Recent outbreaks of food-borne illness associated with the consumption of produce have increased concern over wildlife reservoirs of food-borne pathogens. Wild rodents are ubiquitous, and those living close to agricultural farms may pose a food safety risk should they shed zoonotic microorganisms in their feces near or on agricultural commodities. Fecal samples from wild rodents trapped on 13 agricultural farms (9 produce, 3 cow-calf operations, and 1 beef cattle feedlot) in Monterey and San Benito Counties, CA, were screened to determine the prevalence and risk factors for shedding of several

Rodents of the genus Peromyscus are among the most common North American mammals. Crosses between natural populations of two of these species, P. maniculatus (BW) and P. polionotus (PO), produce parent-of-origin effects on growth and development. BW females mated to PO males produce growth-retarded offspring. In contrast, PO females mated to BW males produce overgrown but dysmorphic conceptuses. Variation in imprinted loci and control of genomic imprinting appear to underlie the hybrid effects. Prior morphological and genetic analyses have focused on placental and post-natal growth. We evaluated the relationships between forest understory structure and the abundance of questing adult and nymphal blacklegged ticks, Ixodes scapularis Say (Acari: Ixodidae), in three Maine towns endemic for Lyme disease, 2001-2003. In fragmented New England woodlands, over-abundant white-tailed deer, Odocoileus virginianus Zimmerman, overbrowse palatable species, allowing browse-resistant exotic-invasive species to replace native forest understory structures. We predicted there would be more ticks in plots dominated by exotic-invasive shrubs (such as Japanese barberry, Berberis thunbergii DC)

The extent to which the biodiversity and community composition of ecosystems affect their functions is an issue that grows ever more compelling as human impacts on ecosystems increase. We present evidence that supports a novel function of vertebrate biodiversity, the buffering of human risk of exposure to Lyme-disease-bearing ticks. We tested the Dilution Effect model, which predicts that high species diversity in the community of tick hosts reduces vector infection prevalence by diluting the effects of the most competent disease reservoir, the ubiquitous white-footed mouse (Peromyscus leucopu

To determine whether rodents that are intensely exposed to the deer tick-transmitted agents of Lyme disease, human granulocytic ehrlichiosis, and human babesiosis are also exposed to deer tick virus (DTV), we assayed serum samples from white-footed mice (Peromyscus leucopus) and meadow voles (Microtus pennsylvanicus) in sites densely infested by deer ticks. To conduct serosurveys, we developed an enzyme-linked immunosorbent assay (ELISA) and Western blot assay by cloning, expressing, and purifying a portion of the DTV envelope glycoprotein (DTV rE) for use as test antigen. Sera from mice and v

Recent investigations have detected polychlorinated biphenyl (PCB) body burdens in wild white-footed mice (Peromyscus leucopus) captured at hazardous waste sites. Insufficient information is currently available to interpret the toxicological significance of these body burdens. In an effort to provide this information, we investigated hepatic changes and PCB body burdens in white-footed mice following a 21-day dietary exposure to a PCB mixture, Aroclor 1254. Dietary concentrations tested were 0, 2.5, 25, 50, and 100 mg Aroclor 1254/kg diet (reported as ppm). Liver weights were significantly inc

Blacklegged ticks (Ixodes scapularis) are vectors for several important human diseases, including Lyme disease, human granulocytic anaplasmosis (HGA), and human babesiosis, caused by Borrelia burgdorferi, Anaplasma phagocytophilum, and Babesia microti, respectively. The continued northward range expansion of blacklegged ticks and associated pathogens is an increasing public health concern in Canada. The Thousand Islands region of eastern Ontario has recently been identified as a new endemic area for Lyme disease in Canada, but the occurrence of other pathogens in ticks in this area has not bee

Stereotyped behavior is commonly observed in neurodevelopmental disorders (e.g., autism, intellectual and developmental disability) and in a wide variety of animal species maintained in restricted environments. Stereotyped behavior can also be induced by psychostimulants, an effect potentiated by repeated intermittent exposure to these drugs (behavioral sensitization). The present study evaluated whether similar neuroadaptations in cortical-basal ganglia circuitry underlie the expression and development of spontaneous stereotypy and psychostimulant-induced sensitization. Sensitization was indu

Lyme disease, caused primarily in North America by the bacterium Borrelia burgdorferi sensu stricto, is the most frequently reported vector-borne disease in North America and its geographic extent is increasing in all directions from foci in the northeastern and north central United States. Several southeastern states, including Virginia and North Carolina, have experienced large increases in Lyme disease incidence in the past two decades, with the biggest changes in incidence occurring in the western portion of each state. We tested the hypothesis that B. burgdorferi s.s. was present in weste

Coexistence of multiple tick-borne pathogens or strains is common in natural hosts and can be facilitated by resource partitioning of the host species, within-host localization, or by different transmission pathways. Most vector-borne pathogens are transmitted horizontally via systemic host infection, but transmission may occur in the absence of systemic infection between two vectors feeding in close proximity, enabling pathogens to minimize competition and escape the host immune response. In a laboratory study, we demonstrated that co-feeding transmission can occur for a rapidly-cleared strai

Effects of Babesia microti Franca on the pattern of feeding time, the body weight of engorged ticks, and the molting rate of the blacklegged tick, Ixodes scapularis Say, were determined. Using the Syrian golden hamster, Mesocricetus auratus (Waterhouse), as an animal model, we found no significant differences in patterns of feeding time determined for both larvae and nymphs that fed on B. microti-infected and uninfected hamsters. However, an infection of B. microti in hamsters delayed engorgement of the ticks. The presence of B. microti had no adverse effects on the body weight of engorged tic

In recent decades, increased incidences of tick-borne diseases, including Lyme disease (Borrelia burgdorferi), Anaplasmosis (Anaplasma phagocytophilum), and Babesiosis (Babesia microti) have occurred in urban areas. The prevalence of the pathogens causing these diseases relies on a complex set of interactions between the tick vector (Ixodes scapularis) and its prominent hosts, white-tailed deer (Odocoileus virginianus) and white-footed mice (Peromyscus leucopus). Additionally, predators of small mammal hosts, such as red fox (vulpes) may influence host-tick interactions. While urbanization alt

Lyme disease, caused primarily by the bacterium Borrelia burgdorferi sensu stricto (s.s.), is the most prevalent vector-borne disease in the United States. Lyme disease spirochetes are maintained through enzootic cycles of pathogen transmission between vertebrate reservoir hosts and tick vectors. Reservoir host targeted strategies have been examined for their ability to disrupt enzootic transmission cycles of B. burgdoferi s.s. within natural environments in the eastern U.S. However, there are no studies in the western U.S. examining the effect of permethrin- and deltamethrin-based topical aca

Ixodes scapularis is considered a significant medical and veterinary arthropod pest, capable of transmitting several pathogens that cause disease in humans and animals. Previous work has identified two distinct populations of I. scapularis in the United States (northern and southern), characterized by differences in their genetics and behavior. This study aimed to characterize and compare the lateral movement and feeding behaviors of nymphal I. scapularis between the northern and southern populations in the United States. Using laboratory-reared ticks from BEI (northern), Oklahoma State Univer

Chiggers are larval mites that pose a significant health risk globally via the spread of scrub typhus. However, fundamental studies into the bacterial microbiome in North America have never been considered. In this investigation, chiggers were collected in the wild from two locally common rodent host species (i.e., Sigmodon hispidus and Peromyscus leucopus) in three different ecoregions of North Carolina (NC), United States to investigate the composition of their bacterial communities, including potential pathogens. DNA was extracted from the chiggers, and the V3-V4 regions of the bacterial 16

Through their potentially devastating impacts on the environment, wildfires may impact pathogen, vector, and host interactions, leading to changing risks of vector-borne disease in humans and other animals. Despite established risks for tick-borne disease and increasing frequency and severity of wildfires in the United States, impacts of wildfire on ticks and tick-borne pathogens are understudied. In 2015, the large Wragg fire extensively burned a long-term field site at Stebbins Cold Canyon University of California Reserve (CC). We characterized the tick, reservoir host and pathogen community

Evolutionary adaptation to extreme environments often requires coordinated changes in multiple intersecting physiological pathways, but how such multi-trait adaptation occurs remains unresolved. Transcription factors, which regulate the expression of many genes and can simultaneously alter multiple phenotypes, may be common targets of selection if the benefits of induced changes outweigh the costs of negative pleiotropic effects. We combined complimentary population genetic analyses and physiological experiments in North American deer mice (Peromyscus maniculatus) to examine links between gene

Rodent species were assessed as potential hosts of Trypanosoma cruzi, the etiologic agent of Chagas disease, from five sites throughout Texas in sylvan and disturbed habitats. A total of 592 rodents were captured, resulting in a wide taxonomic representation of 11 genera and 15 species. Heart samples of 543 individuals were successfully analyzed by SybrGreen-based quantitative PCR (qPCR) targeting a 166 bp fragment of satellite DNA of T. cruzi. Eight rodents representing six species from six genera and two families were infected with T. cruzi. This is the first report of T. cruzi in the pygmy

A mark-release-recapture study was conducted during 2007 through 2010 in six, tick-infested sites in Connecticut, United States to measure changes in antibody titers for Borrelia burgdorferi sensu stricto, Anaplasma phagocytophilum, and Babesia microti in Peromyscus leucopus (white-footed mice). There was an overall recapture rate of 40%, but only four tagged mice were caught in ≥2 yr. Sera from 561 mice were analyzed for total antibodies to B. burgdorferi and A. phagocytophilum by using whole-cell or recombinant (VlsE or protein 44) antigens in a solid-phase enzyme-linked immunosorbent assay

To compare rickettsial infectivity and seropositivity rates against spotted fever group (SFG) rickettsiae, ticks and wild mammals were collected from three areas where Rickettsia rickettsii was thought to be enzootic in Connecticut during 1978-1979, and from four additional sites (with no reported human cases) between 1976 and 1979. Of the 1,001 Dermacentor variabilis examined by the hemolymph test, 59 (5.9%) contained rickettsia-like organisms; direct immunofluorescence tests verified the presence of SFT rickettsiae in 14 specimens. Prevalence of rickettsiae-infected ticks at Newtown, an area
