In its blood stages the malaria parasite, Plasmodium, displays very high lipid metabolism. We present evidence for an abundant long-chain acyl-CoA synthetase (EC 6.2.1.3) activity in Plasmodium knowlesi-infected simian erythrocytes. The activity was found to be 20-fold higher in the schizont-infected (the last parasite stage) than in control erythrocytes. The cosubstrate requirements of the enzyme were similar to those previously reported for acyl-CoA synthetases from other sources. Among the separated reaction products of oleyl-CoA synthetase, only PPi and oleyl-CoA were inhibitory, with Ki o

Bovine tropical theileriosis (Theileria annulata infection) is of great economic importance to several countries of Asia and Africa where millions of cattle are at risk of exposure to this disease. The authors describe symptoms, sequence of appearance of the parasite and pathology of the infection by three virulent strains of the parasite in fully susceptible calves. Symptoms observed were: enlargement of local lymph gland, fever, haemorrhages on visible mucous membranes and sometime on skin, anaemia and jaundice particularly in subacute and chronic forms of the disease. The parasite "schizont

Plasmodium falciparum merozoites invade erythrocytes using various ligand-receptor interactions. Important ligands encoded by the eba and Rh gene families have varying expression levels in different parasite isolates, affecting their vaccine candidacy. Analyses of clinical isolates from endemic areas in Africa have indicated that most variation in these expression profiles exists within each local area, and only minor differences are seen between areas, although comparisons with non-African populations have not previously been performed. To enable this, relative transcript levels of three eba

Theileria annulata infection poses a significant threat to livestock health and production worldwide. This study investigated the prevalence, morphological characteristics, molecular identification, and physiological impacts of T. annulata in cattle and buffaloes across three Egyptian governorates: Beni Suef, Faiyum, and Giza. Blood samples from 300 cattle and 250 buffaloes were analyzed using microscopic examination, molecular techniques, and biochemical assays. Giemsa-stained blood smears revealed intraerythrocytic piroplasms and intralymphocytic schizonts. These morphological findings were

Phosphoinositides (PIPs), are key regulators of membrane identity and vesicular trafficking. By dynamically shaping the lipid composition of intracellular membranes, PIPs help ensure the specificity of cargo delivery. In apicomplexan parasites such as Plasmodium falciparum, the biogenesis of the specialized secretory organelles involved in erythrocyte invasion (named rhoptries, micronemes, and dense granules), remains poorly understood, particularly regarding how proteins are sorted and specifically targeted to their respective destinations. Our hypothesis is that PIPs might play a role in thi

INTRODUCTION: Host gene and protein expression impact susceptibility to clinical malaria, but the balance of immune cell populations, cytokines and genes that contributes to protection, remains incompletely understood. Little is known about the determinants of host susceptibility to clinical malaria at a time when acquired immunity is developing.
METHODS: We analyzed peripheral blood mononuclear cells (PBMCs) collected from children who differed in susceptibility to clinical malaria, all from a small town in Mali. PBMCs were collected from children aged 4-6 years at the start, peak and end of

INTRODUCTION AND IMPORTANCE: Anaplastic lymphoma kinase negative anaplastic large cell lymphoma (ALK-ALCL) is a rare CD30-positive peripheral T-cell lymphoma with a low incidence of ALK-ALCL, which mainly involves lymph nodes and, in a minority of patients, extra-nodal tissues. The etiology is unknown, and the incidence is higher in males.
CASE PRESENTATION: We report a case of primary cutaneous ALK-ALCL. On light microscopy, the cells showed marked heterogeneity with abundant cytoplasm and numerous nuclear schizonts, and immunohistochemistry showed large mesenchymal cells strongly positive f

Antifolates targeting dihydrofolate reductase (DHFR) are antimalarial compounds that have long been used for malaria treatment and chemoprevention (inhibition of infection from mosquitoes to humans). Despite their extensive applications, a thorough understanding of antifolate activity against hepatic malaria parasites, especially resistant parasites, has yet to be achieved. Using a transgenic Plasmodium berghei harboring quadruple mutant dhfr from Plasmodium falciparum (Pb::Pfdhfr-4M), we demonstrated that quadruple mutations on Pfdhfr confer complete chemoprevention resistance to pyrimethamin

Malaria-associated acute respiratory distress syndrome (MA-ARDS) is a deadly complication of malaria, and its pathophysiology is insufficiently understood. Both in humans and in murine models, MA-ARDS is characterized by marked pulmonary inflammation. We investigated the role of hemozoin in MA-ARDS in C57Bl/6 mice infected with Plasmodium berghei NK65, P. berghei ANKA, and P. chabaudi AS. By quantifying hemozoin in the lungs and measuring the disease parameters of MA-ARDS, we demonstrated a highly significant correlation between pulmonary hemozoin concentrations, lung weights, and alveolar ede

Theileriosis is an economically important hemoprotozoal disease with high morbidity and mortality in cattle. The present study reported the pathological features of a natural outbreak of tropical bovine theileriosis due to Theileria annulata in Fars Province, southern Iran. T. annulata was confirmed by the presence of T. annulata piroplasms in the blood smears and also by polymerase chain reaction test. On necropsy, pale mucous membranes and petechial and ecchymotic hemorrhages in the mucosal and serosal surfaces together with lymphadenopathy were observed. The liver was friable, yellowish, an

BACKGROUND: Antibodies in adults living in malaria endemic areas that target specific parasite antigens are implicated in protective immunity to infection and disease. This study aimed to identify, isolate and characterise targets of protective immunity in malaria. A Plasmodium falciparum antigen termed UB05 (Genbank Accession Number DQ235690: PlasmoDB PF10_ 0372) that had been isolated by immunoscreening with semi-immune sera was studied.
METHODS: Polymerase chain reaction, sequencing and bioinformatics were used to analyse the UB05 gene. A specific mouse anti-UB05 antibody was used in paras

Merozoite surface protein-9 of Plasmodium vivax (PvMSP-9) is highly conserved and present in several malaria species. Here, we present the immunogenic properties of two recombinant glutathione S-transferase (GST) fusion proteins comprising the N-terminus (PvMSP-9-Nt) and the second block of tandem repeats (PvMSP-9-RepII) of PvMSP9. These recombinants proteins were used to immunize BALB/c mice. The specificity and subtyping of the antibodies and the cellular immune responses were evaluated by enzyme-linked immunosorbent assay (ELISA) and ELISPOT, respectively, using the recombinant proteins as

Ultrastructure of white blood cells (WBC) were studied in peripheral venous blood from Saudi patients with acute falciparum malaria (AFM) and compared with their counterparts in same patients 2 weeks after chloroquine treatment and full recovery. A counting system was incorporated to determine the rate of abnormal to normal cell type in plastic thick sections during the course of the disease. Neutrophilia, monocytosis, eosinopenia and lymphocytosis were associated with various ultrastructural abnormalities including: (1) Knobby phagocytic polymorphonuclear neutrophils (PMN) and promyelocytes,

Noninfected erythrocytes form rosettes around those infected with trophozoites and schizonts of Plasmodium falciparum in vitro. These rosettes are thought to contribute to the microvascular obstruction which underlies the pathophysiology of severe falciparum malaria. To determine whether the percentage of infected erythrocytes forming rosettes for a parasite isolates in vitro correlates with the in vivo severity of disease, we studied the rosette formation behavior of 35 isolates of P. falciparum from patients with uncomplicated, severe, and cerebral malaria. There was a wide variation in the

Cryptosporidia organisms were identified in 42 of 161 (26%) neonatal, diarrheic calves, over a 32 month period commencing July 1979. Forty of the 161 calves were submitted alive and cryptosporidiosis was diagnosed in 63% (25 of 40) of them. The cryptosporidia infected calves were usually one to two weeks old and came from 26 herds where the typical history was profuse, watery diarrhea in nearly all neonatal calves. The diarrhea usually started around one week of age, was unresponsive to all conventional antidiarrhea therapies, lasted for two or more weeks and was usually fatal. Twenty-nine (69

AIMS: As malaria elimination advances, low parasitaemia infections and declining microscopy expertise have made identification of Plasmodium species challenging. Although microscopy remains the diagnostic gold standard, it is prone to human error and is time consuming. Deep learning algorithms offer a supportive diagnostic framework to enhance rapid, automated, and accurate detection of malaria parasites in peripheral blood smear images.
METHODS AND RESULTS: We collected peripheral blood samples from 35 malaria-positive patients confirmed by microscopy, rapid diagnostic test and nested PCR. A

Although the number of cases of malaria has declined since the introduction of artemisinin-based combination therapy in endemic countries, the emergence of resistant parasites in the field has led to an alarming increase in treatment failure, making the search for novel drug candidates with precise targets critical. Here, we report the identification, from Shionogi's chemical library, of a 2-piperazino-pyrimidine compound (FPSA hereafter), with antimalarial activity; the compound shows a Plasmodium falciparum-killing effect resulting from blockage of merozoite release from infected erythrocyte

Malaria is a parasitic infectious disease that is endemic in many tropical countries. Even though several effective antimalarial agents have been implemented, treatment failure still occurs, and malaria continues to cause neurological complications and death, particularly in severe or drug-resistant cases. Hence, novel therapeutic agents with distinct mechanisms of action, as well as alternative chemical compounds that can overcome resistance, are still needed to improve malaria therapy. This study aimed to investigate the antimalarial activities of Brucea javanica, a tropical plant extracts a

Serine/arginine-rich protein kinases (SRPKs) are cell cycle-regulated serine/threonine protein kinases and are important regulators of splicing factors. In this study, we functionally characterize SRPK1 of the human malaria parasite Plasmodium falciparum. P. falciparum SRPK1 (PfSRPK1) was expressed in asexual blood-stage and sexual-stage gametocytes. Pfsrpk1- parasites formed asexual schizonts that generated far fewer merozoites than wild-type parasites, causing reduced replication rates. Pfsrpk1- parasites also showed a severe defect in the differentiation of male gametes, causing a complete
