BACKGROUND: Hyalomma ticks serve as vectors for a range of pathogens and harbor endosymbiotic bacteria that influence tick biology and the dynamics of pathogen transmission. In the United Arab Emirates, Hyalomma dromedarii and H. anatolicum are prevalent on livestock; yet, limited data exist on their microbial communities. This study aimed to determine the prevalence and phylogenetic relationships of key bacterial endosymbionts and tick-borne pathogens in Hyalomma ticks collected from livestock across all seven Emirates.
METHODS: A total of 671 ticks (532 H. dromedarii and 139 H. anatolicum)

BACKGROUND: Xenosurveillance, which uses blood-feeding insects as biological samplers, is an emerging non-invasive approach for monitoring pathogens circulating among humans, livestock, and wildlife. However, its application to livestock-associated bacterial pathogens at human-animal-wildlife interfaces remains underexplored. We investigated whether mosquito blood meals could be used to detect tick-borne bacterial pathogens circulating in livestock in Kenya.
METHODS: We collected 4673 mosquitoes, belonging to Culex, Anopheles, Aedes, Mansonia, and Coquillettidia genera around livestock enclos

BACKGROUND AND AIM: Zoonotic diseases caused by various blood parasites are important public health concerns that impact animals and humans worldwide. The traditional method of microscopic examination for parasite diagnosis is labor-intensive, time-consuming, and prone to variability among observers, necessitating highly skilled and experienced personnel. Therefore, an innovative approach is required to enhance the conventional method. This study aimed to develop a self-supervised learning (SSL) approach to identify zoonotic blood parasites from microscopic images, with an initial focus on par

Laboratory diagnosis of leishmaniasis shows variable efficacy in detecting infected mammalian hosts and there is a need to identify suitable antigens to improve the accuracy of diagnostic tests. In the present study, a L. infantum hypothetical protein called LiHyQ was evaluated for the diagnosis of tegumentary (TL) and visceral (VL) leishmaniasis using canine and human samples. A collection of dog sera (n=155) were tested and contained samples from asymptomatic (n=20) and symptomatic (n=25) VL animals, from healthy dogs living in endemic (n=25) or non-endemic (n=25) areas of disease, from Leis

BACKGROUND: Because ixodid ticks are vectors of zoonotic pathogens, including Borrelia, information of their abundance, seasonal variation in questing behaviour and pathogen prevalence is important for human health. As ticks are invading new areas northwards, information from these new areas are needed. Taiga tick (Ixodes persulcatus) populations have been recently found at Bothnian Bay, Finland. We assessed seasonal variation in questing abundance of ticks and their pathogen prevalence in coastal deciduous forests near the city of Oulu (latitudes 64-65°) in 2019.
METHODS: We sampled ticks fr

Bartonella vinsonii subspecies berkhoffii is a recently recognized zoonotic pathogen that causes endocarditis, granulomatous rhinitis, and granulomatous lymphadenitis in dogs. Isolation of B. vinsonii (berkhoffii) from blood or tissue samples is frequently unsuccessful; therefore, diagnosis is primarily dependent on serologic or molecular testing modalities. Because previous canine serologic studies have used an indirect immunofluorescence assay (IFA), without Western immunoblot (WI) confirmation, the overall objective of this study was to examine the diagnostic use of WI for confirmation of B

BACKGROUND: Understanding the range of vectors and pathogen diversity in sub-Saharan Africa is crucial for efficient disease prevention and control efforts. Domestic dogs are susceptible to many vector-borne pathogens (VBPs) and serve as sentinels for several human pathogens. Yet, critical surveillance gaps in canine VBPs persist in Central Africa, where the tropical climate and pastoral agro-livelihoods create unique transmission risks.
METHODS: We conducted a cross-sectional survey from September to October 2021, collecting 4238 ticks from 1254 domestic dogs across 56 villages in Chad, Afri

BACKGROUND: Documented changes in spatial and seasonal tick distribution highlight the importance of continuous surveillance. This paper documents results of a year-round sampling campaign in France, including identification of ticks and associated pathogens recovered from dogs and cats, as part of the European project "Protect Our Future Too."
METHODS: Ticks were collected from dogs and cats presented to 35 veterinary practices from 27 administrative French departments between April 2021 and July 2022. DNA extracted from each tick sample was amplified by polymerase chain reaction (PCR) for s

Babesia spp. and Theileria spp. are tick-borne apicomplexan protozoa that can cause disease in animals and humans. Deer are considered reservoirs for a wide variety of Piroplasmida species, including some potentially zoonotic. This study aimed to investigate the occurrence and genetic diversity of piroplasmids in wild deer sampled in four Brazilian states (São Paulo, Mato Grosso do Sul, Paraná and Goiás). For this purpose, extracted DNA samples from 181 deer buffy coat samples (138 Blastocerus dichotomus, 26 Subulo gouazoubira, 4 Mazama jucunda, 3 Mazama rufa and 10 Ozotocerus bezoarticus) wer

UNLABELLED: Protein disulfide isomerase, containing thioredoxin (Trx) domains, serves as a vital enzyme responsible for oxidative protein folding (the formation, reduction, and isomerization of disulfide bonds in newly synthesized proteins) in the endoplasmic reticulum (ER). However, the role of ER-localized PDI proteins in parasite growth and their interaction with secretory proteins remain poorly understood. In this study, we identified two ER-localized PDI proteins, TgPDI8 and TgPDI6, in Toxoplasma gondii. Conditional knockdown of TgPDI8 resulted in a significant reduction in intracellular

BACKGROUND: Theileria equi causes equine piroplasmosis, an economically significant disease that affects horses and other equids worldwide. Based on 18S ribosomal RNA (18S rRNA sequences), T. equi can be classified into five genotypes: A, B, C, D, and E. These genotypes have implications for disease management and control. However, no conventional polymerase chain reaction (PCR) assays are available to differentiate the genotypes of T. equi. To overcome this limitation, we developed and evaluated PCR assays specific for the detection of each T. equi genotype.
METHODS: A pair of forward and re

Cellular reproduction defines life, yet our textbook-level understanding of cell division is limited to a small number of model organisms centered around humans. The horizon on cell division variants is expanded here by advancing insights on the fascinating cell division modes found in the Apicomplexa, a key group of protozoan parasites. The Apicomplexa display remarkable variation in offspring number, whether karyokinesis follows each S/M-phase or not, and whether daughter cells bud in the cytoplasm or bud from the cortex. We find that the terminology used to describe the various manifestatio

BACKGROUND: Lyme disease is a common vector-borne illness in the U.S. caused by Borrelia species spirochetes. Neuroborreliosis has variable presentations, rarely manifesting as meningoradiculitis or "Bannwarth Syndrome", characterized by painful radiculopathy, neuropathy, varying degrees of motor weakness, peripheral facial nerve palsy and cerebrospinal fluid (CSF) lymphocytic pleocytosis. We present a case of Bannwarth Syndrome manifesting with transaminitis and significant weight loss.

BACKGROUND: Plasmodium ovale is relatively unfamiliar to Chinese staff engaged in malaria diagnosis. In 2013, dried blood spots of four unidentified but suspected ovale malaria samples were sent to the National Malaria Reference Laboratory (NMRL) for reconfirmation.
METHODS: Partial and complete, small, subunit ribosomal DNA (SSU rDNA) sequences of four samples were obtained with PCR-cloning-sequencing method. Obtained sequences were analyzed by aligning with each other and with nine SSU rDNA sequences of six known Plasmodium parasites. A phylogenetic tree was constructed based on complete SS

Equine piroplasmosis caused by Theileria equi and Babesia caballi is endemic in Trinidad and Tobago. Transmission occurs by ticks of the family Ixodidae. T. equi can also be transmitted transplacentally; however transplacental transmission of B. caballi is unknown. This study aims to investigate transplacental transmission of equine piroplasmosis from thoroughbred mares naturally infected via the tick vector. Whole blood and serum samples were collected from 117 mares in the fifth month of pregnancy. Blood samples were also collected from each of their foals (89 in total) within the first 36h

Fevers of unknown origin (FUOs) are defined as prolonged fevers of 101 degrees F or greater lasting 3 or more weeks that remain undiagnosed after comprehensive inpatient/outpatient laboratory testing. Tick-borne infections are uncommon causes of FUOs. Any infectious disease accompanied by prolonged fevers can present as an FUO if the diagnosis is not suspected or if specific laboratory testing is not done to confirm the diagnosis. Babesiosis is transmitted by the Ixodes scapularis ticks endemic to areas in the northeastern United States. We present the case of a 73-year-old, non-human immunode

BACKGROUND: Babesiosis and malaria are infectious diseases caused by the intraerythrocytic parasites Babesia and Plasmodium, respectively. While no human red blood cell (RBC) receptors have been shown to be essential for B. microti (Bm) invasion, Duffy (ACKR1) was reported to be essential for P. knowlesi and P. vivax invasion in 1975 and 1976, respectively. This suggests additional medical progress is needed for babesiosis, warranting a detailed analysis.
METHODS: Given similarities in the target cell of infection, data about babesiosis and malaria cases in the US were obtained from the Cente

Because infected dogs are widely considered to be the main domestic reservoir for Leishmania infantum (syn Leishmania chagasi) parasites in Brazil, the diagnosis of canine visceral leishmaniasis (CVL) must be made both accurately and promptly. The present study attempted to standardize a conventional polymerase chain reaction (cPCR) protocol for the detection of L. infantum DNA in canine spleen samples. Quantitative PCR (qPCR) technique was used to confirm the presence of Leishmania DNA in the canine spleen fragments. A comparison was made between the efficacies of these molecular diagnostic t
