A 3 X 3 factorial experiment was conducted to examine how protein content (8, 16 and 32% of kcals from casein) and fat content (12, 24 and 48% of kcals from corn oil) interact to influence prolactin homeostasis in female rats. Weanling Sprague-Dawley rats were assigned to each of nine diets fed ad libitum. Dietary fat had no effect on the time of vaginal opening although first estrus occurred slightly earlier with high fat diets (P less than 0.05). Rats fed diets with protein at 8% of kcal showed a 2-wk delay in vaginal opening and first estrus. After 18-20 wk of feeding, serum and pituitary p

Several experiments were performed to study the changes in the negative feedback of estradiol on gonadotropin secretion around the time of puberty in the female rat. Ovariectomy of juvenile, first diestrus, or adult animals elevated FSH and LH levels 2 and/or 4 days later. Estradiol administered via Silastic capsules, at several dose levels, was much more effective in preventing the postcastration rise of gonadotropins in juvenile than in the older animals. A dose of estradiol that inhibited gonadotropin levels in juvenile rats, but not in adult animals, maintained preovariectomy serum estradi

CONTEXT: Reproductive processes in mammals involve complex interactions between sex steroids and growth factor systems. Understanding these interactions in the uterus during different stages of the reproductive cycle is essential for elucidating the mechanisms of endometrial regulation.
AIMS: This study aimed to investigate the relationship between circulating sex steroids and the expression of genes involved in the insulin-like growth factor 1 (IGF1) system and steroid hormone receptors in the uterine horns of non-pregnant bitches during dioestrus.
METHODS: Uterine horns from 29 bitches wer

The endometrium is a hormonally responsive tissue that undergoes cyclic remodeling. Although endometrial organoids have been established in several species, detailed characterization remains limited. Here, we assessed the structural and molecular fidelity of equine endometrial organoids across reproductive cycle stages and during extended culture. Organoids were generated from biopsies collected during estrus and diestrus and analyzed using histology, immunohistochemistry, electron microscopy, and bulk- and single-cell RNA sequencing. Organoids formed polarized cystic structures composed of co

The mouse estrous cycle drives cyclical remodelling of the endometrium, essential for uterine function and embryo implantation. However, the molecular mechanisms orchestrating these dynamic changes remain incompletely defined. In this study, we preliminarily staged C57BL/6 mice using vaginal smear cytology and confirmed phase-dependent morphological changes, including cyclical endometrial thickening and glandular remodelling, via histological assessment. To elucidate the underlying molecular landscape, we performed comprehensive RNA sequencing of the mouse endometrium across four distinct phas

In this study, we investigated the multigenerational effects of low-dose dibutyl phthalate (DBP) exposure on the reproductive system of female Kunming mice by simulating a long-term environmental exposure scenario for humans, using a food-contamination method for three consecutive generations (F0-F2). Results demonstrated significant reproductive dysfunction across generations, manifested by shortened diestrus intervals (P < 0.05) and prolonged estrus duration (P < 0.05) in F1-F2 generations. The number of pregnancies progressively declined, with numbers for the F1-F2 generations showing marke

Eight non-bred, non-pregnant, regularly cycling Caribbean jennies were examined daily via transrectal ultrasound to define the ovarian and uterine dynamics during four consecutive estrous cycles. Blood samples were collected every other day for progesterone analysis. The mean (±SD) overall inter-ovulatory interval across all donkeys and cycles was 22.93 ± 1.99 days. The maximum follicular diameter was 34.6 ± 2.9 mm. A two-wave pattern was evident in 97% (30/31) of the cycles. The emergence of the future dominant follicle and the largest subordinate follicle of the major primary wave coincided

The progestin-based hormonal contraceptive Depot Medroxyprogesterone Acetate (DMPA) is widely used in sub-Saharan Africa, where HIV-1 is endemic. Meta-analyses have shown that women using DMPA are 40% more likely than women not using hormonal contraceptives to acquire Human Immunodeficiency Virus (HIV-1). Therefore understanding how DMPA increases susceptibility to HIV-1 is an important public health issue. Using C57BL/6 mice and our previously optimized humanized mouse model (NOD-Rag1tm1Mom Il2rgtm1Wjl transplanted with hCD34-enriched hematopoietic stem cells; Hu-mice) where peripheral blood

Growing evidence has revealed the deleterious influence of environmental and food contaminants on puberty onset and development in both animals and children, provoking an increasing health concern. T-2 toxin, a naturally-produced Type A trichothecene mycotoxin which is frequently found in cereal grains and products intended for human and animal consumption, has been shown to impair the reproduction and development in animals. Nevertheless, whether this trichothecene mycotoxin can disturb the onset of puberty in females remains unclear. To clarify this point, infantile female rats were given a

RATIONALE: Psychedelics are considered to be promising therapeutic agents that could effectively treat several psychological disorders. However, these compounds can exert acute adverse effects that limit their practicality.
OBJECTIVES: Across three experiments, the present study examined the acute and delayed effects of psilocybin (PSIL) and its putatively non-hallucinogenic analog, norbaeocystin (NOR), on anxiety-like behavior and the influence of the female estrous cycle on acute response to the compounds.
METHODS: In Experiment 1, rats were treated with PSIL or NOR (0.3-1 mg/kg) and teste

Dimethomorph (DMM), a widely used fungicide recently classified as a reproductive toxicant, has unknown impacts on human ovarian function. Here, mass spectrometry identified DMM in human plasma (up to 3.5 ng/mL) and in follicular fluid (up to 1 ng/mL) for the first time, although at a low concentration. Investigating its effects in primary human granulosa cells (hGCs) and the KGN cell line, we found that DMM significantly reduced cell proliferation (BrdU incorporation) across all tested doses while sparing cell viability at non-cytotoxic concentrations, as measured by the CCK-8 assay. At a con

RATIONALE: Hypocretins (also known as orexins) are neuropeptides that regulate the sleep-wake cycle and modulate a variety of behaviors, including maternal behavior. They act through two receptor subtypes: hypocretin receptor 1 (HcrtR1) and hypocretin receptor 2 (HcrtR2). Hypocretin signaling is dynamically modulated across reproductive states, including the postpartum period, a stage characterized by increased wakefulness and fragmented sleep, likely reflecting the physiological demands of offspring care. Dual orexin receptor antagonists (DORAs), such as suvorexant, are clinically used as hyp

Uterine smooth muscle undergoes electrical, chemical, and mechanical transients during phasic contraction. The spread of electrical activity precedes the mechanical contraction of muscle fibers, and the initiation and coordination of these events is critical to uterine function, in both pregnancy and non-pregnancy. Characterization of non-pregnant activity is scarce, and understanding of the relationship between electrical and mechanical events is not well understood. Isolated uterine horns from virgin female Wistar rats were used in this study. Electrical and mechanical activity were assessed

Phthalates are associated with several reproductive disorders in women and reduce fertility in mice. They are also known to impair hepatic glycogen metabolism. Glucose is a crucial nutrient for the uterus, and glycogen buffers glucose concentration in the endometrium. The objective of this study was to investigate how long-term exposure to di(2-ethylhexyl) phthalate (DEHP) and diisononyl phthalate (DiNP) alters glycogen metabolism in the murine endometrium. Six-week-old female mice were fed chow containing vehicle or DEHP or DiNP at 0.15, 1.5, and 1500 parts per million (ppm) ad libitum for 9

Perimenopause is characterised by irregular reproductive cycles and hormonal fluctuations, notably decreased estradiol (E2), which are linked to increased susceptibility to panic disorder and neuroinflammation. We examined whether panic-related respiratory and behavioural responses to respiratory challenges vary across reproductive stages. Female rats with regular estrous cycles (RC), representing reproductive cyclicity, and rats in periestropause (PE), a stage similar to perimenopause, were exposed to normoxia, hypoxia, and hypercapnia. Ventilation (V̇E) and metabolism (V̇O2) were measured du

Maternal-embryo interactions play a critical role in early mammalian development, with extracellular vesicles (EVs) playing a key role in intercellular communication. Recent studies have focused on the mechanisms by which maternal-derived factors, such as RNA, proteins, and metabolites influence gap junctions, EVs, and direct cell-to-cell interactions, contributing to embryonic development. In this study, using a proteomics approach, we investigated the impact of EVs secreted from porcine endometrial cells (pEECs) and their protein cargoes on embryonic development. We characterized EVs isolate

Deep brain stimulation (DBS) is currently being investigated in patients and preclinical models of posttraumatic stress disorder (PTSD), but differences in behaviour according to sex remain elusive. We exposed female and male rats to fear conditioning and extinction. Thereafter, animals were treated with ventromedial prefrontal cortex DBS, followed by a battery of tests to measure fear and anxiety-like behaviour. As in our prior work, animals with high freezing scores during extinction (weak extinction; WE) were segregated from those with lower freezing scores (non-weak extinction; nWE), since

Social stressors represent highly relevant stress models which remain understudied in females. Concordantly, little is known of the impact of different types of social stress on female reproductive functioning, despite close interactions between the hypothalamic-pituitary-adrenal (HPA) axis and hypothalamic-pituitary-gonadal (HPG) axis. This study provides a comprehensive assessment of reproductive functioning in female rats following exposure to three social stress paradigms. 112 regularly cycling adult female Wistar rats were exposed to 21 days of isolation, crowding, social instability, con
