Global losses due to dairy cattle diseases: A comorbidity-adjusted economic analysis.

An economic simulation was carried out over 183 milk-producing countries to estimate the global economic impacts of 12 dairy cattle diseases and health conditions: mastitis (subclinical and clinical), lameness, paratuberculosis (Johne's disease), displaced abomasum, dystocia, metritis, milk fever, ovarian cysts, retained placenta, and ketosis (subclinical and clinical). Estimates of disease impacts on milk yield, fertility, and culling were collected from the literature, standardized, meta-analyzed using a variety of methods ranging from simple averaging to random-effects models, and adjusted

Rumen fluke infections (Paramphistomidae) in diarrhoeal cattle in western France and association with production parameters.

This study was conducted to assess the impact of rumen flukes (RFs) (Paramphistomatidae) on various production parameters of cattle in Normandy. Faecal and blood samples were taken between 1 January 2010 and 31 December 2019 as part of the routine diagnostic activity for diarrhoea in weaned cattle, including a quantitative parasitological analysis coupled with a search for Johne's disease (paratuberculosis). Information on slaughter and carcass weight, inter-farm movements and mortality was obtained from the French national registration database (BDNI). The study was conducted at two levels us

Rapid and sensitive detection of an intracellular pathogen in human peripheral leukocytes with hybridizing magnetic relaxation nanosensors.

Bacterial infections are still a major global healthcare problem. The quick and sensitive detection of pathogens responsible for these infections would facilitate correct diagnosis of the disease and expedite treatment. Of major importance are intracellular slow-growing pathogens that reside within peripheral leukocytes, evading recognition by the immune system and detection by traditional culture methods. Herein, we report the use of hybridizing magnetic nanosensors (hMRS) for the detection of an intracellular pathogen, Mycobacterium avium spp. paratuberculosis (MAP). The hMRS are designed to

Bacteria in enteric lesions of cattle.

Thirty-nine species of bacteria were isolated from or demonstrated in the abomasal and small and large intestinal mucosa of 23 adult cattle and 41 calves and identified. The bacteria isolated were related to the gross and microscopical lesions. Campylobacters, Clostridium perfringens type A, C sordellii, Actinobacillus lignieresii, Fusobacterium necrophorum, Mycobacterium paratuberculosis, Aeromonas hydrophila and Escherichia coli were all associated with specific lesions. The relationship of other bacteria such as Bacillus licheniformis, Branhamella catarrhalis and Bacteroides vulgatus to les

Safety and Immunogenicity of a Novel Recombinant Simian Adenovirus ChAdOx2 as a Vectored Vaccine.

Adenovirus vectored vaccines are a highly effective strategy to induce cellular immune responses which are particularly effective against intracellular pathogens. Recombinant simian adenovirus vectors were developed to circumvent the limitations imposed by the use of human adenoviruses due to widespread seroprevalence of neutralising antibodies. We have constructed a replication deficient simian adenovirus-vectored vaccine (ChAdOx2) expressing 4 genes from the Mycobacterium avium subspecies paratuberculosis (AhpC, Gsd, p12 and mpa). Safety and T-cell immunogenicity results of the first clinica

Mucosa-associated lymphoid tissues as sites for uptake, carriage and excretion of tubercle bacilli and other pathogenic mycobacteria.

Pathogenic mycobacteria, including those that cause tuberculosis and paratuberculosis, cross mucosal barriers by endocytosis within mucosal lymphoepithelial sites. These entry sites commonly include oropharyngeal and nasopharyngeal tonsils and Peyer's patches. Bacilli discharged at the basolateral surfaces of engulfing epithelial M cells are taken up by professional antigen-presenting cells associated with T lymphocytes of the parafollicular area. Dendritic cells and macrophages in these sites allow mycobacterial replication, due to the permissive immunological environment in lymphoepithelial

Identification of sero-reactive antigens for the early diagnosis of Johne's disease in cattle.

Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of Johne's disease (JD), a chronic intestinal inflammatory disease of cattle and other ruminants. JD has a high herd prevalence and causes serious animal health problems and significant economic loss in domesticated ruminants throughout the world. Since serological detection of MAP infected animals during the early stages of infection remains challenging due to the low sensitivity of extant assays, we screened 180 well-characterized serum samples using a whole proteome microarray from Mycobacterium tuberculosis (MTB), a c

Contribution of respiratory burst activity to innate immune function and the effects of disease status and agent on chemiluminescence responses by ruminant phagocytes in vitro.

The mechanisms of interaction between phagocytes and different bacteria that help resolve lung infections or contribute to lung pathology are poorly defined. Alveolar phagocytes (resident macrophages and recruited neutrophils) make a major contribution to innate immunity by mounting a respiratory burst that helps kill internalised bacteria. However, this ability may be altered during or after exposure to infection. This review considers the application and limitations of a variety of analytical methods for oxygen-dependent mechanisms of respiratory burst in phagocytes initiated by soluble and

Bacterial diseases of farmed deer and bison.

The most important aerobic bacterial diseases of farmed deer and bison include bovine tuberculosis, Johne's disease (paratuberculosis), yersiniosis, leptospirosis, brucellosis, pasteurellosis, anthrax, salmonellosis and colibacillosis. Anaerobic bacterial infections affecting the same animals include necrobacillosis and a number of clostridial diseases such as tetanus, blackleg, malignant oedema and pulpy kidney. The relative importance of these diseases will vary throughout the world according to timing and circumstance, but bovine tuberculosis and Johne's disease are likely to present the mo

Study of animal-borne infections in the mucosas of patients with inflammatory bowel disease and population-based controls.

Crohn's disease may be triggered by an infection, and it is plausible to consider that such an infection may be animal borne and ingested with our food. There has been considerable interest in the past in determining whether Mycobacterium avium subsp. paratuberculosis (M. avium) might be the etiologic agent in Crohn's disease since it causes a disease in cattle that is similar to Crohn's disease in humans. We aimed to determine if there was an association between Crohn's disease and infection with M. avium or other zoonotic agents and compared the findings with those for patients with ulcerati

Herd-level prevalence of selected endemic infectious diseases of dairy cows in Great Britain.

To implement appropriate and effective disease control programs at the national level, up-to-date and unbiased information on disease frequency is needed. The aim of this study was to estimate the prevalence of selected endemic infectious diseases in the population of dairy herds in Great Britain. Bulk milk tank (BMT) samples from 225 randomly selected dairy farms, stratified by region and herd size, were tested for antibodies against bovine viral diarrhea virus (BVDV), bovine herpesvirus type 1, Mycobacterium avium ssp. paratuberculosis, Leptospira Hardjo, Salmonella spp., Coxiella burnetii,

Surveys for disease agents in introduced elk in Arkansas and Kentucky.

Surveys for disease agents were conducted in introduced free-ranging elk (Cervus elaphus nelsoni) in Arkansas and Kentucky. Elk had been captured in Colorado and Nebraska and released in Arkansas during 1981-1985. From 1997 through 2002 elk were captured in Arizona, Kansas, North Dakota, New Mexico, Oregon, and Utah and released in southeastern Kentucky. Specimens were collected from 170 hunter-killed elk in Arkansas during 1998-2006, and 44 elk in Kentucky during 2001-2004. Significant findings included isolation of Mycobacterium avium paratuberculosis from one elk in Kentucky and evidence of

Zoonotic risks of pathogens from dairy cattle and their milk-borne transmission.

Dairy products are major sources of high-quality protein and bioavailable nutrients and dairy production contributes to local, regional and national-level economies. Consumption of raw milk and raw milk products does, however, carry a zoonotic risk, as does direct contact with cattle by farm husbandry staff and other employees. This review will mainly focus on the latter, and deal with it from the standpoint of a well-developed dairy industry, using the example of the Netherlands. With regard to dairy cattle, the main bacterial pathogens are Salmonella spp., Listeria monocytogenes and Leptospi

A comparative study between milk- and serum-based antibody detection assays for Johne's disease in New Zealand dairy cattle.

Dairy cattle are affected by Johne's disease. It is caused by Mycobacterium avium subspecies paratuberculosis (MAP). Suboptimal diagnostic tests add more to the productivity loss resulting from this disease. Agreement between and within different commercial kits is crucial in the decision-making process of disease surveillance programmes. This study compared two ELISAs, that is, Johne's disease commercial antibody detection kits (A and B), using milk and serum samples from New Zealand dairy cattle. These results were also compared with a subset of faecal PCR results. Five scenarios were consid

A review of bovine colostrum preservation techniques.

Preservation of colostrum for neonatal dairy calves has seldom been seldom in recent years, much of the peer reviewed literature having been published in the 1970s and 1980s. First milking colostrum is high in bioactive immune enhancers such as immunoglobulins, lactoferrins, lysozymes and cytokines and is vital to confer passive immunity to newborn dairy calves to promote their health, welfare and future productivity. Bovine colostrum is advisedly restricted from the bulk milk supply for the first 8 milkings post calving due to high somatic cell counts and the risk of antimicrobial residues. A

Idiopathic eosinophilic enteritis in four cattle.

Four cattle had chronic diarrhea and had lost weight but were not anorectic or dehydrated. A diagnosis of eosinophillic enteritis was made on the basis of a combination of clinical and histopathologic findings and exclusion of other diseases. Eosinophilia and hypoalbuminemia were not detected in CBC analyses. Gross and histologic examination of the small intestine revealed edema, diffuse eosinophilic inflammation (> 20 eosinophils/HPF [400 x]), and regional lymphadenopathy. A causative agent was not identified in any of the cattle. Treatment consisting of a 1-month course of systemically admin

Utilising on-farm risk assessment data for the management of Johne's disease in dairy cattle in Northern Ireland.

Johne's disease (JD) causes weight loss, diarrhoea, and reduced milk yields in clinically infected cattle. In 2020, Animal Health and Welfare Northern Ireland (AHWNI) launched a voluntary JD control programme (JDCP) which focuses on bio-exclusion, biocontainment and market reassurance. Authorised veterinary practitioners (AVPs) conduct a Veterinary Risk Assessment and Management Plan (VRAMP) and use this information to make up to three recommendations. Between August 2022 and January 2024, 2274 herds enrolled in the NI JDCP and conducted up to three VRAMPs. This study characterised the JD-rela
