A Bioresistant Nitroxide Spin Label for In-Cell EPR Spectroscopy: In Vitro and In Oocytes Protein Structural Dynamics Studies.

Approaching protein structural dynamics and protein-protein interactions in the cellular environment is a fundamental challenge. Owing to its absolute sensitivity and to its selectivity to paramagnetic species, site-directed spin labeling (SDSL) combined with electron paramagnetic resonance (EPR) has the potential to evolve into an efficient method to follow conformational changes in proteins directly inside cells. Until now, the use of nitroxide-based spin labels for in-cell studies has represented a major hurdle because of their short persistence in the cellular context. The design and synth

Plant-mediated nitrous oxide emissions.

Plants are increasingly recognized as active contributors to nitrous oxide (N2O) fluxes. However, the physiological and biochemical processes underlying these plant-mediated N2O emissions remain poorly defined. This review synthesizes current understanding of direct N2O production pathways within plants, including chloroplast- and mitochondrial-associated processes involving nitrate and nitrite reductases, as well as cytochrome-mediated reactions under specific conditions. Plant-microbe interactions in the rhizosphere and phyllosphere further influence N2O dynamics. Symbiotic nitrogen-fixing b

Loss of S-nitrosoglutathione reductase disturbs phytohormone homeostasis and regulates shoot side branching and fruit growth in tomato.

S-Nitrosoglutathione plays a central role in nitric oxide (NO) homeostasis, and S-nitrosoglutathione reductase (GSNOR) regulates the cellular levels of S-nitrosoglutathione across kingdoms. Here, we investigated the role of endogenous NO in shaping shoot architecture and controlling fruit set and growth in tomato (Solanum lycopersicum). SlGSNOR silencing promoted shoot side branching and led to reduced fruit size, negatively impacting fruit yield. Greatly intensified in slgsnor knockout plants, these phenotypical changes were virtually unaffected by SlGSNOR overexpression. Silencing or knockin

Nitric Oxide Improves Salt Tolerance of Cyclocarya paliurus by Regulating Endogenous Glutathione Level and Antioxidant Capacity.

Cyclocarya paliurus is commonly used to treat diabetes in China. However, the natural habitats of C. paliurus are typically affected by salt stress. Previous studies showed that nitric oxide (NO) level was related to salt tolerance of C. paliurus, and its synthesis was induced by exogenous hydrogen sulfide. However, the effects of different NO donors in alleviating the negative effect of salt stress are still unclear. In the present study, C. paliurus seedlings pretreated with three NO donors (S-nitroso-N-acetylpenicillamine, SNAP and S-nitrosoglutathione, GSNO and sodium nitroprusside, SNP) w

Crystal Structure of Dihydro-Heme d1 Dehydrogenase NirN from Pseudomonas aeruginosa Reveals Amino Acid Residues Essential for Catalysis.

Many bacteria can switch from oxygen to nitrogen oxides, such as nitrate or nitrite, as terminal electron acceptors in their respiratory chain. This process is called "denitrification" and enables biofilm formation of the opportunistic human pathogen Pseudomonas aeruginosa, making it more resilient to antibiotics and highly adaptable to different habitats. The reduction of nitrite to nitric oxide is a crucial step during denitrification. It is catalyzed by the homodimeric cytochrome cd1 nitrite reductase (NirS), which utilizes the unique isobacteriochlorin heme d1 as its reaction center. Altho

The protective efficacy of soursop fruit extract against hepatic injury associated with acetaminophen exposure is mediated through antioxidant, anti-inflammatory, and anti-apoptotic activities.

In the current report, we examined the potential beneficial role of soursop fruit extract (SSFE) on liver injury induced by a single paracetamol (APAP) overdose (2000 mg/kg). Thirty-five Wistar albino rats were randomly divided into five groups as follows: control, SSFE, APAP, SSFE+APAP, and silymarin (SIL)+APAP. APAP intoxication was found to elevate alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and total bilirubin levels. Moreover, it increased the levels of malondialdehyde, nitrites, and nitrates and depleted glutathione, superoxide dismutase, catalase, glutath

MITOCHONDRIAL DYSFUNCTION IN THE AGING HEART IS ACCOMPANIED BY CONSTITUTIVE NO-SYNTHASES UNCOUPLING ON THE BACKGROUND OF OXIDATIVE AND NITROSATIVE STRESS.

We investigated the sensitivity of mitochondrial permeability transition pore (MPTP) opening to its inductor calcium, as well as indicators of oxidative and nitrosative stress in adult and old rat heart mitochondria and heart tissues. The coupling index of constitutive Ca2+/calmodulin-dependent NO-synthase (сNOS) was calculated based on experimentally found parameters. The aging is characterized by oxidative and nitrosative stress, which accompanied by a decrease of the cNOS coupling index and an increased sensitivity of MPTP to calcium. We found that in the heart mitochondria of old rats such

Roles of sodium hydrosulfide and sodium nitroprusside as priming molecules during drought acclimation in citrus plants.

Emerging evidence suggests that the gaseous molecules hydrogen sulfide (H2S) and nitric oxide (NO) enhances plant acclimation to stress; however, the underlying mechanism remains unclear. In this work, we explored if pretreatment of citrus roots with NaHS (a H2S donor) or sodium nitroprusside (SNP, a NO donor) for 2 days (d) could elicit long-lasting priming effects to subsequent exposure to PEG-associated drought stress for 21 d following a 5 d acclimation period. Detailed physiological study documented that both pretreatments primed plants against drought stress. Analysis of the level of nit

Cadmium decreases crown root number by decreasing endogenous nitric oxide, which is indispensable for crown root primordia initiation in rice seedlings.

Cadmium (Cd) is toxic to crown roots (CR), which are essential for maintaining normal growth and development in rice seedlings. Nitric oxide (NO) is an important signaling molecule that plays a pivotal role in plant root organogenesis. Here, the effects of Cd on endogenous NO content and root growth conditions were studied in rice seedlings. Results showed that similar to the NO scavenger, cPTIO, Cd significantly decreased endogenous NO content and CR number in rice seedlings, and these decreases were recoverable with the application of sodium nitroprusside (SNP, a NO donor). Nitric oxide production occurs downstream of reactive oxygen species in guard cells during stomatal closure induced by chitosan in abaxial epidermis of Pisum sativum.

The effects of chitosan (beta-1,4 linked glucosamine, a fungal elicitor), on the patterns of stomatal movement and signaling components were studied. cPTIO (NO scavenger), sodium tungstate (nitrate reductase inhibitor) or L: -NAME (NO synthase inhibitor) restricted the chitosan induced stomatal closure, demonstrating that NO is an essential factor. Similarly, catalase (H(2)O(2) scavenger) or DPI [NAD(P)H oxidase inhibitor] and BAPTA-AM or BAPTA (calcium chelators) prevented chitosan induced stomatal closure, suggesting that reactive oxygen species (ROS) and calcium were involved during such re

Anaerobic inductions of active forms of superoxide dismutases in Escherichia coli.

Escherichia coli growing anaerobically respond to NO3- with a approximately 3-fold induction of active FeSOD and a approximately 5.5-fold induction of an inactive, but activatable form of MnSOD (pro-MnSOD). Paraquat, which mediates anaerobic electron flow to NO3-, increased the induction of pro-MnSOD to approximately 2.5-fold. Strains with defects in the SOD genes or which lacked nitrate reductase activity failed to accumulate active or pro-forms of SODs in response to NO3- +/- PQ++. Diamide caused anaerobic induction of active MnSOD and this effect was also observed in a glutathione-negative

The bacterial strains JAM1T and GP59 of the species Methylophaga nitratireducenticrescens differ in their expression profiles of denitrification genes in oxic and anoxic cultures.

BACKGROUND: Strain JAM1T and strain GP59 of the methylotrophic, bacterial species Methylophaga nitratireducenticrescens were isolated from a microbial community of the biofilm that developed in a fluidized-bed, methanol-fed, marine denitrification system. Despite of their common origin, both strains showed distinct physiological characters towards the dynamics of nitrate ( NO 3 - ) reduction. Strain JAM1T can reduce NO 3 - to nitrite ( NO 2 - ) but not NO 2 - to nitric oxide (NO) as it lacks a NO-forming NO 2 - reductase. Strain GP59 on the other hand can carry the complete redu

Impact of Medications for Stable Angina Pectoris on Osteoporosis: A Review of Current Evidence.

Stable angina pectoris (SAP) and osteoporosis (OP) are both prevalent conditions among the elderly population. Compared to SAP, the prevention and management of OP are often neglected. Furthermore, certain medications used long-term for SAP may exert significant effects on bone metabolism. This review summarizes the impact of commonly prescribed SAP medications on OP. Extensive research indicates that nitrates not only promote vascular and osteogenic coupling via the nitric oxide (NO)-cyclic guanosine monophosphate (cGMP)-protein kinase G (PKG) pathway, enhancing the osteogenic effects of estr

[Stable coronary artery disease--invasive treatment does not replace secondary prevention].

Secondary prevention, i.e. nonsmoking, exercise, weight control, correct nutrition and drugs affecting the prognosis, constitute the basis for the treatment of stable coronary artery disease. The most important drug affecting the prognosis is acetylsalicylic acid. Statin medication can be reduced on the basis of adverse effects only. Angiotensin convertase inhibitor medication is often forgotten in coronary artery disease patients who have undergone cardiac infarction or a temporary stage of cardiac insufficiency. Long-acting nitrate is not the first-line antianginal treatment, beta-blockers a

Pharmacological approaches of refractory angina.

Refractory angina refers to a group of patients with stable coronary atherosclerotic disease and angina symptoms, unresponsive to traditional medical management, while considered to be suboptimal candidates for revascularization procedures. Up to 15% of angina patients are considered to have refractory angina and, taking into account the aging population and the improvements in the treatment of stable coronary artery disease, the incidence of this entity is expected to increase. This review describes traditional and novel pharmacotherapies for symptoms relief and for long-term management of re

Proteomic analysis of Brucella suis under oxygen deficiency reveals flexibility in adaptive expression of various pathways.

Low oxygen tension was proposed to be one of the environmental parameters characteristic of the patho-physiological conditions of natural infections by Brucella suis. We previously showed that various respiratory pathways may be used by B. suis in response to microaerobiosis and anaerobiosis. Here, we compare the whole proteome of B. suis exposed to such low-oxygenated conditions to that obtained from bacteria grown under ambient air using 2-D DIGE. Data showed that the reduction of basal metabolism was in line with low or absence of growth of B. suis. Under both microaerobiosis and anaerobios

Galaxolide, a Synthetic Musk, Disrupts Hepatic Mitochondrial Redox Balance in Mozambique Tilapia.

Galaxolide, a synthetic polycyclic musk, is an emerging contaminant of increasing concern due to its potential risks to both human and aquatic health through direct and indirect exposure. The present study examined the acute and subchronic effects of sublethal and environmentally relevant concentrations of galaxolide on hepatic mitochondrial function in the freshwater fish, Oreochromis mossambicus. The 96-h median lethal concentration (LC50-96 h) of galaxolide in O. mossambicus was determined to be 5.5 mg/L. Subsequently, fish were exposed to a sublethal concentration (550 μg/L; 1/10th of LC50

Helium governs nitric oxide signaling to improve alfalfa salinity tolerance by reestablishing redox and ion homeostasis.

Helium, previously described as "biologically inert", displays great medicinal potential in a number of respiratory ailments via influencing redox signaling. However, whether or how this gas functions in plant biology is still elusive. Here, hydroponic and pot experiments showed that mimicking the responses achieved by nitric oxide (NO)-releasing compound, exogenous helium supply with helium-enriched solution or its fumigation significantly prevented NaCl-induced growth inhibition of alfalfa seedlings. Lower level of Na+/K+ ratio in both root and shoot parts caused by lower efflux of net K+ an
