPR) and endemic biological control in New York in 2006 and 2007. We hypothesized that by using PGPR-treated peppers 1) M. persicae infestations would be reduced via induced resistance, 2) natural enemies would be lured to plants through the elicitation of volatile organic compounds, and 3) yield amount and quality would be improved. Pepper seed was planted in soil containing the PGPR formulation BioYie

The Role of Henosepilachna vigintioctopunctata in Facilitating the Spread of Tomato Brown Rugose Fruit Virus (ToBRFV) Among Hosts.

Tomato brown rugose fruit virus (ToBRFV), a highly stable and mechanically transmissible tobamovirus, poses a significant threat to solanaceous crops worldwide, particularly tomato (Solanum lycopersicum). While its transmission via human activities and contaminated materials is well-documented, the role of common phytophagous insects in its epidemiology remains less understood. Henosepilachna vigintioctopunctata, the Hadda beetle, is a common pest of Solanaceae with a host range that overlaps extensively with that of ToBRFV. This study aimed to quantify the beetle's capacity to act as a mechan

Diet and gallbladder cancer: a case-control study.

Cancer of the gallbladder is rare but fatal, and has an unusual geographic and demographic distribution. Gallstones and obesity have been suggested as possible risk factors. As diet is known to influence both these factors, we carried out the present study to evaluate the possible role of diet in gallbladder carcinogenesis. A case-control study involving 64 newly diagnosed cases of gallbladder cancer and 101 cases of gallstones was carried out. The dietary evaluation was carried out by the dietary recall method based on a preset questionnaire developed specifically for the present study, keepi

Rapid detection of Kenyan tomato leaf curl virus isolates using probe-enhanced loop-mediated isothermal amplification coupled with a modified DNA extraction method.

Plant disease management requires sensitive and user-friendly diagnostic methods. The polymerase chain reaction (PCR) and other methods have helped diagnose diseases, but they are primarily used in high-resource settings, leaving low-resource settings without effective options. A simple probe-enhanced method for the detection of Kenyan tomato leaf curl virus (Kenyan ToLCV) isolates in host crops, including asymptomatic ones, was developed by employing loop-mediated isothermal amplification (LAMP) coupled with a rapid modified DNA extraction method. A modified alkaline polyethylene glycol (APEG

Purification, Structural Characterization, and Anticandidal Activity of a Chitin-Binding Peptide with High Similarity to Hevein and Endochitinase Isolated from Pepper Seeds.

With the emergence of multidrug-resistant microorganisms, microbial agents have become a serious global threat, affecting human health and various plants. Therefore, new therapeutic alternatives, such as chitin-binding proteins, are necessary. Chitin is an essential component of the fungal cell wall, and chitin-binding proteins exhibit antifungal activity. In the present study, chitin-binding peptides isolated from Capsicum chinense seeds were characterized and evaluated for their in vitro antimicrobial effect against the growth of Candida and Fusarium fungi. Proteins were extracted from the s

Developmentally regulated sesquiterpene production confers resistance to Colletotrichum gloeosporioides in ripe pepper fruits.

Sesquiterpenoid capsidiol, exhibiting antifungal activity against pathogenic fungus, is accumulated in infected ripe pepper fruits. In this study, we found a negative relation between the capsidiol level and lesion size in fruits infected with Colletotrichum gloeosporioides, depending on the stage of ripening. To understand the developmental regulation of capsidiol biosynthesis, fungal-induced gene expressions in the isoprenoid biosynthetic pathways were examined in unripe and ripe pepper fruits. The sterol biosynthetic pathway was almost shut down in healthy ripe fruits, showing very low expr

The effects of capsaicin on gastrin secretion in isolated human antral glands: before and after ingestion of red chilli.

BACKGROUND: Capsaicin is known to have regulatory effects on gastrointestinal functions via the vanilloid receptor (VR1). We reported previously that endocrine-like cells in the human antrum express VR1. AIM: To identify VR1-expressing endocrine-like cells in human antral glands and to examine whether stimulation with capsaicin causes release of gastrin, somatostatin, and serotonin. Further, to investigate the effects of a chilli-rich diet. METHODS: Gastroscopic biopsies were received from 11 volunteers. Seven of the 11 subjects agreed to donor gastric biopsies a second time after a 3-week c
