Properties and regional distribution of pyruvate dehydrogenase kinase in rat brain.

A method is described to measure directly in rat brain the activity of pyruvate dehydrogenase kinase (PDHa kinase; EC 2.7.1.99), which catalyzes the inactivation of pyruvate dehydrogenase complex (PDHC, EC 1.2.4.1, EC 2.3.1.12, and EC 1.6.4.3). The activity showed the expected dependence on added ATP and divalent cation, and the expected inhibition by dichloroacetate, pyruvate, and thiamin pyrophosphate. These results, and the properties of pyruvate dehydrogenase phosphate phosphatase (EC 3.1.3.43), indicate that the mechanisms of control of phosphorylation of PDHC seem qualitatively similar i

Organic cerebral reactions in alcoholism: psychobiological aspects of treatment.

The paper considers the rational biological basis for treatment of alcohol withdrawal reactions, alcoholic hallucinosis and the Wernicke-Korsakoff syndrome. Both clinical and experimental data indicate that the alcohol withdrawal reaction reflects a state of hyper-reactivity of the central nervous system. The evidence comprises physiological, electrophysiological and biochemical findings. Magnesium, vitamins and antipsychotics are not specific in the treatment of the alcohol withdrawal reactions per se; rational treatment comprises drugs that may substitute for the suppressive effects of alcoh

[Changes in the activity of alkaline phosphatase and the level of thiamine diphosphatase in simulations of B1 avitaminosis].

Alimentary B1 avitaminosis was attended by a declining activity of alkaline phosphatase in the brain and spleen of rats. A single administration of oxythiamine to rats in a dose of 400 mg/kg produced during the first hours an increased activity of alkaline phosphatase in the liver, brain and, to a lesser extent, in a number of other organs and reduced the thiamine-diphosphate content in the liver, brain and other tissues. The thiamin-diphosphate level in the brain returned back to normal in 12 hours, and in other tissues-towards the 3--5th day. In 24 hours after introduction of oxythiamine the

The influence of hepatic insufficiency due to alcoholic cirrhosis on the erythrocyte transketolase activity (ETKA).

The erythrocyte transketolase activity (ETKA), the stimulated erythrocyte transketolase activity (ETKAS), and the thiaminepyrophosphate effect (TPPE) were measured in 21 alcoholic patients with cirrhosis and hepatic insufficiency, 13 alcoholic patients without cirrhosis and 21 non-alcoholic persons before and after oral treatment with 100 mg of thiamine daily for 2 weeks in order to investigate the influence of hepatic insufficiency on these variables. A statistically significant rise in ETKA and fall in TPPE were found in all three groups. ETKA, ETKAS and TPPE did not differ from each other i

Thiamine Concentration in Human Milk Is Correlated With Maternal and Infant Thiamine Status: A Cross-Sectional Analysis of the Lao Thiamine Study.

The human milk content of some micronutrients including thiamine depends on maternal status, and if low, breastfed infants are at risk of deficiency. Thiamine deficiency remains an important cause of morbidity and mortality among infants in Asia. We aimed to explore correlations between maternal thiamine diphosphate (ThDP) or erythrocyte transketolase activation coefficient (ETKac) and human milk thiamine concentration (MTh) and between MTh and infant ThDP or ETKac among breastfed infants < 6 months of age in northern Lao PDR. Hospitalized infants (aged ≥ 21 days) with symptoms suggestive of t

Amprolium-induced cerebrocortical necrosis (CCN) in dromedary racing camels.

Amprolium was successfully used to induce cerebrocortical necrosis (CCN) in dromedary racing camels, only when they were fed on a barley diet. Camels which were fed on hay ad libitum did not suffer form CCN, although their thiamine pyrophosphate (TPP) reached similar levels as in camels fed on barley. The reason for this phenomenon is discussed. Five camels which suffered from CCN had TPP values of 80-115% and were euthanized on humane grounds when they were in lateral recumbency. Pathohistological investigations revealed a polioencephalomalacia of the dorsal cerebral cortex with oedema and st

Operando 1H nuclear magnetic resonance (NMR) activity assay for pyruvate decarboxylase.

The biological conversion of pyruvate by thiamine pyrophosphate (TPP)-linked enzymes, such as pyruvate decarboxylase (Pdc), is a critical step in microbial production of biofuels such as ethanol. Traditionally, the activity of these enzymes is measured using UV-Vis spectrophotometric coupled assays. However, these assays typically only measure one chemical species. Here, we describe an Operando 1H Nuclear Magnetic Resonance (NMR) activity assay that provides a robust, non-destructive alternative for monitoring Pdc kinetics in real time. This quantitative NMR (qNMR) method allows for the simult

2-Hydroxyacyl-CoA lyase catalyzes acyloin condensation for one-carbon bioconversion.

Despite the potential of biotechnological processes for one-carbon (C1) bioconversion, efficient biocatalysts required for their implementation are yet to be developed. To address intrinsic limitations of native C1 biocatalysts, here we report that 2-hydroxyacyl CoA lyase (HACL), an enzyme involved in mammalian α-oxidation, catalyzes the ligation of carbonyl-containing molecules of different chain lengths with formyl-coenzyme A (CoA) to produce C1-elongated 2-hydroxyacyl-CoAs. We discovered and characterized a prokaryotic variant of HACL and identified critical residues for this newfound activ

Vitamin B2 status in athletes and the influence of riboflavin administration on neuromuscular irritability.

In order to obtain information on the vitamin B2 status in athletes in training, the following aspects were investigated: in one group of athletes, riboflavinuria at rest, during the autumn and winter months (mean values 23.8 and 9.45 mug/h, respectively); in the second group (n = 18), the activity of the nicotinamide-adenine-dinucleotide-phosphate-dependent red blood cell glutathione reductase (EC 1.6.4.2) and its activation by added flavin adenine dinucleotide. In 8 athletes, activation coefficients higher than 1.25 were found, indicating a biochemical deficiency of riboflavin. Erythrocyte t

Which stage of the process of apotransketolase interaction with thiamine diphosphate is affected by the regulatory activity of the donor substrate?

The interaction of thiamine diphosphate (ThDP) with transketolase (TK) involves at least two stages: [formula: see text] During the first stage, an inactive intermediate complex (TK...ThDP) is formed, which is then transformed into a catalytically active holoenzyme (TK* - ThDP). The second stage is related to conformational changes of the protein. In the preceding publication (Esakova, O. A., Meshalkina, L. E., Golbik, R., Hübner, G., and Kochetov, G. A. Eur. J. Biochem. 2004, 271, 4189 - 4194) we reported that the affinity of ThDP for TK considerably increases in the presence of the donor sub

Activation of Thiamine Pyrophosphokinase TPK-1 Contributes to 6-PPD Quinone-Induced Immunosuppression by Inhibiting Mitochondrial UPR in Caenorhabditis elegans.

As an emergent contaminant, 6-PPD quinone (6-PPDQ) causes toxic effects in organisms, including induction of immunosuppression. Thiamine pyrophosphate (TPP) serves as an important cofactor for some metabolisms. We aimed to examine the role of thiamine pyrophosphokinase TPK-1 in modulating 6-PPD-induced immunosuppression. Using Caenorhabditis elegans as the animal model, 6-PPDQ-exposure treatment was applied to nematodes from the L1 larval stage to adult day 3, and innate immune response was assessed by the expression of antimicrobial genes. In nematodes, 6-PPDQ (1-10 μg/L) elevated TPP content

Race explains substantial variance in whole blood thiamine diphosphate concentrations.

Deficiency for thiamine (vitamin B1), traditionally assessed via the activity of the thiamine-dependent enzyme erythrocyte transketolase, has been reported in individuals with alcohol use disorder (AUD) and in people with HIV; concentrations of the metabolically active diphosphate form, however, have yet to be reported in HIV cohorts and results in AUD are equivocal. In this cross-sectional study, samples from 170 AUD, 130 HIV, and 100 healthy control individuals were analyzed to test the hypothesis that AUD and HIV groups relative to healthy controls would show low whole blood thiamine diphos

The role of thiamine in nervous tissue.

The possibility that thiamine (vitamin B1) has a role in nervous tissue that is independent of its well-documented coenzyme function is discussed. After reviewing the localization and metabolism of the vitamin and its phosphate esters, the effects of either thiamine deprivation or antimetabolites of thiamine on conduction and transmission, and the relationship between thiamine triphosphate and the genetic, neurological disease, subacute necrotizing encephalomyelopathy (Leigh's disease), it is suggested that despite the lack of hard evidence, it is likely that the vitamin possesses this alterna
